Effects of Ultrasonic Irradiation on Physical Properties of Silica/PEG Hybrids

  • The effect of ultrasonic radiation is reported for silica-poly(ethylene glycol) system prepared without the solvent using sol-gel processing by varying various parameters such as ultrasonic irradiation time, PEG content and HCl/TEOS molar ratio. The property of sonogel is compared with classic gel which has been prepared with ethanol as a solvent by traditional sol-gel processing. SEM, BET, DTA-TGA, density and Vickers hardness measurements are carried out for analyzing the samples. The gelation time is found strongly dependent on radiation time, PEG content and pH value, and has been discussed on the basis of existing theories. The $SiO_2-10$ & 20 wt% PEG sonogel exhibited superior optical, physical and gel properties as compared to the classic gel, hence, found suitable for device applications. The ultrasonic radiation increased the density and surface area, and also reduced the pore size which is well supported by the shift in the peak of DTA curve. The DTA thermogram was found similar to that of pure silica gel.

Enantiomer Separation of α-Amino Acid Esters as Nitrobenzoxadiazole Derivatives Using Chiral Columns (키랄 컬럼을 사용한 아미노산 에스테르의 니트로벤조옥사디아졸 유도체의 광학분리)

  • A new convenient derivatization method of ${\alpha}$-amino acid esters as nitrobenzoxadiazole (NBD) derivatives for chiral resolution was introduced and the enantiomer separation of ${\alpha}$-amino acid esters as NBD derivatives was performed by normal HPLC using chiral columns based on polysaccharide derivatives. The NBD derivatives were readily prepared by stirring NBD-Cl and ${\alpha}$-amino acid methyl ester HCl with $NaHCO_3$ in ethanol. The performance of Chiralpak IA was superior to the other chiral stationary phases for enantiomer resolution of NBD derivatives of several ${\alpha}$-amino acid methyl esters. Owing to fluorescence detection as well as strong UV absorption, it is expected that the convenient analytical method developed in this study will be very useful for enantiomer separation of ${\alpha}$-amino acid esters as NBD derivatives on polysaccharide-derived chiral columns.

A Study on the Analysis of Amino Acids in Korean Ginseng (韓國人蔘의 年根別 및 貯藏期間別 아미노酸分析)

  • The contents of amino acids were examined in the 3, 4, 5, and 6 year-old roots of fresh ginseng and the 1979, 1980, 1981, and 1982 years' products of white and red ginsengs. Samples extracted with 75% ethanol for free amino acids and hydrolyzed with 6N-HCL for total amino acids were analyzed by Amino Acid Analyzer (Hitachi model KLA-5). The results were summarized as follows: 1. Amino acids from extracted samples were 18 kinds of Tryptophan, Lysine, Histidine, Arginine, Aspartic acid, Threonine, Serine, Glutamic acid, Proline, Glycine, Alanine, Cystine, Valine, Methionine, Isoleucine, Leucine, Tyrosine, and Phenylalanine. 2. Amino acids detected in hydrolyzed samples were 17 kinds execpt Tryptophan of extracted ones. 3. Arginine was the highest quantity of amino acids in ginseng. 4. The content of Tryptophan was 0.5690 mg/g in the 6 year-old fresh ginseng and trace quantities in other samples. 5. The contents of amino acids were increased in fresh ginseng according to cultivation year. 6. The contents of amino acids in white ginseng were slightly decreased but those in red ginseng were not changed during the storage time. 7. The content ratio of free amino acids to total amino acids were 1:3.

  • In order to investigate the effect of Daewhajungeum on the isolated organs, gastrointestinal tract and C.N.S., some kinds of experiments using mice and rats were done. The results were as follows; 1. Daewhajungeum showed the inhibitory effect on the smooth muscle contraction of the isolated ileum and colon induced by acethylchorine chloride and barium chloride in mice. 2. Daewhajungeum showed inhibitory effect on the contraction of the fundus-strip induced by acethylchorine chloride and barium chloride in rats. 3. The preventive effect on pyloric ulcer, indomethacin induced ulcer and ethanol-HCl induced ulcer of mice was recognized. 4. The effect on the acidity of gastric juice was suppressed, but the effect on the pepsin output was not recognized. 5. The transport ability of small and large intestine was significantly decressed. 6. The analgesic effect and the sedative effect were recognized so that the effect of the C.N.S was suggested.

