Nitric oxide 화합물 첨가가 한우 체외수정란의 체외발육에 미치는 효과

  • 소 난포란을 체외에서 성숙, 수정시킨 후, 2~8 세포기 수정란을 체외배양액인 CR$_1$aa에 일정량의 nitric oxide scavenger, Hb와, inhibitor인 L-nitro - L- arginine methyl ester (L-NAME)를 첨가 배양하여 체외발육에 미치는 영향을 검토하고자 실시하였다. 체외수정 후 체외배양 48시간에 난구세포의 제거 유.무에 따라 CR$_1$aa 배양액에 hemoglobin을 0, 1, 5$\mu\textrm{g}$/$m\ell$를 첨가한 구의 상실배기이상 체외발육성적은 난구세포를 제거한 구에서 23.8%, 33.3% 및 26.8%였으며, 난구세포를 제거하지 않은 구에서는 각각 39.5%, 54.8% 및 48.8%로써 난구세포를 제거하지 않은 구에 Hb를 1$\mu\textrm{g}$/$m\ell$를 첨가한 구가 여타구 보다 통계적으로 높은 결과를 얻었다(P<0.05). 체외수정 후 체외배양 96시간 후 난구세포를 제거 유.무에 따라 Hb를 0, 1, 5$\mu\textrm{g}$/$m\ell$를 첨가하였을 때, 상실배기 이상 체외발육성적은 난구세포를 제거한 구에서 각각 28.6%, 46.2% 및 39.1%였으며, 난구세포를 제거하지 않은 구에서는 각각 33.9%, 67.2% 및 46.0%로써, 난구세포를 제거하지 않은 구에 Hb를 1$\mu\textrm{g}$/$m\ell$첨가구가 여타구에 비해 통계적으로 유의하게 높게 나타났다( P<0.05).CR$_1$aa배양액에 L-NAME를 0, 10, 50 및 100mM을 첨가한 구에서 상실배이상 발육된 체외발육성적은 각각 55.6%, 64.9%, 58.8% 및 66.7%로써 L-NAME 첨가구가 무첨가구에 비해 커다란 차이가 나타나지 않았다(P>0.05). 본 실험의 결과로 볼 때, nitric oxide scavenger인 hemoglobin의 첨가는 체외수정 후 체외배양 96시간에 1$\mu\textrm{g}$/$m\ell$ 첨가하는 것이 체외수정란의 체외 발육율을 향상시켰으며, nitric oxide inhibitor인 L-NAME는 첨가 농도에 관계없이 체외수정란의 체외발육율에 영향을 미치지 않는 것으로 나타났다. 본 실험에서 생산된 배반포기 체외수정란의 세포수에는 커다란 차이가 인정되지 않았다.

A Comparison between Food and Nutrition Major, and Non-major, Female University Students in terms of their Nutrient Intakes and Hematological Status, with an Emphasis on Serum Iron (식품영양 전공 및 비전공 여대생 영양섭취상태 및 혈액성상에 관한 비교 연구 - 혈청 철분을 중심으로 -)

  • The purpose of this study was to compare nutrient intakes and the serum iron status of 74 Food and Nutrition major, and 45 non-major, female students at a university in Incheon. This cross-sectional survey was conducted using a self-administered questionnaire and the data were analyzed by the SPSS 10.0 program. The nutrient intake data collected from three-day dietary recalls were analyzed by the Computer Aided Nutritional Analysis Program. Anthropometric data and hematological indices of iron in the blood were measured. Average heights, weights, body fat and mid-upper arm circumference of Food and Nutrition major and non-major female students were 160.3 cm, 53.5kg, 25.8%, 23.7cm and 159.8cm, 55.5kg, 28.9%, 24.8cm, respectively. There were significant differences in body fat percentage and mid-upper arm circumference between the major and non-major students. In all subjects, daily dietary intakes of nutrients- except protein, vitarrlin B1, vitamin C and phosphorus - were lower than the Korean RDA. In particular, calcium and iron intakes of all subjects were under 60% of the Korean RDA. Values of RBC (red blood cell) count, Hb (hemoglobin), Hct (hematocrit), MCV (mean cell volume), MCH (mean cell hemoglobin), and MCHC (mean corpuscular hemoglobin concentration) of the non-major students were significantly higher compared to those of the major students. The diastolic blood pressure of the major students was negatively correlated with MCV, MCH, TS, and serum iron levels. Triceps skinfold thickness, mid-upper arm circumference and waist-to-hip ratios of the non-major students were negatively correlated with TIBC. Fat intake was positively correlated with RBC, Hb, Hct, and TIBC (total iron binding capacity) in the major students. Vitamin C intake was positively correlated with serum iron in the major students. Carbohydrate intake was positively correlated with Hb, Hct, and MCHC in the non-major students. Niacin and iron intakes were positively correlated with Hb and Hct in the non-major students. Therefore, nutrition education is necessary for female university students to improve nutritional status and to practice optimal nutrition strategies. (Korean J Nutrition 35(9) : 952~961, 2002)

