Effect of Oxygen and Shear Stress on Molecular Weight of Hyaluronic Acid Produced by Streptococcus zooepidemicus

  • Dissolved oxygen (DO) and shear stress have pronounced effects on hyaluronic acid (HA) production, yet various views persist about their effects on the molecular weight of HA. Accordingly, this study investigated the effects of DO and shear stress during HA fermentation. The results showed that both cell growth and HA synthesis were suppressed under anaerobic conditions, and the HA molecular mass was only $(1.22{\pm}0.02){\times}10^6 Da$. Under aerobic conditions, although the DO level produced no change in the biomass or HA yield, a high DO level favored the HA molecular mass, which reached a maximum value of $(2.19{\pm}0.05){\times}10^6 Da$ at 50% DO. Furthermore, a high shear stress delayed the rate of HA synthesis and decreased the HA molecular weight, yet had no clear effect on the HA yield. Therefore, a high DO concentration and mild shear environment would appear to be essential to enhance the HA molecular weight.

  • Quantitative measurements have been made by using equilibrium dialysis techniques on the extent of complexation between PCB and dissoved humic acid(HA). This research investigates the effectiveness of activated carbon adsorption for the removal of PCB from organic free water and humid acid background solution by using bench-scale equilibrium and rate tests. It was found that the extent of complexation depended on the pH, calcium concentration, ionic strength, and the concentration of humic acid. When HA was present, activated carbon capacity was greatly reduced due to complexation and competitive adsorption effects and the adsorption characteristics became complicated by the presence of various species such as the unassociated HA, PCB, and PCB-HA complexes.

Characterization of Ha29, a Specific Gene for Helicoverpa armigera Single-nucleocapsid Nucleopolyhedrovirus

  • Open reading frame 29 (ha29) is a gene specific for Helicoverpa armigera single-nucleocapsid nucleopolyhedrovirus (HearSNPV). Sequence analyses showed that the transcription factor Tfb2 motif, bromodomain and Half-A-TPR (HAT) repeat were present at aa 66-82, 4-76, 55-90 of the Ha29 protein respectively. The product of Ha29 was detected in HearSNPV-infected HzAM1 cells at 3 h post-infection. Western blot analysis using a polyclonal antibody produced by immunizing a rabbit with purified GST-Ha29 fusion protein indicates that Ha29 is an early gene. The size of Ha29 product in infected HzAM1 cells was about 25 kDa, which was larger than the presumed size of 20.4 kDa. Tunicamycin treatment of HearSNPV-infected HzAM1 cells suggested that the Ha29 protein is N-glycosylated. Fluorescent confocal laser scanning microscope examination, and Western blot analysis of purified budded virus (BVs), occlusion-derived virus (ODVs), cell nuclear and cytoplasmic fraction, showed that the Ha29 protein was localized in the nucleus. Our results suggested that ha29 of HearSNPV encodes a non-structurally functional protein that may be associated with virus gene transcription in Helicoverpa hosts.

Comparison of Growth, Yield and Yield Components among Rice Cultivars for Organic Farming in No-tillage Paddy

  • Organic farming system in rice paddy is rapidly expanding in Korea. This study was to find out optimum japonica rice cultivars for organic farming. A field research was conducted to evaluate the characteristics of japonica rice cultivars under no-tillage paddy at Doo-ryangmyeon, Sacheon, Gyeongsangnam-do, Korea. The experimental soil was Juggog series (silty clay loam: 56.0% silt, 31.2% clay and 12.8% sand). In experiment, ten lines of Japanese rice cultivars were tested under no-tillage amended with rye (NTR) and no-tillage without cover crop treatment (NTNT). In addition, two Korean japonica rice cultivars as check cultivars were used in this study. The grain yield in NTR was significantly higher in 6.13 Mg $ha^{-1}$ for Kinuhikari, 5.30 Mg $ha^{-1}$ for Komekogane, 5.25 Mg $ha^{-1}$ for Kosihikari, 5.22 Mg $ha^{-1}$ for Mazizbare and 5.12 Mg $ha^{-1}$ for Akitakomachi compared to two Korean rice cultivars (4.57 Mg $ha^{-1}$ for Hwayoungbyeo and 4.00 Mg $ha^{-1}$ for Ilmibyeo) in that order. While, grain yield in NTNT was significantly higher in 4.90 Mg $ha^{-1}$ for Akitakomachi 3.81 Mg $ha^{-1}$ for Hinohikari, 3.74 Mg $ha^{-1}$ for Umezkusi, 3.67 Mg $ha^{-1}$ for Kosihikari and 3.54 Mg $ha^{-1}$ for Dondokuri compared to 3.02 Mg $ha^{-1}$ for Ilmibyeo and 2.36 Mg $ha^{-1}$ for Hwayoungbyeo, respectively. The number of panicle per $m^2$ and grain number per panicle were indispensible for increasing the yield of rice. These results were able to find out optimum japonica rice cultivar Akitakomachi for organic farming in no-tillage paddy.

