Analysis of copy number variation in 8,842 Korean individuals reveals 39 genes associated with hepatic biomarkers AST and ALT

  • Biochemical tests such as aspartate aminotransferase (AST) and alanine aminotransferase (ALT) are useful for diagnosing patients with liver disease. In this study, we tested the association between copy number variation and the hepatic biomarkers AST and ALT based on 8,842 samples from population-based cohorts in Korea. We used Affymetrix Genome-Wide Human 5.0 arrays and identified 10,534 CNVs using HelixTree software. Of the CNVs tested using univariate linear regression, 100 CNVs were significant for AST and 16 were significant for ALT (P < 0.05). We identified 39 genes located within the CNV regions. DKK1 and HS3ST3B1 were shown to play roles in heparan sulfate biosynthesis and the Wnt signaling pathway, respectively. NAF1 and NPY1R were associated with glycoprotein processes and neuropeptide Y receptor activity based on GO categories. PTER, SOX14 and TM7SF4 were expressed in liver. DPYS and CTSC were found to be associated with dihydropyrimidinuria and Papillon-Lefevre syndrome phenotypes using OMIM. NPY5R was found to be associated with dyslipidemia using the Genetic Association Database.

A Model for the $3_{10}$/$\alpha$ Helix Transitions of $\alpha$-Aminoisobutyric Acid-Alanine Oligopeptide ($\alpha$-아미노이소부틸산-알라닌 올리고 펩티드의 $3_{10}$/$\alpha$ 나선 전이에 관한 모형)

  • We suggest a statistical thermodynamic theory for the conformational transition of a synthetic alanine (Ala), ${\alpha}$-aminoisobutyric acid (Aib) alternative oligopeptide, Buo-(Ala-Aib)$_n$-oMe, where the terminal groups Buo and oMe stand for t-butoxy and methoxy, respectively. Pure Aib homo-oligomers have always been found to adopt $3_{10}$ helical conformations, while polyalanine has always $\alpha$ helical conformation. In an organic solvent (e.g. $CD_3$CN) it shows that the length for the $3_{10}$/${\alpha}$ helix transitions of Buo-(Ala-Aib)$_n$-oMe, is 8 at room temperature. In an aqueous solution oligopeptide has always coil conformation at room temperature. In an organic solution, helical structures of the oligopeptide are more stable than coil structure, so we studied the $$3_{10}/\alpha$ helix transitions, considering coiled-conformations, coiled and $3_{10}$ helical conformations, and coiled and $\alpha$ helical conformations by using the zipper model. We determined the values of parameters ($\sigma_A$, $\sigma_T$, $\xi_A$, $\xi_T$) from the relating published data; $\sigma_A$ = 0.00011, $\sigma_T$ = 0.0060, $\xi_A$ = 10.1, $\xi_T$ = 3.90. The distributions of $\alpha$ helical length can be N-2, N-3, N-4, ${\cdots}$, 3, 2, 1 (N = 2n) while those of $3_{10}$ helical length, N-1, N-2, N-3, N-4, ${\cdots}$, 3, 2, 1.

An Analysis Method on Injury Symptoms Utilizing Infrared Thermal Imaging under the Freezing Stress of Hedera helix L. (헤데라 헬릭스 식물의 적외선 열영상에 의한 저온 및 한풍피해에 관한 연구)

  • The experiments, which analyze the injury symptoms and diagnose growth conditions utilizing IRVT and analyzing each parts of H. helix L., had been held under a low temperature. Greenhouse and outdoor growing Genus hedera had been prepared and compared with each Genus hedera's peak and bottom leaves' surface temperature under the experimental categories $-6^{\circ}C$ and $-12^{\circ}C$. As results, analyzing the surface thermal property of peak part leaves' of outdoor growing Genus hedera, at experimental categories $-6^{\circ}C$, $-12^{\circ}C$ were ranged from $-2^{\circ}C{\sim}-7^{\circ}C$ and $-2^{\circ}C{\sim}-15^{\circ}C$. On the other hand, the surface thermal property of bottom part leaves at experimental categories $-6^{\circ}C$, $-12^{\circ}C$ were ranged $-2^{\circ}C{\sim}-11^{\circ}C$ and $-1^{\circ}C{\sim}-12^{\circ}C$. It appears that the thermal properties of leaves' surface on $-6^{\circ}C$ peaks and $-12^{\circ}C$ bottoms were more broadband than bottoms and peaks. It means that the peaks were more sensitive than bottoms, as like $-2^{\circ}C{\sim}-15^{\circ}C$, $-1{\sim}-12^{\circ}C$. Moreover, as similar results had seen to leaves surface temperature added to cold wind conditions. How the cold wind damaged the outdoor growing Genus hedera, analyzed the surface thermal property by IRVT data under $0^{\circ}C$, $-2^{\circ}C$, $-4^{\circ}C$ condition, it resulted to $-6.2^{\circ}C$, $-6.8^{\circ}C$, $-7.5^{\circ}C$. It appeared more $3.5{\sim}6.2^{\circ}C$ low temperature than experimental setting point. In addition, each parts thurmal property of peaks and bottoms was not similar, it referred to each parts' sensitivities of low temperature were different on the peak and bottom leaves surface temperature.

