Betaine is one of the major water-soluble components in Lycii Fructus. In the present study the effect of repeated betaine treatment on the hepatotoxicity and the cytochrome P-4 50-dependent enzyme system was examined in adult female rats. Administrations of betaine (100 or 1,000mg/kg/day, ip) to rats repeatedly for 4 or 9 days did not evoke hepatotoxic response as determined by increases in glutamic pyruvic transaminase(GPT) and glutamic oxaloacetic transaminase(GOT) activities measured 24 hours following the final dose of betaine. The activities of aminopyrine N-demethylase, p-nitroanisole O-demethylase and p-nitrophenol hydroxylase as well as the contents of cytochrome P-450 were determined in hepatic microsomes of rats treated with betaine(1,000mg/kg/day, ip) for 4 or 9 days. Repeated treatment of rats with betaine for a period of 4 days induced a marginal decrease in the contents of cytochrome P-450, but did not influence the activities of p-nitrophenol hydroxylase, p-nitroanisole O-demethylase, or aminopyrine N-demethylase. Extension of the betaine treatment to 9 consecutive days failed to alter the parameters for hepatic drug metabolizing activity determined in the present study. Since repeated large doses of betaine were demonstrated to be tolerated by rats without showing any toxicity or changes in drug metabolizing enzyme activities in the liver, this compound appears to be relatively safe to animals upon long-term ingestion.
Objectives : Positive effects of Ginseng has great research attentions such as anticancer, anti-diabetic, antiaging, liver, immune function, CNS, etc. In this study, we investigated Hydroponic-cultured Ginseng Folium fermented byBacillus subtilisto establish fermentation conditions for enhancing functionality. Methods : Ginseng Folium were cultivated hydroponic-cultured and were extracted with methanol. We inoculateBacillus subtilisfor fermentation by adding to 0%, 3% and 5% sugar respectively and checked antioxidant activities, total phenolic content and total saponin content in 2 days intervals during 11 days. The antioxidant activities were studied by the 1,1-diphenyl-2-picryl hydrazyl(DPPH) radical, 2, 2'-Azino-bis(3-ethylbenzothiazoline-6 sulfonic acid) diammonium salt(ABTS) radical scavenging assay and Reducing power assay. The reducing power was resulted in optical density of 0.7312 with 3% sugar after 9 days of fermentation. and the concentration at 200 ㎍/㎖. Total phenol content was 36.92㎎/g with 3% sugar after 9 days of fermentation, in which crude saponin content wasn't changed, and ginsenoside content such as Rg3, Re and Rb was increased. Activities of GOT and GPT concentration were decreased in rat. Conclusions : This study suggests that hydroponic-cultured Ginseng Folium fermented byBacillus subtilisin 9 days showed significant efficacy of hepato-protection as well as antioxidant compared to the others. In addition, it shows not only improved value but also utilized hydroponic-cultured Ginseng Folium by fermentation.
Effects of sosihotangganogyong on the activity of serum transferase and hepatic lipid peroxide in mice with $CCl_4$ solution were determined. The activity of ALT showed a high value in the mice with $CCl_4$, however in the mice with sosihotang and sosihotangganogyong, these values showed a tendency to rapid recovery compared with those of the mice with $CCl_4$ only and the activity of ALT in the group of sosihotang and sosihotangganogyong showed a low values compared with $CCl_4$ only group on the 21 days after treatment. On the 21 days after treatment, the activities of glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT) and lactic dehydrogenase (LDH) in the mice with sosihotang and sosihotangganogyog were similar to those of control group, however these values of $CCl_4$ only group showed a high values compared with those of other groups. The value of hepatic lipid peroxide in the mice with sosihotang and sosihotangganogyong showed a tendency to rapid decrease and recovery compared with those of $CCl_4$ only group and on the 21 days after treatment, this value showed a similar to those of control group. The activity of serum transferase and the value of hepatic lipid peroxide in the mice with sosihotngganogyong showed a tendency to decrease compared with those of sosihotang group, however these values showed a no significantly deference. Results from this study indicated that the sosihotagganogyong can effectively improve the recovery of liver function in mice with $CCl_4$.
