국내 미기록 콩씨스트선충 기생세균, Pasteuria nishizawae Sayre, Wergin & Nishizawa의 보고 (First Report of Pasteuria nishizawae Sayre, Wergin, & Nishizawa Attcaking Heterodera glycines in Korea)

  • Obligate bacterial parasite attacking Heterodera glycines was firstly found from Chungju soybean field in Korea. Diameters of sporangium and central body were 5.6 ${\mu}{\textrm}{m}$ and 1.9 ${\mu}{\textrm}{m}$ under light microscopy (LM), and 3.9 ${\mu}{\textrm}{m}$ and 1.8 ${\mu}{\textrm}{m}$ under transmission electron microscopy (TEM). Endospore showed cup-shaped with smooth-type exosporium without peg-like thickening in polar area under SEM and TEM. Bacteria completed its life cycle in female of soybean cyst nematode after adhering on cuticle of second-stage juvenile. From these results, the Pasteuria found from Chungju was identified with P. nishizawae.

Heterodera glycines-Induced Syncytium Structures Related to the Nematode Growth and Reproduction in Susceptible Soybean Cultivars

  • The production of soybean cyst nematode (SCN), Heterodera glycines, including female formation and fecundity was much higher in SCN race 14 (R14) than in race 3 (R3) in susceptible soybean cultivars Bragg (intolerant), Lee74 (moderately tolerant), and PI 97100 (highly tolerant). The nematode body was also significantly larger in R14 than in R3 at 20 days after inoculation, but the further nematode growth appeared to be slower in R14 than in R3, resulting in no significant difference between the two races at 30 days after inoculation. Within each race, no significant difference was observed in the growth and reproduction among the soybean cultivars tested. Syncytial areas near the nematode lip regions (infection sites) were measured for each soybean cultivar-SCN race combination. R14 induced significantly larger syncytia than R3. Bragg had relatively larger syncytia than Lee74 and PI 97100, but the difference among the soybean cultivars was minimal or not significantly different. Syncytium occupation in the stelar region differed only between PI 97100 and the other two cultivars, which may be somewhat, but not exactly, related to tolerance levels. Syncytial cytomplasm was degenerated more with R14 and in Bragg than with R3 and in Lee74 and PI 97100, respectively. In light microscopy, degenerated syncytia were characterized by depleted and loose cytoplasm with less plastids than normal-looking (intact) syncytia which had dense syncytial cytoplasm. Electron microscopy revealed that degenerated syncytia contained highly vacuolated cytoplasm with degenerated plastids. The above results suggest that structural characteristics of syncytia may match the nematode growth and reproduction.

콩씨스트선충(線蟲)(Heterodera glycines)의 Race별(別) 분포조사(分布調査) (Race Distribution of the Soybean Cyst Nematode(Heterodera glycines) in Korea)

  • 콩씨스트선충(線蟲)의 race를 판별(判別)하고 그 분포(分布)를 조사(調査)한 결과(結果)를 요약(要約)하면 다음과 같다. 1. 강원도(江原道)에서 채집(採集)한 콩씨스트선충(線蟲)은 race 1과 race 3, 경기도(京畿道)에서 race 3, race 5, race c 그리고 충청남도(忠淸南道)에서 race 1과 race 5가 각각(各各) 판별(判別)되었다. 2. 같은 지역(地域), 적어도 같은 면지역(面地域)에 분포(分布)하는 콩씨스트선충(線蟲)의 race는 동일(同一)하다는 것이 밝혀졌다. 3. 각(各) race의 분포비율(分布比率)을 보면 race 1이 52.4%로 가장 많고, 그 다음으로 race 5가 28.6%, race 3이 14.3% 그리고 race c가 4.8%의 순(順)이었다. 4. 판별(判別)된 4개(個) race 중(中)에서 race 3은 우리나라에서 처음 판별(判別)된 race였다.

