Characterization of Chryseobacterium aquaticum Strain PUPC1 Producing a Novel Antifungal Protease from Rice Rhizosphere Soil

  • Strain PUPC1 produces an antifungal protease as well as plant growth promoting enzymes such as 1-aminocyclopropane-1-carboxylate (ACC) deaminase and phosphatase. Morphological, cultural, and physiological characteristics as well as 16S rRNA gene-sequence-based phylogenetic analysis confirmed the taxonomic affiliation of PUPC1 as Chryseobacterium aquaticum. The optimum growth of PUPC1 was observed at pH 6.0 and $30^{\circ}C$, and maximum protease production was observed in medium B amended with 1% tryptone, 0.5% sucrose, and 0.005% $MnCl_2$. The protease was purified by ammonium sulfate precipitation, Sephadex G-75 gel filtration chromatography, and electroelution from preparative SDS-PAGE. The protease had a molecular mass of 18.5 kDa. The optimum pH and temperature stability of the protease were pH 5.0-10.0 and temperature $40-70^{\circ}C$. Chryseobacterium aquaticum PUPC1 and its protease showed a broad-spectrum antifungal activity against phytopathogenic fungi. Strain PUPC1 also exhibited plant growth promoting traits. The objective of the present investigation was to isolate a strain for agricultural application for plant growth promotion and biocontrol of fungal diseases.

Characterization of Soluble Dietary Fibers from Wax Gourd (Benincasa hispida) Pulp and Peel

  • The physicochemical and in vitro physiological properties of soluble dietary fiber (SDF) from wax gourd (Benincasa hispida) pulp and peel were investigated. The pulp was composed of 11.4% SDF and 24.3% insoluble dietary fiber (IDF), while the peel contained 3.2% SDF and 43.3% IDE The predominant sugar in the SDF of the wax gourd pulp and peel was uronic acid, followed by galactose and rhamnose. The SDFs from the wax gourd pulp and peel gave similar elution patterns, with 4 main neutral sugar and uronic acid peaks eluted by 0.4, 0.5, 1, and 2 M ammonium acetate buffer. The pulp SDF had a much higher glucose retardation index (GRI) than the peel SDF for all measurement times. The pulp SDF showed strong growth-inhibiting activity against Escherichia coli and Clostridium perfringens, whereas the peel SDF produced strong growth-promoting activity against Bifidobacterium longum, Bifidobacterium infantis, and Lactobacillus brevis when compared to glucose.

Molecular Identification and Evaluation of Indigenous Bacterial Isolates for Their Plant Growth Promoting and Biological Control Activities against Fusarium Wilt Pathogen of Tomato

  • In search of an effective biological control agent against the tomato pathogen Fusarium oxysporum f. sp. lycopersici, rhizospheric soil samples were collected from eight agro-ecological zones of Bangladesh. Among the bacteria isolated from soil, 24 isolates were randomly selected and evaluated for their antagonistic activity against F. oxysporum f. sp. lycopersici. The two promising antagonistic isolates were identified as Brevundimonas olei and Bacillus methylotrophicus based on morphological, biochemical and molecular characteristics. These two isolates were evaluated for their biocontrol activity and growth promotion of two tomato cultivars (cv. Pusa Rubi and Ratan) for two consecutive years. Treatment of Pusa Rubi and Ratan seeds with B. olei prior to inoculation of pathogen caused 44.99% and 41.91% disease inhibition respectively compared to the untreated but pathogen-inoculated control plants. However, treatment of Pusa Rubi and Ratan seeds with B. methylotrophicus caused 24.99% and 39.20% disease inhibition respectively. Furthermore, both the isolates enhanced the growth of tomato plants. The study revealed that these indigenous bacterial isolates can be used as an effective biocontrol agent against Fusarium wilt of tomato.

Microbiome of Halophytes: Diversity and Importance for Plant Health and Productivity

  • Saline soils comprise more than half a billion hectares worldwide. Thus, they warrant attention for their efficient, economical, and environmentally acceptable management. Halophytes are being progressively utilized for human benefits. The halophyte microbiome contributes significantly to plant performance and can provide information regarding complex ecological processes involved in the osmoregulation of halophytes. Microbial communities associated with the rhizosphere, phyllosphere, and endosphere of halophytes play an important role in plant health and productivity. Members of the plant microbiome belonging to domains Archaea, Bacteria, and kingdom Fungi are involved in the osmoregulation of halophytes. Halophilic microorganisms principally use compatible solutes, such as glycine, betaine, proline, trehalose, ectoine, and glutamic acid, to survive under salinity stress conditions. Plant growth-promoting rhizobacteria (PGPR) enhance plant growth and help to elucidate tolerance to salinity. Detailed studies of the metabolic pathways of plants have shown that plant growth-promoting rhizobacteria contribute to plant tolerance by affecting the signaling network of plants. Phytohormones (indole-3-acetic acid and cytokinin), 1-aminocyclopropane-1-carboxylic acid deaminase biosynthesis, exopolysaccharides, halocins, and volatile organic compounds function as signaling molecules for plants to elicit salinity stress. This review focuses on the functions of plant microbiome and on understanding how the microorganisms affect halophyte health and growth.

