Development of Sumulation Model for Breeding Schemes of Hanwoo(Korean Cattle) (한우의 개량 체계 모의실험을 위한 모형 개발)

  • A multiple-trait stochastic computer simulation model was constructed to predict the breeding schemes and selection methods on Hanwoo(Korean cattle). The model could be used four kinds of selection criteria (random, phenotype and true or estimated breeding values). At the test run in various population size for 20 years, all estimated parameters of the each simulated populations were resulted similar to input parameters. The deviations between input and output values of parameter in the large population were smaller than in the small population. The simulated results obtained from ten small populations consisted with one sire and ten dams in each population for 500 years were as follows; Inbreeding coefficients of population were similar to theoretical estimating function. Mean values of each traits selected were randomly drifted by generation, but they were converged into a value when inbreeding coefficients came close to one. Additive genetic variances within each population were reduced by generation, and they were converged into zero when inbreeding coefficients came close to one. These results indicated that the simulated populations hold to statistical properties of input parameters.

Characteristics of Rice Hulls, Sawdust, Wood Shavings and Mixture of Sawdust and Wood Shavings, and Their Usefulness According to the Pen Location for Hanwoo Cattle

  • In this study, two experiments were conducted to investigate the physicochemical characteristics (Exp. I) of bedding materials such as rice hulls (RH), sawdust (SD), wood shavings (WS) and sawdust+wood shavings (S+W; 1:1 in volume), and utilization of these beddings except RH (Exp. II) for rearing beef cattle. In Exp. I, the distribution of particle size (%) with $250{\mu}m$ and below $250{\mu}m$ was greater (p<0.05) in SD (30.4) than RH (4.4), WS (18.8) and S+W (20.1). Bulk density ($kg/m^3$) of bedding materials was directly proportional to the percentage of $250{\mu}m$ and below $250{\mu}m$ particles, 178, 46, 112, and 88 for SD, WD, S+W and RH, respectively. Water absorption rate (%) after submersion in water for 24 h was higher (p<0.05) in WS (540.2) compared to SD (270.2), S+W (368.2). The S+W had an intermediate value of the absorption rate between SD and WS, but had an outstanding durability of water absorption capacity. Moisture evaporation rate (%) for 12 h was higher (p<0.05) in WS (75.4) than SD (70.5), S+W (72.2) and RH (57.8). Average ammonia emission ($mg/m^2/h$) for 36 h was higher (p<0.05) in RH (3.15) than SD (1.70), WS (1.63), and S+W (1.73). In Exp. II, thirty six Hanwoo cows were allocated in 9 pens with one side on feed bunk side (Side A) and another side equipped with water supply (Side B) for 3 weeks with duplicated periods. Average moisture concentrations (%) of beddings were higher (p<0.05) in WS (side A, 65.7; side B, 57.9) than SD (side A, 62.5; side B, 52.2) and S+W (side A, 61.6; side B, 50.7). Regardless of types of beddings, moisture concentrations (%) of beddings within a pen were lower (p<0.05) at side B than A, implying longer period of utilization. These results suggest that using S+W would be a better choice than SD or WS alone, considering physicochemical characteristics and economics, and RH is not a suitable material as a bedding for beef cattle.

Estimation of Genetic and Environmental Parameters of Carcass Traits in Hanwoo (Korean Native Cattle) Populations

  • Genetic parameters of carcass weight (CWT), dressing percent (DP), cook loss (CL), eye muscle area (EMA), back fat thickness (BFT), and meat tenderness in terms of mastication (MAS), shear force (SFR) and penetration (PEN) in Korean native cattle were estimated in this study. Effects of sire, location and their interaction on these traits were also evaluated. Sire effects were found to be significant on all the traits studied except for PEN. The CWT and DP were also significantly affected both by location (p<0.01) and by interaction effect between sire${\times}$location (p<0.05). The EMA was significantly (p<0.05) affected by location but not by interaction effect between sire${\times}$location. All the traits were positively correlated ($r_g$ and $r_p$) with each other except between CL and meat tenderness (negatively correlated). Moderate to high genetic correlations between CWT and other important traits were obtained; indicating that selection for CWT would lead to improve carcass quality. Heritability estimates were 0.64, 0.52, 0.37, 0.25, 0.19 and 0.18 for MAS, SFR, CWT, PEN, DP and EMA, respectively.

