Effects of Applying Cattle Manure on Carrying Capacity of Organic Livestock per Unit Area of Summer Forage Crops (우분뇨 시용이 하계사료작물의 단위면적당 유기가축 사육능력에 미치는 영향)

  • This study was carried out to select a proper forage crop, and to estimate the proper level of application of cattle manure and carrying capacity of organic livestock per unit area. Corns and forage sorghum hybrids were cultivated with different types of livestock manures and different amount of them to produce organic forage. For both corns and forage sorghum hybrids, no fertilizer plots had significantly (p<0.05) lower annual dry matter (DM), crude protein (CP) and total digestible nutrients (TDN) yields than those of other plots, whereas the N-P-K (nitrogen-phosphorous-kalium) plots ranked the highest yields, followed by 150% cattle manure plots and 100% cattle manure plots. DM, CP and TDN yields of in cattle manure plots were significantly (p<0.05) higher than those of no fertilizer and P-K (phosphorous-kalium) plots. The yields of in cattle slurry plots tended to be a little higher than those of in composted cattle manure plots. Assuming that corn and forage sorghum hybrids produced from this trial were fed at 70% level to 450kg of Hanwoo heifer for 400g of average daily gain, the carrying capacity (head/year/ha) of livestock ranked the highest in 150% cattle slurry plots (mean 6.0 heads), followed by 100% cattle slurry plots (mean 5.3 heads), 150% composted cattle manure plots (mean 4.7 heads), 100% composted cattle manure plots (mean 4.4 heads), and no fertilizer plots (mean 2.8 heads) in corns (or the cultivation of corns). Meanwhile, in the case of forage sorghum hybrids, 150% cattle slurry plots (mean 6.4 heads) ranked the highest carrying capacity, followed by 150% composted cattle manure plots (mean 4.8 heads), 100% cattle slurry plots (mean 4.4 heads), 100% composted cattle manure plots (mean 4.1 heads), and no fertilizer plots (mean 2.8 heads). The results indicated that the application of livestock manure to cultivated soil could enhance not only DM and TDN yields, but also the carrying capacity of organic livestock as compared with the effect of chemical fertilizers. In conclusion, the production of organic forage with reutilized livestock manure will facilitate the reduction of environmental pollution and the production of environmentally friendly agricultural products by resource circulating system.

Accuracy of Imputation of Microsatellite Markers from BovineSNP50 and BovineHD BeadChip in Hanwoo Population of Korea

  • Until now microsatellite (MS) have been a popular choice of markers for parentage verification. Recently many countries have moved or are in process of moving from MS markers to single nucleotide polymorphism (SNP) markers for parentage testing. FAO-ISAG has also come up with a panel of 200 SNPs to replace the use of MS markers in parentage verification. However, in many countries most of the animals were genotyped by MS markers till now and the sudden shift to SNP markers will render the data of those animals useless. As National Institute of Animal Science in South Korea plans to move from standard ISAG recommended MS markers to SNPs, it faces the dilemma of exclusion of old animals that were genotyped by MS markers. Thus to facilitate this shift from MS to SNPs, such that the existing animals with MS data could still be used for parentage verification, this study was performed. In the current study we performed imputation of MS markers from the SNPs in the 500-kb region of the MS marker on either side. This method will provide an easy option for the labs to combine the data from the old and the current set of animals. It will be a cost efficient replacement of genotyping with the additional markers. We used 1,480 Hanwoo animals with both the MS data and SNP data to impute in the validation animals. We also compared the imputation accuracy between BovineSNP50 and BovineHD BeadChip. In our study the genotype concordance of 40% and 43% was observed in the BovineSNP50 and BovineHD BeadChip respectively.

