DNA 바코드 분석을 통한 소계(小薊) 및 대계(大薊) 기원식물 감별과 종간 유연관계 분석 (Molecular Authentication and Phylogenetic Analysis of Plant Species for Breeae and Cirsii Herba based on DNA barcodes)

  • Objectives : The origin of Breeae Herba (So-gye) and Cirsii Herba (Dae-gye) is differently prescribed in Korean and Chinese modern pharmacopoeia. Since the similar morphological characteristics and chaotic plant names, moreover, the aerial part of Carduus crispus have been used as the Cirsii Herba. To develop a reliable method for correct identification of these herbal medicines and to evaluate the genetic relationship of these closely related plant species, we analyzed sequences of DNA barcode regions. Methods : Thirty-one samples of 6 medicinal plants (B. segeta, B. setosa, C. japonicum var. maackii, C. setidens, C. chanroenicum, and C. crispus) were collected from different habitate and nucleotide sequences of DNA barcode regions (rDNA-ITS, matK, and rbcL) were analyzed after amplification using appropriate primers reported in previous studies. The nucleotides of species-specific authentic marker and phylogenetic relations were estimated based on the entire sequences of DNA barcodes by the analysis of ClastalW and UPGMA, respectively. Results : In comparative analysis of DNA barcode sequences, we obtained specific nucleotides to discriminate the medicinal plant of Breeae/Cirsii Herba in species level and evaluated the phylogenetic relationship of these species. Futhermore, we identified distinct marker nucleotides enough to authenticate respective species. These sequence differences at corresponding positions were avaliable genetic markers to determine the botanical origins of Breeae Herbal as well as Cirsii Herba. Conclusions : These marker nucleotides would be useful to identify the official herbal medicines by providing of definitive information that can identify each plant species and distinguish from unauthentic adulterants and substitutes.

비육돈에 있어서 생약제 혼합물의 급여가 성장율, 영양소 소화율, 혈청학적 변화 및 도체특성에 미치는 영향 (The Effects of Dietary Herbal Plant Mixture on Growth Performance, Nutrient Digestibility, Serological Changes and Carcass Characteristics in Finishing Pigs)

  • 본 연구는 지황, 천궁, 감초, 오미자, 차전자, 산약, 둥글레, 당귀 그리고 황백 등을 배합한 복합생약제제의 비육돈 사료내 첨가가 성장율과 도체특성에 미치는 영향을 조사하기 위해 실시하였다. 시험 1은 개시시 체중 81.58±3.20㎏의 3원교잡종[(Duroc×Yorkshire)×Landrace]비육돈 80두를 공시하여 45일간 사양시험을 실시하였다. 시험설계는 옥수수-대두박 위주의 사료인 1) NC(negative control; antibiotics-free diet), 2) PC(positive control; NC diet added 0.1% of chlortetracycline), 3) NCM(NC diet added 0.1% of herbal plant mixture) 그리고 4) PCM(PC diet added 0.1% of herbal plant mixture)의 4개의 처리구로 하여 처리당 5반복, 반복당 4두씩 완전임의 배치하였다. 시험 1에서는 사양기간동안 일당증체량에서는 대조구에 비해 생약제 첨가구인 HPM과 PHMP 처리구가 높았으며(P<0.08), 무항생제 처리구(NC)에 생약제를 첨가하므로서 체중의 증가 효과를 보였다. (P<0.01). 사료섭취량과 사료효율에 있어서는 PC처리구와 NCM처리구에서 증가를 보였으나 처리구간의 유의적인 차이는 없었다. Total protein에서는 항생제가 없는 대조구에 생약제를 첨가하므로서 증가 효과가 있었다(P<0.04). 시험 2는 개시시 체중 77.36±2.0l㎏의 3원교잡종[(Duroc×Yorkshire)×Landrace] 비육돈 100두를 공시하여 28일간 사양시험을 실시하였다. 시험설계는 옥수수-대두박 위주의 사료인 1) NC(negative control; antibiotics-free diet), 2) PC(positive control; NC diet added 0.1% of chlortetracycline), 3) PM0.1(NC diet added 0.1% of herbal plant mixture) 그리고 4) PM 0.2(NC diet added 0.2% of herbal plant mixture)의 4개의 처리구로 하여 처리당 5반복, 반복당 5두씩 완전임의 배치하였다. 시험 2에서는 사양기간동안 PM0.1 처리구가 다른 처리구들보다 증체량이 증가 되었으나 유의적인 차이는 보이지 않았다. 도체중에서는 대조구가 다른 처리구와 비교하여 높게 평가되었다(P<0.06). 등지방 두께에서는 PC처리구가 다른 처리구보다 높았지만 유의적인 차이는 보이지 않았다. 하지만 도체 등급에서는 생약제를 첨가한 처리구가 대조구 보다 A등급율이 높게 나타났다(P<0.04). 이상의 결과로 종합하여 볼 때 비육돈 사료내 생약제의 첨가는 성장율이나 혈액 내 면역글로불린에 좋은 영향을 미치는 것으로 보인다.

  • Aldose reductase (AR) plays a central role in the development of the diabetic complications. Eighty one Korean herbal medicines have been investigated for inhibitory activities on AR. Among them, fourteen herbal medicines exhibited a significant inhibitory activity compared with 3,3-tetramethyleneglutaric acid (TMG) as positive control. Particularly, Xanthium strumarium (fruit, whole plant), Ilex cornuta (stem and leaf), Vitex rotundifolia(stem), Sophora flavescens (whole plant), Platycarya stobilacea (flower) showed two times more potent inhibitory activity than 3,3-tetramethyleneglutaric acid (TMG).

