Enhanced Yield of Extraction from Gastrodia elata Blume by Ultrasonication and Enzyme Reaction

  • Gastrodia elata Blume (Chunma) belongs to Orchidaceae, which is a perennial parasitic herbaceous plant and grows in the woods of the central provinces of China, Korea and Japan. Recently, the constituents of the tubers of this plant have been investigated by researchers who have revealed the presence of phenolic compounds including gastrodin as a major constituent, together with 4-hydroxybenzaldehyde, 4-hydroxybenzyl alcohol, parishin, 4,4-dihydroxybenzyl sulfoxide, vanillin, vanillyl alcohol, beta-sitosterol, organic acids and polysaccharides, etc. In this study, we used ultrasonicator and two kinds of enzymes for enhancement of extraction yield. We also used electronic nose for the aroma pattern analysis of Chunma extracts. The concentrations of glucose and functional constituents (gastrodin, vanillin, 4-hydroxybenzaldehyde and 4-hydroxybenzyl alcohol) were measured by biochemistry analyzer and HPLC, respectively. Therefore, we showed that the yield of extraction was increased and discomfortable odor was reduced.

Characteristics of Protease Inhibitor Purified from the Eggs of Alaska pollock (Theragra chalcogramma) (명란 단백분해효소 저해제의 특성)

  • Protease inhibitors were purified from the eggs of Alaska pollock (Theragra chalcogramma) using the following purification steps: ammonium sulfate precipitation, ion exchange, gel permeation, and high performance liquid chromatographies (HPLC). The protease inhibitor from the heated eggs of Alaska pollock was not as well purified. In addition, the heated eggs showed lower specific inhibitory activity than the unheated eggs. The purification yields after ammonium sulfate precipitation, ion exchange, and gel permeation chromatographies were 22.7$\%,\;15.3\%$,and $4.4\%$, respectively. There were two kinds of protease inhibitors on the gel permeation chromatography pattern Their molecular weights were estimated to be 66,700 and 16,000 Da, respectively. Both were classified as a cysteine protease inhibitor because of the existence of inhibiting papain, which is one of cysteine proteases.

Determination of Polychlorinated Biphenyls in the Soil by Perchlorination (염소화법에 의한 토양 중 Polychlorinated Biphenyls의 잔유분석)

  • PCBs have been measured using GC-ECD, GC-MS, GC-ELCD, HPLC, TLC, NMR and Immunoassay. The analysis of PCBs using GC-ECD include the peak pattern method as none derivatization and the Perchlorination method as derivatization. This study was conducted to establish the perchlorination method with Sbcls from PCBs to decachlorinated biphenyl(DCB). The aroclor 1242 of PCBs was chlorinated and then, converted into the DCB which showed a single peak in GC-ECD chromatogram. The detection limit of DCB was 2pg. The quantification detection concentration of PCBs extracted with soxhlet was 0.5ng/g in the soil. PCBs were not detected in the suburban soil, but 174ng/g in the soil of industrial complex. Mean PCBs concentration of Shinchun stream at Kumho river and Jinchun stream at Nakdong river was calculated average 낙ngjg in 각e sediment. PCBs concentration in the sediment of Kumho river near 2-7km from conjunction with Nakdong river was average 154ng/g. PCBs concentration in the sediment of Nakdong river near conjunction with Kumho river was average 159ng/g.

Quantitative Comparison of Ginsenosides and Nitrogen Compounds in Korean Ginsengs and Related Origin (고려인삼류와 유사생약중 ginsenoside 및 질소화합물의 함량 비교)

  • To differentiate the quality of Korean ginseng from those of other habitats, the quantitative analysis of free amino acids(FAA) and total amino acids(TAA) in addition to ginsenoside Rb1 and Rg1 was carried out using amino acid analyzer and HPLC, respectively. FAA pattern in Korean ginseng was much different from that of Panax notoginseng. The difference in total content of FAA was also found that Korean ginseng contained 26.3-39.8mg/g while Panax notoginseng contained 6.5mg/g. This FAA content had a tendency to increase with the age of radix. The contents of FAA and TAA in Korean ginseng(6 years old) from Kumsan were shown to be the highest than other ginseng origins tested. The content in the 6 years Panax ginseng from China was about same with that of 4 years Korean ginseng of Kumsan. However, regarding to gisenoside Rbl and Rg1, which have been accepted as the characteristic components of Panax ginseng-Panax notoginseng showed considerably higher content than those of any other ginseng origin.

