Small heat shock proteins (sHSPs) are widely distributed, and their function and diversity of structure have been much studied in the field of molecular chaperones. In plants, which frequently have to cope with hostile environments, sHSPs are much more abundant and diverse than in other forms of life. In response to high temperature stress, sHSPs of more than twenty kinds can make up more than 1% of soluble plant proteins. We isolated a genomic clone, NtHSP18.3, from Nicotiana tabacum that encodes the complete open reading frame of a cytosolic class I small heat shock protein. To investigate the function of NtHSP18.3 in vitro, it was overproduced in Escherichia coli and purified. The purified NtHSP18.3 had typical molecular chaperone activity as it protected citrate synthase and luciferase from high temperature-induced aggregation. When E. coli celluar proteins were incubated with NtHSP18.3, a large proportion of the proteins remained soluble at temperatures as high as $70^{\circ}C$. Native gel analysis suggested that NtHSP18.3 is a dodecameric oligomer as the form present and showing molecular chaperone activity at the condition tested. Binding of bis-ANS to the oligomers of NtHSP18.3 indicated that exposure of their hydrophobic surfaces increased as the temperature was raised. Taken together, our data suggested that NtHSP18.3 is a molecular chaperone that functions as a dodecameric complex and possibly in a temperature-induced manner.
Background: Heat shock proteins play an important role in protection from stress stimuli and metabolic insults in almost all organisms. Methods: In this study, computational tools were used to deeply analyse the physicochemical characteristics and, using homology modelling, reliably predict the tertiary structure of the blunt snout bream (Ma-) Hsp70 and Hsc70 proteins. Derived three-dimensional models were then used to predict the function of the proteins. Results: Previously published predictions regarding the protein length, molecular weight, theoretical isoelectric point and total number of positive and negative residues were corroborated. Among the new findings are: the extinction coefficient (33725/33350 and 35090/34840 - Ma-Hsp70/ Ma-Hsc70, respectively), instability index (33.68/35.56 - both stable), aliphatic index (83.44/80.23 - both very stable), half-life estimates (both relatively stable), grand average of hydropathicity (-0.431/-0.473 - both hydrophilic) and amino acid composition (alanine-lysine-glycine/glycine-lysine-aspartic acid were the most abundant, no disulphide bonds, the N-terminal of both proteins was methionine). Homology modelling was performed by SWISS-MODEL program and the proposed model was evaluated as highly reliable based on PROCHECK's Ramachandran plot, ERRAT, PROVE, Verify 3D, ProQ and ProSA analyses. Conclusions: The research revealed a high structural similarity to Hsp70 and Hsc70 proteins from several taxonomically distant animal species, corroborating a remarkably high level of evolutionary conservation among the members of this protein family. Functional annotation based on structural similarity provides a reliable additional indirect evidence for a high level of functional conservation of these two genes/proteins in blunt snout bream, but it is not sensitive enough to functionally distinguish the two isoforms.
Heat shock proteins (Hsp) 70 are a ubiquitous family of molecular chaperones involved in many cellular processes. A yeast strain, ssa1/2, with two functionally redundant cytosolic Hsp70s (SSA1 and SSA2) deleted shows thermotolerance comparable to mildly heatshocked wild type yeast, as well as increased protein synthesis and ubiquitin-proteasome protein degradation. Since mRNA abundance does not always correlate well with protein expression levels it is essential to study proteins directly. We used a gel-based approach to identify stress-responsive proteins in the ssa1/2 mutant and identified 43 differentially expressed spots. These were trypsin-digested and analyzed by nano electrospray ionization liquid chromatography tandem mass spectrometry (nESI-LC-MS/MS). A total of 22 non-redundant proteins were identified, 11 of which were confirmed by N-terminal sequencing. Nine proteins, most of which were up-regulated (2-fold or more) in the ssa1/2 mutant, proved to be stress-inducible proteins such as molecular chaperones and anti-oxidant proteins, or proteins related to carbohydrate metabolism. Interestingly, a translational factor Hyp2p up-regulated in the mutant was also found to be highly phosphorylated. These results indicate that the cytosolic Hsp70s, Ssa1p and Ssa2p, regulate an abundance of proteins mainly involved in stress responses and protein synthesis.
Cells consistently face stressful conditions, which cause them to modulate a variety of intracellular processes and adapt to these environmental changes via regulation of gene expression. Hyperosmotic and oxidative stresses are significant stressors that induce cellular damage, and finally cell death. In this study, oligonucleotide microarrays were employed to investigate mRNA level changes in cells exposed to hyperosmotic or oxidative conditions. In addition, since heat shock protein 70 (HSP70) is one of the most inducible stress proteins and plays pivotal role to protect cells against stressful condition, we performed microarray analysis in HSP70-overexpressing cells to identify the genes expressed in a HSP70-dependent manner. Under hyperosmotic or oxidative stress conditions, a variety of genes showed altered expression. Downregulation of protein phosphatase1 beta (PP1 beta) and sphingosine-1-phosphate phosphatase 1 (SPPase1) was detected in both stress conditions. Microarray analysis of HSP70-overexpressing cells demonstrated that diverse mRNA species depend on the level of cellular HSP70. Genes encoding Iysyl oxidase, thrombospondin 1, and procollagen displayed altered expression in all tested conditions. The results of this study will be useful to construct networks of stress response genes.