Study on the Prediction of Pressure Drop for Alternative Refrigerants with lubricant in Micro-Fin Tubes (미세휜관내 윤활유를 포함한 대체냉매의 압력강하 예측에 관한 연구)

  • This paper presents a pressure drop correlation for evaporation and condensation of alternative refrigerant with oil in micro-fin tubes. The correlation was developed from a data base consisting of oil-free pure and mixed refrigerants in micro-fin tube; Rl25 R134a. R32 R410a(R32/R125 50/50% mass), R22, R407c(R32/R125/R134a, 23/25/52% mass) and R32/R134a(25/75% mass). The micro-fin tube used in this paper had 60 0.2mm high fins with a 18 helix angle. The cross sectional flow area $(A_c)$ was $60.8 mm^2$ giving an equivalent smooth diameter$(D_e)$ of 8.8mm. The hydraulic diameter $(D_h)$ was estimated to the 5.45mm. The new correlation was obtained by replacing the friction factor and the tube-diameter in Bo Pierre correlation by a friction factor derived from pressure drop data for a micro-fin tube and the hydraulic diameter, respectively. This correlation was also used to predict some pressure data with a lubricant after using a mixing viscosity rule of lubricants and refrigerants. As a result, the new correlation was also well predicted to the measured data within a mean deviation of 19.0%.

Nationalizing Transnationalism: A Comparative Study of the "Comfort Women" Social Movement in China, Taiwan, and South Korea

  • Most literature on the "comfort women" social movement focuses on the case of Korea. These works tend to transpose the meanings generated by South Korean organizations onto the transnational network, assuming certain homogeneity of repertoires and identities among the different social actors that comprise this network. Even though there is some degree of consensus about demands, repertoires, and advocacy strategies at the international level, does this same uniformity exist at the national level? In each country, what similarities and differences are present in the laboratories of ideas, relationships, and identities of social actors in the network? Symbolically and politically, do they challenge their respective societies in the same way? This article compares this social movement in South Korea, China, and Taiwan. My main argument is that the constitutive base for this transnational network is the domestic actions of these organizations. It is in the domestic sphere that these social actors reinforce their agendas, reinvent their repertoires, transform their identities, and expand their submerged networks, allowing national movements to retain their latency and autonomy. Following Melucci's relational approach to the study of social movements, this research is based on a qualitative analysis of institutional documents, participant observation, and open-ended interviews with members of the main social actors.

Structure-Activity Relationships of 9-mer Antimicrobial Peptide analogue of Protaetiamycine, 9Pbw2

  • 9Pbw2 is a 9-mer analog of protaetiamycine derived from the larvae of the beetle Protaetia brevitarsis. Previously, we designed four 9-mer peptide analogues to optimize the balance between the hydrophobicity and cationicity of the peptides and to increase bacterial cell selectivity. Among them, 9Pbw2 has high antibacterial activity without cytotoxicity. The results obtained in previous study suggest that the bactericidal action of 9Pbw2 may be attributed to the inhibition of the functions of intracellular components after penetration of the bacterial cell membrane. In order to understand structure-activity relationships, we determined the three-dimensional structure of 9Pbw2 in 200 mM DPC micelle by NMR spectroscopy. 9Pbw2 has one hydrophobic turn helix from $Trp^3$ to $Arg^8$ and positively charged residues at the N- and C-terminus. This result suggested that positively charged residues from position at the C-terminus in 9Pbw2 may be important for the primary binding to the negatively charged phospholipid head groups in bacterial cell membranes and hydrophobic residues in the middle portion face toward the acyl chains of the hydrophobic lipid in the bacterial cell membrane.

