  • The anti proliferative effects of bile acids and their derivatives on HT -29 human colon cancer cells were investigated. Ursodeoxycholic acid (UDCA) and its synthetic derivatives, HS-1030 and HS-1183, and chenodeoxycholic acid (CDCA) and its synthetic derivatives, HS-1199 and HS-1200 were employed for this study. General evaluations focusing on cell cycle were conducted in HT -29 human colon adenocarcinoma cell line (p53 mutant type). (omitted)

  • Ethanol extracts of the whole herb of Agastachis Herba (A) and of Pueraria Radix (P) alone and of their mixture (A+P) downregulated the cell growth, cyclooxygenase-2 (COX-2) expression, prostaglandin $E_2\;(PGE_2)$, and cGMP production. A, P, and A + P inhibited the cell growth of HT-29 colon cancer cells in a concentration- and time-dependent manner but not the growth of normal colon cell, CCD-112CoN. In addition, they markedly inhibited the productions of $PGE_2$ and cGMP as well as the mRNA expression of COX-2. These data suggest that non-toxic concentration of A, P, and A + P have a significant effect on the in vitro growth of HT-29 cells, specifically through the inhibition of the $PGE_2$ production via COX-2.

감잎의 용매별 추출물의 돌연변이 유발 억제 및 암세포 증식억제 효과 (Inhibitory Effect of Persimmon Leaves on the Mutagenicity in Spore Rec Assay and on the Growth of Human Cancer Cells)

  • 감잎의 메탄을 추출물과 이를 극성이 다른 용매로 더욱 불리한 용매별 획분들의 돌연변이 유발 억제효과를 spore rec assay를 이용하여 검토하고 사람의 결장암 세포인 HT-29와 사람의 위암 세포인 AZ-521의 증식에 대한 저해효과를 실험하였다. 감잎의 메탄을 추출물은 spore rec assay에서 N-methyl- N'-nitro-N-nitrosoguanidine(MNNG) 의 돌연변이성을 61% 정도 억제하였으며, 감잎의 메탄을 추출물에서 효과가 있었던 활성물질을 정제하기 위하여 다시 극성이 다른 용매들로 각각 추출하여 얻은 획분 중에서는 헥산, 클로로포름 및 에틸아세테이트 획분이 강한 돌연변이 유발억제효과를 나타내었다. 그리고 감잎의 메탄을 추출물은 암세포의 증식을 억제시키는 항발암 효과가 관찰되었는데 사람의 결장암 세포인 UT-29와 사람의 위암 세포인 AZ-521에 감잎의 메탄을 추출물을 50$\mu\textrm{g}$/ml와 100$\mu\textrm{g}$/ml 첨가시 이들 암세포의 증식이 각각 90%와 99%가지 크게 억제되었다. 감잎의 메탄을 추출물을 다시 극성이 다른 용매들로 분리한 획분들 중에서는 사람의 결장암 세포인 HT-29와 사람의 위암세포인 AZ-521에 감잎의 클로로포름 획분을 각각 50$\mu\textrm{g}$/ml와 100$\mu\textrm{g}$/ml 첨가시 이들 암세포의 증식 억제율이 100%에 달하였고 감잎의 에틸아세테이트 획분도 사람의 결장암 세포(HT-29)와 사람의 위암세포(AZ-521)에 대해 클로로포름 획분 다음으로 강한 항발암 효과가 관찰되었으며, 헥산 획분도 이들 두 획분(클로로포름 및 에틸아세테이트)보다는 낮았으나 암세포 증식을 억제하는 효과가 관찰되어 항돌연변이 실험에서와 일치하는 결과를 얻었다.

인체암세포증식에 있어 십자화과 채소의 억제효과 (lnhibitory Effect o fVarious Cruciferous Vegetable on the Growth of Human Cancer Calls)

  • 부유세포인 K-562 임체 혈액암세포 및 부착세포인 인체 AGS 위암세포, HT-29 결장암세포 및 MG-63 골육암세포를 이용하여, 10종의 십자회과채소들로부터 추출한 메탄을 추출물의 암세포 성장저해효과를 연구하였다. 모든 십자화과 채소시료는 K-562 인체혈액암세포에 대히 70% 이상의 암세포 증식억제효과를 보였는 데, 특히 브로콜리가 92.9%의 증식억제효과를 나타내 가장 효고가 좋았다. 위암세포인 AGS세포에서는 모든 시료들이 50%이상의 암세포성장 억제효과를 가졌는데, 이 경우 케일, 브로콜리, 냉이가 각각 93.5%, 93.5%, 96.3%의 매우 높은 위암세포의 중식억제효과를 보였다. 또한 사람의 결장암 세포인 HT-29의 경우 양배추, 배추, 케일, 냉이가 각각 82.4%, 72.1%, 79.4%, 95.6$\mid$의 증식억제효과를 보였고, MG-63 골육암세포에 대해서는 케일, 냉이가 각각 79.2%, 88.7%로 가장 높은 저해효과를 보여 일반적으로 십자화과 채소들은 인체암 세포의 성장을 억제하는 것으로 나타났으나 그중 케일과 냉이가 가장 효과가 좋았다.

