Biochemical mechanism of Jijang-Kimch on anti-obesity effect and blood lipid metabolism in obesity model rats (비만모델 랫드에서 지장김치의 혈액 지질대사, 항비만 효능 및 메카니즘)

  • This study investigated action mechanism and biological effect of Jijang-kimch, including its anti-obesity effect and blood lipid-decreasing effect in a high-fat diet-induced obese model animals. There were four treatment groups: CD (chow diet as normal control), HFD (high fat diet as obesity control), HFDCK (HFD plus commercial kimchi extracts), and HFDJK (HFD plus Jijang-kimchi extract). Kimchi extracts were orally administered for 28 days. Body weight, liver, and adipose tissue weight declined in HFDJK compared to those in HFDCK(p<0.05). Blood triglyceride, total cholesterol, low density lipoprotein-cholesterol (LDL-C), and glucose level decreased in CD, HFDJK, and HFDCK compared to those in HFD(p<0.05). Those in HFDJK were lower than those in HFDCK(p<0.05). Sizes of liver and adipose cells increased in HFD, HFDCK, and HFDJ that those in CD(p<0.05). Those in HFDJK were greatly decreased than those in HFDCK(p<0.05). These results indicate that ingestion of Jijang-kimchi in obese model animals has anti-obesity effect by lowering blood lipid and glucose levels and decreasing adipocyte size compared to that of commercial-kimchi.

Inhibitory Effect of Jeju Tea Extracts and Vanadate on Postprandial Hyperglycemia and Hypertension, and In Vitro Study (제주산 녹차, 발효차 추출물과 바나듐의 식후 혈당강하 및 항고혈압 기능에 미치는 In Vitro 효과)

  • The inhibitory effect on α-glucosidase, a marker of postprandial hyperglycemia, and angiotensinconverting enzyme (ACE), a marker of hypertension, was analyzed using non-fermented green tea and three different types of fermented tea, which are popular beverages in modern life. Green tea was mixed with trace amounts of vanadate (50 ㎍/mL), which has insulin-mimetic effects, to investigate the synergistic effect of vanadate on the inhibition of α-glucosidase. The concentration of epigallocatechin gallate (EGCG) and caffeine was also checked. The extracts of green tea and fermented teas showed clear inhibition on α-glucosidase, which caused a decrease in the postprandial glucose levels. The inhibitory effect was most prominent in the 20% fermented tea. Trace amounts of vanadate (50 ㎍/mL)-mixed green tea extract had twice the inhibitory effect on α-glucosidase than the pure tea extract. All teas showed inhibitory effects on ACE. Among those, the effect was most prominent in green tea, which had higher concentrations of EGCG. In contrast, the postprandial glucose-lowering effect and ACE inhibition of the fermented teas, which have a lower level of EGCG, was attributed to some other different functional substances.

The Improvement of Chaga Mushroom (Inonotus Obliquus) Extract Supplementation on the Blood Glucose and Cellular DNA Damage in Streptozotocin-Induced Diabetic Rats (Streptozotocin으로 유발한 당뇨쥐에 있어서 차가버섯(Inonotus Obliquus)의 혈당 및 DNA 손상 개선효과)

  • Mushrooms have become a largely untapped source of powerful new pharmaceutical products that poses anti-inflammatory, and antimutagenic, and antioxidant activities. The antioxidant effects of the mushroom may be partly explained by protecting cellular components against free radical. The aim of this study was to investigate the protective effect of chaga mushroom against diabetes, via the mitigation of oxidative stress and reduction of blood glucose, in streptozotocin-induced diabetic rats. Rats were rendered diabetic by intravenous administration of STZ through tail at a dose of 50 mg/kg. Animals were allocated into four groups with 8 rats each. The control and diabetic control group were fed with standard rat feed. The other diabeic groups, the low chaga extract group and the high chaga extract group were fed ad libitum using 0.5 g/kg and 5 g/kg of chaga mushroom extract, respectively, for 4 weeks. The blood glucose levels in the two chaga extract groups showed a tendency to decrease but did not reach statistical significance after the supplementation. Leukocyte DNA damage, expressed as tail length, was found to be significantly lower in the high chaga extract group than in the diabetic control group (p > 0.05). Plasma level of total radical-trapping antioxidant potential (TRAP) was tend to be higher in the high chaga extract group compared with the diabetic control group. Erythrocyte antioxidant enzyme activities of two groups did not differ. Although we did not obtain beneficial effect on lowering blood glucose levels in the STZ-induced diabetic rats, this results suggest that the chaga mushroom extracts may initially act on protecting endogenous DNA damage in the short-term experiment.

