느릅나무 근피 추출물에 의한 인체 암세포 증식 및 DNA 합성 억제효과 (Effect of Extracts from Root Bark of Ulmus parvifolia on Inhibition of Growth and DNA Synthesis of Human Cancer Cells.)

  • 인체 암세포계(MG-63 골육암 세포, HT-29 인체 결장암세포, K-562 백혈암세포)를 이용하여 느릅나무 근피 메탄올 추출물, 열탕 추출물 및 즙액에 의한 암세포 성장에 미치는 효과를 검토하였다. 느릅나무 근피 메탄올 추출물, 열탕 추출물 및 즙액은 낮은 농도에서부터 인체 골육암 세포의 증식을 억제시켰다. 인체 결장암세포와 백혈암세포에서도 느릅나무 근피 메탄올 추출물, 열탕 추출물 및 즙액은 낮은 농도인 1 mg/ml에서부터 활성을 나타내어 40% 이상으로 암세포 증식 억제효과를 나타내어 앞서의 MG-63 골육암 세포에서 보다 그 증식 억제효과가 높은 것으로 나타났다. 느릅나무 근피 메탄올 추출물, 열탕 추출물 및 즙액을 골육암과 결장암세포에 투여한 2일 후에 세포내의 DNA 합성에 미치는 영향을 측정한 결과, 농도 의존적으로 암세포의 DNA 합성을 저해하는 것을 살펴 볼 수가 있었다. 따라서 느릅나무 근피 추출물은 인체 암세포 증식을 크게 억제하였으며 열탕 추출물에서도 항암활성 효과를 보여 느릅나무 근피 유래 생리활성 물질은 열에 안정한 것으로 여겨진다.

  • 유산균(Lactic acid bacteria)은 Escherichia coli와 Salmonella typhimurium과 같은 병원균에 대한 항균활성을 나타낼 뿐만 아니라 면역 증강효과를 나타내는 등 인체내에서 건강에 이로운 다양한 역할을 수행하는 것으로 알려졌다. 특히, Pediococcus pentosaseus와 몇몇 Lactobacillus 종으로부터 분리한 항균활성을 나타내는 peptide들인 safelac과 lactopad는 몇몇 암세포주의 성장을 억제하는 것으로 나타났다. 이에, 본 연구에서는 HT-29, SW 480 및 Caco-2와 같은 3종류의 인간의 결장암 세포주에 safelac과 lactopad를 투여하여 이들이 항암효과를 나타낼 수 있는 지를 분석하고자 하였다. XTT assay는 safelaf과 lactopad가 HT-29, SW 480 및 Caco-2의 성장을 억제하는 것으로 나타났으며, 특히, 이들 peptide들을 72시간동안 처리했을 때 나타나는 항암효과는 $3.1{\sim}100mg/mL$의 농도범위에서 유의한 결과를 나타내었으며, 분석한 농도 범위에서 용량 의존적인 방식으로 더 강한 효과를 나타내었다. RAW 264.7 세포주는 cytokine인 tumor-necrosis factor(TNF-${\alpha}$)의 생성에 미치는 이들 peptide들의 효과를 조사하기 위한 대식세포의 모델로써 이용되었다. RAW 264.7 세포주에서 TNF-${\alpha}$의 생성은 이들 peptide들에 의해 48시간 배양시 용량에 의존적인 방식으로 영향을 받는 것으로 나타났다. 따라서, 이러한 발견은 safelac과 lactopad와 같은 유산균으로부터 분리한 항균 peptide들이 결장암 세포에 대한 화학적 예방제로서의 잠재성을 갖고 있음을 시사하는 결과로서 주목된다.