Heat Shock 70kDa Protein 1-Like(HSPA1L)는 Heat-shock protein70(HSP70) family에 속하는 chaperone protein으로 polypeptide folding, assembly, protein degradation 등 다양한 biological processes에 관여하고 있다. HSPA1L은 human methyltransferase-like protein 21A(METTL21A)에 의해 lysine residue에 methylation이 일어나게 되는데, 암세포에서 일반적인 HSPA1L은 주로 세포질에서 발견되는 반면 methylated HSPA1L의 경우 주로 핵에서 발견이 됨으로써 HSPA1L methylation이 암 세포 성장에 중요할 역할을 할 것이라 추측되며 anti-cancer drug target으로 주목 받고 있다. 하지만 현재 HSPA1L의 구조가 부분적으로만 밝혀져 있어 HSPA1L와 METTL21A가 어떤 residue들이 interaction 하여 binding을 하는지에 대해서 아직 밝혀 지지 않았다. 이로 인해 anti-cancer drug target으로서의 연구에 제한이 있다. 이번 연구에서는 homology modeling(Galaxy-TBM, Galaxy-refine)을 통해 HSPA1L 전체 구조를 밝혀 낸 후, HSPA1L 와 METTL21A를 protein-protein docking을 통해 binding pose 예측을 하였다. 이러한 binding pose를 protein interaction analysis하여 HSPA1L과 METTL21A binding에 관여하는 중요 residue들을 밝혀 냈다. 이러한 structural information은 methylated HSPA1L와 암 세포 성장간의 연관성, 더 나아가 anti-cancer drug 개발로 까지도 이어 질 수 있을 것이라 생각한다.
본 연구에서는 넙치, Paralichthys olivaceus를 이용하여 oxytetracycline (OTC)처리농도에 따른 체내에서 혈액 생리학적인 변화, 항산화 효소 및 열충격단백질(HSP)등의 변화를 조사하여 항생제의 안정적인 사용에 대한 기초 자료를 제공하고자 본 실험을 수행하였다. 실험구는 각각 0 (대조구), 100, 300 및 500ppm으로 설정하였으며, 1시간 약욕처리 후 순환시켜 0, 1 및 3시간 후에 혈액성상을 분석하였다. 혈액 중 hematocrit(Ht)수치는 전 실험구에서 감소하는 경향을 보였으나 유의한 차이는 나타나지 않았다. Red blood cell (RBC)는 농도 의존적으로 감소하는 경향을 나타내었다. 또한, 혈청 글루코오스 농도는 농도 의존적으로 증가되는 경향을 나타내었으나, 노출 이후 3시간 경과 후에는 이전 대조구 수준으로 감소되었다. 혈중 단백질량은 0시간째 전 실험구에서 감소되었다가 이후 증가하는 경향을 나타내었으나, 처리전의 대조구와 비교하여 유의하게 낮은 값을 나타내었다. SOD 효소 활성은 노출 직후 300 및 500ppm에서는 유의하게 높은 값을 나타내었다. CAT효소활성은 OTC처리 농도에 비례하여 감소하는 경향을 보였으나 유의한 차이는 나타내지 않았다. 그리고 HSP-70의 단백질량은 100 ppm및 500 ppm처리구에서 대조구에 비해 높게 발현되는 것이 확인되었다. 그러나 HSP-70 mRNA는 다소 증가하는 경향은 보였으나, 모든 실험구에서 유의한 차이는 나타나지 않았다.
본 실험에서는 반SP와 thermotolerance 사이에 어떠한 상관관계가 있는지를 알아보기 위하여, 생쥐 SCK 종양세포에서 heat(45$^{\circ}$, 46$^{\circ}C$)처리가 단백질 합성과 세포의 생존에 미치는 영향을 비교하여 보았다. 그 결과 heat 처리를 받은 세포는 HS$\ulcorner$(70K, 87K)를 측이하게 많이 합성했으며, 다음 heat를 받았을 때는 높은 생존율을 꼬였다. 그리고 이러한 thermotolerance가 생성되고 감퇴되는 kinetics는 HSP가 합성되고 감퇴되는 kinetics와 연관성을 보여 주었다. 이러한 결과로 HSP는 heat shock로부터 세포를 보호하는 데 중요한 역할을 할 수 있다고 생 각된다. 아울러, glycerol을 처리하여 HSP의 합성을 봉쇄시켰을 경우에도 열에 대해 저항성을 갖게 되는 실험결과로 미루어, 세포가 갖게 되는 heat resistance에는 (1) HSP의 합성을 초래하지 않는 요인에 의해 유도되는 heatprotection과 (2) 열처리 등의 결과 합성되는 반SP에 의해 유도되는 thermotolerance의 두가지 경우가 있을 것으로 추론할 수 있었다.

