Effect of Ganoderma lucidum Mycelial Fractions on the Functional Recovery of Primary Cultured Hepatocytes. (영지 균사체 분획이 일차배양 간세포 기능회복에 미치는 영향)

  • The cultured mycelia of Ganoderma lucidum were extracted and the extract was separated into six fractions by organic solvent fractionation. The antihepatotoxic activity of all the fractions was evaluated by measuring activities of glutamic pyruvic transaminase (GPT) and glutamic oxaloacetic transaminase (GOT). Among the fractions tested, the high-polarity fractions such as aqueous and n-butanol fractions significantly reduced activities of GPT and GOT in CCl4- and galactosamine-intoxicated rat primary hepatocytes. When intracellular synthetic activities were measured by pulsing the rate primary cultured hepatocytes with [3H]-uridine and [3H]-leucine, activities of DNA, RNA and protein. When direct toxicities of the fractions were measured against human hepatoma(SK-Hep-1), the non-polarity fractions such as n-butanol and ethyl acetate fractions showed potent direct cytotoxicities even at the concentration of 1 $\mu\textrm{g}$/ml. These data showed that Ganoderma lucidum has hepatoprotective and hepatotoxic recovery principles in its mycelia.

Antioxidant and Liver-protective Effects of Caesalpinia sappan (소목 추출물의 항산화 및 간보호작용)

  • The heartwood of Caeralpinia sappan L. (Leguminosae) has been used to activate blood flow, remove blood stasis, reduce swelling and relieve pain in Korean folk medicine. In this study. the antioxidant effects of this crude drug and its hepatoprotective activity on CCl$_4$-induced liver injury in rats were evaluated. The levels of antioxidant enzymes, superoxide dismutase (SOD) and catalase (CAT) reduced by CCl$_4$treatment, were recovered by this crude drug. It is suggested that Caesalpinia sappan L. have antioxidant effect. The increased levels of glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT) by CCl$_4$were also recovered by treatment with this crude drug. These results can be attributed to the agent's antioxidant and membrane - stabilizing actions.

  • The concentration of amino acids, total nitrogen, trehalose, lipids and the activities of respiratory, acid$\cdot$alkaline phosphatase, glutamic oxalozcetic transaminase and glutamic pyruvic transaminase during larval stage in Pine leaf gall midge, Thecodiplosis janensis Uchi. et Inouye were measured using Paper chromatographic method, micro-Kjeldahl method, Thin layer chromatographic method, Warburg's manometric method, Bessey-Lowry method and Reitman-Frankel method, respectively. Healthy specimens )yore chosen as samples of each larval stages; alrva in gall and larva in soil. Amino acids present in the alcoholic extracts were alanine, glutamic acid, glycine, histidine, methionine, proline, threonine, tryptophan and valine. The total nitrogen concentration reached to 31.348mg/g during the larva in gall and the larval stage in soil of the value was decreased to 29.027mg/g. The hemolymph sugar, trehalose value for larva in soil was about two times of the value for larva in gall. Total lipid, phospholipid,monoacylglycerol, triacylglycerol, sterol, free fatty acid and ester cholesterol were identified at larval stages in gall and soil. Triacylglycerol concentration reached high level in contrast with other lipid contents during larvae in gall and larva in soil. Free fatty acid, sterol except decreased lipids during larval stage in soil. Endogenous respiration, succinate of respiratory activities decreased at larval stage in soil compare with larva in gall. The activities of acid phosphatase decreased larval stage in soil but the activities of alkaline phosphatase increased remarkably. The activities of glutamic oxaloacetic transaminase and glutamic pyruvic transaminase reached high level of the larva in gall.

Effect of Lxeris Sonchifolia H. on Serum Lipid Metabolism in Hyperlipidemic Rats (고들빼기의 급여가 고지혈증 흰쥐의 지질대사에 미치는 영향)

  • The effect of lxeris sonchifolia H.(godulbaegi) on lipid metabolism were studied in S. D. rats. Twenty four rats were divided into four groups and fed diets containing 1 % cholesterol, 0.25% sodium cholate, 10% coconut oil and 5% lard(control group)f for 3 weeks. An experimental diet group was added with 5% leaf(PL group) or root powder(PR group) of godulbaegi. The lipid components and enzyme activites of serum and liver were assayed. The concentrations of total cholesterol, triglyceride and phospholipid were significantly lower in the PL and PR groups than in the control group. The levels of HDL-cholesterol were not significant but showed somewhat higher values in the PL and PR groups compaired with the control. The activities of glutamic oxaloacetic transminase(GPT) and glutamic pyruvic transaminase(GPT) were significantly lower in the PL and PR groups compaired with the control. In view of these results Godulbaegi is expected to be an effective food in preventing and improving the hyperlipidemic state.

Protective Effects of the Phenolic Compounds from the Leaves of Hedera rhombea on Hepatic Injury (송악의 페놀성 물질의 간 보호효과)

  • Hedera rhombea (HR) has been used for treatments of hemorrage, chronic catarrh, jaundice, lithisis and convulsion. This study was done to isolate active compounds that have protective effect on liver damage. BuOH and EtOAc fractions of HR recovered serum glutamic pyruvic transaminase (GPT), glutamic oxaloacetic transaminase (GOT) and ${\gamma}-glutamyltranspeptidase$ (${\gamma}-GTP$) activities in CCl4 treated rats. We isolated 7 phenolic compounds from BuOH and EtOAc fractions, which were identified as 3-caffeoyl quinic acid, 3,4-di-O-caffeoyl quinic acid, 3,5-di-O-caffeoyl quinic acid, 4,5-di-O-caffeoyl quinic acid, caffeic acid, methyl 3,4-di-O-caffeoyl quinic acid and methyl 3,5-di-O-caffeoyl quinic acid by chemical and spectral analysis. These compounds reduced significantly serum GOT and GPT elevated by CCl4 treatment in rats, and 3-caffeoyl quinic acid, 3,5-di-O-caffeoyl quinic acid and caffeic acid also showed mild inhibitory activity against human immunodeficiency virus.