Effect of Timing of Ethephon Treatment on the Formation of Female Flowers and Seeds from Male Plant of Hemp (Cannabis sativa L.)

  • Hemp (Cannabis sativa L.) is a dioecious plant, although monoecious plants are bred in some cultivars for fiber or seed production. Recently, hemp has received attention for medicinal use with some cannabinoids, including cannabidiol. Self-fertilization for breeding inbred lines is difficult because of dioeciousness and anemophily in hemp. This experiment was conducted to develop a self-fertilization method by forming female flowers and seeds from male plants of dioecious hemp. To induce the formation of female flowers on male plants, 500 mg L-1 of ethephon was sprayed on plants at soon, seven and fourteen days after primordia formation. The plant ratio of female flowers formation and the number of harvested seeds were increased by ethephon treatment. Female flowers of male plants have 5 stigmas in contrast to the dual stigma of female 1plants. Male plant seeds were lighter and smaller than those from female plants. Although the germination rate was lower than that of normal seeds from female plants, the seeds from male plants germinated to grow seedlings. Thus, we suggest that hemp plants should be treated with ethephon at soon after primordia formation to induce the formation of more female flowers on the male plants.

Characterization of a Novel Cucumber mosaic virus Isolated from Petunia hybrida

  • Petunia hybrida is commonly used in landscapes and interiors for its attractive flower. Virus-like foliar symptoms, including a mosaic with dark green islands surrounding the veins and chlorosis on the leaf margins, were observed on a petunia plant from Icheon, Gyeonggido, Korea. Cucumber mosaic virus (CMV) was identified in the symptomatic petunia by serological testing for the presence of CMV coat protein (CP) with a direct antibody-sandwich-enzyme-linked immunosorbent assay. An agent was mechanically transmitted to indicator plant species including Chenopodium quinoa. Examination of the inoculated plant leaves by RT-PCR analysis and electron microscopy revealed the presence of specifically amplified CP products and spherical virions of approximately 28 nm in diameter, respectively, providing confirmation of a CMV infection. Analysis of CP sequences showed that CMV petunia isolate (CMVYJC) shared 82.5-100% amino acid sequence identity with CPs of representative CMV strains. Phylogenetic analysis of CPs supports that CMV-YJC is a member of CMV subgroup IA (CMV-IA) and has biological properties of CMV-IA on host species. To our knowledge, this is the first report of CMV from P. hybrida in Korea.

약용식물자원 추출물의 항염증활성 (Anti-inflammatory Activity of Medicinal Plant Extracts)

  • The study was conducted to investigate candidate materials as anti-inflammation agent from plant resources. Activities of 33 plant parts extracts with the final concentration of 5${\mu}g/ml$ were evaluated on the several inflammation-related markers such as the release of proinflammatoty cytokine [tumor necrosis factor-alpha (TNF-${\alpha}$) & interleukin-6 (IL-6)], nitric oxide (NO), the expression of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2) and inhibitor of nuclear factor kappa-B alpha ($I{\kappa}-B{\alpha}$) in lipopolysaccharide (LPS)-treated RAW 264.7 cells. The extracts in the final concentration of 10 ${\mu}g/ml$ were also screened on peroxynitrite (ONOO$^-$) scavenging activity. Eleven extracts selected from the screening assay were verified on the inhibition activity on peroxynitrite and total reactive species oxygen (ROS) in the several concentrations. As results, Alpinia officinarum Hance (rhizome), Inula britannica var. chinensis Regel (flower), Ulmus arvifolia Jacq (trunk peel) and Aster scaber Thunb. (aerial part) showed comparatively potent anti-inflammatory activities in vitro cells or chemical level systems, and then these four plant parts should be studied on the antiinflammatory mechanism by further studies.

Effects of Fertilization and Co-Application of Compost Tea on Fruit Growth and Accumulation of Anthocyanin in Omija (Schisandra Chinensis Baillon)

  • This study was conducted to evaluate the influence of fertilization on characteristics of growth and accumulation of anthocyanin in fruit of Omija (Schisandra chinensis Baillon). Nitrogen supply mainly affected growth of fruit and the anthocyanin content in Omija respective of vegetative growth steps. The anthocyanin content in fruit was significantly low in high N supply and non-fertilization. The conjunctive supply of nitrogen and compost tea resulted in a higher anthocyanin content of fruit, total nitrogen content of leaf, and nitrate in soil. This result implies that nitrogen supply to Omija plant, affects the accumulation patterns of anthocyanin in different ways, e.g. it delays the quantitative biosynthesis at low nitrogen supply during fruit maturation or enhances anthocyanin degradation during the final maturation steps.

  • Agaricus bisporus is a popular edible mushroom that is cultivated worldwide. Due to its secondary homothallic nature, cultivated A. bisporus strains have low genetic diversity, and breeding novel strains is challenging. The aim of this study was to investigate the genetic diversity and population structure of globally collected A. bisporus strains using simple sequence repeat (SSR) markers. Agaricus bisporus strains were divided based on genetic distance-based groups and model-based subpopulations. The major allele frequency (MAF), number of genotypes (NG), number of alleles (NA), observed heterozygosity (HO), expected heterozygosity (HE), and polymorphic information content (PIC) were calculated, and genetic distance, population structure, genetic differentiation, and Hardy-Weinberg equilibrium (HWE) were assessed. Strains were divided into two groups by distance-based analysis and into three subpopulations by model-based analysis. Strains in subpopulations POP A and POP B were included in Group I, and strains in subpopulation POP C were included in Group II. Genetic differentiation between strains was 99%. Marker AB-gSSR-1057 in Group II and subpopulation POP C was confirmed to be in HWE. These results will enhance A. bisporus breeding programs and support the protection of genetic resources.