Introduction: Heat shock protein70 (HSP70) is a highly conserved family of proteins produced after a variety of stresses. Many studies reported that the overexpression of HSP70 can improve the prognosis of the patients with sepsis through a reduction of the nitric oxide concentration. However, these results only revealed the effect of HSP70 and nitric oxide. No studies have examined the relationship between HSP70 and nitric oxide. The aim of this study was to evaluate the effect of the overexpression of HSP70 on the expression of inducible nitric oxide synthase and the nitric oxide concentration. In addition, the mechanism of the relationship of HSP70 and inducible nitric oxide synthase (iNOS) in sepsis was examined. Materials and Methods: The experiments were performed on male sprague-dawley rats. Sepsis was induced by a cecal ligation and puncture (CLP). Glutamine (GLN) or saline was administered 1 hour after the initiation of sepsis. Serum and lung tissues were acquired from the rats 12 hours or 24 hours after the initiation of sepsis. The nitric oxide concentration, the expression of HSP70 in lung, and the gene expression of iNOS in lung were analyzed. The three groups, sham operation, CLP and CLP+GLN, were compared. Results: Compared to the other groups, in CLP+GLN, GLN administered after the initiation of sepsis enhanced the expression of HSP70 in the lung at 12 hours ($47.19{\pm}10.04$ vs. $33.22{\pm}8.28$, P=0.025) and 24 hours ($47.06{\pm}10.60$ vs. $31.90{\pm}4.83$, P=0.004). In CLP+GLN, GLN attenuated the expression of iNOS messenger RNA (mRNA) in the lung at 12 hours ($5,513.73{\pm}1,051.60$ vs. $4,167.17{\pm}951.59$, P=0.025) and 24 hours ($18,740.27{\pm}8,241.20$ vs. $9,437.65{\pm}2,521.07$, P=0.016), and reduced the concentration of nitric oxide in the serum at 12 hours ($0.86{\pm}0.48$ vs. $3.82{\pm}2.53$, P=0.016) and 24 hours ($0.39{\pm}0.25$ vs. $1.85{\pm}1.70$, P=0.025). Conclusion: The overexpression of HSP70 induced by the administration of GLN in sepsis attenuates the expression of the iNOS gene but reduces the nitric oxide concentration.
허혈/재관류 뇌손상에서 활성산소류의 역할이 중요시되고 있다. 본 연구에서는 모래쥐의 총경동맥을 묶었다 풀어줌으로써 실험적 허혈/재관류 손상을 유도하고 산화성 스트레스 발생 유무와 이러한 산화성 스트레스가 세포손상으로 연결되는지를 알아보고자 하였다. 해마는 뇌조직 중에서도 특히 산화성 스트레스에 취약한 부분이므로 해마에서 얻은 조직을 대뇌피질에서 얻은 조직과 비교분석하였다. 또한, 이들 부위에서 heat shock protein 70(HSP70)의 발현이 허혈/재관류 손상에 미치는 영향도 검색하고자 하였다. 허혈/재관류에 의한 산화성 스트레스의 지표로써 글루타치온 산화정도, GSSG/(GSH+2xGSSG)를 측정하였을 때 주로 해마에서 산화지표가 상승됨을 관찰하였다. 한편 산화성 스트레스가 세포손상으로 연결되는지를 알아보고자 지질과산화물을 측정하였다. 두 부위 모두에서 지질과산화물 형성의 증가가 있었으며 대뇌피질에서보다 해마에서 더 증가됨을 알 수 있었다. 지질과산화물 형성의 정도나 시간적 변화양상이 글루타치온 산화의 그것들과 유사하였다. 이러한 결과들은 허혈/재관류에 의해 산화성 스트레스가 형성되며 동시에 이러한 산화성 스트레스가 세포 손상을 초래함을 보여준다. 또한 산화성 스트레스 및 산화성 세포손상 정도가 대뇌피질보다는 해마에서 더 큰 것을 알 수 있었다. 그러나, 피질과 해마에서 HSP70의 기초발현(basal level) 정도는 차이가 없었다. 이는 해마의 취약성이 HSP70 발현 결핍에 기인하지 않았음을 나타낸다. 반면 허혈/재관류에 의한 HSP70의 발현유도는 해마조직에서 제대로 이주어지지 않았고 northern blot결과 이는 전사단계에서의 부친에 의한 것으로 나타났다. 이러한 결과들로 볼 때 허혈/재관류에 의한 뇌손상에서 HSP70 유도정도를 측정하는 것이 세포의 취약성을 예측할 수 있는 지표로 유용하게 사용될 수 있을 것으로 사료된다.