Anti-diabetic effect of various fractions of Cordyceps militaris (CM), CCCA (crude cordycepin containing adenosine), CMESS (ethanol soluble supernatant), and cordycepin were evaluated in streptozotocin (STZ) induced diabetic mice, CMESS showed potent inhibitory activity of 34.7% in starch-loaded mice (2 g/kg) while acarbose as a positive standard exhibited 37.8% of inhibition rate. After 3 days administration (50 mg/kg), cordycepin (0.2 mg/kg), and acarbose (10 mg/kg) dramatically reduced blood glucose level (inhibition ratio: 46.9%, 48.4% and 37.5% respectively). CCCA that has high contents of cordycepin (0.656 mg/4 mg) did not influence on reducing blood glucose level. The proliferation of splenocytes and peritoneal macrophages derived from STZ-induced diabetic mice administered samples were evaluated out by addition of mitogens to see the stability of the usage of these herbal medicines. Proliferation of T-lymphocyte was significantly decreased; while NO production was increased more than two fold to STZ control in the cordycepin-administered group. Changes of serum enzyme levels of glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT) were also evaluated. Cordycepin administered group was appeared to acarbose. We conclude that CMESS and cordycepin may be useful tools in the control of blood glucose level in diabetes and promising new drug as an anti-hyperglycemic agent without defects of immune responses and other side effects.
Antitumor activity of Korean mistletoe extract (KM-110) and European commercial mistletoe preparation (Helixor) was investigated. KM-110 showed the cytotoxic effect that it is high for various tumor cell lines and normal splenocytes in comparison with Helixor. Administration of two mistletoe extracts ($100{\mu}g$) to mice did not show any significant changes on the level of glutamic-oxaloacetic transaminase (GOT), glutamic-pyruvate transaminase (GTP), blood creatinine (CRE) and blood urea nitrogen (BUN) in sera. The culture supernatant of macrophages stimulated with KM-110 inhibited effectively tumor growth whereas Helixor had little effect. Administration of KM-110 or Helixor resulted in a effective inhibition of lung metastasis after the i.v. inoculation of colon 26-M3.1 lung carcinoma, B16-BL6 melanoma and L5178Y-ML25 lymphomas. In all cases, the mice treated with KM-110 showed more effective anti-tumor metastatic activity than the mice of Helixor. These results suggest that Korean mistletoe extracts, KM-110 might be used as an alternative methods having antitumor activity like European mistletoe preparation, Helixor.

Clinical-chemical traits are commonly used biomarkers to examine the health status of individuals. There is an appreciable range of normal variation in most clinical-chemical traits and the determining factors of this variation have been relatively uninvestigated in chickens. The aim of this study was to estimate the genetic parameters (i.e., heritability, genetic correlation) for 8 clinical-chemical traits (glucose, total protein, creatinine, high-density lipoprotein cholesterol, total cholesterol, glutamic oxaloacetic transaminase, glutamic pyruvic transaminase and amylase) in an $F_1$ intercross established by purebred breeding among the 5 lines of Korean native chickens. Phenotypic data were collected from approximately 600 $F_1$ animals. The genetic parameters for the clinical-chemical traits estimated by a mixed animal model using the restricted maximum likelihood method were presented. Estimated heritabilities ranged from 8.9% (glucose) to 39.6% (high-density lipoprotein cholesterol). Interestingly, both the sign and the size of the genetic and phenotypic correlations were largely different between the same several pair of clinical-chemical traits. The findings in this study will provide useful information to address issues in both quantitative trait locus study and genetic management in Korean native chickens.
Effect of Quartz porphyty(QP) on functional and morphological changes of the liver and kidney was studied in male common finch and white java sparrow fed with the basal diet(Control group) or experimental diet containing 3.0% QP(QP group) for 14 days. There was not significantly different morphological change of the liver upon light microscopic examination in common finch and white java sparrow between control group and QP group. Morphological change of renal tissue upon light microscopic examination in common finch and white java sparrow was not also significantly different between control group and QP group. The concentrations of serum creatinine, blood urea nitrogen, and nric acid as renal functional parameters of common finch and white java sparrow were not significantly different in the both groups. The activity of glutamic oxaloacetic transaminase(GOT) as hepatic functional parameter in common finch was significantly higher in the QP group($\rho$<0.05), whereas the activity of glutamic pyruvic transaminase(GPT) as hepatic functional parameter in common finch was not significantly different in the both groups. The activities of GOP and GPT in white java sparrow were not significantly different in the both groups. The morphologic findings and functional parameters of the liver and kidney observed in common finch and white java sparrow fed with 3.0% QP diet showed evidence of slightly liver damage accompanied with increased release of enzyme and fatty change of the hepatocytes in common finch, suggested that the tissues in some animals can be damaged by feeding a diet supplemented with 3.0% QP.
