발정주기에 따른 흰쥐 난관섬모세포에 관한 전자현미경적 및 면역조직화학적 연구 (Electron Microscopic and Immunohistological Studies on the Ciliated Cells of Rat Oviduct During Estrous Cycle)

  • The goal of the present study is to investigate the precise variation of tubulin substances in the cytoplasm of oviductal ciliated cells and the morphological changes in cytoplasmic organelles of ciliated cells for ciliogenesis by estrous cycles. The animals used in this study were female rat (Sprague Dawley strain), weighing approximately 200 gm. The ampulla oviducts of these animals (at each of estrous cycle) were rapidly excised. At each stage of estrous cycle, the tissues were used for immunocytochemical study and other were used for electron microscopical study. All specimens were observed by the light and electron microscope. The results obtained are as follows: 1. In the ciliated cells at proestrus, Golgi complex showed $5{\sim}7$ stacked cisternae with numerous saccules and vacuoles. Large amount of fibrous granules were located near the Golgi complex. But at metestrus and diestrus, few fibrous granules were seen. 2. A moderate number of rough endoplasmic reticulum and polyribosomes were scattered in the cytoplasm of ciliated cells at proestrus, but were decrease in number at metestrus and diestrus. 3. At proestrus and estrus, there were a large amount of vesicles in the apical cytoplasm of ciliated cells. 4. Numerous mitochondria were located in the apical cytoplasm at proestrus and estrus, but only a few at metestrus and diestrus. 5. At proestrus and estrus, tubulin substances showed strong reactions in the cytoplasm but weak reactions at metestrus and diestrus. It is suggested consequently that the ciliated cells of the rat oviducts showed no morphological changes of cilia but the ultrastructural organelles of the cells were changed in its shape and location during the entire estrous cycle.

  • 방게의 혈구는 투명혈구, 소과립혈구, 그리고 대과립혈구로 동정되었다. 투명혈구의 모양은 세포질에 비해 큰 핵을 가지며, 세포질에는 난형의 전자밀도가 약한 과립과 소포 및 소포체들이 관찰된다. 소과립혈구는 투명혈구보다 더 작은 핵을 가지며, 세포질에는 잘 발달된 소포체, 골지체, 그리고 작은 원형의 전자밀도가 강한 과립을 갖는다. 대과립혈구는 소과립들이 서로 융합해서 된 전자밀도가 강한 대과립들을 갖는다. 방게의 혈구는 투명혈구로 부터 대과립혈구로 발달한다. 즉 리소솜효소를 갖는 투명혈구의 과립들은 핵막으로 부터 작은 소포들을 형성하며, 이 소포들이 골지체를 통과하면서 전자밀도가 강한 물질로 채워지고, 이 작은 소과립들을 융합하면서 대과립이 된다. 모든 혈구에는 글리코겐 입자들이 산재한다. 대과립혈구들 중 과립의 소멸현상으로 생각되는 이형과립들이 존재한다.

한국산 꿩의 고환수출소관의 구조 (The structure of the ductuli efferentes in the Korean native pheasant(Phasianus colchicus korpowi))

  • The morphology of the ductuli efferentes of the Korean native pheasants were observed in order to obtain a basic data for further studying reproductive physiology and other male genital organs. The mature (14-16 months after hatching) male pheasants were used in this study. The specimens from pheasants were collected on a monthly basis. The general morphological changes of the ductuli efferentes were observed with hematoxylineosin stain, and semithin section by light microscope. The ultrastructural changes of the ductuli efferentes were investigated with ultrathin section by transmission electron microscope. The results obtained are summarized as follows : 1. During the breeding season, the average height of ductuli efferentes epithelium was $23.45{\pm}2.34{\mu}m$ and was largely decreased by $17.85{\pm}2.01{\mu}m$ during the non-breeding season. The thickeness of interstitial tissue was comparatively increased during the non-breeding season. 2. During the breeding season, the epithelial cells of ductuli efferentes were well developed. During the non-breeding season, epithelial layer and lumen of ductuli efferentes, were markedley reduced compared with those of breeding season. 3. Morphological changes of the ductuli efferentes underwent periodic changes paralleling to the spermatogenic cycle. 4. At least two different cell types were identified in the epithelium of ductuli efferentes, namely non-ciliated and ciliated cells. 5. The ciliated cells possess many vesicles, slightly smaller than those of the non-ciliated cells. 6. The ciliated cells contained numerous mitochondria, smooth and rough endoplasmic reticulum, Golgi complex, lysosome, and oval nuclei. The non-ciliated cells had a irregular nuclei and a cytoplasm containing few organelles. 7. During the breeding season, a number of vesicles, rough and smooth endoplasmic reticulum, Golgi complex, and mitochondria were distinctively showed in the epithelial cells but in the non-breeding season only a few observed.

