  • A lactic acid bacteria with ${\beta}$-gal activity was isolated from Kimchi, a traditional fermented vegetable food in Korea. The isolate was identified as a Lactococcus lactis strain and named L. lactis A2. The gene encoding ${\beta}$-gal of L. lactis A2 was cloned as a 5.8 kb PstI fragment. DNA sequencing identified the complete lacA (galactoside acetyltransferase)-lacZ (${\beta}$-galactosidase) genes together with the 3' part of upstream galT (galactose-1-phosphate uridyltransferase), and the 5'region of downstream galE (UDP-galactose-4-epimerase) genes. L. lactis A2 had the same gal/lac operon structure as in L. lactis subsp. lactis 7962. Other genes of the Leloir pathway are most likely to be located in the 5'upstream of the 5.8 kb fragment on the A2 chromosome. Sequences downstream of galE were different from those of L. lactis subsp. lactis 7962.

  • Staphylococcus aureus is a common etiologic agent for Gram-positive sepsis, and its lipoteichoic acid (LTA) may be important in causing Gram-positive bacterial septic shock. Here, we demonstrate that highly purified LTA (pLTA) isolated from Lactobacillus plantarum inhibited S. aureus LTA (aLTA)-induced TNF-${\alpha}$ production in THP-1 cells. Whereas pLTA scarcely induced TNF-${\alpha}$ production, aLTA induced excessive TNF-${\alpha}$ production. Interestingly, aLTA-induced TNF-${\alpha}$ production was inhibited by pLTA pretreatment. Compared with pLTA, aLTA induced a strong signal transduction through the MyD88, NF-${\kappa}B$, and MAP kinases. This signaling, however, was reduced by a pLTA pretreatment, and resulted in the inhibition of aLTA-induced TNF-${\alpha}$ production. Whereas dealanylated LTAs, as well as native LTAs, contributed to TNF-${\alpha}$ induction or TNF-${\alpha}$ reduction, deacylated LTAs did not, indicating that the acyl chain of LTA played an important role in the LTA-mediated immune regulation. These results suggest that pLTA may act as an antagonist for aLTA, and that an antagonistic pLTA may be a useful agent for suppressing the septic shock caused by Gram-positive bacteria.

  • From a genomic library of Lactobacillus reuteri ATCC 55739, one clone, NE347, carrying a pyrH gene encoding UMP kinase, was identified. pNE347 carried a 1.88 kb EcoRI fragment and the pyrH was located in the middle of the insert. pyrH ORF was 723 bp in size and capable of encoding UMP kinase composed of 240 amino acid residues. tsf encoding an elongation factor-Ts and frr encoding a ribosomal recycling factor were present upstream and downstream of pyrH, respectively. When introduced into E. coli KUR1244, a pyrH-negative strain, pNE347 restored the ability to grow at $42^{\circ}C$, indicating that pyrH from L. reuteri synthesized functional UMP kinase in E. coli. Northern blot experiment showed that pyrH and frr were cotranscribed as a 1.4 kb single transcript. pyrH was overexpressed in E. coli by using a pET26b(+) vector, and a major 25 kDa protein band appeared on SDS-polyacrylamide gel.

  • The peels of Carya cathayensis Sarg. (PCCS) were treated under subcritical water conditions ($130^{\circ}C$ to $280^{\circ}C$ for 0 to 120 min). The extract from PCCS included reducing sugar, proteins, and compounds with radical scavenging activity. Addressing the reducing sugar that is a resource of bioethanol, we could maximize the reducing sugar under the subcritical water ($190^{\circ}C$ for 60 min) and obtain 0.24 g/g-sample together with 9.7 units/mg-sample of radical scavenging activity. The obtained extract was estimated to correspond to 1 L of bioethanol/100 g-sample. It was therefore considered that the treatment by subcritical water could yield reducing sugar and natural compounds with radical scavenging activity.

  • This study aimed to compare the efficiency of propylene glycol (PG) and distilled water (DW) as vehicles that would allow diffusion of dye through the root canal system. Human maxillary central incisors were chosen and de-crowned. After enlarging the upper part of the root canal with a Peeso reamer, cementum covering the upper third of the root was removed. The roots were ultrasonically irrigated with 5% NaOCI to remove smear layer.(omitted)

  • Three phytosterols of rare occurrence, schleicheol 2 (1), $7{\beta}$-hydroxysitosterol (2), and $7{\alpha}$-hydroxysitosterol (3), were isolated from the n-hexane fraction of rice (Oryza sativa) bran, for the first time. Some nuclear magnetic resonance (NMR) assignments in the literatures are inaccurate. This study employed two-dimensional NMR experiments to identify exact peak assignments.

  • The abnormal accumulation of collagen is progressive and often results in impairment of liver function, i.e. liver cirrhosis. Collagen synthesis requires several posttranslational events. Prolyl 4-hydroxylase is the key enzyme in collagen synthesis that catalyzes the hydroxylation of peptide-bound proline residues to 4-hydroxyroline.(omitted)
