TBT 노출에 따른 넙치, Paralichthys olivaceus의 혈액학적 성상 및 혈장성분의 변화 (Changes of hematological parameters and plasma constituents in the olive flounder Paralichthys olivaceus exposed to TBT)

  • Juvenile Paralichthys olivaceus (mean length $17.3{\pm}2.2cm$, mean weight $46.5{\pm}6.3g$) were exposed for 10 days to tributyltin (TBT) at various concentrations (0, 1, 2, 4 and $8{\mu}g/L$). Hematological parameters, such as red blood cell (RBC) count, hematocrit (Ht), and hemoglobin (Hb) concentration, were considerably decreased by high TBT concentrations. In plasma constituents, inorganic components such calcium and magnesium were also significantly altered. Plasma calcium was notably decreased, whereas plasma magnesium was increased. The enzyme components, glutamic oxalate transaminase (GOT) and glutamic pyruvate transaminase (GPT), were significantly decreased by TBT exposure. The present findings suggest that TBT exposure can cause significant alterations in hematological parameters and plasma constituents of flatfish Paralichthys olivaceus.

  • Juvenile Anoplopoma fimbria (mean length $15.6{\pm}1.4cm$, mean weight $68.7{\pm}4.3g$) were exposed to 4 months with the different levels of salinity [100 (35.0), 90 (31.5), 80 (28.0), 70 (24.5), 60 (21.0), 50 (17.5), and 40 (14.0) % (psu)] for 4 months. Hematological parameters such as red blood cell (RBC) counts, hematocrit (Ht), and hemoglobin (Hb) concentrations were substantially decreased under salinities of 50% psu or lower. Of the measured inorganic plasma constituents, magnesium was notably decreased, whereas there was no effect on calcium. Among organic plasma components, glucose and cholesterol were significantly increased, and total protein was decreased. Among enzyme plasma components, glutamic oxalate transaminase (GOT), glutamic pyruvate transaminase (GPT), and alkaline phosphatase (ALP) were significantly increased under salinities of 50% psu or lower. Antioxidant responses such as glutathione S-transferase (GST) and glutathione (GSH) were significantly decreased at salinities of 50% psu or lower. The results of this study indicate that salinity affects the blood parameters, plasma constituents, and antioxidant responses of A. fimbria.

  • The water celery, Oenanthe javanica DC., is widely distributed in Korea and the other South East Asian countries. From ancient times, the water celery has widely been used for food or medicine. In herbal medicine, the water celery has a medical actions which are defervescence, diuresis, hypotensor, appetizer. Recently, studies on the water celery are briskly being conducted. For example, studies on the morphologic property and classifications of origin of the water celery, studies on the chemical constituents of the Water celery, studies on the bio activities of the water celery which contains flavonoids. However, there is not studies of hematological changes in dogs administrated the water celery until now. Consicontly, this study was conducted to observe the hematological changes in dogs administrated excessive amount of the water celery concentrated extract for a long period. In this studies, all dogs showed significantly decrease of the RBC, PCV, Hb and MCHC, and increase of the WBC, MCV, GSH and reticulocytes, but did not significantly change in the Met-Hb.

Hematological constituents and ultrastructural changes in dark-banded rockfish, Sebastes inermis, under nitrite stress

  • The acute toxicity and sublethal effects of nitrite on the dark-banded rockfish, Sebastes inermis (mean body weight: $83.3{\pm}7.2$ g), were studied under static conditions for a period of 96 h. The acute toxicity of nitrite was at the 50% lethal concentration ($LC_{50}$) of 700 mg/L. The sublethal effects on selected hematological parameters of the dark-banded rockfish, such as its osmolality, hematocrit, cortisol, alanine aminotransferase (ALT), and aspartate aminotransferase (AST), were measured after 0, 6, 12, 24, 48, 72, and 96 h of exposure to 0, 50, 100, 200, 400, or 700 mg/L nitrite. Sublethal nitrite caused a progressive reduction in the hematocrit of the fish, depending on the nitrite concentration and the exposure period. Exposure to 100-700 mg/L nitrite for 96 h caused a reduction in the hematocrit and an increase in cortisol, ALT, and AST compared with the control levels. Abnormal ultrastructural changes in the gills and liver tissues were observed in fish exposed to 700 mg/L nitrite for up to 96 h compared with the control tissues. Ultrastructural changes included atrophic gill mitochondria and hepatocytes that developed smooth endoplasmic reticulum and atrophic mitochondria. Although no rockfish mortality occurred at 500 mg/L nitrite, all the hematological parameters examined responded adversely to a nitrite dose of 200 mg/L for 96 h. These results show that although the acute toxic concentration of nitrite for the dark-banded rockfish is > 700 mg/L, sublethal concentrations of nitrite also negatively affect its hematological parameters.