선복화 (Inula japonica)추출물의 암세포주에 대한 In Vitro 세포독성 (Cytotoxic Activity of the Inula japonica Extracts Against Several Human Cancer Cell Lines In Vitro)

  • The present study describes the preliminary evaluation of the cytotoxic activity of the extracts from Inula japonica. I. japonica was extracted with methanol, ethanol, acetone, and water, and then cytotoxic activity of these extracts were evaluated. The cytotoxic activity of each extract was assessed by the MTT-dye reduction assay. Both ethanol and acetone extracts from I. japonica showed the cytotoxic activity against the HT-29 human colon cancer cells. Furthermore, the ethanol extract was fractionated with n-hexane, diethyl ether, ethyl acetate, and water according to degree of Polarity, The diethyl ether fraction showed the highest cytotoxic activity against HT-29 cells, but the other fractions showed low cytotokic activity. In addition, diethyl ether layer also showed the cytotoxic activity against various tumor cells, such as human colon carcinoma SW620, human cervix adenocarcinoma HeLa, and human breast adenocarcinoma MCF-7 cells as well as HT-29 cells. These studies support that extracts of I. japonica may be a potential candidate as possible chemotherapeutic agent against human cancer.

Induction of Cytotoxicity and Apoptosis in HT-29 Human Colon Carcinoma Cells by a Gleditsiae Semen Extract

  • Gleditsiae Semen (GS) has been used in both Korea and China as herbal medicine for the treatment of cephalalgia, catharsis, and other diseases. However, the apoptosis of GS against human cancer cells has not previously been investigated. The primary objective of this study was to determine the mechanisms inherent in GS-induced cytotoxicity and apoptosis, using methanolic extract of GS (GSE) in HT-29 human colon carcinoma cells. We found that GSE induced cytotoxicity in HT-29 cells in a dose-dependent manner, and this effect was verified via a lactate dehydrogenase release assay and a colony formation assay. In particular, HT-29 cells showed extensive cell death when treated with $50\;{\mu}g/mL$ of GSE; the calculated $IC_{50}$ value was $20\;{\mu}g/mL$. It induced characteristic apoptotic signs in HT-29 cells, including chromatin condensation and DNA fragmentation, occurring within 6-24 hr when the cells were treated at a concentration of $50\;{\mu}g/mL$. Interestingly, we detected the activation of caspase-3 and -9, but not caspase-8, and apoptotic bodies in GSE-treated HT-29 cells. Collectively, our results indicate that GSE induces apoptosis via a mitochondria-mediated apoptotic pathway, and these findings may be significant with regard to the development of a new drug for the treatment of human colon carcinoma cells.

Chemopreventive Effect of Protein Extract of Asterina pectinifera in HT-29 Human Colon Adenocarcinoma Cells

  • We investigated the effect of protein extract of Asterina pectinifera on the activity of 4 enzymes that may playa role in adenocarcinoma of the colon: quinone reductase (QR), glutathione Stransferase (GST), ornithine decarboxylase (ODC), and cyclooxygenase (COX)-2. QR and GST activity increased in HT-29 human colon adenocarcinoma cells increased that had been exposed to 4 concentrations of the protein extract (80, 160, 200, and $240{\mu}g/mL$). Additionally, 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced ODC activity decreased significantly in cells exposed to the extract in concentrations of $160{\mu}g/mL$ (p<0.05), $200{\mu}g/mL$ (p<0.005), and $240{\mu}g/mL$ (p<0.005). TPA-induced COX-2 activity also decreased in cells exposed to extract concentrations of 10, 20, 40, and $60{\mu}g/mL$. COX-2 expression was also inhibited in cells exposed to this extract. These results suggest that this protein extract of A pectinifera has chemopreventive activity in HT-29 human colon adenocarcinoma cells, and therefore, may have the potential to function as a chemopreventive agent in human colorectal cancer.

