Growth and Energy Budgets of Greenling Hexagrammos otakii Larvae and Juveniles Reared on Different Diets and Temperatures (쥐노래미(Hexagrammos otakii)자치어의 먹이와 수온에 따른 성장 및 에너지 수지)

  • The growth, respiration, ingestion, and ecological efficiencies of Hexagrammos otakii larvae and juveniles reared on different diets and temperatures were estimated. A factorial experiment using two diets [non-enriched hatched Artemia nauplii (NEA) and enriched Artemia nauplii (EA)] crossed with two temperatures [natural seawater temperature (NT, $5.1-8.5^{\circ}C$) and heated seawater (HT, $13{\pm}0.5^{\circ}C$)] was conducted to investigate growth and energy budgets of Hexagrammos otakii larvae and juveniles. The energy used by larvae and juveniles for 6 weeks was calculated using data on ingestion, growth, respiration, excretion, and egestion. In the NT trials, the energy used for growth by larvae and juveniles was 0.121 and 0.129 Kcal in the NEA and EA treatments, respectively. Assimilation efficiency at NT were 78.0% and 80.2% in the NEA and EA trials, respectively. Gross growth efficiency ($K_1$) were 60.5% and 62.3% and net growth efficiency ($K_2$) was 77.56% and 77.71% in the NEA and EA trials, respectively. In the HT trials, the energy used for growth by juveniles was 0.189 and 0.212 Kcal in the NEA and EA trials, respectively. Assimilation efficiency at HT was 86.4% and 95.1% in the NEA and EA trials, respectively. Values of $K_1$ and $K_2$ at HT were 69.5% and 80.43%, respectively, in the NEA trials, and 73.9% and 77.66%, respectively, in the EA trials. These results suggest that enriched Artemia nauplii and higher temperatures ($13{\pm}0.5^{\circ}C$) are effective for rearing larvae and juveniles of H. otakii.

Two Cases of Human Thelaziasis and Brief Review of Korean cases

  • The present study was performed to describe 2 cases of human thelaziasis (HT) which occurred in Gyeongsangnam-do and to briefly review the previously reported Korean cases. A 58-year old woman, residing in Hadong-gun, Gyeongsangnam-do, came to Gyeongsang National University Hospital (GNUH) complaining of foreign body sensation and itching of the right eye in March 2000. Total 6 adult nematodes of Thelazia callipaeda (2 males and 4 females) were detected in her right eye. A 80-year old man, residing in Jinju-si, Gyeongsangnam-do, came to GNUH complaining of foreign body sensation, itching, and pain of the right eye in December 2007. A total of 5 worms (4 females and 1 degenerated) were removed from his right eye. We analyzed characteristics of the total 39 Korean HT cases reported to date, including the present 2 cases. Most of the cases (71.8%) occurred in Seoul and Gyeonggi-do before 2000, and 21 cases (53.8%) were males and 18 (46.2%) were females. The prevalence was higher in younger ages below 30 years (48.7%) than 31-60 years (41.0%) and over 61 years (10.3%). The seasonal prevalence showed a higher incidence in autumn (43.6%) than in other seasons. Most of the cases (94.9%) were conjunctival sac infections and only 2 (5.1%) were intraocular cases. The present 2 HT cases are the first reported cases in Gyeongsangnam-do. Some characteristics of Korean HT cases were analyzed.

lnhibitory Effect o fVarious Cruciferous Vegetable on the Growth of Human Cancer Calls (인체암세포증식에 있어 십자화과 채소의 억제효과)

  • The anticarcingenic effect of methanol extracts from such cruciferous vegetables as cabbage, red cabbage, Korean cabbage, kale, cauliflower, broccoli, radish root, leafy radish, rape leaves and shepherd’s purse on the growth of human K-562 leukemia cells, MG-63 osteosarcoma cells, HT-29 colon cancer cells and AGS gastric cancer cells were studied. All of cruciferous vegetables inhibited more than 70% of the growth of K-52 leukemia cells and more than50% fo rhe growth fo AGS gastric cancer cells. Particularly, kale, broccoli and shepherd’s purse showed inhibition rates of 93.5%, 93,5% and 96.3% on the growth of AGS gastric cancer cells, respectively. In case of HT-29 colon cancer cells, the methanol extracts of cabbage, kale and shepherd’purse exhibited 82.4%, 72.15, 79.4% and 95.6% of inhibitory effects, respectively. The cabbage, kale, cauliflower and shepherd’s purse extracts also highly suppressed the proliferation of MG-63 cells. Generally the 10 cruciferous vegetable we studied strongly decreased the growth of various human cancer cells in vitro, however, kale and shepherd’s showed the most effective vegetable among them.