  • SBovine tuberculosis is a chronic bacterial disease of animals and humans caused by Mycobacterium bovis. Besides the classical intradermal tuberculin test, a number of blood and serum tests have been used. The purpose of this study was to establish seroprevalence of M. bovis. The sera were screened using the ELISA technique. A total seroprevalence of 65.8% in positive cattle, suspect 36.0%, negative 5.9% in TB-infected herds by PPD and dairy cattle is 3.0%, Hanwoo is 1.6% in TB-free herds. The deer of seroprevalence is 55.0% in TB-infected herd and 7.7% in TB-free herds.

Forage and Cattle Productivities of Intensive Grazing System (집약방목지에서의 목초 및 가축생산성에 관한 연구)

  • This experiment was conducted to establish the intensive Grazing system through the improved pasture utilization of the Hanwoo beef cattle. Intensive grazing period, 2 to 3 days was compared to conventional rotation-grazing period, 5 to 6 days as control. The intensive grazing system which has 2 to 3 days of grazing period, increased forage DM yield(7.33t/ha) by 17% compared to control(6.28t/ha), and contributed to stabilization of the seasonal productivity through the increased DM yield after August. Plant height, feed intake and efficiency of grazing was increased in intensive rotation.

Identification of the Proteins Expressed at Optimum Marbling in Hanwoo Loin (한우육의 육질에 중요 영향을 미치는 근내지방축적 최적기에 발현되는 단백질 동정)

  • Proteins from the loin tissues age ranged from 0 to 24 months of ten Korean cattle were extracted, separated and compared on two dimensional(2 D) gels to identify the proteins whose expression is highly correlated to marbling. We also compared the difference of loin proteins between castrated and non castrated bull cows on two dimensional gels. As the marbling in the loin of the cattle is optimized at 18 to 24 months, eight proteins expressed significantly higher level in 24 month than in 0 or 6 month were selected in terms of isoelectric points(pIs) and molecular weights. Using these values, we searched the Swiss Prot database via the ExPASy molecular biology server with TagIdent program. The proteins with the nearest molecular weights and isoelectric points were selected from the lists. These possible candidates were confirmed by N terminal microsequencing of the eight selected proteins. Three proteins, myoglobin, hemoglobin and ATPase, whose N termini were not blocked could be microsequenced and found to be exactly matched to the selected candidates. It is suggested that the proteins increasingly expressed in marbling periods can be involved in meat color, lipid transport and flavor improvement.

Determining the Authenticity of Labeled Traceability Information by DNA Identity Test for Hanwoo Meats Distributed in Seoul, Korea (DNA 동일성 검사를 통한 서울지역 유통 한우육의 표시 이력정보 진위 판별)

  • Beef traceability systems help prevent the distribution of Hanwoo (Korean native cattle) meat as imported beef. In particular, assigning a traceability number to each cattle can provide all information regarding the purchased Hanwoo meat to the consumers. In the present study, a DNA identity test was conducted on 344 samples of Hanwoo meat from large livestock product stores in Seoul between 2021 and 2022 to determine the authenticity of important label information, such as the traceability number. Traceability number mismatch was confirmed in 45 cases (13.1%). The mismatch rate decreased to 11.3% in 2022 from 14.7% in 2021, and the mismatch rate was higher in the northern region (16.9%) than in the southern region (10.2%). In addition, of the six brands, B and D showed satisfactory traceability system management, whereas E and A showed poor traceability system management, with significant differences (P<0.001). The actual traceability number confirmation rate was only 53.9% among the mismatch samples. However, examination of the authenticity of label information of the samples within the identified range revealed false marking in the order of the traceability number (13.1%), sex (2.9%), slaughterhouse name (2.2%), and grade (1.6%); no false marking of breed (Hanwoo) was noted. To prevent the distribution of erroneously marked livestock products, the authenticity of label information must be determined promptly. Therefore, a legal basis must be established mandating the filling of a daily work sheet, including the traceability number of beef, in partial meat subdivisions. Our findings can be used as reference data to guide the management direction of traceability systems for ensuring transparency in the distribution of livestock products.

