Implication of embB Gene Mutation in Ethambutol-Susceptible Clinical Isolates of Mycobacterium tuberculosis (임상에서 분리된 에탐부톨 감수성균에서의 embB 유전자 돌연변이에 대한 고찰)

  • Background : Ethambutol(EMB) is one of the first-line drugs included in short-course anti-tuberculosis therapy. The point mutations in embB gene have been speculated to be associated EMB resistance. However, detection of embB mutations at these positions have been observed in both EMB-susceptible isolates; thus, it remains controversial whether these mutations are associated with EMB resistance Methods : The 36 M. tuberculosis isolates were selected from clinical isolates which tested susceptible to EMB and resistant to at least one drug. DNA extracted from the isolates was analyzed by amplifying embB gene. The PCR products were purified and directly sequenced. We reviewed the history of past drug susceptibility test results. Results : Out of 36 EMB-susceptible strains, 3 strains (8.3%) had a mutation in codon 306 or 406 of the embB gene. These three strains had at least isoniazid resistance. They grew at 1.0 mcg/ml of EMB in Lowenstein-Jensen media. The patients of the strains were continuously smear-positive for over 3 years despite taking TB therapy. One strain had been EMB-resistant in past drug susceptibility tests. Conclusion : EMB-susceptible strains containing embB mutation may be caused by decreased viability in vitro test not by itself.

Expression of Neuregulins and Their Receptors During the Differentiation of Rat Hippocampal HiB5 Cells

  • Differentiating HiB5 cells, a rat hippocampal cell line, expressed neuregulins and showed constitutive activation of a neuregulin receptor, ErbB2, suggesting development of a neuregulin autocrine loop. RT-PCR analyses indicated that HiB5 cells produced SMDF and NDF, but not GGF, during the differentiation. None of neuregulin isoforms were detected in proliferating HiB5 cells. The neuregulins in HiBS cells, at least in part, are the $\beta$-isoforms of which the most of neuronal neuregulin isoforms are. The expression of SMDF and NDF was enhanced by PDGF and bFGF that promote cell survival and differentiation, suggesting a close relationship between the synthesis of neuregulins and the differentiation process. HiB5 cells have ErbB2 and ErbB4, but not ErbB3 receptors. Constitutive tyrosine phosphorylation of ErbB2 was detected in HiB5 cells that had not been exposed to exogenous GGF.

The Effect of Hydrophobicity and Amphiphilicity on the Mastoparan B-induced Antibacterial Activity and Hemolytic Activity (Mastoparan B의 항균 활성 및 용혈 작용에 미치는 소수성도와 양친매성의 영향)

  • Tetradecapeptide, Mastoparan B(MP B) and its [$Ala^2$]-, [$Ala^4$]-, [$Ala^6$]-, [$Ala^9$]-MP B derivatives were synthesized, and then their antibacterial and hemolytic activities were assayed to examine the effect of hydrophobicity and amphiphilicity on the MP B-induced those activities. MP B and more hydrophobic [$Ala^2$]-, [$Ala^4$]-MP B showed stronger antibacterial activity and less hydrophobic [$Ala^6$]-MP B than MP B did similar or weaker activity, so more hydrophobic [Ala]-MP B derivative had stronger activity. But more hydrophobic [$Ala^9$]-MP B than MP B showed weaker activity because of its Trp substitution by Ala. On the other hand, [$Ala^2$]- and [$Ala^4$]-MP B showed 100.0% and 69.4% hemolytic activity, but [$Ala^6$]-MP B did the weakest activity(6.1%) and [$Ala^9$]-MP B, weaker activity(26.0%) than MP-B. Therefore, more hydrophobic [Ala]-MP B derivative had stronger activity and the effect of amphiphilicity on the activity was weak.

Effectiveness of Computed Tomography for Blow-out Fracture

  • Purpose: This study assessed the association between eye symptoms (enophthalmos or diplopia) and site of damage, volume, deviated inferior rectus muscle (IRM) and type of fracture with computed tomography (CT). The intent is to anticipate the prognosis of orbital trauma at initial diagnosis. Methods: Forty-five patients were diagnosed with fractures of the inferior wall of one orbit. Fracture area, volume of displaced tissue, deviated IRM, and type of fracture were evaluated from coronal CT by one investigator. The association of those variables with the occurrence of eye symptoms (diplopia and enophthalmos) was assessed. Results: Of 45 patients, 27 were symptom-free (Group A) and 18 had symptoms (Group B) of enophthalmos and/or diplopia. In Group B, 12 had diplopia, one was enophthalmos, and five had both. By CT measurement, group A mean area was $192.6mm^2$ and the mean volume was $673.2mm^3$. Group B area was $316.2mm^2$ and volume was $1,710.6mm^3$. The volume was more influential on symptom occurrence. Each patient was categorized into four grades depending on the location of IRM. Symptom occurrence and higher grade were associated. Twenty-six patients had trap-door fracture (one side, attached to the fracture), and 19 had punched-out fracture (both sides detached). The punched-out fracture was more strongly associated with symptoms and had statistically significantly higher area and volume. Conclusion: In orbital trauma, measurement of fracture area and volume, evaluation of the deviated IRM and classification of the fracture type by coronal CT can effectively predict prognosis and surgical indication.