Proximate Analysis, Fatty Acid Composition of Lycopus lucidus Turcz. and Its Cytotoxic Effect in Cancer Cell Lines (택란의 일반성분, 지방산 조성 및 세포독성 효과)

  • In this paper, we investigate to determine quality characteristics, fatty acid composition and cytotoxic effect of extracts and fractions from whole Lycopus lucidus Turcz. roots. Additionally, we evaluated cytotoxic activity against the growth of human fibrosarcoma cells (HT-1080) and human gastric adenocarcinoma (AGS), human colon cancer cell (HT-29) lines using a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Acetone+methylene chloride (A+M) and methanol (MeOH) extracts from L. lucidus Turcz. were obtained through solvent extraction. Then we further fractionated both extracts with n-hexane, 85% aq. MeOH, n-butanol (n-BuOH) and water. In fatty acid composition, L. lucidus Turcz. contained 33.2% of 18:1n-9 and 1.81% of 18:3n-3, respectively. The incorporation of treatment with A+M and MeOH extracts and n-hexane, 85% aq. MeOH, n-butanol (n-BuOH) and water fractions dose-dependently increased cytotoxicity against the growth of HT-1080 and AGS, HT-29 cancer cells (p<0.05). The A+M extract had a higher inhibitory effect on the growth of all cancer cells in comparison to MeOH extract. Among the fractions, the 85% aq. MeOH and n-hexane fractions showed a higher inhibitory effect after proliferating the three cancer cells. These results suggest that the 85% aq. MeOH and n-hexane fractions have a potential to inhibit the growth of human cancer cell lines.

Correlation and Chronology of the Marine Terraces and Thalassostatic Terraces in the Yeongdeok Coast, South Eastern Korean Peninsula (영덕 일대의 해성단구와 해면변동단구의 대비와 편년)

  • The Yeongdeok 53m marine terrace (Y53mT), Y43mT, Y33mT, Y24mT, Y19mT and Y11mT distributed along the Yeongdeok coast, southeastern Korean Peninsula are well compared with the thalassostatic terraces of the high terrace 1 (ℓHT1 ; 51m of the relative heights from the river floor), high terrace 2 (ℓHT2 ; 43m), middle terrace 1 (ℓMT1 ; 32m), middle terrace 2 (ℓMT2 ; 25m), lower terrace 1 (ℓLT1 ; 18m) and lower terrace 2 (ℓLT2 ; 10m) respectively, developed along the lower reaches of the Chucksan-cheon and Obo-cheon rivers, judging from the comparison of paleosols (red soils) between the above marine and thalassostatic terraces. Using the Y19mT of the MIS 5e as the key surface, we propose that the terraces of the Y53mT and ℓHT1, Y43mT and ℓHT2, T33mT and ℓMT1, Y24mT and ℓMT2, Y19mT and ℓLT1, and Y11mT and ℓLT2 have been formed at the MIS 11, 9, 7e and 7a (or 7a), 5e and 5a respectively. The red soils have been developed at the Y19mT and ℓLT1 and above them, but not on the Y11mT and ℓLT2 surfaces.

Inulae Flos Induces Cell Cycle Arrest and Autophagy of HT-1080 Human Fibrosarcoma Cells in 2D and 3D Cell Culture Model (선복화의 2D 및 3D 세포배양모델에서 HT-1080 인간 섬유육종 세포의 세포주기정지 및 자가포식 유도 효과)

  • Objective : Inulae Flos(IF) has been used to treat arthritis, sever furuncle, fear and palpitation, vomiting, stroke, asthma and cough in Korean Medicine. Although the anticancer activity of IF has been reported, the molecular mechanism is still not well understood. In this study, we investigated the growth inhibitory activity of an ethanol extract of IF in HT-1080 human fibrosarcoma cells and its underlying mechanisms using two-dimensional (2D) and three-dimensional (3D) cell culture system. Methods : HT-1080 cells were cultured with IF for 9 days in 3D cell culture. To check an inhibition of cell prolifelation by IF, MTT assay was performed. DNA contents were measured using flow cytometry. Western blotting was used to evaluate the regulation of cell cycle- and autophagy-related proteins. Acridine orange staining was performed to confirm autophagy, and DCF-DA staining was performed to confirm the occurrence of ROS. Results : IF controlled a spheroid formation and decreased a cell viability in 3D cell culture. IF-induced cell proliferation inhibition was associated with a distinct increase of S and G2/M phase cell distribution in 2D cell cultre. In addition, IF significantly induced autophagy and generated reactive oxygen species(ROS). Interestingly, IF-induced cell cycle arrest and autophagy were recovered after pre-treatment of N-acetyl-L-cysteine, ROS scavenger. Conclusion : Our results indicate that IF induced ROS-mediated cell cycle arrest and autophagy and it may potentially useful for human fibrosarcoma treatment.