The Effect of HP4060 on the Brain Neurotransmitter and Hot Flushes in the Female Sprague-Dawley Rat

  • In this study, the effects of HP4060, a pomegranate extract, on Sprague-Dawley (SD) female rats were investigated. SD rats used in the experiment were divided into 3 groups: a control group, 100 mg HP4060/kg rat powder fed group, and 25 mg HP4060/kg rat liquid fed group. After 20 days of administration, the changes of the brain neurotransmitters were measured. The data showed that the concentration of the serotonin and the norepinephrine were increased, whereas that of the epinephrine was decreased in HP4060 administered groups. In addition, the improving effect of HP4060 on depression symptom of menopause women were shown by increased immobility time of the SD rates in a separate experiment. The uterus weight of HP4060 fed groups were also shown to be increased. In order to monitor toxic effect of HP4060, glutamic-oxaloacetic transaminase (GOT) and glutamic-pyruvic transaminase (GPT) levels were measured, and the results showed that no significant difference in GOT and GPT levels among experimental groups implying no significant toxic side effects of HP4060. According to these results, it seems clear that HP4060 may improve symptoms of hot flush and depression caused by menopause without significant level of toxic effects.

A clinical study on the changes of serum enzymes after open heart operation under the cardiopulmonary bypases (개심술후 혈청효소 변화에 관한 연구)

  • The changes of serum creatine phosphokinase [CPK], serum lactic dehydrogenase [LDH], serum glutamic oxaloacetic transaminase [GOT] and serum glutamic pyruvic transaminase [GPT] were studied in a total of 82 cases who underwent open heart operation under the cardiopulmonary bypass at the Department of Thoracic and Cardiovascular Surgery, Chungnam National University Hospital. The results were as follows 1. The average values of CPK and COT after the operation were reached to the maximal values on the first postoperative day, which were returned to the normal range until the fifth postoperative day. The average values of LDH and CPT after the operation were reached to the maximal values on the first and second postoperative day respectively, which were significantly increased until the seventh postoperative day. 2. In the relationship of the serum enzymes and duration of the extracorporeal circulation, the values on the group over 90 minute of the extracorporeal circulation were more increased than on the group below 90 minute of the extracorporeal circulation. 3. In the relationship of the changes of the serum enzymes and congenital heart diseases and acquired heart diseases, there were no significantly differences in the values between the two groups.

Effect of Quartz Porphyry Supplemented Diet on Clinical Chemical Parameters of Serum in White Java Sparrow (맥반석 첨가 식이가 백문조의 혈액 임상화학지수에 미치는 영향)

  • Effect of Quartz porphyry(QP) on clinical chemical parameters of serum in male white java sparrow by feeding the basal diet(Control group) containing 3.0% QP(QP group) for 14 days was studied. Body weight gain, feed intake, the weight of the gizzard and the heart were not significantly different in the both groups. However, the weights of kidney and liver in the QP group were higher than in the control group. The contents of triglyceride, cholesterol, total protein, glucose, creatinine, blood urea nitrogen, uric acid and albumin, and the activities of glutamic oxaloacetic transaminase(GOT) and glutamic pyruvic transaminase(GPT) in serum were not significantly different in the both groups. The activity of alkaline phosphatase(ALP) in serum was higher in the QP group than in the control group.

  • Water extracts of 21 traditional Korean herbal drugs were prepared, and a dose range of 100 mg/kg to 400 mg/kg was administered orally into mice once a day for five days. Changes of serum enzyme activities of glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), blood urea nitrogen (BUN), alkaline phosphatase, body weight changes and histo pathological examination of various organs were investigated. Water extract of Ephedra Herba caused severe body weight loss at a dose of 100 mg/kg and death from a dose level of 200 mg/kg by oral administration. Angelica koreanae Radix and Anthrisci Radix showed a slight body weight loss and damages to liver and kidney.

A Study for the Correlations between Obesity and Blood Parameters at the Worksite (직장인에서 비만과 혈액지표와의 상관성에 관한 연구)

  • Obesity, a state of having excessive body fat enough to attack one's health, is most blamed for causing chronical degenerative illness such as hypertension, diabetes mellitus as well as arteriosclerosis. According to many studies, the program for the weight control was the most demanded at the worksites. This study was designed to investigate the correlation between the degree of obesity and blood parameters of the employees at the worksites. The results of periodic medical examination for 571 employees(male: 418, female: 153) were analyzed; Age, BMI, systolic blood pressure(SBP), diastolic blood pressure(DBP), hemoglobin(Hb), fasting blood sugar(FBS), total-cholesterol(TC), serum glutamic oxaloacetic transaminase (sGOT), serum glutamic pyruvic transaminase(sGPT) and $gamma$-glutamyl transpeptidase($gamma$-GTP). Pearson correlation coefficient was used to find out the correlation between BMI and each blood parameters. In the case of male, 27.3% of the subjects were over-weight. The subject's age and their blood levels of uric acid, FBS, sGOT and sGPT did not show any significant difference regardless of BMI's, while SBP, DBP, Hb, TC and $gamma$-GTP levels of the overweight group were significantly higher than those of normal group. In the case of female, 9.2% of the subjects were over-weight. This study shows that the implementation of weight control program is required at the worksites