A Study of the International Color Sensibility through the Analysis of the Ethnic Color Preference (민족적 색채(Ethnic color)기호의 분석을 통한 국가별 색채감성)

  • The purpose of this study is to confirm the international unique color sensibility according to the ethnic color preferences. The existing studies about color sensibility were investigated to analyze the international color sensibility. The countries were chosen according to its, strong regional and racial color. Also, the documents and websites about environment color such as structure color, natural feature color, traditional folk costume color and customary color names were investigated, and then, the international color sensibility was analyzed by using the color image scale. As a result of the analysis about the differences of color sensibility, internationally distinguished color sensibility was discovered. There were differences not only for the preference trend of hue but also for the tone or contrast of color among the selected countries. Especially, Great Britain had a strong preference for G categories that they preferred the warm-grayish color image. Russia has a preference for R, G, and B categories with the preference for the warm-clear image. Netherlands had a preference for R, Y, and PB categories and it preferred the cool-hard-grayish, warm-soft-clear image. Italy had a preference for R and Y categories and it preferred the warm-clear image. Morocco had a preference for R and B categories and it preferred the warm and cool, clear image. Japan had a preference for R, G categories and it preferred the warm-grayish image. Korea had a preference for R and B categories and it preferred the warm-soft-clear, and cool-clear image. With these results, the researcher concludes that the integrated analysis of the environment color and the traditional racial color factors are very persuasive methods to comprehend the international color sensibility.

The Exception Case about the Diagnose Forward Head Posture using the CranioVertebra Angle, CranioRotation Angle and Cobb angle : a Case Report

  • PURPOSE: The purpose of present study was to introduces an exceptional case in measurement methods (CVA, CRA and Cobb angle) to identify the FHP with verified reliability and validity. Subjects: Three males aged 30 years were recruited: A Normal, B and C who have FHP. METHODS: All the subjects were measured CVA, CRA and Cobb angle with the Photogrammetry and Radiography. RESULTS: The results revealed that it is not enough for measurement methods to identify the FHP using CVA, CRA and Cobb angle. On Photogrammetry values; CVA had $65^{\circ}$, CRA was $148^{\circ}$ of Normal subject A and CVA had $61^{\circ}$, CRA was $149^{\circ}$ of FHP subject B and CVA had $51^{\circ}$, CRA was $149^{\circ}$ of FHP subject C. On Radiography values; CVA had $73^{\circ}$, CRA was $148^{\circ}$ and Cobb was $50^{\circ}$ of Normal subject A and CVA had $70^{\circ}$, CRA was $150^{\circ}$ and Cobb was $53^{\circ}$ of FHP subject B and CVA had $61^{\circ}$, CRA was $153^{\circ}$ and Cobb was $31^{\circ}$ of FHP subject C. CONCLUSION: The reliable CVA, CRA and Cobb angle use methods from the previous studies might not be suitable for the diagnose the FHP. We think that it is necessary to have more detailed evaluation methods and the radiography is also needed for clear evaluations because of some possible exceptions.

Analysis of Prognostic Factors according to WHO Classification for Thymic Epithelial Tumor (흉선종의 WHO 분류에 따른 예후인자의 임상적 고찰)

  • Background: Primary thymic epithelial neoplasm is a type of mediastinal tumors that have various biologic and morphologic features. In this study, we reclassified 59 cases of thymic epithelial tumors by the new WHO classification. We inquired whether the new WHO classification has independent prognostic relevance by analyzing clinical characteristics of thymic epithelial tumors including Masaoka's clinical stage. Material and Method: From December 1986 to August 2003, 59. patients who underwent surgery in the Keimyung University Dongsan Medical Center with definite diagnosis of thymic epithelial tumor were studied. We analyzed the histologic subtype (WHO classification). clinical stage (Masaoka's clinical stage) and patient's characteristics (sex, age, myasthenia gravis, tumor size, invasion. recurrence, metastasis) as prognostic factors. We analyzed the relationship between histologic subtype and clinical stage. Result: 32 patients were male and 27 were female. Mean age was 50.1$\pm$14.2. From WHO A to C, all thymic epithelial tumors were reclassified by the new WHO classification. Six patients (10.2%) had Type A, 7 (11.9%) had Type AB, 7 (11.9%) had Type B$_1$, 10 (16.9%) had Type B$_2$ and 7 (11.9%) had Type B$_3$, 22 (37.3%) had Type C. Two factors were shown by multivariate analysis to be associated with a favorable prognosis: completeness of resection (p=0.003) and non-invasiveness (p=0.001). The overall 5-year survival of the 59 patients was 53%, subtype A and AB were 92.3%, B$_1$ and B$_2$ were 70.2%, and B$_3$ and C were 26.1%. The association between histologic subtype and invasive behavior (stage) was statistically significant (p<0.001). Conclusion: The WHO classfication is not only a histologic classfication of the thymic epithelial tumors but also a significant prognostic factor that influence the survival of thymic epithelial tumors.