Discrimination study between carcass yield and meat quality by gender in Korean native cattle (Hanwoo)

  • Objective: The aim of this study was to identify a distribution pattern of meat quality grade (MQG) as a function of carcass yield index (CYI) and the gender of Hanwoo (bull, cow, and steer) to determine the optimum point between both yield and quality. We also attempted to identify how pre- and post-deboning variables affect the gender-specific beef quality of Hanwoo. Methods: A total of 31 deboning variables, consisting of 7 pre-deboning and 24 post-deboning variables from bulls (n = 139), cows (n = 69), and steers (n = 153), were obtained from the National Institute of Animal Science (NIAS) in South Korea. The database was reconstructed to be suitable for a statistical significance test between the CYI and the MQG as well as classification of meat quality. Discriminant function analysis was used for classifying MQG using the deboning parameters of Hanwoo by gender. Results: The means of CYI according to 1+, 1, 2, and 3 of MQG were 68.64±2.02, 68.85±1.94, 68.62±5.88, and 70.99±3.32, respectively. High carcass yield correlated with low-quality grade, while high-quality meat most frequently was obtained from steers. The classification ability of pre-deboning parameters was higher than that of post-deboning parameters. Moisture and the shear force were the common significant parameters in all discriminant functions having a classification accuracy of 80.6%, 71%, and 56.9% for the bull, cow, and steer, respectively. Conclusion: This study provides basic information for predicting the meat quality by gender using pre-deboning variables consistent with the actual grading index.

Crude Protein Requirements for Maintenance of Hanwoo Steers by Protein Balance Trial (단백질 균형시험에 의한 한우 거세우의 유지 조단백질 요구량)

  • This experiment was conducted to determine crude protein requirements for maintenance (CPm) of growing Hanwoo steers. Eighteen Hanwoo steers (173.7kg) were allocated randomly to diets with three levels of crude protein in the concentrate by replacing ground corn with corn gluten meal: 0% (LCP), 7.2% (MCP) and 14.2% (HCP), respectively. Hanwoo steers in each treatment were fed diets containing rice straw (20%) and concentrates (80%). CP intake was 5.5, 7.8 and 10.7g/BW0.75 for LCP, MCP and HCP, respectively. Protein balance was -0.05, 2.36 and 4.07g/BW0.75 for LCP, MCP and HCP, respectively. Intercept of the regression equation between CP intake and retained CP indicated that CPm was 5.56g/BW0.75. The estimate of CPm adopted by Korean Feeding Standard (2002) was almost 20% higher than that adopted by Japanese Feeding Standard for Beef Cattle (2000); this would need to be verified with more feeding trial data set including protein requirements for growth.

Cellularity of Adipose Tissue Obtained from Different Sex and Growth Stages of Hanwoo Cattle and Sheep

  • In order to understand the effects of sex or age on cellular characteristics of adipocytes from Hanwoo and sheep, samples were obtained from omental, subcutaneous, intermuscular and intramuscular adipose tissue depots of bulls, steers, heifers and cows in Hanwoo, and perirenal, omental and subcutaneous adipose tissues of fetal lambs, suckling lambs and wethers in sheep. In case of Hanwoo, mean diameter, surface area and volume of adipocytes from each depot were obtained by multisizer II (Coulter Co., UK). Osmium-fixed adipocytes were sized and counted using $560{\mu}m$ aperture. For samples obtained from sheep, cellularity was measured by using microscope and MCV program of Texas Instrument. Bulls had less subcutaneous and kidney fat than steers even though their slaughter and carcass weight were heavier. The amounts of fat from cows were greater in subcutaneous, kidney and internal organs than heifers. Steers had larger adipocytes in subcutaneous, intermuscular and intramuscular adipose tissues than bulls, although the differences were significant only for the subcutaneous adipose tissue depots. Adipocytes appeared to be largest in omental and smallest in intramuscular adipose tissue, although there were no significant differences among tissues. In a comparison of heifers and cows, significant site effects (p<0.05) were shown in adipocyte diameter, surface area and volume, and adipocyte appeared to be largest in omental tissue. Statistical difference (p<0.05) was only shown in cell volume of intramuscular tissue which was higher in cow than heifer. Intramuscular adipose tissue tended to have relatively greater numbers of cells per gram tissue and reflect lesser maturity of intramuscular adipose tissue relative to other adipose tissues. In sheep, regardless of adipose tissue depots, wethers had the greater adipocyte diameters than those at any other growth stage of sheep. Within adipose depots, the ranking of cell size was the greatest in the omental tissue of wether and the lowest in the renal and subcutaneous adipose tissue depots of fetal lamb. The cell size of adipocyte became larger with age, especially from fetal to suckling lamb due to a rapid hypertrophy of both perirenal and subcutaneous adipocytes during the suckling period.