TBT 노출에 따른 돌돔, Oplegnathus fasciatus의 성장 및 혈액성분의 변화 (Changes of Growth and Hematological Constituents in the Rock Bream Oplegnathus fasciatus Exposed to TBT)

  • 본 실험은 TBT에 노출된 돌돔의 성장 및 혈액학적 변동을 검토하기 위하여 실시하였다. 돌돔은 TBT 0, 1, 2, 4, $8{\mu}g/L$ 농도에서 4주간 노출하였다. 돌돔의 성장은 $8{\mu}g/L$ TBT 농도에서 유의한 감소를 나타냈다. RBC count, hematocrit 값, hemoglobin 농도, 혈청 glucose 농도는 $8{\mu}g/L$ TBT 농도에서 유의한 감소를 나타낸 반면, 총 단백질 농도는 유의한 증가를 나타냈다. 또한 GOT 및 GPT 활성은 $8{\mu}g/L$ TBT 농도에서 유의한 증가를 나타냈다. 따라서 해역에 TBT 농도 $8{\mu}g/L$ 이상의 존재할 경우에는 돌돔의 성장 및 혈액학적 변동이 예상된다.

The water celery is widely distributed in Korea, Japan, china, Taiwan, Indonesia, Malaysia and the other countries. From ancient times, the water celery has widely been used for food or medicine. In herbal medicine, the water celery has a medical actions which are defervescence, diuresis, jaundice, edema, hypotensor, haematic, angiostasis, sthenia, appetizer. Recently, studies on the water celery are briskly being conducted. For example, studies on the morphologic property and classifications of origin of the water celery, studies on the chemical constituents of the water celery which contains vitamins, proteins and amino acids, studies on the bio activities of the water celery which contains flavonoids. However, there is not studies of hematological changes in dogs administrated the water celery until now. Consicontly, this study was conducted to observed the hematological changes in dogs administrated excessive amount of the water celery crude juice for a long period. In this studies, all dogs showed significantly decrease of the PCV and Hb, and increase of the WBC, GSH and reticulocytes, but did not significantly change in the RBC, MCV, MCHC and Met-Hb.

Alterations of growth performance, hematological parameters, and plasma constituents in the sablefish, Anoplopoma fimbria depending on ammonia concentrations

  • Juvenile Anoplopoma fimbria (mean length $16.8{\pm}2.2cm$, and mean weight $72.8{\pm}5.4g$) were exposed for 2 months with different levels of ammonia (0, 0.25, 0.50, 0.75, 1.00, and 1.25 mg/L). Growth performances such as daily length gain, daily weight gain, condition factor, and hepatosomatic index were significantly decreased by ammonia exposure. Hematological parameters such as red blood cell (RBC) count, hematocrit, and hemoglobin were also significantly decreased. In plasma inorganic components, calcium and magnesium were significantly decreased by ammonia exposure. In plasma organic components, there was no alteration in cholesterol and total protein. In enzyme plasma components, glutamic oxalate transaminase (GOT) and glutamic pyruvate transaminase (GPT) were significantly increased. The results of this study indicated that ammonia exposure can induce significant growth reduction and blood biochemistry alterations of A. fimbria.

Management of Radiation Injuries by Panax ginseng Extract

  • Chemical radiation protection is an important strategy to protect living beings against the deleterious effects of radiation. In the present study, the radioprotective effect of hydro-alcoholic extract of Panax ginseng extract (PGR-HAE) was studied on radiation-induced deleterious alterations in Swiss albino mice. Oral administration of such extract (25 mg/kg b wt/day/animal) for 5 consecutive days, half an h. before whole-body exposure to 6 Gy gamma radiation, enhanced the 30 days survival and also inhibited the radiogenic sickness, weight loss and life shortening. PGR-HAE ameliorated radiation induced depletion in blood constituents at different necropsy intervals between 12 h to 30 d, and significantly increased the number of femoral spleen colony forming units that survived after irradiation. Furthermore, it checked depletion of glutathione and antioxidant enzymes (superoxide dismutase, catalase, and glutathione S-transferase) as well as elevation of lipid peroxidation (LPO) level in blood and liver. The significant reduction in the yield of LPO demonstrates that PGR-HAE protects the membranes against radiation-induced oxidative damage. These findings conclude that such plant extract provides significant radioprotection, and it may be potentially valuable in the prevention of injuries caused during planned and unplanned radiation exposure.

