Sarcocystosis in a 30 month old Hanwoo(Bos taurus oreanae)

  • Unusual yellowish-green intramuscular granulomatous lesions were found in a carcass of Hanwoo(slaughtered, 30 month old). Those were 1-3 mm in diameter, oval shaped, and paralleled with muscle fibers. Histologically, severe inflammation, eosinophilic granulomas and necrosis were observed in the muscle tissue. We also observed sarcocysts in the muscle cells. In a polymerase chain reaction, we identified 900bp length, sarcocystis specific fragment. It would be diagnosed as sarcocystosis in Hanwoo.

  • To apply and evaluate the effectiveness of genetic markers on Hanwoo traceability systems, samples of 33 Hanwoo individuals from Korean elite sire families were used, and five microsatellite markers were selected finally, which were located on chromosomes different chromosomes with the end sequencing of 100 HW-YUBAC that were recorded in the NCBI by Yeungnam University. Ten major microsatellite markers were selected from alleles amplified, their frequencies, H(Heterozygosity) and PIC(Polymorphism information content) with Hardy-Weinberg equilibrium. Next, in order to evaluate the power of the markers selected on the individual animal identification, the match probability(MP) and the relatedness coefficient(R) were computed.

  • Bovine genome samples were collected from meat of three different beef breeds (Hanwoo, Holstein and imported beef breed) that are commercially merchandized in Korean beef market. Operon B (OPB)-kits were used as random primers (3, 7, 10, 11, 12, 14) in random amplified polymorphic DNA (RAPD) method on whole genome. Each primer provided characteristic bands that were highly polymorphic. Each single primer could provide relatively efficient polymorphic band patterns among breeds. However, use of two or more primers in combination is recommended to improve resolution of experiments with higher molecular weight bands of DNA. In our experiments, OPB-11 resolved well between beef cattle breeds and Holstein. And OPB-7, 12 and 14 could be combined with OPB-11 to identify Hanwoo beef from the other two kinds of beef.

  • The study investigated consumption patterns and inclination of consumers of Hanwoo meat being more expensive than imported meat and/or beef cattle meat based on the author's business experience at the shop in Budang Seongnam. The author who has managed shop firstly investigated customers' inclination and/or propensity. The author heard specific customer's story at neighboring shopping center, and each customer's buying method, occupation, financial power and apartment size and others, and forecast visiting customer's buying and demand upon part of the beef that customer asks for. The aut hor who sold out limited scope of beef product at limited area thought that he did narrow scope of business. The author would make effort to sell product enough to meet customer's taste by better quality product from point of view of customers. The author would make effort to supply good quality beef products to the customers who relied upon the author's butcher's.

화염소독을 이용한 한우송아지 설사병 방제효과 (Prevention Effects of Hanwoo Calf Diarrhea by Disinfect with Flame)

  • For the prevention of Hanwoo calf diarrhea, the floor in a cattle shed was flamed using liquefied petroleum gas (LPG) in an attempt to substitute for the chemical disinfection method. Before flaming, litter on the floor was removed and cleaned. Outbreak rate of diarrhea in calves vaccinated with ScourGuard 3K® was decreased from 70% to 30% and the effect of the flame disinfection lasted for three to four months.

한우 번식우 농가의 번식실태 및 번식장애 치료에 관한 연구 (Studies on the Reproductive Performance and Treatment of Reproductive Disorder in Hanwoo)