Antagonistic and Plant Growth-Promoting Effects of Bacillus velezensis BS1 Isolated from Rhizosphere Soil in a Pepper Field

  • Pepper (Capsicum annuum L.) is an important agricultural crop worldwide. Recently, Colletotrichum scovillei, a member of the C. acutatum species complex, was reported to be the dominant pathogen causing pepper anthracnose disease in South Korea. In the present study, we isolated bacterial strains from rhizosphere soil in a pepper field in Gangwon Province, Korea, and assessed their antifungal ability against C. scovillei strain KC05. Among these strains, a strain named BS1 significantly inhibited mycelial growth, appressorium formation, and disease development of C. scovillei. By combined sequence analysis using 16S rRNA and partial gyrA sequences, strain BS1 was identified as Bacillus velezensis, a member of the B. subtilis species complex. BS1 produced hydrolytic enzymes (cellulase and protease) and iron-chelating siderophores. It also promoted chili pepper (cv. Nockwang) seedling growth compared with untreated plants. The study concluded that B. velezensis BS1 has good potential as a biocontrol agent of anthracnose disease in chili pepper caused by C. scovillei.

간호사의 개인적 특성과 건강상태 인지, 신체상, 건강증진행위간의 관계 (The Relationship of Individual Characteristics, Perceived Health Status, Body Image, and Health Promoting Behavior in Hospital Nurses)

  • Objective: The purpose of this study were to: (1) describe the individual characteristics, perceived health status, body image, and health promoting behaviors of staff nurses working in a metropolitan hospital and (2) determine the relationship of perceived health status, body image, and health promoting behaviors. Methods: Data were collected from 311 staff nurses working at one of university affiliated hospitals using a self-administered questionnaire for perceived health status(SF 36 V2), body image(BAQ), and health promoting behaviors (HPLP-II) and using the report of year 2002 employee physical check-up results for health related characteristics. Results: Most of nurses were within the normal range of BMI, total cholesterol, and liver enzymes but 42.2% had systolic BP above 120mmHg and 37.3% hemoglobin below 12g/dL. Although 96.4% of BMI score indicated 'underweight' or 'normal', 'feeling fat' showed the highest. Among health promoting behaviors the most frequently reported one was spiritual growth and the least one was engagement in physical activity. In the correlational analysis, health promoting behaviors had the positive relationships with perceived health status, vitality, mental health, attractiveness, strength and fitness (p=.000 - .004). Conclusion: These findings provide information that is relevant in designing interventions to enhance health promoting behaviors among nurses working in a hospital.

In vivo와 In vitro 평가모델을 利用한 韓藥抽出物의 毛髮成長 및 促進에 미치는 實驗的 硏究 (Extracts for the Hair Growth Stimulation using In vivo and In vitro Test Models)

  • To screen the effective materials for hair loss treatment, several natural extracts were tested using in vivo and in vitro test models. Firstly, all test materials were applicated onto the back skin of C57BL/6 mouse and then hair growth promoting effect was measured using hair growth index. As a result, Prunus mume, black bean, Brassica campestris subsp. black sesame and Rubi Fructus showed potent hair growth promoting effect, ranking as 1.5-2.0 of hair growth index. However, there were no plant extracts, which have remarkable potential of growth promotion of human hair dermal papilla cells cultured in vitro. In the experiments of 5${\alpha}$-reductase type Ⅱ inhibition assay, Prunus mume, Eriobotryae Folium showed effective potential to inhibit the activity of 5${\alpha}$-reductase type Ⅱ. To investigate the possible involvement of the effect of several plant extracts on the gene expression of growth factors in human hair dermal papilla cells, RT - PCR analyses were performed. However, there were no plant extracts, which have profound effect on the gene expression of several growth factors such as IGF-I, KGF, HGF and VEGF in the dermal papilla cells. Another tests for inhibition of microbial such as P. acne were also carried out to find whether these plant extracts have anti -microbial activities. Rubi Fructus showed anti -microbial effects on Propionibacterium acnes, which is believed as a pathogen of acne. Together, these results showed several plant extracts can be used for hair growth promotion.