Pullulan Production from Starch Hydrolysate by Aureobasidium pullulans SH8646

  • Pullulan was produced from starch hydrolysate with Aureobasidium pullulans SH8646. We could measure the correct amount of pullulan produced without the interference of starch from the culture supernatant by using a bacterial $\alpha$-amylase treatment and ethanol: acetone (1:1) precipitation. When 5% acid-hydrolyzed starch was used as a carbon source, the dry cell weights obtained were similar irrespective of DE values of starch hydrolysates. The dry cell weights of those on the starch hydrolysate media prepared with 0.1 N HC1 treatment, were slightly higher (9.5~10.5 g/l) than those on the starch hydrolysate media prepared with 1.0 N HCl (8.5~9.5 g/l). And among the starch hydrolysates showing DE values lower than 50, maximum pullulan production of 15 g/l was obtained at DE 30~40 starch hydrolysate but those showing DE values higher than 50, the pullulan production was increased with the increase of the DE value of starch hydrolysates. From the media containing 5%, 10%, and 15% starch hydrolysate (DE 25, 45, and 75), about 20~34% pullulan yield was obtained and the maximum pullulan yield of 34% (17g/l) was obtained from 5% DE 75 starch hydrolysate. The pullulan yields from starch hydrolysate media were much lower than those from glucose, maltose, maltotriose, and sucrose media.

Antigastritic and Antiulcerative Effects of Cirsium japonicum var. ussuriense Extract and Fractions (엉겅퀴(Cirsium japonicum var. ussuriense)추출물 및 분획물의 항위염 및 항위궤양 효과에 대한 연구)

  • Cirsium japonicum var. ussuriense (CJ) is used as a folk medicine for diuretic, arthritis, dyspepsia, and bleeding in Korea. Until now, more than fifteen kinds of CJ were identified and among them, Cirsium japonicum var. nakaianum Nakai was selected as experimental material. Both that from The Medical Herb Garden of Seoul National University located at Gayang-dong, Koyang city and native that from a hillock near Dangjin, Chungnam were collected and analyzed to determine whether it is varied in efficacy on their habitats. Extracts and fractions of CJ had a strong effect of antibacterial activity on H. pylori and antioxidant effects. And the CJ from Garden and Dangjin were showed to be more efficient in acute gastritis through the HCl${\cdot}$ethanol-induced gastric lesion test and chronic gastritis through the indomethacin-induced gastric lesion test, respectively. This study proved that the effects of CJ are varied by their habitats. Also, these results suggest that the CJ has potentials for use as functional food and medicine.

Antigastritic and Antiulcerative Action of the Extract of Zingiberis Rhizoma (생강추출물의 항위염 . 항궤양 작용)

  • The rhizoma of Zingiber officinale has been used as antiemetic, expectorants, stomachache relieving drugs and digestive accelerators. From the observation of antigastritic action of the methanol extract of the rhizoma, it was fractionated with hexane, chloroform, ethyl acetate and butanol, followed by bioassay on antigastritic and antiulcerative activity. The hexane and the chloroform fraction reduced significantly HCl ethanol induced gastric lesion at the dose of 370 and 210 mg/kg, p.o., respectively. On the gastric ulceration and gastric secretion in pylorus-ligated rats, the hexane fraction decreased the volume of gastric secretion and acid output, and also increased pH at the dose of 370 mg/kg, i.d.. It showed considerable curative ratio of acetic acid induced ulcer without inhibition of indomethacin induced gastric lesion. The methanol extract showed low acute toxicity with minimum lethal dose of more than 3000 mg/kg, p.o. in mice. In conclusion, Zingiberis rhizoma exhibited antigastric and antiulcerative activity which might be attributable to inhibition of gastric secretion. It is revealed that the active component may be present in the hexane fraction.

Preparations of Universal, Functionalized Long-Chain Alkylthiol Linkers for Self-assembled Monolayers (자기조립단분자막을 위한 보편적이고 기능화된 긴 사슬 알킬티올 연결자의 제조)

  • In this research, the preparation processes for making a series of $\omega$-mercapto alkylamine 1 and $\omega$-mercapto alkanoic acid 2 useful for studying of the self-assembled monolayer(SAM) are described. The preparation methods of the first goal materials, $\omega$-mercapto alkylamines 1 were carried out as follows: First, $\omega$-phthalimide alkanol 3 was synthesized from commercially available potassium phthalimide derivatives and $\omega$-bromoalkanol in DMF at $80{^{\circ}C}$ via substitution reaction. After refluxing $\omega$-phthalimide alkanol 3 with hydrazine hydrate in ethanol followed by treating with c-HCl, $\omega$-aminoalkanol 4 was obtained in 76-98% yield, accompanied with side-product 5. Bromination of hydroxyl moiety of $\omega$-aminoalkanol 4 using aqueous hydrobromic acid furnished $\omega$-bromoamine 6 in 34-97% yields. Substitution reaction 6 with thiourea in 95% ethanol gave $\omega$-aminoalkanthiuronium 7, which was treated with aqueous strong base and aqueous strong sulfuric acid gave desired products, $\omega$-mercapto alkylamines 1 through overall 5 steps. The second target material, $\omega$-mercapto alkanoic acid 2 was prepared via 2 steps. $\omega$-bromo alkanoic acid was reacted with thiourea to give $\omega$-thiourea alkanoic acid 7 in 69-85%, which was treated with aqueous strong base and strong acid to furnish $\omega$-mercapto alkanoic acid 2 in 50-98%. The fabricated long-chain alkylthiol(LCAT) can be used as linkers to immobilize protein, enzyme and various kinds of biomolecules on the surface of metallic materials(Au, Pt, Ti) by SAM, and can be useful chemical tools for the application study on the surface modification of metallic materials.