  • Using the alkalophillic Bacillus sp. SH-8 and its mutant Bacillus sp. SH-8M capable of growing at the neutral pH, the amino acid compositions of the cell wall and cell membrane were studied at varying cultivation pH's. The pattem of protein electrophoresis was also tested. It was elucidated that the amino acids consisting of the cell wall were alanine, glutamic acid, lysine, aspartic acid, and meso-diaminopimelic acid. There was not any significant difference in the amino acid compositqon betweeo`two straqns regardless of the culture pH. As the results of HPLC ssay, glutamic acid and aspartic aciu accounted for more than 50% in the amqno acid composytqon of the cell wall. By the isolatqon of the crude cell membrane and the SDS-PAGE analysis, it was found that there was a considerable difference qn the protein pattern when the straqns were cultured at the neutral pH. In addition, by the two dimensional gel electrophoresis, it was confirmed that there was a difference in the protein patterns between two strains cultivated at the neutral pH medium but no difference at the alkaline medium.

  • Rhus Verniciflua Stokes has been used for long history to treat blood stasis and cancer in Oriental Medicine. But urushiol, a constituent of Rhus Verniciflua Stokes has T-cell mediated allergic reaction, so it has made clinical limitation. Nexia is allergen removed extract of Rhus Verniciflua Stokes and its extraction process as well as quality control is needed to be standardized. The optimized extraction condition is as follow; temperature is $90{\sim}95^{\circ}C$, volume of slovent 10 times, extraction time >6 hours, pressure $, slovnet water or EtOH(60%), yield 8.1${\sim}$9.8(w/w). HPLC pattern and isolation study showed Nexia contain fisetin(>13.0%), fustin(>7.0%) and no urushiol.

Study of quercetin and kaemferol contents variation in leaves of Cudrania tricuspidata associated with cultivating locations, collecting seasons and drying methods

  • In order to study the content variation of Quercetin and Kaempferol associated with Varieties (with thorn, without thorn), Cultivating Locations, Collecting Seasons, Leaves of Cudrania tricuspidata (carrier) Bureau containing higher content of pharmaceutical components than the other parts were tested after collecting 6 varieties including w or w/o thorn from 4 locations such as Cheongsong, Milyang, Goesan and Gochang at 3 seasons, spring, summer and fall in 2015 and 2016. As a result, Kaempferol content associated with Varieties between with thorn and without thorn, collecting Locations and Years have no difference statistically in their mean values but the differences associated with Seasons were statistically significant. The content was higher at spring and then gradually declined at summer and fall. The content variation in Year 2015 which was higher at spring and lower at summer and then recovered at fall abnormally, shows similar pattern of sunshine variation of the year. The content variation of Quercetin and Kaempferol seems to be come from climate fluctuation specially in Sunshine hours rather than Varieties, Locations. The variation associated with drying methods for utilizing leaves are statically significant. Natural Dry was shown the least variation because heat input while drying makes degradation of the content.

Effect of Cellobiose Octaacetate, Avicel, and KC-flock on Production of Avicelases from Penicilliurn verruculosum (Penicillium verruculosum의 Acicelase 생성에 대한 Cellobiose Octaactate와 Avicel 및 KC-flock 의 영향)

  • During the cultivation of Penkillium uerrmulosum in the media containing cellobiose octaacetate (COA), avicel, or KC-flock as an inducer and as a sole carbon source for 21 days, cellulolytic activity and SDS-PAGE pattern of proteins in the culture broth were investigated. Protein concentration and cellulolytic activity were highest in the COA medium. As cultivation period was increased, protein content and avicel hydrolytic activity of culture broth were increased as similar extent but neither $\beta$-glucosidase nor CMC hydrolytic activity was correlated to protein content. When crude proteins from the culture broth were separated on DEAE column by HPLC, distribution of avicel-hydrolytic activities were well correlated with that of major proteins. From those results it was suggested that three major proteins having 60 K, 68 K, and 76 K of Mr. were avicel-hydrolytic enzymes.

Comparison of Higenamine Extraction from Unprocessed and Processed Aconite Roots

  • Higenamine is a cardiotonic constituent of Aconite root, one of the most important oriental traditional medicine. Since Aconite root contains toxic aconitine alkaloids, variously processed roots have been often used. Much works have been done with the chemical significances concerning with the toxic aconitine alkaloids during the processing periods. However, effects of processing on higenamine have not yet been previously studied. In this paper, the extract pattern and the amounts of higenamine extracted with water from unprocessed and processed Aconite roots were compared. R-(+)-isomer was the only higenamine enantiomer detected although racemic higenamine was reported to be separated from Aconitum spp. Sonication for 1 hour resulted in higher higenamine extraction $(12.3\;{\mu}g/g)$ than boiling water extraction for 3 hours $(6.7\;{\mu}g/g)$ of unprocessed Aconite root. Extraction of not only higenamine but also most of the other components of unprocessed Aconite roots were reduced with boiling in water. Similarly, reduced extraction was observed with extracts of all three processed Aconite roots (Kyung-Po-Aconite root, Dang-Po-Aconite root and Huk-Peon-Aconite root) by either sonicated extraction or boiling water extraction.