  • The optimum forest road density was calculated with the method which is used in Europe on the investigation site in Korea. The economical optimum forest road density at the minimum total transport cost was 10.51 m/ha. The total transport cost was calculated 235,354 won/ha per year. The forest road construction cost amounted to 99,693 won/ha per year in case of the depreciation period of 30 years and the interest rate of 3%, the forest road maintenance cost amounted to 14,502 won/ha per year, the logging cost amounted to 99,564 won/ha per year, the cost of footpaths amounted to 18,142 won/ha per year, the cost by the loss of the production area amounted to 3,454 won/ha per year.

Biomass and Net Primary Productivity of Pinus densiflora Natural Ecosystem in Kangwondo, Korea (강원도산(江原道産) 소나무천연림생태계(天然林生態系)의 Biomass 및 Net Primary Production에 관(關)한 연구(硏究))

  • Dimension analysis was used to estimate biomass and net primary production(NPP) in a 36-year-old Japanese red pine (Pines densiflora S. et Z.) natural forest in Kangwondo. Best estimation was made by the equation model of $Wt=aD^bH^c$ where Wt is weight in kg, D is DBH in cm, and H is total tree height in m. Total aboveground biomass was estimated at 198.82 t/ha. Organic matter was distributed in the Japanese red pine stand as follows ; bolewood 68.8%, live branches 16.5%, bolebark 5.4%, foliage 4.6%, dead branches 3.7%, and cones 0.6%. Net primary production was estimated at 15.87 t/㏊/yr and was distributed: bolewood 44.5%, live branches 30.9%, foliage 14.1%, current twig 7.1%, and bolebark 3.3%. Leaf efficiency was estimated at 1.876. The power equation for biomass and IVPP yielded similar results as the results for Pines densiflora in Japan.

Properties of β-carotene-loaded chitosan/hyaluronic acid nanocapsules: solubility and redispersibility (베타카로틴 함유 키토산/하이알루론산 나노캡슐의 용해도 및 재분산성 특성)

  • To improve the solubility of β-carotene, three types of β-carotene-loaded nanocapsules were prepared using chitosan (CS) and two cross-linkers, sodium tripolyphosphate (TPP) and hyaluronic acid (HA), alone or in combination (CS-TPP, CS-TPP-HA, and CS-HA). The entrapment efficiency of all nanocapsules significantly increased with an increase in TPP and HA, with the efficiency ranging from 95% to 99%. The solubility of β-carotene was significantly improved by CS nanoencapsulation before and after lyophilization and during storage. CS/HA nanoencapsulation significantly improved (by 11-fold) the water solubility of β-carotene. In particular, CS/HA nanoencapsulation was the most effective in terms of not only the solubility of β-carotene, but also the redispersibility ratio. Therefore, CS/HA encapsulation could be useful for improving the solubility of poorly soluble active ingredients, such as β-carotene.

Soil Loss and Water Runoff in a Watershed in Yeoju (소유역(小流域)에서 토양(土壤) 유실(流失) 및 물 유출양상(流出樣相))

  • Soil loss and runoff were investigated in a small watershed located in Sangeo-ri, Yeoju-eup, Yeoju-gun, Kyonggi-do. The watershed with the area of 35 ha consists of forest, grassland, uplands and mulberry. V-notch type water tank. flow-meter, automatic water sampler and rain gauge were installed at the main outlet stream. Out of $1.037.9Mg\;35ha^{-1}$ of total annual rainfall. 17.9% was lost via run-off. The total amount of soil eroded was $152.2Mg\;35ha^{-1}$, of which $78.6Mg\;35ha^{-1}$ was suspended load and $73.6Mg\;35ha^{-1}$ ha was sediment load. The soil losses under different land uses were $16.02Mg\;ha^{-1}$ for upland annual Crops. $2.69Mg\;ha^{-1}$ for mulberry field, $0.58Mg\;ha^{-1}$ for grassland and $0.55Mg\;ha^{-1}$ for forest. The predicted soil loss by Universal Soil Loss Equation was approximately 20% underestimated in forest, grassland and uplands, and 32% underestimated in mulberry field.