The Effect of the Experience of Diabetes Education on Knowledge, Self-Care Behavior and Glycosylated Hemoglobin in Type 2 Diabetic Patients (당뇨교육경험이 제2형 당뇨환자의 지식, 자가간호행위, 당화혈색소에 미치는 영향)

  • Purpose: This study was to identify the effect of the experience of diabetes education on knowledge, self-care behavior and glycosylated hemoglobin (HbA1C). Further, this study was held to examine about patient's preferred methods of education and re-education frequency. Methods: 166 type 2 diabetes patients from two hospitals in Incheon participated in this study. Data were analyzed by using descriptive analysis, t-test, ANOVA, Scheffe's test and multiple regression analysis. Results: 72.3% patients needed re-education and the average interval of re-education was 8.53 months. Patients preferred education methods were lectures, practical training, and studying from pamphlet. Depending on the frequency of diabetes education, there were significant differences in the level of diabetes knowledge (F=10.88, p<.001) and self-care behaviors (F=4.59, p=.012), but there was not significant difference with HbA1C (F=1.53, p=.220). As to how much the diabetes education helped managing diabetes, there was a significant difference in the level of self-care behaviors (t=2.01, p=.049), but there were not significant differences in level of knowledge (t=1.10, p=.275) and HbA1C (t=-.33, p=.746). The experience of diabetes education was a significant factor which influenced patient's knowledge (t=3.93, p<.001) and self-care behaviors (t=2.21, p<.001). But HbA1C was not influenced by the experience of diabetes education (t=-1.68, p=.096). Conclusion: It is necessary to provide diabetes education with appropriate interval and methods and subjects that reflect the needs of patient through the study results.

Aerobic and Graduated Treadmill Exercise Decreases Blood Glucose Levels, Lipid Levels and Oxidative Stress in an Animal Model of Type 1 Diabetes Mellitus

  • Purpose: Exercise has been shown to be a simple and economical therapeutic modality that may be considered as an effective aid for diabetic mellitus. For example, exercise training increases insulin sensitivity in type 2 diabetes. But we found no reported of how exercise affect type 1 diabetes. This study investigated the impact of aerobic and graduated treadmill exercise regimens on body weight, glucose and insulin concentrations, lipid profiles, and oxidative stress indicators in rats with streptozotocin (STZ) induced diabetes. Glycosylated hemoglobin ($HbA_{1c}$) was determined as an indicator of glucose control during exercise. Methods: In our study, a total of 40 rats were used. Three groups of 10 rats each were given STZ to induce diabetes. The remaining 10 rats became the normal group. After 28 days we determined biochemical parameters such as glucose, glycosylated hemoglobin ($HbA_{1c}$), insulin concentration, serum total cholesterol (TC), triglycerides (TG), and high-density lipoprotein (HDL). Superoxide dismutase (SOD) and catalase activities were also measured. Results: Concentrations of blood glucose and $HbA_{1c}$ in the moderated exercise groups were significantly decreased after 28 days compared with the control group (p<0.05). There was a significant reduction in serum TC and TG in the experimental groups. The activity of SOD increased significantly by 17.70% and 48.25% respectively. Conclusion: These results indicate that physical training and exercise training affects body weight, fasting blood glucose, $HbA_{1c}$, insulin, lipid profiles, and antioxidant status in rats with streptozotocin-induced diabetes. We suggest that graduated treadmill exercise may have therapeutic, preventative, and protective effects against diabetes mellitusby improving glycemic control, oxidant defenses, and lipid metabolism.

