백회와 사신총 혈위에 적합한 침구 소자의 개발 (The Development of Acupuncture- Moxibustion Device for 4 points around GV20(Baihui))

  • Objectives This paper focused on developing the acupuncture-moxibustion device for head. Disadvantages of the existing devices for head moxibustion are its hume and inconvenience in operating because of the hair. Methods The proposed acupuncture-moxibustion device for GV20(Baihui) is 10mm long as needle body and 6mm long as needle root whose feature are 1~2 turns spiral coil. The spiral coil contains the moxa in order not to drop the moxa while operating Results The direct temperature on the burning moxa were measured by TK-305 infrared thermometer. But the direct temperature on the needle of acupuncture-moxibustion device were not able to measure by TK-305 infrared thermometer. By definition equation, the indirect temperatures on the needle of acupuncture-moxibustion device were able to be calculated. Conclusion A very safe, small and cheap acupuncture-moxibustion device for GV20(Baihui) of head is proposed. It will be useful to generate the heat shock protein and to operate in preventing and curing the Alzheimer's disease.

Molecular Cloning and Characterization of a Heat Shock Protein 70-related cDNA from Olive flounder (Paralichthys olivaceus)

  • The complete nucleotide sequence of olive flounder (Paralichthys olivaceus) hsp70-related rDNA was determined by RT- and RACE-PCR methods. A full-length of hsp70-related cDNA has an open reading frame of 1.95 kb encoding 650 amino acids with a calculated molecular weight of 71.1 kD. A corresponding hsp70-related protein contains a number of conserved elements including an ATP-binding domain, a nuclear localization signal and the carboxyl terminal motif, EEVD, which may have a role in chaperone function. Comparison of nucleotide and predicted amino acid sequence between olive flounder hsp70-related gene and hsp/hsc70 genes of other species revealed a high similarity with the cognate form of these genes. These results indicated that we recovered likely to be a olive flounder cognate hsc70 gene.

SIRT1 inhibitor에 의한 Hsp90 inhibitor의 Hsp90 샤페론 기능 억제 및 항암제 내성세포의 Hsp90 inhibitor에 대한 세포독성 증강 (SIRT1 Inhibitor Enhances Hsp90 Inhibitor-mediated Abrogation of Hsp90 Chaperone Function and Potentiates the Cytotoxicity of Hsp90 Inhibitor in Chemo-resistant Human Cancer Cells)

  • 본 연구는 Hsp90 inhibitor 및 SIRT1 inhibitor의 병용처리가 항암제 다제내성(MDR) 인간 암세포의 증식 억제에 효과적임을 밝혔다. SIRT1 활성 억제가 Hsp90 inhibitor인 17-AAG의 세포 독성의 효과를 증강시켰으며, 이로 인해 Hsp90 inhibitors에 대한 내성을 극복시킬 수 있음을 인간 자궁암세포인 HeyA8의 MDR 변이주인 HeyA8- MDR 세포에서 확인하였다. SIRT1 inhibitor는 Hsp90 inhibitor에 의한 Hsp90 샤페론 기능 억제를 증강시키며, ubiquitin ligase CHIP의 발현 증강을 유발하여, Hsp90 client protein 인 mutant p53 (mut p53)의 분해를 촉진시킨다. Mut p53 의 발현 감소는 암세포의 Hsp90 inhibitor 내성 획득의 가장 중요한 원인으로 지적되는 heat shock factor 1 (HSF1)/heat shock proteins (Hsps)의 발현 억제와 관련됨을 알 수 있었으며, 이는 항암제 다제내성 세포에서 SIRT1 inhibitor에 의하여 Hsp90 inhibitor에 대한 감수성이 증강되는 분자적 기전임을 밝혔다. 그러므로, SIRT1 억제에 의한 mut p53/HSF1 발현 감소가 MDR 암세포의 Hsp90 inhibitors 내성 극복에 매우 유효함을 시사하는 결과를 얻었다.

흰쥐 C6 신경교종 세포에서 Venlafaxine 그리고 Dexamethasone 처리가 열충격 단백질 70의 발현에 미치는 영향 (Effects of Venlafaxine and Dexamethasone Treatment on HSP70 Expression in Rat C6 Glioma Cells)

  • Object:The intracellular action of the antidepressant, venlafaxine, was studied in C6-gliomas using heat shock protein 70(HSP70) immunocytochemistry and HSP70 Western blots because HSP70 is associated with stress and depression. Methods:To examine how the glucocorticoid affects the expression of HSP70 in nerve cells, the rat C6 glioma cell was treated with dexamethasone for 6 hours. In addition, venlafaxine was administered to the experimental groups of C6 glioma cells for 1, 6, 24, and 72 hours each, after which the expression of HSP70 was investigated. Finally, venlafaxine and dexamethasone were simultaneously administered to the experimental groups for 1, 6, 24, and 72 hours, followed by an investigation of the expression of HSP70. Results:The short term(1 hour) venlafaxine treatment significantly increased the level of HSP70 expression. The short term treatment of venlafaxine with dexamethasone also increased the level of HSP70 expression but this reduction was not statistically significant. The long term(72 hours) venlafaxine with dexamethasone treatment significantly reduced the level of HSP70 expression. The long term treatment of venlafaxine also reduced the level of HSP70 expression but this reduction was not statistically significant. Dexamethasone(10uM, 6hours) did not affect the level of HSP70 expression compared with controls. Conclusion:Venlafaxine increases the expression of HSP70 at short term treatment, but prolonged treatment with dexamethasone suppresses the expression of HSP70.