한국 서해산 암컷 덕대 Pampus echinogaster (Basilewsky)의 난형성과정 중 난모세포 발달과 난모세포 내에서의 난황형성과정에 관한 미세구조적 연구 (Ultrastructural Studies on Oocyte Development and Vitellogenesis in Oocytes During Oogenesis in Female Pampus echinogaster in western Korea)

  • The ultrastructural study on oocyt development and the process of vitellogensis in the oocytes during oogenesis in female Pampus echinogaster were investigated by electron microscope observations. In the previtellogenic phase, in particular, several intermitochondrial cements appear in the cytoplasms of the chromatin nucleleolus oocyte and perinuclear oocyte. The number of intermitochondrial cements are associated with the multiplication of the number of mitochondria in the early developmental stage. In the early vitellogenic phase, the Golgi complex in the cytoplasm of the yolk vesicle oocyte is involved in the formation of yolk vesicles containing carbohydrate yolks. At this time, many pinocytotic vesicles containing yolk precursors (exogenous substances) by pinocytosis are observed in the cytoplasm near the region of initial formation of the zona pellucida. In the late vitellogenic phase, two morphological different bodies, which formed by the modified mitochondria, appeared remarkably in the yolked oocytes. The one is the multivesicular bodies and another is yolk precursors. The multivesicular bodies were transformed into the primary yolk globules, while yolk precursors were connected with exogeneous pinocytotic vesicles near the zona pellucida. After the pinocytotic vesicles were taken into yolk precursors, the yolk precursors were transformed into the primary yolk globules. Thereafter, primary yolk globules mixed with each other, eventually, they developed into secondary and tertiary yolk globules. In this study, vitellogenesis of this species occurred by way of endogenous autosynthesis and exogenous heteogenesis. Vitellogenesis occurred through the processes of endogeneous autosynthesis, involving the combined activity of the Golgi complex, mitochondria and multivesicular bodies formed by modified mitochondria. However, the process of heterosynthesis involved pinocytotic incorporation of extraovarian precursors (such as vitellogenin in the liver) into the zona pellucida (by way of granulosa cells and thecal cells) of vitellogenic oocytes.

Fine Structural Observations on Spermatogenesis of the Goldeye Rockfish, Sebastes thompsoni (Teleostei: Scorpaenidae)

  • Fine structural changes of the germ cells during spermatogenesis in the goldeye rockfish, Sebastes thompson; were examined by means of the transmission and scanning electron microscopy. A spermatogonium has a large nucleus with a single nucleolus in the interphase. Primary spermatocytes are characterized by the formation of chromatin clumps and presence of the synaptonemal complex in the nucleus. The nucleoplasm of secondary spermatocytes is more condensed than that of primary spermatocytes, and the cytoplasm contains numerous mitochondria, endoplasmic reticulum and Golgi complex. The nuclei of spermatids in metamorphosis show sickle-like shape as the nucleoplasm becomes more condensed. In the cytoplasm of spermatids, the proacrosomal granules are not found at all. A spermatozoon consists of head, neck and tail. The acrosome is absent in the head. Four to five cytoplasmic collars are observed in the posterior portion of the head of spermatozoon. The well developed axonemal lateral fins are observed in the flagellum of spermatozoon.

  • These experiments were performed to know ultrastructural changes of the cumulus expansion in virot. SEM:In expanded oocyte-cumulus complex, the cell surface are characterized by the presence of many evaginations:they are relatively short and round shape. The mucous extracellular material were deposited between cumulus cells. TEM:In compact cumulus cells, golgi apparatus and rough endoplasmic reticulum developed. In expanded cumulus cells, rough endoplasmic reticulum decreased and the smooth endoplasmic reticulum increased. Also, there were numbers of mitochondria. Extracellular mucous material which is presumed to be hyaluronic acid appears when cumulus cell were expanded. In expanded cumulus cell, numbers of smooth endoplasmic reticulum help cumulus cell to develop in steroidogenic cell.

Testosterone과 Cyclic AMP-theophyllin이 흰쥐 부정소의 미세구조 및 몇 가지 효소활성에 미치는 영향 (Effect of Testoeterone and Cyclic AMP-theophylline on Ultrastructure and Several Enzymes' Activities in Rat Epididymis)