Cobalt Chloride Induces Necroptosis in Human Colon Cancer HT-29 Cells

  • Necroptosis, also known as "programmed necrosis", has emerged as a critical factor in a variety of pathological and physiological processes and is considered a cell type-specific tightly regulated process with mechanisms that may vary rather greatly due to the change of cell line. Here we used HT-29, a human colon cancer cell line, to establish a necroptosis model and elucidate associated mechanisms. We discovered that cobalt chloride, a reagent that could induce hypoxia-inducible $factor-1{\alpha}(HIF1{\alpha})$ expression and therefore mimic the hypoxic microenvironment of tumor tissue in some aspects induces necroptosis in HT-29 cells when caspase activity is compromised. On the other hand, apoptosis appears to be the predominant death form when caspases are functioning normally. HT-29 cells demonstrated significantly increased RIPK1, RIPK3 and MLKL expression in response to cobalt chloride plus z-VAD treatment, which was accompanied by drastically increased $IL1{\alpha}$ and IL6 expression, substantiating the notion that necrosis can induce profound immune reactions. The RIPK1 kinase inhibitor necrostatin-1 and the ROS scavenger NAC each could prevent necrosis in HT-29 cells and the efficiency was enhanced by combined treatment. Thus by building up a necroptosis model in human colon cancer cells, we uncovered that mechanically RIP kinases collaborate with ROS during necrosis promoted by cobalt chloride plus z-VAD, which leads to inflammation. Necroptosis may present a new target for therapeutic intervention in cancer cells that are resistant to apoptotic cell death.

인삼과 계피 혼합물에 의한 in vitro에서 암세포 증식억제의 상승 효과 (Synergistic Effect of Panax ginseng and Cinnamoum Blume Mixture on the Inhibition of Cancer Cell Growth in vitro)

  • 인삼과 계피 추출물의 인체 직장암 세포(HT-29), 인체 간암 세포(HepG2)및 인체 결장암 세포(HRT-18)의 증식에 미치는 영향을 in vitro에서 확인하였다. 암세포의 배양액에서 인삼 추출물 또는 계피 추출물의 효과는 농도에 비례하여 암세포의 증식을 억제하였다. 세포증식의 억제 정도는 인삼과 계피 추출물을 단독으로 첨가한 것보다 병용하여 첨가한 경우 현저한 상승 효과를 나타내어, 낮은 인삼 농도에서도 HT-29, HepG2및 HRT-18암세포의 증식을 효과적으로 억제하였으며 심지어는 사멸시켰다. 인삼과 계피의 혼합물은 세포주기 중 G1 단계에서 S단계로의 진행을 지체시킴으로써 암세포의 증식을 억제하는 것으로 나타났다.

Potential Chemoprevention Activity of Pterostilbene by Enhancing the Detoxifying Enzymes in the HT-29 Cell Line

  • Detoxifying enzymes are present in most epithelial cells of the human gastrointestinal tract where they protect against xenobiotics which may cause cancer. Induction of examples such as glutathione S-transferase (GST) and its thiol conjugate, glutathione (GSH) as well as NAD(P)H: quinoneoxidoreductase (NQO1) facilitate the excretion of carcinogens and thus preventing colon carcinogenesis. Pterostilbene, an analogue of resveratrol, has demonstrated numerous pharmacological activities linked with chemoprevention. This study was conducted to investigate the potential of pterostilbene as a chemopreventive agent using the HT-29 colon cancer cell line to study the modulation of GST and NQO1 activities as well as the GSH level. Initially, our group, established the optimum dose of 24 hours pterostilbene treatment using MTT assays. Then, effects of pterostilbene ($0-50{\mu}M$) on GST and NQO1 activity and GSH levels were determined using GST, NQO1 and Ellman assays, respectively. MTT assay of pterostilbene ($0-100{\mu}M$) showed no cytotoxicity toward the HT-29 cell line. Treatment increased GST activity in the cell line significantly (p<0.05) at 12.5 and $25.0{\mu}M$. In addition, treatment at $50{\mu}M$ increased the GSH level significantly (p<0.05). Pterostilbene also enhanced NQO1 activity significantly (p<0.05) at $12.5{\mu}M$ and $50{\mu}M$. Hence, pterostilbene is a potential chemopreventive agent capable of modulation of detoxifiying enzyme levels in HT-29 cells.