내독소 투여 직후 가해진 열충격이 백서의 급성폐손상에 미치는 영향 (The Effect of Heat Co-treatment on Acute Lung Injury of the Rat Induced by Intratracheal Lipopolysaccharide)

  • 연구배경: 열 전처치는 조직 내에 열충격단백질의 생성을 유도하며 이러한 열 전처치가 내독소로 유도된 쥐의 급성폐손상을 감소시키고 패혈증에 의한 사망률을 감소시킨다고 알려져 있다. 그러나, 폐손상을 유발하는 원인에 노출된 후 가해진 열처치가 폐손상에 미치는 효과에 대하여는 아직까지 잘 알려져 있지 않다. 따라서 본 연구는 내독소 투여 직후 시행한 열충격이 내독소에 의해 유발된 쥐의 급성폐손상에 미치는 영향과 그에 따른 염증성 및 항염증성 사이토카인에 미치는 영항을 알아보고자 하였다. 방 법: 대조군은 백서의 기관지 내로 생리식염수를 투여하였고 열처치 대조군은 생리식염수 투여 직후 열처치를 시행하였다. 내독소군은 열처치 없이 내독소를 기관지내로 투여하였다. 열 전처치군은 내독소 투여 18시간 전에 열 전처치를 시행하였고, 열 동시처치 군은 내독소 투여 직후 열처치를 시행하였다. 내독소 투여 후 6시간에 기관지폐포세척을 시행하여 기관지폐포세척액 내의 호중구 백분율을 측정하였고 폐를 적출하여 myeloperoxidase(MPO)의 활성도를 측정하였으며 기관지폐포세척액과 혈청에서 LDH(lactic dehydrogenase), 단백질, IL(interleukin)-$1{\beta}$, TNF(tumor necrosis factor)-${\alpha}$ 및 IL-10를 측정하였다. 또한 각군에서 폐 조직 내 HSP72의 표현정도를 관찰하였다. 결 과: 1) 내독소군, 열 전처치군 및 열 동시처치군 모두에서 기관지폐포세척액 내의 호중구 백분율, 폐조직이 MPO, 혈청 및 기관지폐포세척액내의 $IL-1{\beta}$, $TNF-{\alpha}$ 및 IL-10이 대조군에 비해 증가하였다(각 p<0.05). 2) 열 동시처치군은 폐조직의 MPO와 기관지폐포세척액 단백질의 농도가 내독소군과 차이가 없었고 기관지폐포세척액의 LDH가 내독소군에 비해 증가하였다(p<0.05). 3) 열 동시처치군의 혈청 $TNF-{\alpha}$의 농도는 내독소군과 비교 시 증가하였다(p=0.01). 결 론: 내독소 투여 직후 시행한 열충격은 내독소로 유도되는 폐손상을 감소시키지 못하며 염증성 사이토카인의 농도를 증가시켰다.

Expression of Heat Shock Protein and Antioxidant Genes in Rice Leaf Under Heat Stress

  • We have previously investigated the proteome changes of rice leaves under heat stress (Lee et al. in Proteomics 2007a, 7:3369-3383), wherein a group of antioxidant proteins and heat shock proteins (HSPs) were found to be regulated differently. The present study focuses on the biochemical changes and gene expression profiles of heat shock protein and antioxidant genes in rice leaves in response to heat stress ($42^{\circ}C$) during a wide range of exposure times. The results show that hydrogen peroxide and proline contents increased significantly, suggesting an oxidative burst and osmotic imbalance under heat stress. The mRNA levels of chaperone 60, HSP70, HSP100, chloroplastic HSP26, and mitochondrial small HSP responded rapidly and showed maximum expression after 0.5 or 2 h under heat stress. Transcript levels of ascorbate peroxidase (APX), dehydroascorbate reductase (DHAR) and Cu-Zn superoxide dismutase (Cu-Zn SOD) showed a rapid and marked accumulation upon heat stress. While prolonged exposure to heat stress resulted in increased transcript levels of monodehydroascorbate reductase, peroxidase, glyoxalase 1, glutathione reductase, thioredoxin peroxidase, 2-Cysteine peroxiredoxin, and nucleoside diphosphate kinase 1, while the transcription of catalase was suppressed. Consistent with their changes in gene expression, the enzyme activities of APX and DHAR also increased significantly following exposure to heat stress. These results suggest that oxidative stress is usually caused by heat stress, and plants apply complex HSP- and antioxidant-mediated defense mechanisms to cope with heat stress.