  • 본 연구는 경상북도 영주지역의 한우 사육농가를 대상으로 사육규모와 형태, 사양관리 방법 등 번식우의 사양관리 형태와 번식우의 번식장애 발생률을 조사하였으며, 번식장애우에 대하여 proge-sterone제재인 control internal drug releasing for cattle(CIDR)를 처리하여 치료효과를 구명하기 위하여 수행하였다 번식장애우에 대한 CIDR 처리는 7일간 질내에 삽입하였으며, 제거 하루 전에 PGF$_2$$\alpha$를 투여하여 황체를 퇴행 발정을 유기하고 발정 확인 후 10시간 간격으로 인공수정을 실시 임신의 유무로 치료 판정한 결과는 다음과 같다. 1. 경북 영주지방의 127호의 한우 사육농가에서 사육하는 총 1,061두의 번식우의 번식상황을 조사한 결과 번식장애우는 209두로서 발생률은 19.7%이었다. 사육규모별 번식장애 발생율은 10두 이하, 10~20두 및 20두 이상 농가에서 각각 37.5, 14.7 및 13.6%로서 사육규모가 커질수록 발생률이 유의적으로(P<0.05) 낮아지는 경 향이 었다. 2. 번식우의 농가 사육형태별 번식장애 발생률은 계류사육이 30.4%로서 군사의 14.7%보다 유의 적으로(P<0.05) 발생률이 높았으며, 군사의 경우 두당 사육면적이 넓을수록 번식장애 발생률이 낮아지는 경향이었다. 3. 번식장애우의 산차별 분포를 보면 미경산우가 50.7%로 가장 많았으며 산차가 증가할수록 번식장애우에서 차지하는 비율이 감소하는 경향이었다. 4. 번식장애우의 요인별 분포는 무발정이 65.1%, 미약발정 29.2%, 난소낭종 4.3% 그리고 기타1.4%의 순으로 많이 분포되었다. 5. 209두의 번식장애우에 CIDR를 처리하고 인공 수정한 결과 157두가 임신되어 75.1%의 치료효과를 얻었다. 번식장애우의 신체충실 지수별 치료효과는 신체충실 지수가 1, 2 및 3인 경우 수태 율이 각각 85.7, 84.9 및 86.8%로서 유의 적으로(P<0.05) 높았으며, 전체적으로 다소 야윈 경우가 과비된 경우에 비하여 치료 효과가 좋았다. 본 연구결과를 종합하면, 한우 사육농가의 번식장애 발생률은 19.7%로서 사육규모가 커서 농가의 관심도가 높을수록 또한 계류보다는 넓은 면적에서 군사하는 경우가 번식장애 발생률이 낮았으며, 요인별로는 발정과 관련된 장애가 많았다. 이러한 번식장애우도 농가에서 손쉽게 이용할 수 있는 progesterone 제재인 CIDR를 처리함으로서 75.1%를 치료할 수 있으며 치료 효과를 높이기 위해서는 신체충실지수를 1, 2 및 3으로 유지하기 위한 적정 사양관리가 필요하다고 사료된다.

Fast Microchip Electrophoresis Using Field Strength Gradients for Single Nucleotide Polymorphism Identification of Cattle Breeds

  • A microchip electrophoresis (ME) method was developed using a programmed field strength gradients (PFSG) for the single nucleotide polymorphism (SNP) based fast identification of cattle breeds. Four different Korean cattle (Hanwoo) and Holstein SNP markers amplified by allele-specific polymerase chain reaction were separated in a glass microchip filled with 0.5% poly(ethyleneoxide) ($M_r$ = 8 000 000) by PFSG as follows: 750 V/cm for 0 - 14 s, 166.7 V/cm for 14 - 31 s, 83.3 V/cm for 31 - 46 s, and 750 V/cm for 46 - 100 s. The cattle breeds were clearly distinguished within 45 s. The ME-PFSG method was 7 times and 5 times faster than the constant electric field ME method and the capillary electrophoresis- PFSG method, respectively, with a high resolving power ($R_s$ = 5.05 - 9.98). The proposed methodology could be a powerful tool for the fast and simultaneous determination of SNP markers for various cattle breeds with high accuracy.

