The red seaweed, Gracilaria tikvahiae McLachlan, was cultivated in open water farms in urbanized estuaries of Long Island Sound (26-30 psu of salinity) and New York City (20-25 psu), USA in 2011. Plants were harvested monthly from summer (August, $24^{\circ}C$) to fall (November, $13^{\circ}C$) and analyzed for total nitrogen, protein, and amino acid content. On a dry matter (DM) basis, nitrogen and protein significantly increased over the harvest period until October and then plateaued. Nitrogen increased from $22{\pm}1g\;kg^{-1}$ DM in August to $39{\pm}3g\;kg^{-1}$ DM in October (p < 0.001). Protein increased from $107{\pm}13g\;kg^{-1}$ DM in August to $196{\pm}5g\;kg^{-1}$ DM in November (p < 0.001). With two exceptions, amino acid concentrations expressed on a crude protein (CP) basis were similar over the harvest period. Essential amino acids accounted for $48{\pm}1%$ of all amino acids present with lysine and methionine averaging $56{\pm}2g\;kg^{-1}$ CP and $18{\pm}1g\;kg^{-1}$ CP, respectively. Histidine was underrepresented among essential amino acids and averaged $13{\pm}1g\;kg^{-1}$ CP. Taurine ranged from 2.1 to $3.2g\;kg^{-1}$ DM. With its moderate levels of lysine, methionine and taurine, ocean farmed G. tikvahiae has the potential of overcoming many nutrient deficiencies currently associated with terrestrial plant ingredients in alternative feeds for fish and shrimp.
The bio-molecules involved in defense mechanisms can be used as efficient biomarkers for physiological changes in organisms caused by both of internal and external stress. Thus, the expression level of genes which encoding such molecules serve as critical 'early warning system' for environmental assessment as well as health diagnosis of biological organisms. In this study, Cytochrome P450, Heat shock protein 90, Ubiquitin and ${\beta}$-actin gene were isolated for the first time from a red alga Gracilaria textorii. The quantitative differential gene expression analyses of three genes, GteCYP1A, GteHsp90 and Gte-UB, were carried out in G. textorii sporophytes collected from two different localities, polluted Sujeong (Masan, Korea) and potentially unpolluted Danggeum (Daemaemuldo Is., Korea). The transcripts of all three tested genes were highly expressed in the Sujeong population. The results suggest: 1) the Sujeong site was more polluted than the Danggeum site; 2) G. textorii could be applicable to marine environment monitoring in coastal regions.

The red seaweed Gracilaria verrucosa has been used for the production of bioethanol. Pretreatment for monosaccharide production was carried out with 12% (w/v) G. verrucosa slurry and 500 mM HNO3 at 121℃ for 90 min. Enzymatic hydrolysis was performed with a mixture of commercial enzymes (Cellic C-Tec 2 and Celluclast 1.5 L; 16 U/ml) at 50℃ and 150 rpm for 48 h. G. verrucosa was composed of 66.9% carbohydrates. In this study, 61.0 g/L monosaccharides were obtained from 120.0 g dw/l G. verrucosa. The fermentation inhibitors such as hydroxymethylfurfural (HMF), levulinic acid, and formic acid were produced during pretreatment. Activated carbon was used to remove HMF. Wild-type and adaptively evolved Saccharomyces cerevisiae, Candida lusitaniae, and Kluyveromyces marxianus were used for fermentation to evaluate ethanol production.

Two different molecular markers, the plastid rbcL and mitochondrial cox1 genes, were used to define the taxonomic position of the northwest Pacific Ocean species currently named Gracilaria chorda. We analyzed both genes (1,222 bp for rbcL and 1,245 bp for cox1) from 18 specimens collected in Korea, Japan, and China. Phylogenetic reconstruction revealed that this organism should be classified in the genus Gracilariopsis, rather than in the Gracilaria. Thus, Gracilariopsis chorda (Holmes) Ohmi is the legitimate name for Gracilaria chorda Holmes. Within the species, the sequences differed by 8 bp (0.7%) in rbcL and 5 bp (0.4%) in cox1. Six haplotypes of cox1 tended to be geographically organized. Gp. chorda is characterized by coarse, elongate terete axes, short filiform branchlets usually at irregular intervals, an abrupt transition in cell size from medulla to cortex, cystocarps without tubular nutritive cells connecting the gonimoblast to the upper pericarp, and relatively large gonimoblast cells of the cystocarp in the specimens collected from Wando in southern Korea.
Gracilaria verrucosa and Gelidium amansii collected from Wando, Yeosoo and Namhae districts Gracilaria sp. imported from Manila, and Ecklonia cava from Cheju island were investigated to raise the extraction yield of agar or alginic acid. The results are summarized as follows. In agar extraction from domestic seaweeds, Gelidium sp. showed 8-10% yield increase by gamma-irradiation whereas Gracilaria sp. exhibited no effect. In Manila seaweed, gamma-irradiation (1.5 Mrad) caused 25-30% increase in agar extraction and the properties of agar sample by these methods were acceptable. In alginic acid extraction from Ecklonia sp., gamma-irradiation (0.3 Mrad) showed 6% yield increase.

