구강편평세포암종에서 미슬토 추출물의 Apoptosis 유도 효과에 대한 실험적 연구 (AN EXPERIMENTAL STUDY ON MISTLETOE EXTRACT-INDUCED APOPTOSIS IN ORAL SQUAMOUS CELL CARCINOMA)

  • This study was performed to investigate mistletoe extract-induced apoptosis in oral squamous cell carcinoma. In vivo study, HN22 cells were xenografted in nude mice. After tumor was experimentally induced, mistletoe extract was directly injected on the tumor mass. The specimens were evaluated using light and transmission electron microscopes. In vitro study, HN22 cells were cultured and exposed to mistletoe extract. The cells were evaluated using transmissin electron microscope. To evaluate apoptotic cells, flow cytometric analysis was done. The results were obtained as follows: 1. Light microscopic view of tumor mass showed necrosis at 2-4 weeks. 2. Transmission electron micrographs of tumor mass showed apoptosis and necrosis. 3. In TEM view of cell lines, necrosis and apoptosis were shown with mistletoe extract at $300{\mu}g/ml$, apoptosis was shown with mistletoe extract at $100{\mu}g/ml$. 4. In flow cytometric analysis, early and late apoptosis was shown when using caspase-3Ab and annexin-V, but no significant change was noted when using mebstain and Apo2.7 Ab. In this study, mistletoe extract induced necrosis and apoptosis in the tumor mass was induced by HN22 cells, early and late apoptosis in vitro study. Mistletoe extract was likely to induce cell death in oral squamous cell carcinoma through apoptosis.

수신증의 정도와 요로감염 및 자연경과, 산전초음파상의 수신증 여부와의 관련성에 대한 임상적 고찰 (Clinical study of urinary tract infection, natural courses, and prenatal ultrasonographic results according to the grades of hydronephrosis)

  • 목 적 : 신초음파검사는 수신증의 발견을 향상 시켰지만, 수신증의 임상 경과 및 치료에 대해서는 많은 이견이 있다. 이에 저자들은 수신증의 정도에 따른 환아들의 임상적 특징을 연구하였다. 방 법 : 1998년 4월부터 2007년 12월까지 신초음파검사에서 수신증이 발견된 총 367명의 환아들 중에 추적 관찰된 207명을 대상으로 후향적으로 조사하였다. 신우전후직경에 따라 대상을 grade 1-4까지 분류하고, 분석을 위해 이를 다시 mild 군 및 severe 군으로 나누었다. 결 과 : 추적기간 동안 요로감염은 207명 중 91명(44%)의 환아에서 총 128회 발생하였고, 수신증 정도에 따라서는 grade 1이 89명 중 35명(39.3%), grade 2는 88명 중 41명(46.5%), grade 3은 22명 중 11명(50%), grade 4는 8명 중 4명(50%)으로 요로감염의 발생 유무에 대해서는 mild 군과 severe 군 사이에 유의한 차이는 없었다. 그러나 요로감염의 누적 빈도 및 년당 발생한 요로감염의 횟수를 비교 해볼 때 severe 군이 mild 군보다 유의하게 높았다(P=0.041). Mild 군은 175명 중 103명(58%)이 완해를 보여 28명 중 3명(10.7%)의 완해를 보였다(P<0.001). 또한 산전초음파에서 수신증의 발견될 확률은 심한 신우전후직경을 나타낸 환아에서 더 높았다(P<0.001). 결 론 : 저자들은 수신증의 정도가 심할수록 자연완해는 더 적게 일어나며, 요로감염은 더 빈번하게 발생하는 것을 알게 되었다. 따라서 수신증을 가진 환아에 대해 신장 예후를 증진시키기 위하여 예방적 항생제 등의 치료 지침의 확립이 필요하다.

분열효모에서 hnRNP-유사 단백질인 THO4가 생장 및 mRNA 방출에 미치는 영향 (Effect of hnRNP-like protein THO4 on growth and mRNA export in fission yeast)

