Bisphenol A의 노출에 따른 조피볼락(Sebastes schlegeli)의 혈액학적 영향 (In vivo effects of bisphenol A exposure on haematological parameters in Korean rockfish, Sebastes schlegeli)

  • 본 연구는 우리나라 주요 양식어류인 조피볼락을 대상으로 bisphenol A가 혈액에 어떠한 영향을 끼치는지를 조사하였다. 실험어를 대상으로 어체중 kg 당 0.1, 1 및 10 mg BPA를 0, 3, 7 및 12일째에 복강주사 한 후, 15일째에 그 영향을 조사하였다. BPA에 노출된 조피볼락은 적혈구수와 헤모글로빈의 농도가 유의한 감소를 나타내었고, 혈청 클로라이드, 칼슘, 혈당, 빌리루빈 및 요소질소의 농도는 증가하는 경향을 나타내었고, 콜레스테롤은 감소하였다. 또한, 혈청 전이효소와 젖산탈수소효소의 활성은 증가하였다.

  • Tiliacora racemosa Colebr. belonging to the family Menispermaceae, is the biggest storehouse of diphenyl bisbenzylisoquinoline (DBBI) alkaloids. Exhaustive chemical processing of the root of T. racemosa by the application of modern separation techniques yielded a DBBI alkaloid which was identified as tiliacorine using sophisticated spectroscopic methods (UV, IR, $^1H-NMR$, MS). Haematological study with tiliacorine proved that there was no abnormal haematological results in comparison with the normal values. Chronic toxicity study with tiliacorine revealed that the alkaloid is devoid of any hepatotoxic and nephrotoxic action.

Effects of solanum macrocarpon (African eggplant) on haematological parameters of wistar rats exposed to urban air pollution

  • Our study investigated the effects of Solanum macrocarpon (African eggplant) on haematological and biochemical parameters of male albino rats exposed to urban air pollution ($O_3$, $PM_{10}$, CO, $SO_2$, $NO_2$ and VOC). Male albino Wistar rats were exposed for 63 days either to urban air pollution without treatment (A); oral supplementation with Solanum macrocarpon given at 36 and 75 mg/kg body weight (BW), representing C and D respectively and exposed to air pollution; oral supplementation with Solanum macrocarpon given at 75 mg/kg BW after exposure to air pollution (B); or kept in animal house without exposure to air pollution (E). Animals exposed to air pollution showed significant alterations in haematological and biochemical parameters signaling that the blood and organs were badly injured. There were significant elevations in white blood cells (WBC) and its indices, reduction in red blood cells (RBC) and significant depletions of non-enzymic antioxidants, total protein and increase in lipid peroxidation values. Solanum macrocarpon supplementation in the feed of animals halted significantly the deleterious effects of air pollution, with co-administration during exposure given better results.

  • The effect of an ethanolic extract of the plant Trianthema portulacastrum L. on the $CCI_4$-induced chronic hepatocellular damage of Swiss albino mice has been investigated. The normal mice received olive oil (0.2 ml/mouse) for five weeks. The $CCI_4$ control mice, on the other hand, received $CCI_4$ (0.05 ml/mouse) in olive oil for five weeks. The extract was administered at the dose of 100 mg/kg or 150 mg/kg for five weeks by gastric intubation in addition to $CCI_4$ treatment. The $CCI_4$ administraction alone caused hepatocellular necrosis, severe anemia, leucopaenia, lymphocytopaenia, neutrophilia, eosinophilia and haemoglobinaemia along with the alterations of plasma albumin and globulin. The administration of plant extract (at 100 or 150 mg/kg) restored the $CCI_4$-induced alterations of the haematological parameters to the normal level. The extract of T. portulacastrum elicited a marked protection against $CCI_4$-induced hepatotoxicity as indicated by the several haematological parameters, related indices of formed elements, and different fractions of plasma protein. We also observed the dose-dependent antihepatotoxic effect of the extraction on these mice. The 150 mg/kg of extract was found to be more effective in normalizing the toxic effects of $CCI_4$ on the above parameters of mice. These results suggest that the hepatoprotective effect of T. poltulacastrum could be caused by its critical involvement in modulating several factors associated with erythropoiesis, and the boosting of general immunity of the host.

Effect of Working on Physiological, Biochemical and Haematological Parameters in Hariana Bullocks

  • An experiment was conducted on eight bullocks of Hariana cattle study draft efficiency using different appliances, changes in their physiological, biochemical and haematological parameters, and how the body tries to maintain these within normal physiological range. Blood collection was done at pre-exercise, three hours after exercise and two hours post exercise in summer and winter seasons of the year 1990-91. Average pulse rate and respiration rate per minute were found to be $63.09{\pm}0.78,\;25.60{\pm}0.54,\;97.21{\pm}2.51,\;63.37{\pm}2.70,\;64.05{\pm}1.90,\;25.33{\pm}1.20$, respectively, at pre-exercise, three hours exercise and two hours post exercise rest. The seasonal differences were found to significant and the values were higher in summer than in winter. After two hours post exercise rest Hariana bullocks regained their normal values for all the parameters, except that pulse rate, respiration rate and serum sodium during summer did not reach the pre - exercise levels.