Ethanol Extract of Oldenlandia diffusa - an Effective Chemotherapeutic for the Treatment of Colorectal Cancer in Humans -Anti-Cancer Effects of Oldenlandia diffusa-

  • Objectives: Oldenlandia diffusa is traditionally used to relieve the symptoms of and to treat various diseases, but its anti-cancer activity has not been well studied. In the present study, the authors investigated the anti-cancer effects of an ethanol extract of Oldenlandia diffusa (EOD) on HT-29 human adenocarcinoma cells. Methods: Cells were treated with different concentrations of an EOD, and cell death was assessed by using a 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide (MTT) assay. Analyses of the sub G1 peak, the caspase-3 and -9 activities, and the mitochondrial membrane depolarizations were conducted to confirm cell death by apoptosis. Also, intracellular reactive oxygen species (ROS) generation was determined using carboxy-H2DCFDA (5-(and-6)-carboxy-20,70-dichlorodihydrofluorescein diacetate). Results: EOD inhibited the proliferation of HT-29 cells for 24 hours by $78.6%{\pm}8.1%$ at $50{\mu}g/mL$, $74.4%{\pm}4.6%$ at $100{\mu}g/mL$, $65.9%{\pm}5.2%$ at $200{\mu}g/mL$, $51.4%{\pm}6.2%$ at $300{\mu}g/mL$, and by $41.7%{\pm}8.9%$ at $400{\mu}g/mL$, and treatment for 72 hours reduced the proliferation at the corresponding concentrations by $43.3%{\pm}8.8%$, $24.3{\pm}5.1mV$, $13.5{\pm}3.2mV$, $6.5{\pm}2.3mV$, and by $2.6{\pm}2.3mV$. EOD increased the number of cells in the sub-G1 peak in a dose-dependent manner. The mitochondrial membrane depolarization was elevated by EOD. Also, caspase activities were dose-dependently elevated in the presence of EOD, and these activities were repressed by a pan-caspase inhibitor (zVAD-fmk). The ROS generation was significantly increased by EOD and N-acetyl-L-cysteine (NAC; a ROS scavenger) remarkably abolished EOD-induced cell death. In addition, a combination of sub-optimal doses of EOD and chemotherapeutic agents noticeably suppressed the growth of HT-29 cancer cells. Conclusion: These results indicate that EOD might be an effective chemotherapeutic for the treatment of human colorectal cancer.

Nitric Oxide on the MMP-2 expression by human gingival fibroblasts (치은섬유아세포의 MMP 발현에 대한 Nitric Oxide의 영향)

  • It has been suggested that increased number and activity of phagocytes in periodontitis lesion results in a high degree of reactive oxygen species (ROS) such as superoxide anion, hydrogen peroxide, nitric oxide and peroxynitrite. There are few reports on the relationship between ROS and MMPs expressions in gingival fibroblast. We studied to elucidate whether and how ROS, especially nitric oxide affects the MMP expression. Human gingival fibroblasts and HTl080 cells (human fibrosarcoma sell line as reference) were grown in DMEM supplemented with 10 mM HEPES, 50 mg/L gentamicin, and 10% heat inactivated fetal bovine serum with addition of various reactive oxygen species (ROS). Culture media conditioned by cells were examined by gelatin zymography. HT1080 cells expressed proMMP-2 and proMMP-9, but human gingival fibroblasts (HGF) produced only proMMP-2. Hydrogen peroxide upregulated MMP-9 expression in HT1080 cells, whereas in human gingival fibroblast SNP treatment showed marked increase in MMP-2 level compared to other ROS. These results suggest that the effects of ROS on MMPs expressions are cell-type specific. RT-PCR for MMP-2 and TIMP-2 m-RNA were performed using total RNA from cultured cells under the influence various kinase inhibitors. In HT1080 cells, treatment with FPTI III (Ras processing inhibitor) and LY294002 (PI3-kinase inhibitor) resulted in inhibition of MMP-2 and MMP-9 expressions, suggesting that Ras/P13-kinase pathway is important for MMPs expression in HT1080 cells. In gingival fibroblasts, treatment with FPTI III and PDTC (NF-kB inhibitor) showed marked decrease in MMP-2 regardless of the of SNP , suggesting that Ras/NF-kB could be the key pathway for NO-induced MMP-2 expression in gingival fibroblasts. This study showed that ROS, especially nitric oxide, could be the critical mediator of periodontal disease progression through control of MMP-2 expression in gingival fibroblasts possibly via Ras/NF-kB pathway.