Angelicae gigantis Radix aqua-acupuncture solution (AGRAS) and Angelicae gigantis Radix water-extracted solution (AGRWS) were prepared and tested for their organ toxicities and chemopreventive potentials. The organ-toxicity of AGRAS to male ICR mice was studied by the measurements of glutamic oxaloacetic transaminase (GOT), glutamic pyruvate transaminase (GPT), lactate dehydrogenase (LDH) and alkaline phosphatase (ALP-s) activities after injection of AGRAS for 7 days. The activities of GOT GPT and LDH were decreased, but the activity of ALP-s was not changed with AGRAS. When AGRAS was administered once daily for 10 days before the tumor implantation, AGRAS exerted antitumor activity by inhibiting the growth of Ehrich ascites tumor cells (EATC) in viva. The inductions of quinone reductase (QR), glutathione (GSH) and glutathione S-transferase (GST) and inhibition of polyamine metabolism were tested for the chemopreventive potentials of AGRAS and AGRWS. AGRAS was potent inducer of QR activity in murine hepatoma Hepalclc7 cells. In cultured rat Ac2F cells, AGRAS was also significantly induced QR activity GSH levels were increased about 1.3 fold with AGRAS. In addition the activity of GST was increased about 2.5 fold with AGRAS at the concentration of $0.1{\;}{\times}{\;}$. The effects of AGRAS and AGRWS were tested on the growth of Acanthamoeba castellanii. Proliferation of Acanthamoeba castellanii in a broth medium was inhibited by AGRAS and AGRWS at the concentration of $1{\;}{\times}{\;}and{\;}5{\;}{\times}{\;}$, respectively: These results suggest that AGRAS has chemopreventive potential by inducing QR activity increasing GSH and GST levels and inhibition of polyamine metabolism.
Objectives: The purpose of this study was to investigate anti-obesity effect of Scutellariae Radix extract combined with metformin. Methods: Male C57BL/6 mice, 4 weeks of age, were used to set high-fat diet induced obesity model. They were grouped NOR (normal control), HFD (high fat diet control), MET (metformin, 100 mg/kg/day), MH2 (metformin 50 mg/kg/day+Scutellariae Radix 200 mg/kg/day), and HG4 (Scutellariae Radix 400 mg/kg/day). MET, MH2, and HG4 were orally administered for 10 weeks. Body weight was measured every week. Fasting blood glucose, triglyceride, total cholesterol (TC), high density lipoprotein, glutamic oxaloacetic transaminase (GOT), and glutamic-pyruvic transaminase (GPT) were measured before sacrifice. Oral glucose tolerance test (OGTT) was also performed. Organ weight and internal fat weight were measured after sacrifice. Results: MH2 group showed a reduction of body weight when compared with HFD group. MH2 group showed stable blood level control which was calculated areas under the curves by OGTT. TC, GOT, GPT level, internal fat, and organ weight in MH2 group reduced. Conclusions: The combined treatment of Scutellariae Radix and Metformin has impact on treating obesity. Further studies are needed to evaluate the effect of Metformin and herbal medicine combination therapy.
To elucidate the protective effects of Lactobacillus acidophilus-fermented milk against cadmium toxicity, the effects of administration of L acidophilus-fermented milk on hematological values and histopathological changes in cadmium-treated rats were investigated. The experimental rats were divided into 2 groups that were consisted of the one group administered with cadmium alone, and the other group administered with cadmium mixed with the fermented milk. Each group was orally administered with different doses of cadmium such as $1.7{\mu}g/g$ bw/day, $3.4{\mu}g/g$ bw/day, $6.8{\mu}g/g$ bw/day, and $13.6{\mu}g/g$ bw/day, respectively, for 1 to 8 weeks. Hematological values and enzyme activities, histopathological changes of kidney tissues were examined for the experimental groups. The values of RBC, WBC, and Hb in the groups administered with cadmium mixed with the fermented milk showed no significant differences to those of the groups administered with cadmium alone, but Hct showed significant reducing values. The activities of glutamic oxaloacetic transaminase(GOT) and glutamic pyruvic transaminase (GPT) in serum were significantly reduced than those of the groups administered with cadmium alone, at the low dose of cadmium treated groups. But alkaline phophatase(ALP) and lactate dehydorgenase(LDH) were significantly reduced at the high dose of cadmium treated groups. In histopathological study, a severe acute tubular necrosis of the convoluted tubules and distalation of tubules were showed in the groups administered with cadmium alone, but the kidney tissues of the groups administered with cadmium mixed with the fermented milk were similar to those of the normal group. In conclusion, the above results would suggest that L acidophilus-fermented milk has reducing effects on cadmium toxicity, at the low dose of cadmium administration.