Analysis Microstrip Patch Antenna of MIMO Structure (MIMO 구조의 마이크로스트립 패치 안테나 분석)

  • This study proposed a patch antenna with a MIMO structure which is applicable for wireless communication equipment by combining a single patch antenna with a multi port. The proposed MIMO patch antenna was designed through the TRF-45 substrate with a relative permittivity of 4.5, loss tangent equal to 0.0035 and dielectric high of 1.6 mm, and the center frequency of the antenna was 2.45 GHz in the ISM (Industrial Scientific and Medical) band. The proposed MIMO patch antenna had a 500 MHz bandwidth from 2.16 ~ 2.66 GHz and 24.1% fractional bandwidth. The return loss and VSWR were -62.05 dB, 1.01 at the ISM bandwidth of 2.45 GHz. The Wibro band of 2.3 GHz was -17.43 dB, 1.33, the WiFi band of 2.4 GHz was -31.89 dB, 1.05, and the WiMax band of 2.5 GHz was -36.47 dB, 1.03. The radiation patterns included in the bandwidth were directional, and the WiBro band of 2.3 GHzhad a gain of 4.22 dBi, the WiFi band of 2.4 GHz had a gain of 4.12 dBi, the ISM band of 2.45 GHz had a gain of 4.06dBi, and the WiMax band of 2.5 GHz had a gain of 3.9 6dBi.

The Fundamental Properties of Alkali-Activated Slag Cement (AASC) Mortar with Different Water-Binder Ratios and Fine Aggregate-Binder Ratios (물-결합재 비와 잔골재-결합재 비에 따른 알칼리 활성화 슬래그 모르타르의 기초특성)

  • This study investigates the fundamental properties of the water-binder (W/B) ratio and fine aggregate-binder (F/B) ratio in the alkali-activated slag cement (AASC) mortar. The W/B ratios are 0.35, 0.40, 0.45, and 0.50, respectively. And then the F/B ratios varied between 1.00 and 3.00 at a constant increment of 0.25. The alkali activator was an 2M and 4M NaOH. The measured mechanical properties were compared, flow, compressive strength, absorption, ultra sonic velocity, and dry shrinkage. The flow, compressive strength, absorption, ultra sonic velocity and dry shrinkage decreased with increases W/B ratio. The compressive strength decreases with increase F/B ratio at same W/B ratio. Also, at certain value of F/B ratio significant increase in strength is observed. And S2 (river sand 2) had lower physical properties than S1 (river sand 1) due to the fineness modulus. The results of experiments indicated that the mechanical properties of AASC depended on the W/B ratio and F/B ratio. The optimum range for W/B ratios and F/B ratios of AASC is suggested that the F/B ratios by 1.75~2.50 at each W/B ratios. Moreover, the W/(B+F) ratios between 0.13 and 0.14 had a beneficial effect on the design of AASC mortar.

Determination of Phenolic Compounds in Adzuki bean (Vigna angularis) Germplasm

  • The aim of this study was to evaluate 30 phenolic compounds in adzuki bean germplasm. Adzuki 21653 had the highest content of total phenolics compounds ($6597\;{\mu}g\;g^{-1}$) while 104372 had the lowest concentration. The average total phenolic content of Japanese ($2432\;{\mu}g\;g^{-1}$) adzuki beans was higher than that of Korean ($2256\;{\mu}g\;g^{-1}$) adzuki beans. The average total phenolic contents were $2507\;{\mu}g\;g^{-1}$ in small sized adzuki beans from Japan and $2459\;{\mu}g\;g^{-1}$ in those from Korea. In large sized adzuki beans, the average total phenolic contents were $1315\;{\mu}g\;g^{-1}$ in Japanese seeds and $1232\;{\mu}g\;g^{-1}$ in Korean seeds. The average total phenolic contents in medium seeds were $2369\;{\mu}g\;g^{-1}$ in Japanese adzuki beans and $1397\;{\mu}g\;g^{-1}$ in Korean ones. In small seeds, the total phenolic contents of adzuki beans varied from $524\;{\mu}g\;g^{-1}$ to $6597\;{\mu}g\;g^{-1}$ in Japanese ones and from $375\;{\mu}g\;g^{-1}$ to $6569\;{\mu}g\;g^{-1}$ in Korean ones. Japanese and Korean adzuki beans were divided into landraces and wild adzuki beans. In this study, the wild adzuki beans showed higher contents of total phenolics than the native varieties. Specifically, the wild adzuki beans from Korea had the highest concentration of phenolics ($3403\;{\mu}g\;g^{-1}$). All adzuki bean germplasms were measured for their color and were classified into four groups accordingly: A; L < 30, +a, +b; B; L < 30, +a, -b, C; L > 50, +a, +b, D; L > 50, +a, -b. Especially, group B had the highest concentration of total phenolic compounds ($2827\;{\mu}g\;g^{-1}$), whereas group C had the lowest concentration ($1882\;{\mu}g\;g^{-1}$).