크롬 노출에 따른 숭어, Mugil cephalus의 혈액성분 변화 (Changes of Hematological Constituents in the Mullet, Mugil cephalus Exposed to Chromium)

  • 크롬은 여러 산업으로 배출되어 수 환경을 오염시키는 주요 중금속 중 하나이며, 특히 Cr(VI)은 수용액 내에서 주로 크롬산 이온으로 존재하며, 생체막을 쉽게 통과하여 산화 스트레스 및 생체 축적을 일으킨다. 따라서 본 연구는 숭어를 대상으로 그들의 혈액성분의 변동에 미치는 크롬의 영향을 검토하였다. RBC count 및 Ht 값은 $50{\mu}g/L$ Cr 농도에서 유의적인 감소를 나타냈으나, Hb 농도의 유의한 변동은 관찰되지 않았다. 혈청 칼슘농도는 $50{\mu}g/L$ Cr의 농도에서 유의적인 감소를 보였고, 마그네슘농도는 $100{\mu}g/L$ Cr의 농도에서 유의적인 증가를 보였다. Glucose 및 total protein 농도는 가장 높은 크롬농도인 $200{\mu}g/L$ Cr에서 유의적인 증가를 보였으나, 중성지방인 triglyceride 농도는 노출기간 동안 유의한 변화가 관찰되지 않았다. GOT 및 GPT 활성은 $100{\mu}g/L$ Cr 이상의 농도에서 유의적인 증가를 보였고, ALP활성은 가장 높은 크롬농도인 $200{\mu}g/L$ Cr에서 유의적인 증가를 보였다. 이상의 결과는 숭어의 혈액성상과 혈청 무기성분은 크롬 농도 $50{\mu}g/L$ 이상, 혈청유기성분과 효소성분은 크롬농도 $100{\mu}g/L$이상에서 변동이 나타나는 것으로 결론 내릴 수 있다.

Blood Biochemical Profile and Rumen Fermentation Pattern of Goats Fed Leaf Meal Mixture or Conventional Cakes as Dietary Protein Supplements

  • The expediency of replacing cost prohibitive and often inaccessible traditional protein supplements prompted the monitoring of hematological parameters was carried out in female goats at 0, 30, 60 and 90 days post feeding. Rumen environment was (3), respectively fed supplements containing either a leaf meal mixture (LMTM) of Leucaena leucocephala-Morus alba-Tectona grandis (2:1:1) or traditional protein supplements groundnut cake (GNC) or soybean meal (SBM) and wheat straw as basal diet. The periodic monitoring of hematological parameters was carried out in female goats at 0, 30, 60 and 90 days post feeding. Rumen environment was studied in bucks in a $3{\times}3$ switch over design. Rumen liquor was collected at 0, 2, 4, 6 and 8 h post feeding after 4 weeks of feeding. The goats fed on LMTM or GNC had similar dry matter intake (g/kg $W^{0.75}$), which was significantly (p<0.05) higher than SBM. Except for packed cell volume (PCV), none of the blood biochemical constituents (Hemoglobin, serum glucose, total protein, serum albumin (A) and globulin(G), A:G ratio, alkaline phosphatase, transaminases) varied significantly due to replacement of 50% dietary protein by LMTM throughout the experiment. GNC group had significantly higher level of PCV than other treatments. However, the level of serum total protein (p<0.01) tended to increase from 60th day onwards irrespective of dietary treatments. The average rumen pH was significantly higher (p<0.001) on SBM followed by LMTM and GNC, respectively. Total volatile fatty acid (TVFA) production was comparable in goats given LMTM or GNC supplements, the corresponding values were significantly different (p<0.001) when compared with SBM. The ammonical-N, total-N and TCA-precipitable-N (mg/100 ml SRL) did not differ significantly among dietary treatments. It may be concluded that supplementing wheat straw with LMTM based concentrate had no adverse effect on voluntary intake, blood biochemical profile and rumen fermentation pattern of the goats.
