Test and Analysis of Thermal Ratcheting Deformation for 316L Stainless Steel Cylindrical Structure (316L 스테인리스강 원통 구조물의 열라체팅 변형 시험 및 해석)

  • In this study, the progressive inelastic deformation, so called, thermal ratchet phenomenon which can occur in high temperature structures of liquid metal reactor was simulated with thermal ratchet structural test facility and 316L stainless steel test cylinder. The thermal ratchet deformation at the reactor baffle cylinder of the liquid metal reactor can occur due to the moving temperature distribution along the axial direction as the sodium free surface moves up and down under the cyclic heat-up and cool-down transients. The ratchet deformation was measured with the laser displacement sensor and LVDTs after cooling the structural specimen which is heated up to 55$0^{\circ}C$ with steep temperature gradients along the axial direction. The temperature distribution of the test cylinder along the axial direction was measured with 28 channels of thermocouples and was used for the ratchet analysis. The thermal ratchet deformation was analyzed with the constitutive equation of nonlinear combined hardening model which was implemented as ABAQUS user subroutine and the analysis results were compared with those of the test. Thermal ratchet load was applied 9 times and the residual displacement after 9 cycles of thermal load was measured to be 1.79mm. The ratcheting deformation shapes obtained by the analysis with the combined hardening model were in reasonable agreement with those of the structural tests.

Analysis of Elastic-Plastic J Integrals for 3-Dimensional Cracks Using Finite Element Alternating Method (유한요소 교호법을 이용한 삼차원 균열의 탄소성 J 적분 해석)

  • SGBEM(Symmetric Galerkin Boundary Element Method)-FEM alternating method has been proposed by Nikishkov, Park and Atluri. In the proposed method, arbitrarily shaped three-dimensional crack problems can be solved by alternating between the crack solution in an infinite body and the finite element solution without a crack. In the previous study, the SGBEM-FEM alternating method was extended further in order to solve elastic-plastic crack problems and to obtain elastic-plastic stress fields. For the elastic-plastic analysis the algorithm developed by Nikishkov et al. is used after modification. In the algorithm, the initial stress method is used to obtain elastic-plastic stress and strain fields. In this paper, elastic-plastic J integrals for three-dimensional cracks are obtained using the method. For that purpose, accurate values of displacement gradients and stresses are necessary on an integration path. In order to improve the accuracy of stress near crack surfaces, coordinate transformation and partitioning of integration domain are used. The coordinate transformation produces a transformation Jacobian, which cancels the singularity of the integrand. Using the developed program, simple three-dimensional crack problems are solved and elastic and elastic-plastic J integrals are obtained. The obtained J integrals are compared with the values obtained using a handbook solution. It is noted that J integrals obtained from the alternating method are close to the values from the handbook.

Development of a Flow Analysis Code Using an Unstructured Grid with the Cell-Centered Method

  • A conservative finite-volume numerical method for unstructured grids with the cell-centered method has been developed for computing flow and heat transfer by combining the attractive features of the existing pressure-based procedures with the advances made in unstructured grid techniques. This method uses an integral form of governing equations for arbitrary convex polyhedra. Care is taken in the discretization and solution procedure to avoid formulations that are cell-shape-specific. A collocated variable arrangement formulation is developed, i.e. all dependent variables such as pressure and velocity are stored at cell centers. For both convective and diffusive fluxes the forms superior to both accuracy and stability are particularly adopted and formulated through a systematic study on the existing approximation ones. Gradients required for the evaluation of diffusion fluxes and for second-order-accurate convective operators are computed by using a linear reconstruction based on the divergence theorem. Momentum interpolation is used to prevent the pressure checkerboarding and a segregated solution strategy is adopted to minimize the storage requirements with the pressure-velocity coupling by the SIMPLE algorithm. An algebraic solver using iterative preconditioned conjugate gradient method is used for the solution of linearized equations. The flow analysis code (PowerCFD) developed by the present method is evaluated for its application to several 2-D structured-mesh benchmark problems using a variety of unstructured quadrilateral and triangular meshes. The present flow analysis code by using unstructured grids with the cell-centered method clearly demonstrate the same accuracy and robustness as that for a typical structured mesh.