국내 콩 씨스트선충의 HG type 분석 (Studies on HG Type of Heterodera glycines in Korea)

  • 콩 저항성품종 육성의 기초자료로 이용하고자 국내 13개 콩 씨스트선충 집단을 대상으로 HG type를 조사하였다. 국내 콩 씨스트선충 HG type는 0, 2, 5, 2.5, 1.2.7, 2.5.7로 총 6개의 HG type가 발견되었다. 그중 HG type 2.5가 30.8%로 가장 많았으며 다음으로 HG type 2.5.7(23.0%)이었다. 각 indicator별로는 PI 88788 가해 집단이 76.9%, PI 209332 가해 집단이 61.5%, PI 548316('Cloud') 가해 집단이 30.8%, PI 548402('Peking') 가해 집단이 7.7%였고 PI 90763, PI 437654, PI 89772를 가해할 수 있는 콩 씨스트선충 집단은 없었다. PI 90763, PI 437654, PI 89772은 국내 모든 콩 씨스트선충 집단에 대해 저항성으로 나타나 저항성품종 육성 교배 모본으로 이용될 수 있을 것이다.

  • 전국적으로 발생하며 대두에 많은 피해를 주고 있는 콩씨스트 선충의 방제에 보다 능률적인 효과를 얻기 위하여 본선충의 피해를 구명하고 저항성 대두 품종을 선발하여 품종육성에 기여코자 장려품종 8품종을 포함 한 64품종을 공시 저항성을 검정하였다. 1. 장단백목을 공시하여 콩씨스트선충에 대한 피해(감수율)를 조사한 결과, 접종시험(Pot)에서 $y=36.2-0.63\times(r=-0.74)$이었고, 포장시험에서는 $y=10.3-0.32\times (r=-0.56)$ 으로서 선충의 밀교와 대두 수양과는 부의 상관관계가 있었다. 2. 장려품종은 모두 저항성에 약 하였고 PI-84751, PI-90763, Southern-Proripic, 금강소입, 백태등은 비교적 강하였으며 그외의 모든 공시 품종들은 중간성일 감수성으로 나타났다. 3. 저항성 품종과 감수성 품종 間에 선충의 발육에 차가 있었고 특히 여성충수에 있어서 차가 많았다

  • 대두 씨스트 선충포에 있어서 대두 품종별 선충저항성정도를 검정하여 내선충성 대두 품종육성시 기초자료로 삼고져 본 시험을 실시하였던 바 그 결과를 요약하면 다음과 같다. 1. 지표품종에 대한 평균 종실중 감소율은 저항성 품종은 38\sim44%, 감수성 품종은 6972%로 차리가 뚜렷하였으며 전중 및 협수도 같은 경향이었다. 2. 공시된 품종중 저항성정도가 $\ulcorner$최강$\lrcorner$인 품종은 5, $\ulcorner$$\lrcorner$에 속하는 것이 8품종으로써 이를은 모두 중ㆍ만생종이었다. 저항성 품종으로 유망시 되는 것으로는 울산, 강림, pickett 71, Forrest, Bedford 등이었다. 3. 선충포에 있어서 종실중, 전중 및 협수의 감소율을 조생종을 62\sim75%, 중생종은 39\sim65%, 만생종은 29\sim36%로써 생육일수가 짧은 품종일수록 저항성이 약하게 나타났다. 이 원인으로서는 선충의 분포범위와 대두 뿌리의 발생심도 및 발생량, 가해시기와 대두의 개화기 및 회복가능기간이 밀접히 관여하고 있는 것으로 고찰되었다. 4. 내선충성 품종선파을 위한 유의 상관 형질은 전중, 협수, 100입중, 주전장이었으며 특히 전종과 협수는 모두 +0.9 이상의 고도의 상관계수를 보였다.