Induction of Heme Oxygenase-1 by Traditional Herb Mix Extract Improves MKN-74 Cell Survival and Reduces Stomach Bleeding in Rats by Ethanol and Aspirin in vivo

  • Chinese herb medicines have traditionally been used to treat or alleviate the symptom of various diseases. The rationale for use of certain herbs to certain disorder is now getting unveiled by modern technology. In the present study, we investigated whether herb mix extract(HMX), which is alleged to be useful for gastric ulcer, protects stomach from oxidative stress. Rats were allowed to normal diet with and without HMX (1, 5, 10 mg/kg) for 30 days. To induce gastric ulcer, ethanol (75%, 1.5 ml) or acidified aspirin (100 mg/kg in 0.2 N HCl) was administered by oral route in 24 h-fasted rats and examined the gastric ulceration(bleeding) by measuring the size 1 h after the treatment. Results indicated the area of gastric bleeding was significantly less in HMX fed rats than in normal diet fed ones, and it was dependent on the duration and amount of HMX. To investigate the underlying mechanism by which HMX protects stomach from oxidative stress, expression of enzymes like heme oxygenase (HO), cyclooxygenase (COX), and inducible nitric oxide (iNOS) were investigated in MKN-74 cells, where aspirin or H. pylori was introduced. The results were compared with RAW 264.7 cells to check if there's cell specificities exist. The expression of HO-1 but not COX-2, iNOS was significantly increased by HMX. Furthermore, HO-1 inhibitor, SnPP IX reduced the HO-1 activity and reversed the survival rate in HMX-treated MKN-74 cells. There's no difference between RAW 264.7 cells and MKN-74 cells. We, thus, concluded that HMX is beneficial for protection from oxidative injury, and induction of HO-1 by HMX in gastric cells is, at least, responsible for protection from oxidative stress such as ethanol, aspirin and possibly H. pylori infection.

Effects of Ethylacetate Fraction of Plantain (Plantago asiatica L.) on Experimentally-Induced Gastric Mucosal Damage and Gastric Ulcers in Rats (질경이가 실험적으로 유발된 흰쥐의 위염 및 위궤양에 미치는 영향)

  • Plantain has been used for antidiarrhea, antihemorrhage and the remedy of indigestion. Plantain was extracted with ethanol and fractionated systemically with n-hexane, chloroform, ethylacetate (EtOAC) and n-butanol. Antioxidant index (AI was expressed as induction period of oil containing various fractions/induction period of oil of 600 ppm) of EtOAC fraction was the highest among fractions in vitro. The protective effects of the EtOAC fraction of plantain (PE) administered 1 mL orally or intraduodenally on experimentally induced gastritis, gastric ulcer and gastric secretion were evaluated in rats. Sprague-Dawley rats weighing 250∼300 g were divided into 4 groups; negative control group (CON), PE 200 mg/kg treated group (PEL), PE 400 mg/kg treated group (PEH) and positive control group (cimetidine 100 mg/kg-CMT or omeprazol 100 mg/kg treated group-OMT), respectively, PE significantly suppressed HCl-ethanol induced gastric lesions and indomethacin-induced gastric ulcers (administered subcutaneouly) in rats. Specially PE 400 mg/kg showed significantly inhibitory effect, which was more potent than that of 100 mg/kg of commercial drug, cimetidine, and elevated an inhibitory effect to be close to the level in inhibitory ratio of omeprazol administered group in Shay's ucler. On gastric secretion in pylorus ligated rat, PE 200 mg/kg and 400 mg/kg decreased the gastric volume and acid output, but did not show an apparent effect on pepsin activity. In addition, PE 400 mg/kg depressed gastric ulcers induced by water immersion stress and duodenal ulcers induced by cysteamine administered subcutaneouly. These results suggest that the ethylacetate fraction of plantain can be used in prevention and treatment of experimentally induced gastric mucosal damage and ulcers.