Optimal Growth Conditions for the Two Euryhaline Cyanobacterial Clones, Anabaena sp. CB-MAL21 and CB-MAL22 Isolated from Mankyeong Estuary, Korea

  • As a result of the 2-year monthly monitoring of the phytoplankton community at 3 stations in Mankyeong Estuary, Korea, we learned that cyan bacterial species of the genus Anabaena occurred at most sampling points with huge salinity differences (0.1-32.5 psu). We isolated several clones of Anabaena spp. from the monitoring stations, and screen out two euryhaline and nitrogen-fixing Anabaena clones, CB-MAL21 and CB-MAL22. The two clones were grown under various environmental gradients such as temperature (20, 30, 35 and 40$^{\circ}C$), salinity (0, 2, 5, 15 and 30psu), and $PO_4^{3-}$-P concentration (0, 1.6, 8.0, 40 and 200 ${\mu}M$M). Growth of CB-MAL21 and CB-MAL22 was measured by daily monitoring of chlorophyll fluorescence from each experimental culture for more than three serial transfers. Both the two experimental clones did not grow at 0psu. Maximal growth rates of the two clones were markedly reduced at lower $PO_4^{3-}$-P concentrations showing negligible growth at 0 and 1.6 ${\mu}M$M. However, growth of CB-MAL21 was not affected by low $NO_3^--$ concentration in culture media, showing the nitrogen-fixing ability. Maximum biomass yields of the two clones decreased dramatically at 35 and 40$^{\circ}C$. Optimal growth conditions for the two experimental clones were determined to be 20-30$^{\circ}C$, 40 ${\mu}M$M $PO_4^{3-}$-P, and wide salinity range from 5.0 to over 30psu. Best growth of CB-MAL21 was shown at (20$^{\circ}C$-15psu), which is less saline and cooler condition than those (i.e., 30$^{\circ}C$-30psu) for the best growth of CB-MAL22. The euryhaline and nitrogen-fixing CB-MAL21 strain thus can be a candidate laboratory culture for the future cyan bacterial marine biotechnology in temperate coastal waters.

A New Double-Stranded RNA Mycovirus from Pleurotus ostreatus (ASI 2504)

  • A new virus with a dsRNA genome was isolated and characterized from the Suhan-:neutari strain (ASI 2504) of Pleurotus ostreatus, which was characterized as long and slightly bent with small caps on the stipe of fruit body. Thirty nm isometric viruses with three dsRNA segments (approximately 2.0, 1.84 and 1.82 kb in sizes) were isolated by ultracentrifugation in sucrose gradients. Western analysis of protein extracted purified viruses with anti-virus polyclonal antibody confirmed that viruses have two specific proteins (36 and 68 kDa). Computer analysis of 2.0 kb segment shows that high. sequence identity with RNA-dependent RNA polymerase (RdRp) of partitiviruses, respectively. When compared to other dsRNA mycoviruses in a phylogenetic analysis, OMDV was most related to Pleurotus ostreatus virus 1.

Differential protein expression in avian liver in response to invasion by Salmonella gallinarum

  • Salmonella gallinarum is a pathogen that is capable of causing disease in Korean native chicken. Although Salmonella gallinarum is important world-wide pathogens of poultry, little is understood of the mechanisms of pathogenesis of Salmonella gallinarum in the chicken. This study was to investigate chicken liver proteins affected by infection of Salmonella gallinarum in Korean native chicken. The differentially expressed proteins of chicken livers were identified by using 2-dimensional electro- phoresis (2D-E) and mass spectrometry (MS). We detected more than 300 protein spots on silver stained 2D gels using pH 3∼10 gradients. Three differentially expressed protein spots were analyzed by MALDI-TOF MS and MS/MS. The obtained MS and MS/MS data were searched against a protein database using the Mascot search engine. Further researches on the identified proteins can give valuable information of mechanism of pathogenesis in chicken.

  • Heterogeneous hydraulic conductivity in a flow domain is generated under the assumption that it is a random variable with a lognormal, spatially-correlated distribution. The hydraulic head and the conductivity in a groundwater flow system are represented as a stochastic process. The method of Monte Carlo Simulation (MCS) and the finite element method (FEM) are used to determine the statistics of the head and the logconductivity. The second moments of the head and the logconductivity indicate that the cross-covariance of the logconductivity with the head has characteristic distribution patterns depending on the properties of sources, boundary conditions, head gradients, and correlation scales. The negative cross-correlation outlines a weak-response zone where the flow system is weakly responding to a stress change in the flow domain. The stochastic approach has a potential to quantitatively delineate the zone of influence through computations of the cross-covariance distribution.