The Change of Blood Properties of HK Phenotype Jindo dogs Administrated with Excessive Amount of Water Celery Extracts (과량의 미나리 생즙 투여가 HK phenotype 진도견의 혈액상의 미치는 영향)

  • This study was conducted to investigate changes in blood properties of high potassium (HK) phenotype Jindo dogs (15kg$\pm$2kg) after daily oral administration with water celery extracts (10 ml/kg) for 7 days. Blood samples were collected for three days in a row before administration of water celery extracts. After water celery extracts administration, blood samples were collected at 3h, 6h, 9h and then on daily basis until day 10 post administration (PA). At day 15, final sample was collected. Blood samples were analyzed on the basis of red blood cell (RBC), white blood cell (WBC), packed cell volume (PCV), hemoglobin (Hb) concentration, mean corpuscular volume(MCV), mean corpuscular hemoglobin concentration(MCHC), gluthathione concentration(GSH) and met-hemoglobin(Met-Hb) concentration. The significant changes (p<0.01, p<0.05) of RBCs were shown at 3 h to day 5, and days 7 and 9 after administration. PCV values were decreased form 3 h to day 10 after administration. Mean Hb concentration showed significant increase as 3 h to day 3, and day 6 to day 9 after administration. The significant changes (p<0.05) of WBCs were shown at 9 h and day 1 after administration. The increased numbers of MCV were detected at days 6 to 9 after administration (p<0.05, p<0.01). The significant changes of MCHC were shown at 9h and day 1 after administration. The significant increases (p<0.01, p<0.05) of GSH concentration were detected at days 1, 6 and 7 after administration. In Met-Hb concentration, the significant increases (p<0.05) occurred at only 9h and day 7 after administration, The significant increases (p<0.01, p<0.05) of reticulocyte were detected at days 2, 4, 5, 6 and 7. Data from blood samples collected at day 15 after administration showed that all of blood analysis results returned to normal level, compared to controls.

Electrochemistry of Hemoglobin in the Chitosan and TiO2 Nanoparticles Composite Film Modified Carbon Ionic Liquid Electrode and Its Electrocatalysis

  • Direct electron transfer of hemoglobin (Hb) in the chitosan (CTS) and $TiO_2$ nanoparticles (nano-$TiO_2$) composite films was achieved by using a room temperature ionic liquid of 1-butyl-3-methylimidazolium hexafluorophosphate ($BMIMPF_6$) modified carbon paste electrode (CILE) as the basal electrode. UV-Vis and FT-IR spectroscopy indicated that Hb in the film retained the native structure. Electrochemical investigation indicated that a pair of well-defined quasi-reversible redox peaks of Hb heme Fe(III)/Fe(II) was obtained with the formal potential located at -0.340 V (νs. SCE) in pH 7.0 phosphate buffer solution (PBS). The electrochemical parameters such as the electron transfer coefficient (α), the electron transfer number (n) and the standard electron transfer rate constant ($k_s$) were got as 0.422, 0.93 and 0.117 $s^{-1}$, respectively. The fabricated CTS/nano-$TiO_2$/Hb/CILE showed good electrocatalytic ability to the reduction of trichloroacetic acid (TCA) and hydrogen peroxide ($H_2O_2$), which exhibited a potential application in fabricating a new kind of third generation biosensor.

Studies on the Blood Protein Polymorphisms of Deer: Cervus nippon, Cervus unicolor (녹(鹿)의 혈청단백(血淸蛋白)에 관한 연구(硏究))

  • 한국(韓國), 대만(臺灣), 일본(日本)에서 사육(飼育)하고 있는 꽃사슴(Cervus nippon) 129두(頭)와 물사슴(Cervus unicolor) 7두(頭)에서 hemoglobin, transferrin, albumin carbonic anhydrase, slow-${\alpha}_2$ 및 amylase형(型)을 전기영동(電氣泳動)에 의(依)하여 분석(分析)한 결과(結果) 다음과 같은 결론(結論)을 얻었다. 1. cellulose acetate에 의(依)한 전기영동(電氣泳動)이 starch gel에 의(依)한 전기영동(電氣泳動)보다 hemoglobin형(型) 분리(分離)에 있어서 더 간편하며 시간이 적게 걸리고, 선명(鮮明)할 뿐만 아니라 영구보존(永久保存)이 가능(可能)하다. 2. 꽃사슴의 hemoglobin형(型)은 $Hb^F$, $Hb^{FS}$, $Hb^S$형(型)으로 분리(分離)되었으나 물사슴에 있어서는 전부(全部) $Hb^F$형(型)으로 나타났다. 3. hemoglobin ${\beta}$ chain은 4가지형(型) 즉 ${\beta}$-1, ${\beta}$-2, ${\beta}$-3 및 ${\beta}$-4로 분리(分離)되었다. 4. hemoglobin ${\alpha}$ chain은 ${\alpha}_1$${\alpha}_1{\alpha}_2$형(型)으로 분리(分離)되었다. 5. slow-${\alpha}_2$형(型)은 A형(型)과 AB형(型)으로 분리(分離)되었으며, 꽃사슴에 있어서는 AB형(型)이 12% 출현(出現)하였으나 물사슴에서는 전부(全部) A형(型)으로 AB형(型)은 없었다. 6. albumin형(型)에서는 F형(型)과 S형(型)으로 분리(分離)되었으며 꽃사슴은 전부(全部) F형(型)이였고, 물사슴은 전부(全部) S형(型)이였다. 7. transferrin형(型), carbonic anhydrase형(型) 및 amylase형(型)은 전부(全部) 각각(各各) 1종류(種類)의 형(型)이였다.