Effects of cultivated wild ginseng pharmacopuncture on lowering lipid and oxidative capacity in biochemical and molecular biological study in obese rats (산양산삼약침의 지질강하 및 항산화효과에 대한 생화학적 및 분자생물학적 검토)

  • Objectives : This study was carried out to identified the effects of distilled cultivated wild ginseng pharmacopuncture to the obesity. Methods : Cultivated wild ginseng pharmacopuncture was administered on the points of chung-wan(CV12), $Ch'{\breve{o}}nch'u$(ST25), and Chok-samni(ST36) on lowering lipid and oxidative capacity in biochemical and molecular biological aspects were investigated in obese rats fed with high fat diet. Results : 1. The contents of plasma ${\beta}-lipoprotein$ showed a tendency to decrease in the pharmacopuncture groups compared to the control group. In the pharmacopuncture groups, the values of ST25 and ST36 pharmacopuncture groups showed lower value. 2. The contents of plasma free fatty acids showed a tendency to decrease in pharmacopuncture groups compared to the control group. However, in the pharmacopuncture groups, the values were not significantly different. 3. Plasma triglyceride and glucose showed lower value in the ST25 pharmacopuncture groups compared with the other groups. 4. The activity of AST showed a tendency to decrease in the pharmacopuncture groups. However, the activity of ALT was not significantly different in all the treatment groups. 5. Plasma total cholesterol and LDL-cholesterol showed lower value in the ST25 and ST36 pharmacopuncture groups and HDL-cholesterol showed higher value in the CV12 pharmacopuncture groups than that of the other treatment groups. 6. Liver total cholesterol values didn't show significant difference in all the treatment groups, and triglyceride showed lower value in the pharmacopuncture groups. 7. The contents of plasma TEARS showed lower value in the ST25 pharmacopuncture group and contents of liver TBARS showed a tendency to decrease in the pharmacopuncture groups. However these values didn't show significant difference in the pharmaco puncture groups. 8. Liver super oxide dismutase activity showed higher value in the ST25 and ST36 pharmacopuncture groups, and the value of liver glutathione peroxidase and catalase activity showed a tendency to increase in the pharmacopuncture groups. However, these values showed no significant difference in the pharmacopuncture groups. 9. Expression of apo-B and E mRNA in liver cells was lower in the ST25 pharmacopuncture group than that of the other treatment groups. However, expression of $TNF-{\alpha}$ and leptin mRNA in adipose cell showed no difference among all the treatment groups. 10. ST25 pharmacopuncture group showed a good histological character of liver. It showed similar to that of normal group. However other treatment groups and control group showed slight vasodilation and slight fat accumulation. Conclusion : These results indicate that distilled cultivated wild ginseng pharmacopuncture suppressed adipose tissue mass and lipid peroxidation, and increased antioxidant capacity.

  • The lipid lowering and antioxidant effects of Gansoo(BL18), Pungji(GB20) and Eumnungcheun(SP9) acupuncture in rats fed high fat diet were analyzed in biochemical and molecular biological aspects. The results obtained from this study are as follows : 1. In the body weight reduction, all acupuncture groups showed a high reduction compared to those of control group and in acupuncture groups, Gansoo(BL18) and Pungji(GB20) acupuncture groups showed a high reduction. 2. The concentration of plasma triglyceride, total cholesterol and LDL-cholesterol with acupuncture groups showed a little decrease and in acupuncture groups, Gansoo(BL18) and Pungji(GB20) groups showed a low values compared to those of other acupuncture groups. However, the tendency of HDL-cholesterol concentration showed no significant different. 3. The concentration of plasma ${\beta}-lipoprotein$ and free fatty acids showed a lowest values in the Gansoo(BL18) and Pungji(GB20) acupuncture groups and the glucose concentration showed to decrease in all treated acupuncture groups. 4. The concentration of liver total cholesterol and triglyceride in Gansoo(BL18) and Pungji(GB20) acupuncture groups showed a lower values than those of control group. 5. In all the acupuncture groups, the plasma glutamic oxaloacetic transaminase(GOT) activity showed a little decrease. In the glutamic pyruvate activity(GPT), Gansoo(BL18) and Pungji(GB20) acupuncture groups showed a lower values than those of control groups. However the values of eumneungcheun acupuncture only group showed no significant difference to those of control group. 6. The plasma and liver Thiobabituric acid reactive substance (TBARS) concentration in Gansoo(BL18) and Pungji(GB20) acupuncture groups were a lower than those of control group. However the values of eumneungcheun acupuncture group showed no significant difference to control group. 7. The superoxide dismutase (SOD) activity in Gansoo(BL18) acupuncture group and Glutathione peroxidase (GSH-Px) activity in Gansoo(BL18) and Pungji(GB20) acupuncture groups showed a high values. The catalase (CAT) activity in all the acupuncture groups showed a higher values than those of control group. 8. In acupuncture groups, DNA expression of Apo-B and Apo-E showed a tendency to decrease, however DNA expression of leptin showed no significant difference in all treatment groups. DNA expression of $TNF-{\alpha}$ showed a increase in acupuncture groups. These results indicate that Gansoo(BL18) and Pungji(GB20) (especially Gansoo(BL18)) acupuncture affect the lipid metabolism and showed possibility of lowering adipose tissue mass and lipid peroxidation.