Antiproliferative properties of luteolin against chemically induced colon cancer in mice fed on a high-fat diet and colorectal cancer cells grown in adipocyte-derived medium

  • Purpose: Obesity and a high-fat diet (HFD) are risk factors for colorectal cancer. We have previously shown that luteolin (LUT) supplementation in HFD-fed mice markedly inhibits tumor development in chemically induced colon carcinogenesis. In this study, we evaluated the anticancer effect of LUT in the inhibition of cell proliferation in HFD-fed obese mice and HT-29 human colorectal adenocarcinoma cells grown in an adipocyte-derived medium. Methods: C57BL/6 mice were fed a normal diet (ND, 11.69% fat out of total calories consumed, n = 10), HFD (40% fat out of total calories consumed, n = 10), HFD with 0.0025% LUT (n = 10), and HFD with 0.005% LUT (n = 10) and were subjected to azoxymethane-dextran sulfate sodium chemical colon carcinogenesis. All mice were fed the experimental diet for 11 weeks. 3T3-L1 preadipocytes and HT-29 cells were treated with various doses of LUT in an adipocyte-conditioned medium (Ad-CM). Results: The weekly body weight changes in the LUT groups were similar to those in the HFD group; however, the survival rates of the LUT group were higher than those of the HFD group. Impaired crypt integrity of the colonic mucosa in the HFD group was observed to be restored in the LUT group. The colonic expression of proliferating cell nuclear antigen and insulin-like growth factor 1 (IGF-1) receptors were suppressed by the LUT supplementation in the HFD-fed mice. The LUT treatment (10, 20, and 40 µM) inhibited the proliferation and migration of HT-29 cells cultured in Ad-CM in a dose-dependent manner, as well as the differentiation of 3T3-L1 preadipocytes. Conclusion: These results suggest that the anticancer effect of LUT is probably due to the inhibition of IGF-1 signaling and adipogenesis-related cell proliferation in colon cancer cells.

대장암세포주 HT29에서의 Treculia africana 추출물의 항산화 및 항암 활성 분석 (Anti-oxidative and Anti-cancer Activities of Treculia africana Extract in Human Colon Adenocarcinoma HT29 Cells)

  • Treculia africana Decne는 빵나무종으로 뽕나무과, Treculia 속에 속하는 식물로서, 이 식물의 다양한 부위에서 추출한 물질은 항염증, 항균등의 효과를 가지고 있어 백일해의 치료등 다양한 질환에서 민간요법으로 사용되어 왔다. 하지만 정확한 생리활성에 관한 연구는 보고된 바가 없다. 따라서 본 연구에서는 T. africana Decne 메탄올추출물(META)을 사용하여 항산화능 및 인체 대장암 세포주인 HT29에 대한 항암활성에 관하여 분석하였다. DPPH radical scavenging activity를 통해 분석한 결과, META의 IC50가 4.53 μg/ml로 강력한 항산화능을 보유하 였음을 확인하였다. 또한 META 처리에 의해 HT29의 생존율이 감소함과 더불어 IC50가 66.41 μg/ml로 강력한 세포사멸효과를 나타냈다. META 처리에 의해 HT29의 subG1 세포비율 및 Annexin V+ 세포의 비율이 증가하고, DAPI로 염색된 apoptotic body가 증가하였다. 또한 apoptosis 관련 단백질인 Fas, Bax, cytochrome c의 발현이 증가하였으며, 이는 caspase 3, 8, 9를 활성화 시켜 최종적으로 PARP가 분해되어 apoptosis가 유도되었음을 확인하였다. 이러한 결과들을 통해 META는 강한 항산화 활성과, 대장암세포에서 apoptosis 유도에 의한 높은 항암활성을 보유한 물질임을 확인하였다.

Anticancer Effect of COX-2 Inhibitor DuP-697 Alone and in Combination with Tyrosine Kinase Inhibitor (E7080) on Colon Cancer Cell Lines

  • Colorectal cancer remains one of the most common types of cancer and a leading cause of cancer death worldwide. In this study, we aimed to investigate effects of DuP-697, an irreversible selective inhibitor of COX-2 on colorectal cancer cells alone and in combination with a promising new multi-targeted kinase inhibitor E7080. The HT29 colorectal cancer cell line was used. Real time cell analysis (xCELLigence system) was conducted to determine effects on colorectal cell proliferation, angiogenesis was assessed with a chorioallantoic membrane model and apoptosis was determined with annexin V staining. We found that DuP-697 alone exerted antiproliferative, antiangiogenic and apoptotic effects on HT29 colorectal cancer cells. For the antiproliferative effect the half maximum inhibition concentration ($IC_{50}$) was $4.28{\times}10^{-8}mol/L$. Antiangiogenic scores were 1.2, 0.8 and 0.5 for 100, 10 and 1 nmol/L DuP-697 concentrations, respectively. We detected apoptosis in 52% of HT29 colorectal cancer cells after administration of 100 nmol/L DuP-697. Also in combination with the thyrosine kinase inhibitor E7080 strong antiproliferative, antiangiogenic and apoptotic effects on HT29 colorectal cancer cells were observed. This study indicates that DuP-697 may be a promising agent in the treatment of colorectal cancer. Additionally the increased effects observed in the combination with thyrosine kinase inhibitor give the possibility to use lower doses of DuP-697 and E7080 which can avoid and/or minimize side effects.
