Studies on the Isolation of δ-Endotoxin and Plasmids in Bacillus thuringiensis (Bacillus thuringiensis의 내독소(內毒素)와 플라스미드의 분리에 관한 연구)

  • Delta-endotoxin in Bacillus thuringiensis var. finitimus, HD-1, HD-9 and HD-73 strains were isolated by NaBr, CsCl and Renografin density gradients. The purity of the toxin was about 98%. The purified o-endotoxin was analyzed by SDS-PAGE, electron microscopic observation and bioassay. According to SDS-PAGE, the molecular weight of subunits of the o-endotoxin were about 66,000 and 130,000 daltons. The shapes of the crystal toxin observed by TEM except finitimus strain were bipyramidal. When the purified endotoxin was bioassayed against tobacco horn worm, the entomocidal activities ($1{\mu}g/ml$) of HD-1 and HD-73 strains were, respectively, 60% and 100% at nine days after treated. The molecular weights of the plasmids isolated from B. thuringiensis were various from 0.5 to 120 Kb. The numbers of plasmids in HD-1, HD-9 and HD-73 strains were 12, 3 and 11, respectively, but B. thuringiensis var. finitimus strain had no plasmid.

Environmental Impact Assessments along with Construction of Residential and Commercial Complex (주거단지 건설이 하천에 미치는 생태영향평가)

  • The integrative ecological approaches of chemical assessments, physical habitat modelling, and multi-metric biological health modelling were applied to Gwanpyeong Stream within Gap-Stream watersheds to evaluate environmental impacts on the constructions of residential and commercial complex. For the analysis, the surveys conducted from 45 sites of reference streams within the Gap-Stream watershed and 3 regular sites during 2009 - 2010. Physical habitat health, based on the habitat model of Qualitative Habitat Evaluation Index(QHEI) declined from the headwaters(good - fair condition) to the downstream(poor condition). Chemical water quality, based turbidity and electric conductivity(EC), was degraded toward to the downstream, and especially showed abrupt increases, compared to the values of control streams(CS). Also, concentrations of chlorophyll-a in the downstreams were greater compared to the control stream(CS), indicating an eutrophication. Biological health conditions, based on the Index of Biological Integrity(IBI) using fish assemblages, averaged 19.3 which is judged as a fair condition by the biological criteria of the Ministry of Environment, Korea. The comparisons of model metric values in sensitive species and riffle-benthic species on the Maximum Species Richness Line(MSRL) of 45 reference streams indicated a massive disturbances in all sampling locations. Also, tolerance guild and trophic guild analyses suggest that dominances of tolerant species and omnivores were evident, indicating a biological degradation by habitat disturbances and organic matter pollutions. There was no distinct longitudinal variations of IBI model values from the headwater to the downstream in spite of slight chemical and habitat health gradients among the sampling sites. Overall, integrative ecological health(IEH) scores, based on the chemical, physical, and biological parameters, were low compared to the 45 reference streams due to physical and chemical disturbances of massive constructions of the residential and commercial complex. This stream, thus showed a tendency of typical urban streams which are disturbed in the chemical water quality, habitat structures, and biological integrity. Effective stream management plans and restoration strategies are required in this urban stream for improving integrative stream health.

No-reference Image Quality Assessment With A Gradient-induced Dictionary

  • Image distortions are typically characterized by degradations of structures. Dictionaries learned from natural images can capture the underlying structures in images, which are important for image quality assessment (IQA). This paper presents a general-purpose no-reference image quality metric using a GRadient-Induced Dictionary (GRID). A dictionary is first constructed based on gradients of natural images using K-means clustering. Then image features are extracted using the dictionary based on Euclidean-norm coding and max-pooling. A distortion classification model and several distortion-specific quality regression models are trained using the support vector machine (SVM) by combining image features with distortion types and subjective scores, respectively. To evaluate the quality of a test image, the distortion classification model is used to determine the probabilities that the image belongs to different kinds of distortions, while the regression models are used to predict the corresponding distortion-specific quality scores. Finally, an overall quality score is computed as the probability-weighted distortion-specific quality scores. The proposed metric can evaluate image quality accurately and efficiently using a small dictionary. The performance of the proposed method is verified on public image quality databases. Experimental results demonstrate that the proposed metric can generate quality scores highly consistent with human perception, and it outperforms the state-of-the-arts.