Effects of ${\alpha}-linolenic$, eicosapentaenoic and docosahexaenoic acids administration on lowering of triacylglycerol level in the hepatic and serum of rats (n-3계 지방산 투여가 성장기 흰쥐의 간장 및 혈청 Triacylglycerol 농도에 미치는 영향)

  • We studied the difference effects of dietary ${\alpha}-linolenic\;acid\;({\alpha}-LA,\;18:3\;n-3)$, eicosapentaenoic acid (EPA, 20:5 n-3) and docosahexaenoic acid (DHA, 22:6 n-3) on the lowering of triacylglycerol in the liver and serum on lipid metabolism in rats. Rats were fed semipurified diets containing 10% fat with constant polyunsaturated/monounsaturated/saturated fatty acids (1:1:1) and n-6/n-3 ratio (1:2). EPA (98%) and DHA (98%) were added in diets as the ethyl esters. The concentration of liver triacylglycerol was significantly lower in rats fed both EPA and DHA than in those fed ${\alpha}-LA$. The concentration of liver phospholipid was significantly higher in rats fed DHA than in those fed ${\alpha}-LA$ and EPA. Both EPA and DHA reduced serum triacylglycerol concentration compared with ${\alpha}-LA$, but this effect was more pronounced in the EPA diet. The activity of phophatidate phosphohydrolase in the liver microsome was significantly lower in rats fed both EPA and DHA than in those fed ${\alpha}-LA$. but, there was no significant difference on the activities of diacylglycerol acyltransferase among the three groups. The concentration of liver triacylglycerol were correlated with changes in the microsomal phosphatidate phosphohydrolase activity (r=0.84). Hepatic NADPH generating enzyme, glucose-6-phosphate dehydrogenase was more effective to reduce the activity in rats fed both EPA and DHA than in those fed ${\alpha}-LA$. In conclusion, EPA or DHA reduced the hepatic triacylglycerol concentration by inhibiting microsomal phosphatidate phosphohydrolase, thereby inhibiting synthesis of triacylglycerol in the liver.

Comparison of blood biochemical characteristics and dietary intake by sex in gastric cancer patients over 40 years in Korea based on 7th (2016-2018) Korea National Health and Nutrition Examination Survey: a cross-sectional study (국내 40세 이상 위암 환자의 성별에 따른 혈액생화학적 특성, 영양섭취비교: 제7기(2016-2018) 국민건강영양조사 자료를 이용한 횡단연구)

  • Objectives: The purpose of this study was to compare the sex-associated differences in the dietary intake of gastric cancer patients in Korea. Methods: Data from the Korea National Health and Nutrition Examination Survey (KNHANES) the 7th (2016-2018) were analyzed in the present study. The subjects included 122 gastric cancer patients aged over 40 years (75 male, 47 female). General characteristics (age, marital status, household income, education, food security, comorbidities, alcohol drinking, and smoking), anthropometric characteristics (height, weight, body mass index, and blood pressure), blood biochemical characteristics [fasting plasma glucose (FPG), blood urea nitrogen (BUN), creatinine, triglyceride, total cholesterol, and HDL-cholesterol)], and quantity and quality of dietary intake were compared between male and female participants. Results: Males had higher rates of having a spouse, prevalence of hypertension, alcohol drinking, and smoking than females. The proportion of males with a normal range of FPG, BUN, and HDL-cholesterol was lower than that in females. The total cholesterol levels above the normal range were higher in females than in males. We also found that females had a higher percentage of intakes below the estimated energy requirement (EER) and intakes below the estimated average requirement (EAR) for carbohydrates, niacin, phosphorus, and iron than males. The index of nutritional quality (INQ) for phosphorus and folate, nutrient adequacy ratio (NAR) for vitamin C, thiamine, niacin, folate, calcium, and phosphorus, and the mean adequacy ratio (MAR) were lower in females than males. Conclusions: In Korean gastric cancer patients, management of comorbidities such as diabetes and hypertriglyceridemia, lowering FPG, and raising HDL-cholesterol level management is required for males, whereas management of lowering total cholesterol and raising hematocrit is required for females. The quantitative and qualitative nutritional intakes were poor in gastric cancer patients, especially in females, who had a lower nutritional intake than males. We suggest that nutritional interventions are needed to improve the overall nutritional intake in both male and female gastric cancer patients. In particular, we propose that support is urgently needed for females whose nutritional intake is lower than that of males. In addition, family, social, and national support for nutritional management of female gastric cancer patients is highly necessary.