대장 상피세포에서 p-Hydroxycinnamic Acid의 항염증 효과와 염증성 장질환에 대한 치료 효과 (Anti-inflammatory Effect of p-Hydroxycinnamic Acid on HT-29 Intestinal Cells and Its Therapeutic Effect of Immune Bowel Disease)

  • Inflammatory bowel disease (IBD) is a chronic inflammatory disorder on the large intestine that has been considered as an incurable not only in Western society but also in Eastern Asia in recent years. Despite enormous efforts to develop novel therapeutics for this disease, strategy using bioactive compounds from natural product is still considered as important. p-hydroxycinnamic acid (HCA) is an intermediate substance found in several plants and has been known to possess anti-inflammation but little evidence is reported whether HCA has an inhibitory effect on intestinal inflammation. In the present study, we observed HCA does not show cytotoxic and apoptotic in HT-29 cells. Quantitative PCR analysis revealed that HCA effectively blocks the activity of HT-29 cells stimulated with TNF-α treatment. HCA inhibits translocation of p65 and MAPK pathways in activated HT-29 cells by TNF-α treatment. Besides, oral administration of HCA attenuates manifestation of DSS-induced inflammatory disease in vivo. Histological analysis exhibited that oral administration of HCA recovers IBD symptoms. The expression of pro-inflammatory cytokines were reduced by oral administration of HCA on intestinal tissues. Therefore, these results suggest that HCA has a potent anti-inflammatory effect on intestinal cells as well as show a therapeutic potential for treating IBD in vivo.

Epithelial-mesenchymal Transition is Associated with Acquired Resistance to 5-Fluorocuracil in HT-29 Colon Cancer Cells

  • 5-Fluorouracil (5-FU) is commonly used for the therapy of colon cancer; however, acquired resistance to 5-FU is a critical barrier to successful treatment and the primary cause of chemotherapy failure. Epithelial-mesenchymal transition (EMT) is a process whereby cells undergo alterations in morphology and molecular characteristics promoting tumor progression and metastasis. Accumulating evidence shows that transition from epithelial to mesenchymal phenotype in cancer cells is associated with their resistance to chemotherapy. However, it is still poorly understood whether EMT is involved in acquired resistance to 5-FU. In this study, we developed an in vitro cell model, 5-FU-resistant HT-29 colon cancer cells, and characterized the differences in cellular morphology and molecular alterations between parental and resistant cells. In accord with mesenchymal-like morphology of 5-FU-resistant HT-29 cells, the expression of the mesenchymal marker fibronectin was significantly increased in these cells in comparision with that in the parental cells. Of interest, we also found a marked increase in the expression of EMT-inducing transcription factors Twist, Zeb1, and Zeb2. Finally, 5-FU-resistant cells showed enhanced migration in comparison with parental HT-29. Taken together, these results indicate that EMT could be associated with 5-FU resistance acquired by HT-29 cells. A specific role of each transcription factor found in this study will require further investigation.

5-Fluorouracil과 Capsaicin의 병용에 의한 HT-29 대장암세포 사멸 증진 효과 (Combined Treatment with 5-Fluorouracil and Capsaicin Induces Apoptosis in HT-29 Human Colon Cancer Cells)