Association Study Between the Polymorphisms of Exostosin-1 Gene and Economic Traits in Hanwoo (한우 Exostosin-1 유전자의 SNP 탐색 및 경제형질 관련성 분석)

  • The aim of this study was to identify the polymorphism on exostosin-1 (EXT1) gene and to associate with economic traits in Hanwoo (Korean cattle). We sequenced for detection of single nucleotide polymorphism (SNP) with 24 unrelated individuals and identified four SNPs (T272196A, C272359T, G290964A and A302092G). Relationship between the genotypes of 583 Hanwoo individuals by PCR-RFLP and economic traits were analyzed by general linear model. In EXT1 gene, there were four SNPs associated with economic traits such as eye muscle area breeding value, marbling score breeding value, backfat and thickness breeding value (p<0.05 to p<0.01). In conclusion, this study indicates an important role of EXT1 gene in determining the meat quality or economic characteristics in Hanwoo.

Study on the Carcass Yield Grade Traits and Prediction of Retail Product Weight in Hanwoo Beef (한우도체의 육량등급 요인 특성과 판매 정육량 추정)

  • Analyses were conducted to estimate carcass component of yield grade factors by sex and live weight class and to develop the prediction equation of retail product weight by sex in Korean native cattle (Hanwoo). Data from 42,113 Hanwoo carcasses were used to estimate the traits of yield grade factor and an additional 1,066 carcasses were used to develop the prediction equation of retail meat weight. The average of fasting weight of cow, bull and steer were 529 kg, 596 kg, and 634 kg respectively. Carcass weight (CW), backfat thickness (BFT), loineye area (REA), Index score of wholesale meat and yield grade were significantly (p<0.01) affected by sex and live weight. The lean meat percentage, fat percentage and bone percentage based on the weight of cold carcasses were significantly different (p<0.05) between sex groups. The equation of predicting the retail meat product from this study could be expressed as a multiple regression $Y=-4.18+0.63{\times}CW\;(kg)-0.17{\times}BFT\;(cm)+0.16{\times}REA\;(cm^2)$, $R^2=0.93$. Among the independent factors, the BFT was the highest contributor to the prediction equation. Using the equation from this study should allow for rapid, precise and cost-effective assessment of the retail product in Hanwoo beef carcasses.

Aroma Pattern Analysis of Hanwoo Beef (M. longissimus) using Electronic Nose during Refrigerated Storage (전자 코를 이용한 한우 등심육의 냉장저장 중 향기 패턴 분석)

  • This study was carried out to investigate aroma patterns of Hanwoo (Korean cattle) beef using electronic nose during refrigerated storage, and to compare these results with chemical quality (pH, TBARS). The M. longissimus muscle from Hanwoo carcasses after 24 hrs postmortem was obtained and stored at 5${\pm}$1$^{\circ}C$ for 7 days. Sensitivity (dR/RO) values among electronic nose data were changed differently during refrigerated storage, and showed significant difference on the 7th day of storage (p<0.01). The dR/RO from SY/G, SY/AA, SY/Gh, SY/gCTl, SY/gCT decreased but those from SY/LG, T30/1, P10/1, P10/2, P40/1, T70/2, PA2 increased during storage for 7 days. Mapping these data using PCA (principal component analysis) showed that the 1st day data were present in the middle of the right side, the 3rd day data were present in bottom part of this area and the 7th day data spread out more widely on the left side. In case of DFA (discriminant factor analysis), the flock clustered round and located in different side clearly comparing with PCA plot. In analysis of correlation coefficients among electronic nose data and chemical quality data, there was significant correlation among sensor data (p<0.001). But pH and TBARS were not significantly correlated with electronic nose data. Consequently, PCA and DFA plot by electronic nose data showed difference during refrigerated storage and there were significant correlations among sensors. Therefore it will be possible to detect separate aroma patterns of Hanwoo beef using electronic nose.