  • Testosterone과 cyclic AMP(cAMP)가 부정소에 미치는 영향을 조사하기 위하여 흰쥐의 복강내로 testosterone 및 cAMP를 투여하여 acid phosphatase(ACP),alkaline phosphatase(ALP), lactate dehydrogenase(LDH)의 활성도를 측정하였으며 상피세포내 미세구조의 변화를 전자현미경으로 관찰하였다. 그 결과 거세한 실험군에서 ACP활성도는 거세 후 3일째까지 감소하기 시작하였으며 ALP활성도는 5일째,LDH활성도는 7일째부터 각각 감소하였다. 또한 거세 후 14일째 부터 testosterone을 7일간 투여한 경우 ACP와 ALP의 활성도는 5일간 계속 투여하였을 때,LDH의 활성도는 7일간 계속 투여하였을 때만 각각 유의성있게 증가하였다. cAMP 및 cAMP-theophyllin을 투여한 경우 LDH 및 ACP의 활성도가 증가하였으나 그 증가폭은 큰 의미가 없었다. ALP의 활성도는 두 가지의 경우에서 모두 증가를 나타내었다. 미세구조를 관찰한 결과는 거세한 실험군에서 골지체가 점차 변형되어 층상구조가 없어지며 미토콘드리아도 파괴되었고 상당수 존재하던 부동모 (stereocilia)도 파괴되었다. 이 경우 testosterone투여시 파괴되었던 고리지체, 미토콘드리아 및 부동모가 회복되었고 cAMP 및 cAMP-theophyllin 을 투여하였을 때는 골지체 및 여러 세포기관들이 대조군과 같은 수준으로 회복되었으나 부동모는 거세군과 같은 수준으로 관찰되었다. 이상의 결과는 부정소액에서 정자성숙시 나타나는 효소활성도 증가와 유사하게 나타났는데, 이것은 정자↔부정소액↔부정소 상피세포 와 같은 상호작용을 통하여 정자성숙이 이루어지며 이는 상피세포에서 합성된 물질이 부정소내 강으로 방출된다는 것을 의미하며 cAMP를 처리하였을 경우 효소활성도 증가 및 여러 세포소기관의 회복 등 testosterone과 유사한 작용을 나타내는 것은 cAMP가 웅성호르몬 작용시 화확적으로 전달자로 작용할 수 있다는 가능성을 나타내는 것으로 생각된다.

  • 담수산과 해수에 적응된 guppy 아가미 상피내 점액세포의 미세구조를 주사 및 투과 전자현미경으로 관찰하고, 점액질 조성의 변화를 조직화학적으로 분석하여 다음의 결과를 얻었다. 점액세포는 아가미궁에서 돌출되어 형성된 일차층판상피에 주로 위치하였다. 미성숙점액 세포에는 과립형질내세망과 Golgi 복합체가 잘 발달되어 있었으며, 성숙 점액세포는 거의 점액과립들로 가득 채워져 있었다. 담수산 guppy 아가미 상피내 점액세포의 점액질은 중성점액질과, 산성점액질인 sialomucin 및 sulfomucin을 다량 포함하고 있었으며, 바닷물에 적응시킨 경우에서는 sialomucin과 sulfomucin이 약간 감소하였다. 바닷물에 적응된 guppy의 아가미 상피내 점액세포의 수는 담수산과 비교하여 3배이상 감소하였다. 이는 서식환경을 해수로 옮긴 결과, 변화된 삼투적 스트레스에 대응한 결과일 뿐 아니라 담수보다는 높은 염분농도인 해수환경의 감소된 감염기회와도 연관된 결과라고 생각된다.

  • The posterior hypothalamus of the hibernating greater horseshoe bats (Rhinolophus ferrumequinum korai Kuroda) were observed with an electron microscope. The posterior hypothalamus is known to be closely related to the reflex responses activated by cold, and the following observations were obtained in the cellular type of nerve cells: there are three types of neurons in the posterior hypothalamus. 1. The first type of neuron was the largest, ovoid or conical in shape, the nucleus was elliptic and the nuclear envelope had many deep invaginations. The cell organelles were well developed, in particular there was an abundance of variously shaped mitochondria, and the Golgi complex and the polysomes were observed in the cytoplasm. 2. The second type of neuron was moderate in size, ovoid or elliptic in shape, the nucleus was located nearer to the plasma membrane and the nuclear envelope had. a few invaginations. The cytoplasm was rich in amount compared with that of the third type of neuron, and the cell organelles, especially the rough endoplasmic reticulum were well developed. Also lipofuscin pigments were observed. 3. The third type of neuron was the smallest in size and round in shape. The nucleus and the nucleolus were observed in the central portion of the cell body and the nuclear envelope had a few invaginations. The cytoplasm was small compared with those of the first and second types, but the rough endoplasmic reticulum, the mitechondria and the polysomes were relatively well developed. The cytoplasm was characterized by the presence of membrane-bound small bodies with a single membrane containing a fine particular substance around the rough endoplasmic reticulum and the Golgi complexes.

  • The oval shaped pericardial cells are clustered along the lateral sides of the heart and irregularly connected with the heart. The cells are bounded by a basement membrane. The basement membranes of the connected two peicardial cells are irregularly linked each other there-fore funnels are formed. The multiple invaginations of the cell membrane are observed and septate junctions develope at the part of enterance of the cell membrane. The coated pits are appeared in the inner side of the invaginated cell membrane. The coated vesicles, tubular and spherical shaped vesicle, Golgi complex containing high electron densed material in the cisternae and mitochondria are observed in the cytoplasm and lysosomes are remarkably well developed. The whirled membrane structures in the multiformed complex bounded by single membrane are linked with low electron densed granules and spherical shaped small granules having high electron density with $0.03{\mu}m$ in diameter are located between the whirled membrane in a row and gradually secretes the granules and then they produced the multilamellar body. The lysosomal regions of cytoplasm of pericardial cell are appeared negative reaction to the acid phosphatase and according to the results of the electrophoresis, lipoproteins having acid phosphatase activity are contained. The axon is contacted with the pericardial cells.