Fish Distribution and Compositions Along with Altitude and Longitudinal Distance from the River Mouth in the Tamjin River Basin, Korea (탐진강수계 고도 및 하구로부터 거리에 따른 어류구성 및 분포특성)

  • This study is focused on fish fauna and distributional features along with physical gradients and stream orders in the Tamjin River basin, Korea. Fish sampling and investigation were conducted for 2 years from 2009 to 2010. Total number of fish species collected were 47 species of 16 family. The group of cyprinidae was mostly abundant. Both fish abundance and biotic index were declined with increasing altitude and longitudinal distance from the river mouth. Two species of Zacco temminckii and Pungtungia herzi were prevalent in most of sampling sites and showed no altitudinal and longitudinal differences. Most other species, however, were spatially restricted to specific regional altitudes. Several species including Zacco platypus, Squalidus gracilis majimae and P. herzi were broadly distributed from river mouth to 50 km. Eight species including Z. temminckii and Rhynchocypris oxycephalus only appeared in over 10 km from the river mouth. Both Odontobutis platycephala and Pseudobagrus koreanus were classified as "upstream dewelling species" observed only in over 30 km from the river mouth. High proportion of tolerant and omnivorous species were more dominated in the downstream zone of the dam, whereas intolerant and insectivorous species were abundant in the upstream zone of the dam. The variable of stream order showed positive relationship between number of fish species and total individuals collected. Sensitive species including insectivore had a declining tendency with increasing stream order, while tolerant specie including omnivore species had relatively increased with stream order. Overall, our study suggested that fish distribution considerably depends on altitudinal gradient and longitudinal distance from the river mouth as well as physical habitat.

Development of Two-dimensional Finite Volume Model Applicable to Mixed Meshes (혼합격자의 적용이 가능한 2차원 유한체적모형의 개발)

  • In this study, 2D finite volume model, which can apply to the mixed meshes that is effective to treat the complicated topography such as a natural river, is developed. To do so, an algorithm for finding the neighbouring cell of a computational cell is introduced, and fluxes are computed using the HLLC approximate Riemann solver at each interface between a computational cell and it's neighbouring cells. Moreover, in order to numerically treat the bed slope which has important effect on the balance between flux gradients and sourte terms, different formula to compute the bed slope for rectangular and triangular mesh are applied. The developed model is applied to analyze dam-break in an experimental channel with $90^{\circ}$ bend and Malpasset dam-break in France. The two cases consist of mixed meshes and the suggested method is validated for the experimental channel and natural channel by comparison with the experimental data, field data and computed results.

Leukocyte Sequestration and Free Radical-Mediated Lung Injury in Ovine Cardiopulmonary bypass Using Membrane Oxygenator (양에서 막형 산화기를 사용하여 심폐바이패스할 경우 백혈구격리 및 자유라디칼로 중재되는 폐손상)

  • Background: Complement activation with transpulmonary leukocyte sequestration is considered a main mediator leading to ischemia-reperfusion lung(I-R) injury. We studied the role of leukocytes in the formation of I-R injury in ovine cardiopulmonary bypass(CPB) model with a membrane oxygenator. Material and Method: Five sheep were used. CPB circuitry consisted of a roller pump(American Optical Corp., Greenwich, CT, USA) and a membrane oxygenator(UNIVOX-IC, Bentley, Baxter Health Corp, Irvine, CA, USA). The CPB time was fixed at 120 min. Ten minutes after the start of CPB, total CPB was established. Thereafter a total CPB of 100 min was performed, followed by another 10 min of partial CPB. The CPB was discontinued and the animals were fully recovered. For measuring left and right atrial leukocyte counts, blood samples were taken before thoracotomy, 5 min and 109 in after the start of CPB, and 30 min and 120 min after weaning. C3a was measured before thoracotomy, 109 min after the start of CPB, and 30 min and 120 min after weaning. Plasma malondialdehyde(MDA) was checked before thoracotomy, 109 min after the start of CPB, and 30 min after weaning. One to two grams of lung tissue were taken for water content measurement before thoracotomy, 109 min after the start of CPB, and 30 min after weaning. Lung biopsy specimens were examined by light and electron microscopy. Result: Of 5 animals, 4 survived the experimental procedures. Of these, 3 animals survived on a long-term basis. No significant differences in transpulmonary gradients of leukocyte were found and no significant complement activation was expressed by C3a levels. MDA level did not show significant changes related to lung reperfusion despite an increase after the start of CPB. On both light and electron microscopic examinations, mild to moderate acute lung change was observed. Interstitial edema, leakage of erythrocytes into the alveolar space and endothelial cell swelling were the main findings. Water content of the lung showed a slight increase after the start of CPB, but there was no statistical significance. Conclusion: These findings indicate that ischemia-repersusion lung injury may not be from complement activation-leukocyte sequestration but from another source of oxygen free radicals related to CPB.