Improved pregnancy rate and sex ratio in fresh/frozen in vivo derived embryo transfer of Hanwoo (Bos taurus coreanae) cows

  • This study aimed to assess the effects of embryonic developmental stage, quality grade, and fresh or frozen/thawed conditions on the pregnancy rate and sex ratio of live offspring in Hanwoo (Bos taurus coreanae) cows. The quality and developmental stage of in vivo-derived (IVD) transferred embryos were evaluated using the standard criteria of the International Embryo Technology Society. The recipient cows were synchronized using conventional (estradiol benzoate and progesterone) protocols before embryo transfer. Embryos were transferred to 297 cows, and pregnancy was monitored for 60-70 days after embryo transfer. The pregnancy rates of fresh and frozen/thawed embryos were 56.90% and 52.49%, respectively. Pregnancy rates varied according to embryo quality (56.18% for grade 1 vs. 36.67% for grade 2). Pregnancy rates also varied by developmental stage and cryopreservation (67.86% vs. 63.49% for stage 4-1, 64.00% vs. 54.72% for 5-1, and 50.00% vs. 47.83% for 6-1, in fresh embryos vs. frozen/thawed embryos, respectively). For stage 7-1, the pregnancy rates were 72.73% for fresh embryos and 20.00% for frozen/thawed embryos. In 66 fresh embryos, the sex ratio of live offspring was 5:5, whereas it was 4(female):6(male) for frozen/thawed embryos among the 95 frozen/thawed embryos. The miscarriage rate was approximately 3% higher for frozen/thawed embryos than for fresh embryos (18.1% for fresh vs. 21.1% for frozen). Seasonal fertility rates were 33.3% in spring, 55.67% in summer, 52.8% in autumn, 60.0% in winter. The following male-to-female ratios were observed in different seasons: 6.7:3.3 in spring, 4.0:6.0 in summer, 5.5:4.5 in autumn, and 3.3:6.7 in winter. The current data revealed no significant differences in pregnancy rates between fresh and frozen/thawed IVD embryos. However, there was a lower pregnancy rate with advanced-stage frozen/thawed embryos (stage 7-1). The current study provides comprehensive results for the better optimization of embryo transfer in Hanwoo cattle to obtain the desired fertility rate, pregnancy rate, and sex ratio of calves. These results provide important insights into the factors that influence the viability and success of IVD embryo transfer in Hanwoo cows and may have practical applications for improving breeding programs and reducing production costs.

Development of Natural Alkaloid-feed Additive for Enhancing Yield Grade of Hanwoo Steers (거세한우 육량향상을 위한 알칼로이드계열 천연물 이용 사료첨가제 개발)

  • This study was indicated that both potato leaves and stems contained certain amount of alkaloids compounds which digested in the rumen of Hanwoo steers. Three candidate plant byproducts such as potato, tomato, and eggplant were collected and dried for making feed additives. Among the candidate byproducts, potato extract was contained 2.05±0.27mg/g of α-chaconine and 0.60±0.08mg/g of α-solanine. These glycoalkaloids was potentially activated as functional compounds to skeletal muscle in beef cattle. However, tomato and eggplant byproducts were not detected any alkaloids compounds. After potato byproducts collected and dried for 24 hours, dry matter of potato byproduct were used as feeding study for evaluating active dose titrations. For evaluating dietary rate of rumen microbes, potato dry matter were treated with in situ hybridization for 72 hours using annulated Hanwoo steers. The glycoalkaloids contained potato dry matter were digested until 12 hours after treatment. Blood concentration of glycoalkaloids were not detected after 24 hours treatment of potato dry matter. These data indicated that potato byproduct were digested with rumen microbes for 24 hours after treatments.