  • 진화적으로 보존된 TREX 복합체의 구성요소인 Yra1/ALY는 hnRNP-유사 단백질의 REF (RNA와 방출인자 결합단백질) 계열에 속하는 단백질로서 전사, 핵 RNA의 안정성, mRNA의 핵에서 방출 등 여러 과정에 관여하는 것으로 알려져 있다. 분열효모 Schizosaccharomyces pombe의 게놈에는 2개의 REF 단백질을 암호화하고 있다. 이미 mRNA 방출인자로 알려진 Mlo3 이외에 THO 복합체의 구성인자로 예측되는 Tho4이 존재한다. 본 연구에서 tho4 (SPBC106.12c) 유전자의 결실이 생장과 mRNA의 방출에 영향을 주지 않는다는 것을 알았다. 그러나 tho4 유전자를 과발현시키면 생장이 늦어지고 $poly(A)^+$ RNA가 핵 안에 약간 축적되었다. ${\Delta}tho4$ ${\Delta}mlo3$${\Delta}tho4$ ${\Delta}mex67$ 이 중 돌연변이는 어느 것도 추가적인 생장 결함을 보이지 않았다. 또한 Yeast two-hybrid와 공동침전(Co-immunoprecipitation) 분석에서 Tho4는 THO/TREX 복합체의 다른 구성인자들과 어떠한 상호작용도 보이지 않았다. 이와 같은 관찰결과들은 S. pombe의 Tho4 단백질이 기대와는 다르게THO/TREX 복합체의 구성인자가 아니며, mRNA 방출에도 직접 관여하지 않음을 의미한다.

  • We synthesized ethylene-styrene copolymer by coordination polymerization using 2 types of pyrazolato metallocene catalysts ((1,2,4-Me3C5H4)TiCl2(3,5-iPr2C3HN2), (tert-BuC5H4) TiCl2(3,5-iPr2C3HN2)). We observed the effects of styrene concentration on the catalytic activity, yield, molecular weight and molecular weight distribution.

약침치료(藥鍼治療)를 통한 요통환자(腰痛患者)의 호전도(好轉度)에 관한 임상적(臨床的) 관찰(觀察) (Clinical Observation about the Extent of Improvement of Low Back Pain Patient through Medi-acupuncture Therapy)

  • After 45 patients examined as to the result of medical treatment among the ones who came to Dept. of Pain Clinic, Oriental Medical Hospital, Chunju Woosuk University complaining low back pain chiefly from Dec/19/1994 to Feb/7/1995 for 50 days or so were observed clinically, the results were obtained as follows: 1. In duration of the case history, acute stage(37.8%) was the most predominant, and subacute stage(35.6%) and chronic stage(26.7%) were revealed in turn. 2. In opinions of radiation, Spondylosis(26.7%) was the most predominant, Scoliosis(15.6%) and HIVD(11.1%) were revealed in order, and 3 cases were revealed to be negative. 3. In the radiating pain of the lower limb, the radiating pains of the left lower limb were the most predominant and those of the right lower limb and those of both lower limbs were revealed in turn. By the way, 31.1% of patients didn't suffer from the radiating pain of the lower limb. 4. In the grade of the seriousness of subsective symptom, Grade 2(66.7%) was the most predominant, and Grade 3, Grade 4, and Grade 1 followed it in order. 5. In the period of the treatments of medi-acupuncture, 2-5 day treatments(31.1%) were major and 6-10 day treatments(26.7%), 16-20 day treatments(11.1%), 26-30 day treatments(11.1%), over 31 day treatments(11.1%), 11-15 day treatments(4.4%), and 21-25 day treatments(4.4%) followed it by turns. Thus 2-10 day treatments are 57.8% and under 30 day treatments are 57.8% of all. 6. In the frequency of use of each medi-acupuncture, V was most frequency used in 41 cases(91.1%), and 11 cases of HN(24.4%), 8 cases of MOK(17.8%), 4 cases of OK(8.9%), 2 cases of B(4.4%), and a case of I(2.2%) were revealed in turn. 7. In the effect of treatments, 10 cases(22.2%) were excellent, 25 cases(55.6%) were good, 5 cases(11.1%) were fair, and 5 cases(11.1%) were poor. As the result, 88.9% of all changed for the better and all of these were improved within the third trial. 8. The effect of the treatments per durations was 100% in acute stage, 93.8% in subacute stage, and 66.7% in chronic stage. According to that, it was reavealed that the rate of treatments decreased as it came near to the chronic stage. 9. The effect of treatments per radiating pains was 87.5% in the radiating pains of the left lower limb, 81.8% in those of the right lower limb, 100% in those of both lower limbs, and 92.9% in case that patients have no radiating pains. So in the rate of treatments on radiating pains of the lower limbs, the case of both sides or no radiating pains was higher than that of one side. 10. In the effect of treatments per the condition of patients, Grade 4 showed 100% of improvement, Grade 3 showed 90.9% of improvement, Grade 2 showed 86.7% of improvement, and Grade 1 showed 100% of improvement. 11. In the effect of treatments per contents of treatments, the group treated with medi-acupuncture therapy, herb medication therapy, acupuncture therapy, and physiothrapy at the same time(Group 1) showed 100% of improvement, the group treated with medi-acupuncture therapy, acupuncture therapy, and physiotherapy simultaneously(Group 2) showed 73.7% of improvement, and the group treated with medi-acupuncture therapy and acupuncture therapy at the same time(Group 3) showed 100% of improvement. 12. 2-5 day treatments showed 78.6% of improvement, 6-10 day treatments showed 91.7% of improvement, and over 31 day treatments showed 100% of improvement. As the result, genarally the longer the period of treatment was, the better the effect of treatment was. 13. When only V was used, the rate of treatment was 96.2%. When only HN was used, the rate of treatment was 100%. When only MOK was used, the rate of treatment was 100%. When V and HN were used at the same time, the rate of treatment was 33.3%. When V and MOK were used at the same time, the rate of treatment was 100%. When V and OK were used at the same time, the rate of treatment was 100%. When V, HN, and MOK were used at the same time, the rate of treatment was 100%. When V, OK, and MOK were used at the same time, the rate of treatment was 100%. When V, MOK, and B were used at the same time, the rate of treatment was 100%. When V, HN, OK, and I were used at the same time, the rate of treatment was 100%. When V, HN, and B were used at the same time, the rate of treatment was 100%.