Effects of Applying Cattle Manure on Carrying Capacity of Organic Livestock per Unit Area of Summer Forage Crops (우분뇨 시용이 하계사료작물의 단위면적당 유기가축 사육능력에 미치는 영향)

  • This study was carried out to select a proper forage crop, and to estimate the proper level of application of cattle manure and carrying capacity of organic livestock per unit area. Corns and forage sorghum hybrids were cultivated with different types of livestock manures and different amount of them to produce organic forage. For both corns and forage sorghum hybrids, no fertilizer plots had significantly (p<0.05) lower annual dry matter (DM), crude protein (CP) and total digestible nutrients (TDN) yields than those of other plots, whereas the N-P-K (nitrogen-phosphorous-kalium) plots ranked the highest yields, followed by 150% cattle manure plots and 100% cattle manure plots. DM, CP and TDN yields of in cattle manure plots were significantly (p<0.05) higher than those of no fertilizer and P-K (phosphorous-kalium) plots. The yields of in cattle slurry plots tended to be a little higher than those of in composted cattle manure plots. Assuming that corn and forage sorghum hybrids produced from this trial were fed at 70% level to 450kg of Hanwoo heifer for 400g of average daily gain, the carrying capacity (head/year/ha) of livestock ranked the highest in 150% cattle slurry plots (mean 6.0 heads), followed by 100% cattle slurry plots (mean 5.3 heads), 150% composted cattle manure plots (mean 4.7 heads), 100% composted cattle manure plots (mean 4.4 heads), and no fertilizer plots (mean 2.8 heads) in corns (or the cultivation of corns). Meanwhile, in the case of forage sorghum hybrids, 150% cattle slurry plots (mean 6.4 heads) ranked the highest carrying capacity, followed by 150% composted cattle manure plots (mean 4.8 heads), 100% cattle slurry plots (mean 4.4 heads), 100% composted cattle manure plots (mean 4.1 heads), and no fertilizer plots (mean 2.8 heads). The results indicated that the application of livestock manure to cultivated soil could enhance not only DM and TDN yields, but also the carrying capacity of organic livestock as compared with the effect of chemical fertilizers. In conclusion, the production of organic forage with reutilized livestock manure will facilitate the reduction of environmental pollution and the production of environmentally friendly agricultural products by resource circulating system.

Accuracy of Imputation of Microsatellite Markers from BovineSNP50 and BovineHD BeadChip in Hanwoo Population of Korea

  • Until now microsatellite (MS) have been a popular choice of markers for parentage verification. Recently many countries have moved or are in process of moving from MS markers to single nucleotide polymorphism (SNP) markers for parentage testing. FAO-ISAG has also come up with a panel of 200 SNPs to replace the use of MS markers in parentage verification. However, in many countries most of the animals were genotyped by MS markers till now and the sudden shift to SNP markers will render the data of those animals useless. As National Institute of Animal Science in South Korea plans to move from standard ISAG recommended MS markers to SNPs, it faces the dilemma of exclusion of old animals that were genotyped by MS markers. Thus to facilitate this shift from MS to SNPs, such that the existing animals with MS data could still be used for parentage verification, this study was performed. In the current study we performed imputation of MS markers from the SNPs in the 500-kb region of the MS marker on either side. This method will provide an easy option for the labs to combine the data from the old and the current set of animals. It will be a cost efficient replacement of genotyping with the additional markers. We used 1,480 Hanwoo animals with both the MS data and SNP data to impute in the validation animals. We also compared the imputation accuracy between BovineSNP50 and BovineHD BeadChip. In our study the genotype concordance of 40% and 43% was observed in the BovineSNP50 and BovineHD BeadChip respectively.

Discrimination study between carcass yield and meat quality by gender in Korean native cattle (Hanwoo)

  • Objective: The aim of this study was to identify a distribution pattern of meat quality grade (MQG) as a function of carcass yield index (CYI) and the gender of Hanwoo (bull, cow, and steer) to determine the optimum point between both yield and quality. We also attempted to identify how pre- and post-deboning variables affect the gender-specific beef quality of Hanwoo. Methods: A total of 31 deboning variables, consisting of 7 pre-deboning and 24 post-deboning variables from bulls (n = 139), cows (n = 69), and steers (n = 153), were obtained from the National Institute of Animal Science (NIAS) in South Korea. The database was reconstructed to be suitable for a statistical significance test between the CYI and the MQG as well as classification of meat quality. Discriminant function analysis was used for classifying MQG using the deboning parameters of Hanwoo by gender. Results: The means of CYI according to 1+, 1, 2, and 3 of MQG were 68.64±2.02, 68.85±1.94, 68.62±5.88, and 70.99±3.32, respectively. High carcass yield correlated with low-quality grade, while high-quality meat most frequently was obtained from steers. The classification ability of pre-deboning parameters was higher than that of post-deboning parameters. Moisture and the shear force were the common significant parameters in all discriminant functions having a classification accuracy of 80.6%, 71%, and 56.9% for the bull, cow, and steer, respectively. Conclusion: This study provides basic information for predicting the meat quality by gender using pre-deboning variables consistent with the actual grading index.