Association Analysis between Insulin-like Growth Factor Binding Protein 3 (IGFBP3) Polymorphisms and Carcass Traits in Cattle

  • The insulin-like growth factor binding protein 3 (IGFBP3) has been investigated as a candidate gene for growth promoting effects in beef cattle and a modulator of IGF bioactivity. Previously, we have reported twenty two sequence variants discovered in Korean native cattle (Hanwoo). In this study, we examined the association between gene-specific polymorphisms of IGFBP3 and cold carcass weight (CW) and marbling score (MS) among Korean native cattle. Among twenty two polymorphisms, four common polymorphic sites (-854G>C, -100G>A, +421G>T and +3863C>A) were genotyped in our beef cattle (n = 437). Statistical analysis revealed that one common polymorphism in the promoter region (-854G>C) showed putative associations with MS (p = 0.03). IGFBP3 variation/haplotype information analyzed in this study will provide valuable information into strategies for the production of a commercial line of beef cattle.

Concentration Differences in LH, FSH and Progesterone Secretion among Seasonal Changes in Hanwoo and Holstein Heifers in Daegwallyeong

  • The objective of present study was to investigate the effect of seasons on reproductive performance of Hanwoo and Holstein heifers. Heat stress in summer or cold stress in winter stress to Hanwoo and Holstein heifers may bring reproduction failure, which would pose an important economic loss, even around Daegwallyeong region located in high mountainous area. Seasonal differences in the serum levels of LH, FSH and progesterone ($P_4$) in response to environmental factors (hot and cold) out of 20 pubertal Hanwoo heifers in Daegwallyeong, Gangwon Province and 20 non-lactating Holstein heifers in Chonan city of Republic of Korea at 2-3 years of age were compared. Blood samples for hormonal analysis were from jugular vein after detection of estrus repeatedly over four seasons within four-week intervals (Spring: May to June, Summer: July to August, Autumn: October to November and Winter: January to February). In Hanwoo heifer population, averages of LH and FSH concentration in spring and in summer were greater compared to those in winter (p<0.05). LH or FSH levels tended to be greater (p=0.06) in spring and less (p=0.09) in winter compared to the levels in autumn. Only in summer, cattle seemed to show lower LH or FSH secretion (p<0.05). Similar to the results in Hanwoo heifers, the serum concentrations of LH and FSH in Holstein heifers decreased further by heat stress in summer when P 4 levels were high during luteal phase. The results demonstrate significant effect of summer heat on reproduction of Hanwoo or Holstein heifers. Although parameters indicating the extent of heat stress were not measured in this study, we suggest that serum hormone levels could be considered as successful indicators of summer heat stress condition for Hanwoo and Holstein heifers even under rather cool summer climate.

축분 첨가제를 한우분에 첨가시 pH와 유해성 병원균에 미치는 영향에 관한 연구 (Effects of Manure Additives on pH and Pathogen Populations in Hanwoo (Korean native cattle) Manure)

  • The objective of this study was to evaluate the effect of applying alum (aluminum sulfate) and aluminum chloride on pH and pathogen populations of Hanwoo manure. A total of 36 steers (8 months old and averaging 300 kg in weight) were used in this trial and allotted to 9 pens (3 replication pens per group with 4 steers per experimental unit, $5{\times}8$ m). Chemical additives were applied as a top dressing with garden rake to a depth of 1 cm of manure with wood shavings in each treatment. The chemical amendments were control (without chemical amendments), 50 g of alum and 50 g of aluminum chloride/kg of Hanwoo manure. The experiment was carried out for 4 weeks. Adding alum and aluminum chloride to Hanwoo manure reduced (P < 0.05) pH compared to untreated controls during the 4-wk period. Both levels of the alum and aluminum chloride treatments tested decreased (P < 0.05) Escherichia coli and Salmonella enterica populations in Hanwoo manure at 2 and 4 weeks. It appears that the reduction in pathogen populations was primarily associated with the lower manure pH. If more strict environmental regulations are put into effect regarding pathogen populations from Hanwoo facilities, treating Hanwoo manure with alum and aluminum chloride may be a good management practice.