  • Fluorouracil (5-FU) is one of the most widely used chemotherapeutic drugs in the treatment of advanced colorectal cancer patients. Capsaicin (N-vanillyl-8-methyl-alpha-nonenamide), a spicy component of hot pepper, is a homovanillic acid derivative that preferentially induces cancer cells to undergo apoptosis. The purpose of the present study is to examine whether capsaicin enhances the anticancer effect of 5-fluorouracil in HT-29 human colon cancer cells by inducing apoptosis, and whether PPARgamma is involved in the capsaicin action in combination treatment with 5-FU. Treatment of the cells with either 5-FU or capsaicin alone for 48 h had little effect on the cell viability up to $50{\mu}M$ concentration, whereas co-treatment of the cells with capsaicin in the presence of 5-FU for 48 h significantly decreased the cell viability in a concentration-dependent manner. In addition, caspase-3 activity, a marker enzyme for apoptosis, was significantly increased by the combined treatment with 5-FU and capsaicin compared to the 5-FU or capsaicin alone treatment. Also, treatment with troglitazone, a peroxisome proliferator-activated receptor gamma ($PPAR{\gamma}$) agonist, further enhanced the effect of the combination treatment on the cell viability and caspase-3 activity, and bisphenol A diglycidyl ether (BADGE), a $PPAR{\gamma}$ antagonist, blocked the effect of the combination treatment. These results suggest that the combination treatment of HT-29 cells with 5-FU and capsaicin induces apoptotic cell death at relatively low concentration than each drug alone, and the combination treatment may be associated with the $PPAR{\gamma}$ pathway activation.

  • Berberine, an isoquinoline alkaloid, has a wide range of pharmacological effects, including anti-inflammation. It has been reported that berberine inhibits experimental colitis through inhibition of IL-8, and that inhibitory effect of berberine on inflammatory cytokine expression is mediated through peroxisome proliferator activated receptor (PPAR)-$\gamma$. In this study, we examined the effects and action mechanism of berberine on the tumor necrosis factor (TNF)-$\alpha$-induced monocyte adhesion to HT29 human colonic epithelial cells, which is commonly used as an in vitro model of inflammatory bowel disease (IBD). Berberine significantly inhibited the TNF-$\alpha$-induced monocyte adhesion to HT29, which is similar to the effect of PDTC, a nuclear factor (NF)-$\kappa$B inhibitor. However, ciglitazone and GW, the ligands of PPAR-$\gamma$, did not suppress the TNF-$\alpha$-induced monocyte adhesion to HT29 cells. In addition, TNF-$\alpha$-induced chemokine expression and NF-$\kappa$B transcriptional activity were significantly inhibited by berberine in a concentration-dependent manner. The results suggest that inhibitory effect of berberine on colitis is mediated through suppression of NF-$\kappa$B and NF-$\kappa$B-dependent chemokine expression.

Probiotics Inhibit Lipopolysaccharide-Induced Interleukin-8 Secretion from Intestinal Epithelial Cells

  • It has been suggested that probiotics could be useful for the prevention of symptomatic relapse in patients with inflammatory bowel disease (IBD). Interleukin (IL)-8 has been well recognized as one of the pro-inflammatory cytokines that could trigger inflammation and epithelial barrier dysfunction. In this study, the anti-inflammatory effects of probiotics were investigated using a human epithelial cell line (HT-29). Probiotics from infant feces and kimchi were tested for their cytotoxicity and effects on adhesion to epithelial cells. The present results show that seven strains could form 70 % adhesion on HT-29. The probiotics used in this study did not affect HT-29 cell viability. To screen anti-inflammatory lactic acid bacteria, HT-29 cells were pretreated with live and heat-killed probiotics, and lipopolysaccharide (LPS) ($1{\mu}g/mL$) was then added to stimulate the cells. The cell culture supernatant was then used to measure IL-8 secretion by ELISA, and the cell pellet was used to determine IL-8 and toll-like receptor (TLR-4) mRNA expression levels by RT-PCR. Some probiotics (KJP421, KDK411, SRK414, E4191, KY21, and KY210) exhibited anti-inflammatory effects through the repression of IL-8 secretion from HT-29 cells. In particular, Lactobacillus salivarius E4191, originating from Egyptian infant feces, not only decreased IL-8 mRNA expression, but also decreased TLR-4 expression. These results indicate that Lactobacillus salivarius E4191 may have a protective effect in intestinal epithelial cells.