Let-7c miRNA Inhibits the Proliferation and Migration of Heat-Denatured Dermal Fibroblasts Through Down-Regulating HSP70

  • Wound healing is a complex physiological process necessitating the coordinated action of various cell types, signals and microRNAs (miRNAs). However, little is known regarding the role of miRNAs in mediating this process. In the present study, we show that let-7c miRNA is decreased in heat-denatured fibroblasts and that inhibiting let-7c expression leads to the increased proliferation and migration of dermal fibroblasts, whereas the overexpression of let-7c exerts an opposite effect. Further investigation has identified heat shock protein 70 as a direct target of let-7c and has demonstrated that the expression of HSP70 in fibroblasts is negatively correlated with let-7c levels. Moreover, down-regulation of let-7c expression is accompanied by up-regulation of Bcl-2 expression and down-regulation of Bax expression, both of which are the downstream genes of HSP70. Notably, the knockdown of HSP70 by HSP70 siRNA apparently abrogates the stimulatory effect of let-7c inhibitor on heat-denatured fibroblasts proliferation and migration. Overall, we have identified let-7c as a key regulator that inhibits fibroblasts proliferation and migration during wound healing.

넙치, Paralichthys olivaceus에서의 Oxytetracyline 처리 농도가 생리ㆍ생화학적 반응에 미치는 영향 (Effects of Different Levels of Oxytetracycline on Physiological and Bio- chemical Responses in Olive Flounder, Paralichthys olivaceus)

  • 본 연구에서는 넙치, Paralichthys olivaceus를 이용하여 oxytetracycline (OTC)처리농도에 따른 체내에서 혈액 생리학적인 변화, 항산화 효소 및 열충격단백질(HSP)등의 변화를 조사하여 항생제의 안정적인 사용에 대한 기초 자료를 제공하고자 본 실험을 수행하였다. 실험구는 각각 0 (대조구), 100, 300 및 500ppm으로 설정하였으며, 1시간 약욕처리 후 순환시켜 0, 1 및 3시간 후에 혈액성상을 분석하였다. 혈액 중 hematocrit(Ht)수치는 전 실험구에서 감소하는 경향을 보였으나 유의한 차이는 나타나지 않았다. Red blood cell (RBC)는 농도 의존적으로 감소하는 경향을 나타내었다. 또한, 혈청 글루코오스 농도는 농도 의존적으로 증가되는 경향을 나타내었으나, 노출 이후 3시간 경과 후에는 이전 대조구 수준으로 감소되었다. 혈중 단백질량은 0시간째 전 실험구에서 감소되었다가 이후 증가하는 경향을 나타내었으나, 처리전의 대조구와 비교하여 유의하게 낮은 값을 나타내었다. SOD 효소 활성은 노출 직후 300 및 500ppm에서는 유의하게 높은 값을 나타내었다. CAT효소활성은 OTC처리 농도에 비례하여 감소하는 경향을 보였으나 유의한 차이는 나타내지 않았다. 그리고 HSP-70의 단백질량은 100 ppm및 500 ppm처리구에서 대조구에 비해 높게 발현되는 것이 확인되었다. 그러나 HSP-70 mRNA는 다소 증가하는 경향은 보였으나, 모든 실험구에서 유의한 차이는 나타나지 않았다.

Salicylic Acid and Wounding Induce Defense-Related Proteins in Chinese Cabbage

  • The response of plants to pathogens and wounding is dependent upon very sensitive perception mechanisms. Although genetic approaches have revealed a variety of resistance genes that activate common defense responses, defense-related proteins are not well characterized in plants. Therefore, we used a proteomic approach to determine which defense-related proteins are induced by salicylic acid (SA) and wounding in Chinese cabbage. We found that SA and wounding induce pathogenesis-related protein 1a (PR1a) at both protein and mRNA levels using proteomics and Northern blot analysis, respectively. This indicates that our proteomic approach is useful for identifying defense-related proteins. We also identified several other proteins that are induced by SA or wounding. Among the seven SA-induced proteins identified, four may be defense-related, including defense-related protein, phospholipase D (PLD), resistance protein RPS2 homolog, and L-ascorbate peroxidase. Out of the six wounding-induced proteins identified, three may be defense-related: heat shock cognate protein 70 (HSC70), polygalacturonase, and peroxidase P7. The precise functions of these proteins in plant defense responses await further study. However, identification of the defense-related proteins described in this study should allow us to better understand the mechanisms and signal transduction pathways involved in defense responses in Chinese cabbage.