Effect of Cervus Elaphus-Herbal Acupuncture on Focal Ischemia Induced by Inserted Intraluminal Filament in MCA of Rats (녹용 약침이 국소 뇌허혈 모델 백서에서 방사형 미로과제 학습 및 뇌신경 보호에 미치는 영향)

  • Objectives : Cervus Elaphus(CE) has been used to medication for ischemic stroke in the Oriental Medicine. So this study was planned to investigate the effects of CE herbal acupuncture therapy(CE-HAT) on the focal ischemia-induced by intraluminal filament insertion in the rats. Materials and methods : The focal ischemia was induced by middle cerebral artery occlusion for 2hours. The groups divided into 6 groups, normal(intachess group), control(no theray group after ischemia-induced), CE-HAT1(Cervus Elaphus-herbal acupuncture therapy group at LR1, HT9 after ischemia-induced), CE-HAT2(Cervus Elaphus-herbal acupuncture therapy group at LU8, KI7 after ischemia-induced), CE-HAT3(Cervus Elaphus-herbal acupuncture therapy group at LR1, HT9, LU8, KI7 after ischemia-induced), CE-HAT4(Cervus Elaphus-herbal acupuncture therapy group at LR1, HT9, HT3, KI10 after ischemia-induced). CE-HAT was carried out during 3 weeks after focal ischemia-induced. Eight-arm radial maze was used for the behavioral task and neuropretective effect of CE-HAT was observed by Cresyl violet, AchE, ChAT-stain. Results : The error rate in the eight-arm radial maze task was significantly decreased in CE-HAT1, CE-HAT2, CE-HAT4 on 3days, CE-HAT4 on 4days, CE-HAT2, CE-HAT4 on 5days, CE-HAT3, CE-HAT4 on 6days. The rate of correct choice was significantly increased in CE-HAT4. The density of neurons in the hippocampal CA1 was significantly increased in CE-HAT1, CE-HAT2, CE-HAT3, CE-HAT4, compared to control group. The density of ChAT in the hippocampal CA1 was significantly increase in CE-HAT4. The density of ChAT in the hippocampal CA1 was significantly increased CE-HAT1. Conrlusions : These results suggest that the Cervus Elaphus-herbal acupuncture therapy could be used as a medication for controlling the stroke induced by deficiency.

Extraction and Composition of Bound Lipids in Naked Barley (쌀보리 결합지질의 추출과 그 조성에 관한 연구)

  • Bound lipids(BL) of naked barley(Hordeum vulgare L.) were extracted by different methods and the composition of BL was determined by the procedures of column chromatography, thin layer chromatography and gas chromatography. For the extraction, after free lipids were removed from barley flour by petroleum ether(PE) extraction and then BL were extracted from PE treated flour by the solvent systems of water-saturated butanol(WSB) at $25^{\circ}C$(WSB-LT) and at $95^{\circ}C$ (WSB-HT). BL were extracted by WSB-HT with higher extraction yield as 1.5% as dry basis of flour. The contents of neutral lipids(NL), glycolipids(GL) and phospholipid(PL) in BL were $20.7{\sim}35.5%$, $28.7{\sim}32.4%$, $32.1{\sim}50.6%$, respectively with particularly higher content of PL in WSB-HT as 50.6%. Digalactosyl-diglycerides $(40.2{\sim}44.8%)$, monogalactosyl-diglycerides $(20.3{\sim}31.1%)$, sterly glycerides$(11.2{\sim}15.2%)$ and cereb rosides$(11.6{\sim}12.9%)$ were observed in GL. Of the PL in BL, lysophosphatidyl choline, phosphatidyl choline and phosphatidyl serine, and phosphatidyl ethanolamine were the major components. The predominent fatty acids of NL, GL and PL were linoleic and palmitic acids, however, no significant difference was observed in the composition of fatty acids between two extraction methods.