NMR Structural Analysis and 3D Homology Modelling of APG8a from Arabidopsis thaliana

  • The gene coding for APG8a (At4g21980), a protein from Arabidopsis thaliana, is involved in the autophagy process. The protein is an interesting candidate for structure determination by NMR spectroscopy. Toward this end, APG8a has been produced recombinantly in Escherichia coli and typical NMR experiments such as $^{15}N-HSQC$, HNCA, HN(CO)CA, CBCA(CO)NH, HCCH-TOCSY, HNCO were performed. The backbone resonances, HN, N, CA, CB, and C' were sequence-specifically assigned, and the secondary structures including 3 $\alpha$ helices and $4\beta$ strands were deduced based on the assignments. Due to the intrinsic flexibility or the effect of the denaturant, the backbone resonances were not fully observed. Since the structure calculation by NMR data was not possible, the 3-dimensional model was built based on the sequence homology, and compared with the NMR results. The overall structure of the model could explain and complement the NMR derived secondary structures.

부상여재반응기에서 ASM3를 이용한 질산화 공정 모사 (Modelling of Nitrogen Oxidation in Aerated Biofilter Process with ASM3)

  • 하폐수의 생물학적 고도처리 공정의 설계와 운전 최적화에 필요한 자료를 제공하기 위하여 ASM3를 이용하여 부상여재반응기(ABF)에 대한 해석을 수행하였다. 대상 반응기로서는 수중의 부유물질 제거와 질산화 반응을 위해 이용되는 호기적 부상여재반응기로 하였다. 본 연구에서는 하폐수 고도처리공정 중 활성슬러지의 생흡착반응을 이용한 전체 시스템 중에 흡착조를 거친 유출수를 부상여재를 이용하여 질산화시키는 공정에 대한 해석을 목적으로 하였다. 민감도 분석과 영향평가를 위해서 상용 수처리 공정해석 프로그램인 GPS-X(V 5.0)을 이용하였다. 민감도 분석을 통해 공정거동에 영향이 큰 매개변수($Y_A$, $k_{sto}$, ${\mu}_A$, $K_{A,HN}$)가 결정되었고, 70일간의 pilot plant 운전자료를 이용한 동적 시뮬레이션을 통해 최적화하였다. 최적화된 매개변수 값들은 $Y_A$가 0.14, $k_{sto}$는 3.5/d, ${\mu}_A$는 2.7/d, $K_{A,HN}$는 1.1mg/L로 결정되었다. 최적화된 매개변수 값들을 이용하여 시뮬레이션을 수행하였고, 부상여재반응기의 수리학적 체류시간(HRT)을 10분에서 4시간으로 변화시키면서 TN, $NH_4{^+}-N$, $NO_3{^-}-N$의 변화를 예측하여 실측값과 비교한 결과 HRT가 10분 정도로 매우 짧은 운전조건에서도 60%에 가까운 질산화율을 보여주고 있었다. 짧은 HRT에서 우수한 질산화 능력을 보인 부상여재의 특성을 이용하여 유량변동이 매우 큰 현장에 부상여재반응기가 효과적으로 적용될 수 있다고 판단되었다.