Numerical simulation of dimensional changes during sintering of tungsten carbides compacts

  • During sintering of very porous green bodies, as obtained by compaction of hard powders - such as tungsten carbide or ceramics - or by injection moulding, important shrinkage occurs. Due to heterogeneous green density field, gravity effects, friction on the support, thermal gradients, etc., this shrinkage is often non-uniform, which' may induce significant shape changes. As the ratio of compact dimension to powder size is very high, the mechanics of continuum is relevant to model such phenomena. Thus numerical techniques, such as the finite element method can be used to simulate the sintering process and predict the final shape of the sintered part. Such type of simulation has much been developed in the last decade firstly for hot isostatic pressing and next for die compaction. Finite element modelling has been recently applied to free sintering. The simulation of sintering should be based on constitutive equations describing the thermo-mechanical behaviour of the material under any state of stress and any temperature which may arise within the sintering body. These equations can be drawn either from experimental data or from micromechanical models. The experiments usually consist in free sintering and sinter-forging tests. Indeed applying more complex loading conditions at high temperature under controlled atmosphere is delicate. Micromechanical models describe the constitutive behaviour of aggregates of spheres from the deformation of two-sphere contact either by viscous flow or grain boundary diffusion. Such models are not able to describe complex microstructure and mechanisms as observed in real materials but they can give some basic information on the formulation of constitutive equations. Practically both experimental and theoretical approaches can be coupled to identify the constitutive equations. Such procedure has been performed for modelling the sintering of compacts obtained by die pressing of a mixture of tungsten carbide and cobalt powders. The constitutive behaviour of this material during sintering has been described by a linear viscous constitutive model, whose functions have been fitted from results of free sintering and sinter-forging experiments. This model has next been introduced in ABAQUS finite element code to simulate the sintering of heterogeneous green compacts of various geometries at constant temperature. Examples of simulations are shown and compared with experiments.

  • We calculated line profiles of 32 Cyg with an assumed $str{\"{o}}mgren$ sphere at orbital phases 0.06 and 0.78. The wind models with $Alfv\'{e}n$ waves show diminished line profiles compared to those of the models using power velocity laws. The reduced density of absorbers in HII region produce weak profiles, but line profiles at ${\phi}=0.06$ do not show the differences except in the red edge of absorption. At ${\phi}=0.78$ however, we could reproduce the line profiles of Alfven waves model by the power law models with two velocity gradients. It suggests that the power law model with 2 acceleration regions could reduce the errors in the theoretical line formation with no consideration of wind acceleration mechanism.nism.

  • The influence of alpha-fetoprotein (AFP) on the bone marrow (BM) natural suppressor (NS) cells of intact Ehrlich carcinoma -bearing CBA mice was studied. Bone marrow NS cells were fractionated into three fractions by isopycnic centrifugation on percoll gradients: NS1 (${\rho}$=1.080 g/ml), NS2 (${\rho}$=1.090 g/ml) and NS3 (1.100> ${\rho}$ > 1.090 g/ml). These fractions were highly different in their sensitivity to known NS cell inductors (interleukin (IL)-2, IL-3 or histamine). None of the NS fractions isolated from the intact mice spontaneously produced antiproliferative activity, however, they showed a high level of NS (antiproliferative and natural killer cell inhibitory) activity under the influence of AFP. A single injection of AFP to intact mice led to an increase of spontaneous NS activity and the inhibition of natural killer cell activity. NS activity, especially NS2, was increased in when tumor cells were subcutaneously inoculated three days after AFP injection. In the AFP-treated mice, the tumor mass at 14 days was 60% larger than that in the untreated mice. Our data confirmed that AFP is a tumor marker that can inhibit cancer immunity and plays a role in cancer pathogenesis.