Effects of Herbal Acupuncture(Crataegus Pinnatifida) at $BL_{21}$ on the Obese Rats Induced by High Fat Diet (위유(胃兪)의 산사(山査) 약침(藥鍼)이 고지방(高脂肪) 식이(食餌)로 유발(誘發)된 비만백서(肥滿白鼠)에 미치는 영향(影響))

  • Objectives : This research was performed to investigate the effect of herbal acupuncture(Crataegus pinnatifida) at WI-SU($BL_{21}$) on weight gain, food intake, food efficiency, serum of lipid concentrations, liver function and cardiovascular risk of rats fed high fat diet for 8weeks. Methods : Experimental groups were divided into high fat diet group(Control), high fat diet and saline in $BL_{21}$ treated group(Saline), high fat diet and Crataegus pinnatifida-herbal acupuncture in $BL_{21}$ treated group(HA-1, 0.08mg/ml/g), high fat diet and Crataegus pinnatifida-herbal acupuncture in $BL_{21}$ treated group(HA-2, 0.04mg/ml/g), high fat diet and Crataegus pinnatifida-herbal acupuncture in $BL_{21}$ treated group(HA-3, 0.016mg/ml/g), Herbal acupuncture was bilaterally treated at Crataegus pinnatifida-herbal acupuncture in both $BL_{21}$ 1time (0.2cc) per 4days, total 14 times in 56days. Results : Body weight was decreased in HA-1 and food efficiency was decreased in HA-1, HA-3. Feed intake was increased in HA-3. The level of serum total cholesterol was decreased in Saline, HA-1, HA-2, HA-3. The level of serum HDL-cholesterol was increased in HA-1, but LDL-cholesterol was not significant. The level of serum triglyceride was decreased in HA-1, HA-2, HA-3. and the level of serum Total Bilirubin was decreased in HA-1. The level of serum Atherogenic index was decreased in HA-1, HA-2, HA-3. and the level of serum HTR was increased in HA-3. The evel of serum AST, ALT were increased in saline, and the level of serum r-GTP was not significant. Conclusions : Crataegus pinnatifida acupuncture in Wi-Su($BL_{21}$) can control the body weight, feed intake and feed efficiency ratio, total cholesterol, HDL-cholesterol, triglyceride, total bilirubin, and prevention cardiovascular risk. And Crataegus pinnatifida acupuncture is not injurious on liver function.

Effect of Drainage Culvert Spacing on Forage Crops Production in Poorly Drained Paddy Field Converted to Upland Crop Cultivation (배수불량 논에서 암거배수처리가 동.하계 사료작물생산에 미치는 영향)

  • This experiment was conducted to compare the agronomic characteristics, productivity of silage com and barley cropping, forage sorghum and barley cropping in accordance with Drainage Culvert Spacing at poorly drained paddy field in National Institute of Animal Science, at Seonghwan in Korea, March 2006 to May 2007. The emergency and flowering date were no different among treatments. Emergency rate and flowering date were 90% and July 26 in silage com, 91% and July 21 in forage sorghum, 92% and April 27 in barley, respectively. Dry matter yield was high in line with 3 m drainage culvert spacing (24,389 kg/ha) > 5 m (23,543 kg/ha) > 7 m (21,527 kg/ha) > 0 m (14,132 kg/ha). In cropping systems, dry matter yield of forage sorghum and barley (22,111 kg/ha) was higher than silage com and barley (19,684 kg/ha). Crude protein and TDN yield were high in line with 3 m (2,365 and 15,394 kg/ha) > 5 m (2,255 and 14,513 kg/ha) > 7 m (1,884 and 13,747 kg/ha) > 0 m (995 and 8,682 kg/ha). In cropping systems, crude protein and TDN (total digestible nutrients) yield of forage sorghum and barley cropping system (2,165 and 13,582 kg/ha) was higher than silage com and barley cropping system (1,576 kg/ha and 12,482 kg/ha), respectively. Consequently proper drainage culvert Spacing at poorly drained paddy field was 5 m with forage sorghum and barley cropping system.