인위적인 토양산성화에 대한 사방오리나무와 물오리나무 묘목의 생리반응 및 생장특성 (Physiological Responses and Growth Performance of Alnus firma and Alnus hirsuta Seedlings to Artificial Soil Acidification)

  • To obtain basic information for evaluating resistance of soil acidification for growth, net photosynthesis, $N_2$ fixation rate and nutrient status of Alnus firma and Alnus hirsuta seedlings grown in brown forest soil acidified with $H_2SO_4$ or $HNO_3$ solution were investigated (control (pH=5.9), LN (pH=5.0; Low levels of Nitric acid), HN (pH=4.0; High levels of Nitric acid), LS (pH=5.0; Low levels of Sulfuric acid), and HS (pH=4.0; High levels of Sulfuric acid)). The shoot dry weight of Alnus firma and Alnus hirsuta and the root dry weight of Alnus hirsuta seedlings grown in the HN, LS and HS were significantly less than that of the seedlings grown in the control. The Chlorophyll a/b ratio in leaves of Alnus firma at LN, LS and HS was significantly lower than that of control. The concentration of N in leaves of Alnus hirsuta at HS was significantly higher than that of control. The net photosynthetic rate of Alnus firma and Alnus hirsuta seedlings at LN and HN was higher than that of control. Based on the results, we concluded that the negative effects of soil acidification due to sulfate deposition are greater than those of soil acidification due to nitrate deposition on growth, net photosynthesis and $N_2$ fixation rate of Alnus firma more than Alnus hirsuta.

기공체조(氣功體操)가 DITI로 촬영한 상(上), 중(中), 하단전(下丹田)의 체표온도(體表溫度)에 미치는 영향(影響) (Effects of Qigong therapy on the thermal changes of upper, middle, lower $Danj{\breve{o}}n$(Ex-HN3, CV17, CV4) examined by Digital Infrared Thermographic Imaging(DITI))

  • Objectives : It is the object of Qigong therapy to promote the circulation of Qi and blood, and to relieve mentality by way of warming lower Danjon. In this study, to prove that Qigong therapy could actually subside heat on upper or middle Danjon and warm the temperature on lower Danjon, we observed the thermal changes of upper, middle, lower Danjon before and after Qigong therapy and compared them. Methods : We selected 16 patients, treated Qigong therapy and examined by D.I.T.I.(Digital Infrared Thermographic Imaging) before and after Qigong therapy, among patients who visited. Qigong clinic, Kangnam Korean hospital, Kyunghee University(Daechi-2dong, Kangnam-Gu) from april to october, 1999. We watched the difference of temperature among upper, middle, lower Danjon before and after Qigong therapy, and used student T-test(paired type, 2 tail) for proving effects of Qigong therapy statistically. Conclusions 1. The difference of temperature$({\Delta}T)$ between upper(Ex-HN3) and lower Danjon(CV4) significantly decreased about $0.55^{\circ}C$ after Qigong therapy(p<0.01). 2. The difference of temperature${\Delta}T$ between middle(CV17) and lower Danjon(CV4) significantly decreased about $0.39^{\circ}C$ after Qigong therapy(p<0.05). 3. The difference of temperature${\Delta}T$ between upper(Ex-HN3) and middle Danjon(CV17) decreased about $0.25^{\circ}C$ after Qigong therapy, but it was not statistically significant.

Photodissociation of Methane at Lyman Alpha (121.6 nm)

  • Laser induced fluorescence studies of hydrogen atom using four wave mixing technique are reported for the photodissociation of CH4 and its isotopomers at Lya (121.6 nm). The source of dissociating and probe radiation is one and the same (delay time??20 nsec). The average translational energy of ejected hydrogen atoms (50 Kcal/mol) reveals that CH4 + hn??CH3 + H(2S) and CH4 + hn??CH2(a1A1) + H2(1Sg) are the main dissociation processes. The absolute quantum yield for CH4 and CD4 are the same, FH(CH4) = FD(CD4) = 0.31 0.05. If one divides the experimental H/D ratios from the isotopomers CH3D, CH2D2, CHD3 by the isotopic H/D ratios, a value 2 is obtained in all three cases. Overall, the heavier D atoms are more likely than the H atoms to remain attached to the carbon atom.