Image Segmentation Using Anisotropic Diffusion Based on Diagonal Pixels (대각선 방향 픽셀에 기반한 이방성 확산을 이용한 영상 분할)

  • Anisotropic diffusion is one of the widely used techniques in the field of image segmentation. In the conventional anisotropic diffusion [1]-[6], usually 4-neighborhood directions are used: north, south, west and east, except the image diagonal directions, which results in the loss of image details and causes false contours. Existing methods for image segmentation using conventional anisotroplc diffusion can't preserve contour information, or noises with high gradients become more salient as the umber of times of the diffusion increases, resulting in over-segmentation when applied to watershed. In this paper, to overcome the shortcoming of the conventional anisotropic diffusion method, a new arusotropic diffusion method based on diagonal edges is proposed. And a method of watershed segmentation is applied to the proposed method. Experimental results show that the process time of the proposed method including diagonal edges over conventional methods can be up to 2 times faster and the Circle image quality improvement can be better up to $0.45{\sim}2.33(dB)$. Experiments also show that images are segmented very effectively without over segmentation.

  • The cyst of Scrippsiella trochoidea from the surface mud in Masan Bay was germinated in the incubator for the culture. This species was one of dinoflagellates responsible for the early spring bloom in the southern coastal water of Korea. The culture experiments were carried out under the various gradients of environmental factors to know their effects on the population growth of this dinoflagellate. With respect to the effects of environmental factors on the growth, it was proved that the maximum cell growth was occurred at 4,000 lux of light intensity, salinity $30\%0$ and temperature $20^{\circ}C$. When 0.25ml/ml of the filtrates of Skeletonema costatum culture medium and the supernatants of soil extracts were added to growth medium as organic growth stimulants, both materials enhanced the population growth. In the fatty acid composition of S. trochoidea, $C_{16:0}$ was the major component, and $C_{18:0},\;C_{18:1},\;C_{22:0}\;and\;C_{22:1}$ were a minor components.

EFFECTS OF THE RING CURRENT ON ULF WAVES IN THE MAGNETOSPHERE (지구자기구의 극초저주파수 파에 대한 RING CURRENT의 효과)

  • A three-dimensional box model has been developed to study the MHD wave coupling in the magnetosphere. In this model, the effects of the ring current are included by assuming the pressure gradients in the MHD equations. It is found that the axisymmetric ring current may play an important role in producing spectral noises in compressional waves, while field line resonances have no such disturbances. These results may explain the current observational characteristics that compressional cavity modes hardly appear in the satellite experiment, while field line resonances often occur. Our numerical resluts also suggest that any discrete spectral peaks such as the global cavity modes can hardly occur where the pressure distribution of the ring current becomes important. The continuous band of transverse waves is found to be unperturbed until the ring current becomes significantly asymmetric with respect to the dipole axis. In addition, our results in the absence of the pressure gradient are found to be consistent with the previous results from the box-like and dipole models.

  • The cross-correntropy concept can be expressed with inner products of two different probability density functions constructed by Gaussian-kernel density estimation methods. Blind algorithms based on the maximization of the cross-correntropy (MCC) and a symbol set of randomly generated N samples yield superior learning performance, but have a huge computational complexity in the update process at the aim of weight adjustment based on the MCC. In this paper, a method of reducing the computational complexity of the MCC algorithm that calculates recursively the gradient of the cross-correntropy is proposed. The proposed method has only O(N) operations per iteration while the conventional MCC algorithms that calculate its gradients by a block processing method has $O(N^2)$. In the simulation results, the proposed method shows the same learning performance while reducing its heavy calculation burden significantly.