Refolding of Bacillus macerans Cyclodextrin Glucanotransferase Expressed as Inclusion Bodies in Recombinant Escherichia coli

  • This research was undertaken to restore the biological activity of cyclodextrin glucanotransferase (CGTase) of Bacillus macerans origin expressed as inclusion bodies in recombinant Escherichia coli. The optimum concentration of urea used as a denaturant was 8 M. The supplementation of 0.5 M urea into a dialysis buffer increased the refolding efficiency by preventing any protein aggregation. The influence of the protein concentration, temperature, and pH were also investigated. The protein concentration was found to be the most important factor in the refolding efficiency. The optimum temperature was 15-$25^{\circ}C$ and the optimum pH was 6.0. The maximum specific activity of the CGTase refolded under the optimum conditions was 92.2 U/mg, corresponding to 72% of the native CGTase. A comparison of the secondary structure between the native and the refolded CGTase showed that the relative ratio of the $\alpha$-helix content in the native to the refolded CGTase was 1:0.82.

Transcriptional Control of Lactate Dehydrogenase A-Gene Expression during the Pre-replicative Phase of Regenerating Rat Liver (백서 재생간조직의 낙산탈수소효소 A-유전자 발현의 전사활성)

  • Transcriptional rate of lactate dehydrogenase A-gene(LDH-A) during the prereplicative phase of regenerating rat liver was determined by in vitro run-off transcription assay. The results show that the transcription rate of LDH A-gene increases between 12 hours and 15 hours peaking at 13 hours after partial hepatectomy of rat liver. The increased rate of LDH A-gene transcription was interfered after DL-propranolol treatment intraperitoneally injected twice at 1 hour and 8 hours after partial hepatectomy indicating that the transcriptional control of LDH A-gene expression may be mediated by beta adrenergic receptor and cAMP as a second messenger. And also was it shown that the temporally increased rate of LDH A-gene transcription was maximum one hour after the second cAMP-surge which is known to play an important role for the initiation of DNA replication during regeneration of rat liver. And the transcriptional rate of LDH A-gene was decreased to the basal level at the time period when the hepatocytes proliferate rapidly suggesting that the induced LDH Aisozyme may be required for the initiation of DNA replication during regeneration of rat liver. These data may be supporting for the hypothesis suggesting that the induced LDH A-isozyme during the pre-replicative phase of regenerating rat liver may play bifunctional roles as a glycolytic enzyme and a helix destablizing protein as well.

Small Molecules Targeting for ESX-Sur2 Proteins' Interaction

  • It's been known that overexpression of the oncoprotein Her2 (eu/ErbB2), transmembrane receptor protein, occurs in human breast cancer. Her2-positive breast cancer patients who have Her2 overexpression show less therapeutic efficacy with enhanced metathesis and increased resistance to chemotherapy. So far, a humanized monoclonal antibody against Her2 protein called Herceptin is the only drug approved by Food and Drug Administration for treatment of Her2-overexpressing breast tumors. However, antibody therapy of Herceptin may not be ideal method for therapeutic intervention of Her2 protein expression. The therapeutic intervention of Her2 protein expression may be more efficiently achieved by inhibiting the expression of Her2 gene rather than by down-regulating the Her2 protein already overexpressed. Here, we found that the interaction of two proteins of ESX (an epithelial-restricted transcription factor) and DRIP130/CRSP130/Sur2 (a Ras-linked subunit of human mediator complexes) mediates the expression of Her2 gene. The association of ESX with Sur2 is mediated by a small hydrophobic face of 8-amino acid helix in ESX, suggesting that the ESX-Sur2 interaction can be a new novel target for Her2-positive cancer. The process to develop potent ESX-Sur2 interaction inhibitors targeting for Her2-positive cancer therapeutics will be discussed.

  • The effects of an inserted coil on flow . patterns and heat transfer performance for a horizontal rotating heat pipe have been studied experimentally. Especially, the present study is to see an internally inserted helical coil inside a RHP would lead to the same kind of results as internal fins. Visualization test conducted for an acryl tube, charged water with at a volumetric rate of 20%. When the flow kept pool regime at a low rpm(less than 1,000rpm), the movement of coil forced the water to flow in axial direction. But this pumping effect of coil disappeared, when the pool regime changed to annular one which could be created by increasing rpm. The pumping effects for RHP with an inserted coil resulted in the enhancement in both condensation heat transfer coefficient and heat transport limitation, as obtained in case of using internal fins. But all these effects became negligible in the range of higher rpm(above 1,000-1,200) with the transition of flow regime to annular flow.