충남지역 두류작물 재배지 식물기생선충 감염현황 (Incidence of Plant-parasitic Nematodes from Legume Fields in Chungnam Province)

  • To survey the incidence of plant-parasitic nematodes, 187 soil samples from leguminous plant fields in Chungnam province, Korea were assessed from June to August 2018. As the result, four nematode genera - the cyst nematode (Heterodera spp.), root-knot nematode (Meloidogyne spp.), root-lesion nematode (Pratylenchus spp.), spiral nematode (Helicotylenchus spp.) - were detected. Among them, the incidence of cyst nematode was the highest in Taean (73%) and lowest in Hongseong (10%). The cyst nematode was detected from the adzuki and kidney bean fields as well as soybean fields. The incidence of cyst nematode in upland fields was higher than in paddy fields. In addition, the extracted cyst nematodes consisted of two species, which was identified as Heterodera glycines and H. sojae. The incidence of H. glycines (22%) was higher than H. sojae (10%), and a few of the fields (5%) was mixed with two cyst nematodes. This result suggested that we should consider that the cyst nematode is the major plantparasitic nematode when controlling the nematodes in soybean fields in Chungnam province.

Inheritance of Cyst Nematode Resistance in a New Genetic Source, Glycine max PI 494182

  • Worldwide, cyst nematode(Heterodera glycines Ichinohe) is the most destructive pathogen of cultivated soybean. In the USA, current annual yield losses are estimated to be nearly a billion dollars. Crop losses are primarily reduced by the use of resistant cultivars. Nematode populations are variable and have adapted to reproduce on resistant cultivars over time because resistance primarily traces to two soybean accessions. It is important to use diverse resistance sources to develop new nematode resistant cultivars. Soybean PI 494182 is a recent introduction from Japan and found to be resistant to multiple nematode populations. It is yellow seeded and maturity group 0. We have determined inheritance of resistance in PI 494182 using $F_{2:3}$ families derived from cross PI 494182 X cv. Skylla. Skylla is a susceptible parent. Three nematode populations, races 1, 3, and 5, corresponding to HG types 2.5.7, 0, and 2.5.7 were used to bioassay 162 $F_{2:3}$ families in greenhouse experiments. Based on Chi-square tests, a two-gene model is proposed for resistance to race 1 and a three-gene model is proposed for conditioning resistance to both races 3 and 5. Correlation coefficient analysis indicated that some genes conditioning resistance to races 1, 3, and 5 are shared or closely linked with each other. These results will be useful to soybean breeders for developing soybean cultivars for broad resistance to nematodes.

Characterization of Heterodera sojae Virulence Phenotypes in Korea

  • The white soybean cyst nematode Heterodera sojae, isolated from the roots of soybean in Korea, is widespread in most provinces of the country and has the potential to be as harmful to soybean as H. glycines. Determining the virulence phenotypes of H. sojae is essential to devising management strategies that use resistant cultivars. Consequently, virulence phenotypes of 15 H. sojae populations from Korea were determined on seven soybean lines and one susceptible check variety. Two different HS types were found to be present in Korea; the more common HS type 2.5.7, comprising 73.3% of the H. sojae populations and the less common HS type 0, constituting only 26.7% of the tested populations. Considering the high frequency of H. sojae adaptation to soybean indicator lines, the PI 88788 group may not be a possible source of resistance while PI 548402, PI 90763, PI 437654, and PI 89772 can be used as resistance sources for soybean breeding programs aimed at developing H. sojae-resistant soybean cultivars in Korea.

PCR-RFLP를 이용한 국내 분포 씨스트선충 4종의 동정 (Identification of Four Cyst Nematodes using PCR-RFLP in Korea)

  • To identify four cyst nematodes (Heterodera schachtii, H. trifolii, H. glycines, H. sojae) that are economically important plant-parasitic nematodes in Korea, restriction fragment length polymorphism (RFLP) by 8 endonucleases (PstI, VspI, AlwI, RsaI, MvaI, EcoRI, Eco72I, Hinf I) was performed based on sequence difference of mitochondrial DNA cytochrome c oxidase subunit I (COI) gene. As a result, species-specific DNA band patterns by RsaI endonuclease were observed in H. schachtii. The specific patterns was in H. trifolii by 3 endonucleases (VspI, AlwI, Hinf I), and was in H. glycines by Hinf I. While, H. sojae was not digested by 4 endonuclease (VspI, AlwI, RsaI, Hinf I). This study showed that four cyst nematodes could be distinguished using RFLP by 4 endonucleases (RsaI, VspI, AlwI, Hinf I) based on the sequence difference of COI gene.