A Study on Nutritional Status after Gastrectomy of Gastric Cancer Patients in Jeonbuk Province (전북지역 위암환자의 위절제술 전.후 영양상태 평가)

  • The purpose of this study was to investigate the nutritional status before and after gastrectomy of gastric cancer patients in Jeonbuk Province. The subjects were 136 patients with gastrectomy of gastric cancer. We assessed the nutritional status before and after an operation by general characteristics (age, clinicopathological stage, type of operation, method of reconstruction), anthropometric characteristics (height, weight, skeletal muscle mass, body fat mass, body mass index (BMI), percent body fat, arm muscle circumference AMC, arm circumference AC) and biochemical characteristics (hemoglobin Hb, hematocrit Hct, mein corpuscular volume MCV, mean corpuscular hemoglobin MCH, total lymphocyte count TLC, serum albumin, serum transferrin). Mean ages were 60.0 yrs in males and 58.8 yrs in females. Age, clinicopathological stage, types of operation and reconstruction methods were not significantly different between males and females. Weight, skeletal muscle mass, body fat mass, BMI, percent body fat, AMC and AC significantly deteriorated by gastrectomy. There were severe weight losses in males and females after gastrectomy. Hemoglobin, Hct, MCV, MCH, TLC, albumin and transferrin significantly deteriorated by gastrectomy. After gastrectomy, subjects who were assessed as malnounrished in Hb and Hct were increased in male and those who were assessed as malnounrished in Hb were increased in females. These results suggest that a nutrition intervention, specially for energy, protein and iron, is necessary to improve the nutritional status of gastric cancer patients with gastrectomy.

Electrochemical Investigation of Bovine Hemoglobin at an Acetylene Black Paste Electrode in the Presence of Sodium Dodecyl Sulfate

  • Electrochemical behaviors of bovine hemoglobin (Hb) at an acetylene black paste electrode based on the enhancement effect of sodium dodecyl sulfate (SDS) were investigated. In the optimal conditions, a very weak reduction peak was observed at an acetylene black paste electrode for hemoglobin in the absence of SDS. However, the reduction peak current increased remarkably after the addition of 4.0 × 10?4 mol L?1 SDS, suggesting that SDS exhibits obvious enhancement effect to the determination of hemoglobin. All the experimental parameters, such as pH value, concentration of SDS, accumulation time and accumulation potential were optimized for hemoglobin analysis. The proposed method possesses high sensitivity (detection limit is 3.0 × 10?9 mol L?1), wide linearity (6.0 × 10?9 to 6.0 × 10?7 mol L?1), rapid response and low cost. Finally, the method was successfully employed to determine hemoglobin in a spiked sample.

A study on biomarker for biomonitoring of 1,3-butadiene inhalation exposure (1,3-부타디엔 호흡기 노출의 생체지표 대사물질에 대한 연구)

  • The purpose of this study is to investigate the appropriate metabolite as biomarker for the biomonitoring of 1,3-butadiene(BD) inhalation exposure. We measured the hemoglobin adducts which were extracted from the blood of the ICR mice inhalation exposure with 100ppm and 500ppm 1,3-butadiene for 2 weeks(5 hr/day ${\times}$ 5 days/week). Hemoglobin adducts were the (N-2-hydroxy-3 -butenyl) valine (HB Val) and (N-2,3,4-trihydroxy-butyl)valine (THB Val). Body weights of the exposure groups were significantly lower from 11 exposure post-day in 100ppm BD inhalation mice and from 7 exposure post-day in 500ppm BD inhalation mice than in control. The levels of HB Val are 0.8~1.7pmol/mg globin for 100ppm BD inhalation exposure, and 2.1~4.4 pmol/mg globin for 500ppm BD inhalation exposure. The levels of THB Val are 15.0~22.0 pmol/mg globin in 100ppm BD inhalation exposure, and 34.8~45.7 pmol/mg globin for 500ppm BD inhalation exposure. So the levels of THB Val and HB Val are proportional relationship with BD exposure level. THB Val is 12.9~18.8 times higher level that HB Val in 100ppm BD exposure group and 10.4~16.6 times higher level than HB Val in 500ppm BD exposure group. We concluded that THB Val is an appropriate metabolite as biomarker for the biomonitoring for BD inhalation exposure.