Effect of Bulnesia sarmienti Ethanol Extract on Plasma Levels of Glucose and Lipid in Streptozotocin Induced Diabetic Rats (Bulnesia sarmienti 에탄올 추출물이 streptozotocin 유발 당뇨 흰쥐의 혈당 및 혈장 지질수준에 미치는 영향)

  • This study examined the effects of Bulnesia sarmienti ethanol extract on lipid metabolism in streptozotocin (STZ)-induced diabetic rats. Sprague Dawley rats were induced with diabetes mellitus by injecting STZ (45 mg/kg) into the tail vein, and were then divided into 5 groups: normal, STZ-control, and three experimental diabetic groups. The rats were fed a basal diet along with B. sarmienti ethanol extract for 8 weeks. According to the results, after 8 weeks of consuming the experimental diets, the body weights and food efficiency ratios of the B. sarmienti ethanol extract-fed groups were not different from those of the control group. However, the concentrations of plasma total-cholesterol, LDL-cholesterol, and triglycerides were significantly lower in the B. sarmienti ethanol extract groups as compared to those of the control group. Although a blood glucose control effect was not observed after the eight weeks of B. sarmienti ethanol extract treatment in this type-l diabetes model, it was discovered that the experimental diets were highly effective at lowering levels of serum lipids, including triglycerides, total-cholesterol, and LDL-cholesterol.

Red Ginseng Alters Lipid Metabolism through AMPK Activation in Liver and Adipose Tissues of High-Fat Diet-Fed Mice (홍삼 복합 추출물의 AMPK 활성화를 통한 고지혈증 개선)

  • Hyperlipidemia is known as a glucose and lipid metabolism-related disorder that is increasing in incidence in modern society. Red ginseng (RG) is a natural herb candidate with a positive effect on regulation of cholesterol and lipids. To observe the effects of RG on regulation of lipids, cholesterol, glucose, and oxidative stress, we examined the in vitro and in vivo effects of Chamdahan RG on differentiated 3T3-L1 adipocytes and high-fat diet-fed mice. RG ($50{\mu}g/mL$) significantly inhibited lipid synthesis in 3T3-L1 cells. In addition, a low concentration of RG (880 mg/kg/d) resulted in the lowest total blood cholesterol level. Moreover, high density lipoprotein-cholesterol quantity increased in RG-treated groups, consequently lowering the cardiovascular risk factor and atherosclerosis index. Moreover, RG increased activity of AMP-activated protein kinase, as a regulator of lipid and cholesterol synthesis, in adipose and liver tissues. Cumulatively, this paper suggests that RG has a positive effect on reducing the amounts of cholesterol and lipids and may be a good candidate for treating hyperlipidemia.

  • These experiment were conducted to investigate the cultural condition of the lipase production by Rhizopus japonicus. The results obtained were as follows: 1. Soybean meal and ammonium sulfate were the most effective in the lipase production as organic and inorganic nitrogen sources, respectively. 2. The lipase production was strongly inhibited, when added as carbon sources xylose, glucose, fructose, galactose, maltose, soluble starch, and dextrin causing the lowering of pH of the medium during culture. Sucrose did not inhibit the lipase production, but not caused any effect when added. 3. $K_2HPO_4$ as phosphate salt and $MgSO_4{\cdot}7H_2O$ as magnesium salt were the most effective in the lipase production. 4. The addition of olive oil, soybean oil, and coconut oil respectively increased the enzyme production and especially 1% olive oil increased it by 50%. 5. The enzyme production increased slightly on the addition of yeast extract to $0.05{\sim}0.07%$. 6. The optimum composition of the medium for the lipase production by Rhizopus japonicus was in the composition of soybean meal 2%; $K_2HPO_4{\cdot}$ 0.5%; $(NH_4)_2SO_4$ 0.1%; $MgSO_4\;7H_2O$ 0.05%; yeast extract 0.05%; olive oil 1%. The maximum production of the lipase was attained by the incubation far 48hrs under the optimum incubation condition.