The red alga Gracilaria vermiculophylla, a species native to the waters of Korea and Japan, has invaded marine coastal areas of Europe and the Americas, thriving in conditions that differ from those of its native habitat. In recent years, G. vermiculophylla has been discovered in the Long Island Sound (LIS) estuary growing alongside the native congener Gracilaria tikvahiae. The goal of this study was to determine whether the two strains of G. vermiculophylla from different regions of the world have evolved genetic differences (i.e., ecotypic differentiation) or if the physiological performance of the strains simply reflects phenotypic plasticity. Two strains of G. vermiculophylla (isolated in Korea and LIS) and a strain of the LIS native G. tikvahiae were grown for four weeks under temperatures ranging from 20 to $34^{\circ}C$ using a temperature gradient table (all other environmental conditions were kept constant). At the end of each week, wet weight of each sample was recorded, and thalli were reduced to the original stocking density of $1gL^{-1}$ (excess biomass was preserved for tissue carbon and nitrogen analysis). Generally, the growth rates of Korean G. vermiculophylla > LIS G. vermiculophylla > G. tikvahiae. After one week of growth G. tikvahiae grew 9.1, 12.0, 9.4, and 0.2% $d^{-1}$, at temperatures of 20, 24, 29, and $34^{\circ}C$, respectively, while G. vermiculophylla (LIS) grew 6.6, 6.2, 5.7, and 3.6% $d^{-1}$. G. vermiculophylla (Korea) grew 15.4, 22.9, 23.2, and 10.1% $d^{-1}$, much higher than the two strains currently inhabiting the LIS. On average, the LIS G. vermiculophylla strain contained 4-5% DW N, while the Korean strain and G. tikvahiae had more modest levels of 2-3% N DW. However, tissue N content declined as temperature increased in LIS and Korean G. vermiculophylla. The non-native haplotype may have evolved genetic differences resulting in lower growth capacity while concentrating significantly more nitrogen, giving the non-native a competitive advantage.
Imported seaweeds Gracilaria sp. and Gelidium sp. from the Philippines and Gracilaria sp. from Brazil were subjected to the examination of yield and quality of agar according to various pretreatment conditions. As pretreatment condition for alkali-treated Gracilaria sp. from the Philippines, $0.05{\sim}0.1%$ concentration of sulfuric acid was proper to obtain the higher yield of agar for which a reverse proportionality was shown between the acid concentration and the strength of extraction condition. Highest yield from Gelidium sp. from the Philippines was obtained by pretreating with 5% sodium hydroxide at $90^{\circ}C$ for one hour, followed by 0.1% sulfuric acid treatment. Gracilaria sp. from Brazil gave the highest yield of agar by pretreatments with 1% sodium hydroxide followed by 0.05% sulfuric acid, both at room temperature for one hour. Acid treatment of alkali-treated Gracilaria sp. from the Philippines brought about the decrease of total nitrogen, total sulfur and crude ash, the increase of jelly strength and no marked change in gelation point and gelation ability. In general, acid-treatment conditions to give rise to high yield of agar improved the quality of product.
Hainan is the second largest and southernmost island of China, and it harbors many species of the family Gracilariaceae. In this study, we conducted molecular and morphological analyses of gracilarian specimens collected from Hainan Island, to clarify their taxonomic identity and phylogenetic relationships. Five species of Gracilariaceae in Hainan were determined through maximum likelihood and Bayesian analysis of rbcL sequence data. Based on the phylogenetic trees and morphology, Hainan specimens were identified as three species of Gracilaria, one species of Gracilariopsis, and one species of Hydropuntia, namely, G. salicornia, G. tenuistipitata, Gracilaria sp., Gp. bailiniae and H. edulis. This is the first report of the molecular phylogeny of Gracilariaceae on Hainan Island, and it helps to clarify the taxonomy and distribution of gracilarian species in the Asia-Pacific region.

Indonesia is well-known as biggest producer of seaweed especially for Eucheuma and Gracilaria and also has huge potential resources and capability to develop seaweed cultivation and product. There are several provinces which have potential resources and have been contributing on seaweed production. The next challenge about seaweed production is using integrated system on brackishwater and marine aquaculture. Furthermore, about 2,000,000 ton of potential seaweed production is not explored yet. This article also tries to figure out some related aspects which are technical, economical and forecasting aspect. There is a disease which named "ice-ice" is one of the main problem and giving a new challenge in developing of problem solving for seaweed cultivation method. Economical parameters are also main important key to find out the feasibility of seaweed cultivation industry. In addition, the seaweed cultivation and production in Indonesia also have potential performance on biofuel resources as a part for solving the world problem on energy demand.