Subacute Oral Toxicity Assesment of Ethanol Extract of Mariposa christia vespertilionis Leaves in Male Sprague Dawley Rats

  • The term Butterfly tea refers to decoction of Mariposa christia vespertilionis leaves which is widely consumed by cancer patients throughout Malaysia and has gained a huge popularity among Malaysians, not only cancer patients but also researchers to discover the real potential of this plant. Herein, the study is aimed at evaluating the possible toxicity in 28-day subacute oral toxicity of ethanolic extract M. christia vespertilionis in male Sprague Dawley rats. The 28-day subacute toxicity study was conducted to detect the no-observed adverse effect level (NOAEL). In this study, a total of 30 rats were divided into the control, 5% DMSO (vehicle), low dose (75 mg/kg), medium dose (125 mg/kg) and high dose (250 mg/kg) groups. The extract was administered daily from day 1 until day 28. At the end of the study, the animals were humanely sacrificed and assessed for the effect extract of Mariposa christia vespertilionis leaves on body weight and relative organ weights and haematological, biochemical and histopathological parameters. The haematological and serum biochemical parameters for the assessment of kidney and liver injuries were carried out. Results of haematological and serum biochemistry results showed no changes in the control and treated groups. In the histopathology, evaluation of kidney tissues in all treated groups showed no significant (p > 0.05) lesions. In contrast to kidney, liver tissues showed significant differences (p < 0.05) in lesions observed in low dose (430 mg), medium dose (700 mg) and high dose (1480 mg) groups with very mild, mild and mild to moderate lesion of hepatic necrosis, in the respective groups, and very mild hepatic degeneration and hepatitis were scored in all three groups.

Pentachlorophenol impact assessment of haematological parameters in olive flounder, Paralichthys olivaceus

  • The toxic effects of pentachlorophenol (PCP) on haematological parameters in olive flounder (Paralichthys olivaceus) after chronic exposure to dietary PCP (0.1, 0.5 and 1.0 mg/kg diet) for 2, 4 and 6 weeks were studied. A significant decrease in total RBC count, haemoglobin concentration and hematocrit value was noted in fish exposed to PCP compared to the non-exposed fish. The PCP treatment group showed significantly lower concentration of serum total protein and albumin, and significantly higher serum chloride, magnesium and total bilirubin levels compared with those in the control group. However, PCP had no major effects on serum glucose, total cholesterol, phosphate and calcium ions in flounder. These results demonstrated that PCP have induced adverse haematological impacts in the olive flounder, Paralichthys olivaceus. Because we found damages in blood-forming function and disruption in blood homeostasis due to chronic exposure to PCP, it is needed to develop further experimental studies for the risk assessment of this environmental pollutant.

Haematological Parameters Induced by Benzo(a)pyrene Exposure as a Toxicity Biomarker in the Fanned Red Sea Bream, Pagrus major

  • Farmed red sea breams, Pagrus major, were fed for 60 days with pellets containing different concentrations of benzo(a)pyrene (0, 0.2, 2, 20 mg/kg) to generate a biomarker of the chemical toxicity in the fish. The fish exposed to the chemical concentrations did not show any significant difference in the weight gain, conditioning, factor, and hepatosomatic index. However, some haematological parameters, such as glucose, calcium, magnesium, GOT (glutamic oxalate transaminase), and GPT (glutamic pyruvate transaminase) were influenced by the chemical exposure. Of them, two enzymes, GOT and GPT, increased significantly 60 days after the exposure in a way of concentration dependence (P<0.05). In the study of ecotoxicological biomarker, sensitivity to adverse environments is one of the key available factors. The fish changes in GOT and GPT were an earlier and reliable sign of the fish response against the chemical exposure, rendering the two enzymatic factors as a useful biomarker at least to benzo(a)pyrene exposure in the farming waters.

Blood haematology, muscle pH and serum cortisol changes in pigs with different levels of drip loss

  • Objective: An experiment was conducted to study the blood haematology, muscle pH, and serum cortisol changes in pigs with different levels of drip loss. Methods: Two groups (low and high) of 20 animals were selected from 100 pigs based on drip loss. All [$Duroc{\times}(Large\;White{\times}Landrace)$] pigs were slaughtered according to standard slaughtering procedures. At exsanguinations, blood samples were taken for the haematological parameters and serum cortisol analysis. The muscle samples were taken from longissimus dorsi muscle to evaluate the muscle pH and drip loss. Results: Haematological parameters of low drip loss group showed higher content of white blood cells and monocytes than high drip loss group (p<0.05). The low drip loss group had higher muscle pH at 45 min (p<0.05) and 24 h (p<0.001) post-mortem than the high drip loss group. However, there was no significant difference in serum cortisol levels (p>0.05). Conclusion: Drip loss is mainly affected by the muscle pH decline after slaughter and also might be affected by white blood cells and monocytes.

  • The purpose of the present study was to investigate the effects of non-watering, fasting and non-watering fasting for 16-17 hrs on haematological and biochemical parameters in SD rats. 1. Liver weight was decreased in the order of in the fasting group, non-watering fasting group and non-watering group in both sexes. 2. In fasting group and non-watering fasting group, haematologic parameters of male (HGB and MCH) and femal (MCHC) were trends to decrease compared with normal feeding & watering group. 3. In the differential leucocyte counts, there were no significant differences compared with noraml feeding % watering group.