The Promoting Effect of Rumex japonicas Houttuyn ethanol extract on Hair Growth

  • Objectives: This study was conducted to evaluate the hair growth-promoting effects by Rumex japonicas Houttuyn ethanol extract (RJHEE) in C57BL/6N mice and HaCaT cells. Methods: The hair growth effect was examined by topical application of RJHEE on the shaved dorsal skin of C57BL/6 mice. Six-week old mice were depilated and separated in 4 groups; CON (vehicle treatment), MXD (2% Minoxidil), and RJHEE (2% and 4%). The treatments were applied daily for 17 days. The hair growth was determined photographically and the hair density, thickness and length were identified by Folliscope. In dorsal skin tissue, the expression of hair growth-related protein was analyzed by Western blotting. In HaCaT cells, the cell proliferation and the protection against H2O2-induced cell damage by RJHEE were analyzed. Results: Our results indicate that RJHEE promote the hair growth, hair density, thickness and length. RHE activate the Wnt/𝛽-catenin signaling and induced the expression of cell survival-related proteins, such as pERK/ERK and Bcl-2/Bax. In HaCaT, RJHEE accelerated the cell proliferation and protected the H2O2-induced cell damage. Conclusions: Our results strongly suggest that RJHEE promotes hair growth by regulating the activation of Wnt/𝛽-catenin signaling and cell survival signaling and protects oxidative stress-induced hair damage. Therefore, RJHEE has a hair growth activity and can be useful for the treatment of alopecia.

물억새 뿌리로부터 Agrobacterium sp. BE516 균주의 분리 및 식물생육촉진활성 (Isolation of Agrobacterium sp. BE516 from the Root of Miscanthus sacchariflorus and Its Plant Growth Promoting Activity)

  • 비식량 바이오매스 대상작물인 억새의 생육을 촉진하는 균주를 선발하기 위하여 충북 청원군 대청호 주변에서 서식하는 물억새 뿌리로부터 64균주를 분리하였다. 분리균의 식물생육촉진 활성을 확인하기 위하여 옥신, 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase 생선능 및 기타 배양조건에 따른 생육 특성을 조사한 결과, 8균주가 선발되었으며 Agrobacterium sp. BE516 균주가 억새 줄기의 생육을 2배 이상 증가시켜 가장 활성이 뛰어난 균주로 선발되었다. Agrobacterium sp. BE516 균주는 식물호르몬 indole acetic acid를 64 ${\mu}g/mL$ 생산하고, 염 및 건조 등의 환경 스트레스 저항성 ACC deaminase 를 생산하며, $4-15^{\circ}C$, pH 4.0 및 4% NaCl 농도에서 생육하였다. 따라서, Agrobacterium sp. BE516 균주가 간척지와 같은 조건불리지역에서 비식용 바이오매스 대상작물인 억새의 생육을 촉진시키기 위한 미생물비료제 개발에 유용한 균주임을 확인하였다.

근권에 존재하는 Bacillus 속 균주들의 식물 생장 촉진 활성 특성 (Plant Growth-Promoting Activity Characteristics of Bacillus Strains in the Rhizosphere)

  • 본 연구에서는 토양 및 근권에 존재하는 Bacillus 속의 식물 생장 촉진 활성, 식물 병원성 곰팡이의 생장 억제활성, 미네랄 가용화능 및 세포 외 효소활성을 확인해 보고자 하였다. 식물 병원성 곰팡이에 대한 항진균 활성에서 DDP257은 10종의 병원성 곰팡이에서 항진균 활성이 모두 나타났다. 식물 생장 촉진 인자인 indole-3-acetic acid 생성능에서는 ANG20이 70.97 ㎍/ml로 가장 높게 나타났다. 추가적으로 1-aminocyclopropane-1-carboxylate deaminase 생성능 조사에서는 총 10종에서 생성능을 확인하였고, 질소 고정능과 siderophore 생성능 조사에서는 대부분의 분리균주에서 활성을 확인하였다. 이후 분리된 균주에 대하여 phosphate, calcite, zinc과 같은 미네랄 가용화능을 확인하였으며 세포외 효소활성에서도 대부분의 효소에서 활성이 나타났다. 특히 alkaline phosphatase, esterase (C4), acid phosphatase, naphtol-AS-BI-phosphohydrolase에서 선별된 균주 모두 유사한 활성을 보였다. 이는 Bacillus 속이 다양한 유기물과 항생물질 및 세포 외 효소를 분비함으로써 이러한 결과가 나타난 것으로 판단된다. 따라서, 본 연구 결과를 통해 토양의 환경 개선에 기여하는 균주를 활용하여 미생물 제제로써의 가능성을 제시한다.
