Adventitious Rooting of Cherry Dwarfing Rootstock 'Gisela 5' in Semi-hardwood Cuttings (체리 왜성대목 '기셀라 5' 반경지 삽목 발근 특성)

  • The first commercial planting of sweet cherry in Korea is thought to be done in early 1920s. The planting area of sweet cherry in Korea is about 500 ha in 2017. Although sweet cherry is considered a minor fruit in Korea, it is one of premium fruits in market and popular among children and women, especially among pregnant women. The import of sweet cherry fruits has increased dramatically in recent years. Seedlings of Prunus lannesiana or clones of 'Colt' (P. avium × P. pseudocerasus) were usually used as sweet cherry rootstocks in Korea. During recent decades new dwarfing rootstocks for cherry such as 'Gisela' series were developed in Germany. Among them, several 'Gisela' series have been mainly used in international nurseries. In this study, we investigated optimum rooting condition of a dwarfing rootstock 'Gisela 5' in summer season cuttings. Among eight soil conditions tested, saprolite + commercial nursery soil (1:1) and saprolite + vermiculite (1:1) showed higher rooting percentage than others, showing 93.6% and 88.9%, respectively. More than 95% of 'Gisela 5' rootstocks produce roots when it was treated with IBA for several seconds just before cuttings, irrespective of concentrations (500 to 2000 mg·L-1).

  • Based on the results in this study, here we propose a systematic micropropagation process for 'Gisela 5' that is one of the important dwarfing cherry rootstocks. When the apical tips detached from newly developed shoot in spring season were cultured on the half strength MS media with 0.5 mg/L IBA and 0.5 ~ 1.0 mg/L BA, the cultures scored the highest acquisition rate at 90% for normal shoot with vigorous growth and without hyperhydricity. As next step, the young shoots maintained in vitro well multiplied on the full strength MS medium supplemented with 0.5 mg/L IBA and 0.5 mg/L BA, in which multiplication rate was approximately nine-fold. Given the half strength MS medium containing 2.0 mg/L IBA, each transplanted shoot further developed robust roots. Finally, the plantlets were easily acclimatized in the compost consisted of vermiculite, perlite, and peatmoss in the proportion of 1:1:1. We expect that the results are useful for cherry cultivation and its rootstock production.

Throat Carriage Rate and Antimicrobial Resistance of Streptococcus pyogenes In Rural Children in Argentina

  • Objectives: The aim of this study was to determine the prevalence of asymptomatic carriers of group A ${\beta}-hemolytic$ streptococci (GAS) in children living in a rural community and to investigate the association between episodes of acute pharyngitis and carrier status. Methods: Throat swabs were collected from September to November 2013 among children 5-13 years of age from a rural community (Maria Ignacia-Vela, Argentina). The phenotypic characterization of isolates was performed by conventional tests. Antimicrobial susceptibility was assayed for penicillin, tetracycline, chloramphenicol, erythromycin, and clindamycin (disk diffusion). The minimum inhibitory concentration was determined for penicillin, cefotaxime, tetracycline, and erythromycin. Results: The carriage of ${\beta}-hemolytic$ streptococci was detected in 18.1% of participants, with Streptococcus pyogenes in 18 participants followed by S. dysgalactiae ssp. equisimilis in 5. The highest proportion of GAS was found in 8 to 10-year-old children. No significant association between the number of episodes of acute pharyngitis suffered in the last year and the carrier state was detected (p>0.05). Tetracycline resistance (55.5%) and macrolide-resistant phenotypes (11.1%) were observed. Resistance to penicillin, cefotaxime, or chloramphenicol was not expressed in any streptococcal isolate. Conclusions: The present study demonstrated significant throat carriage of GAS and the presence of group C streptococci (S. dysgalactiae ssp. equisimilis) in an Argentinian rural population. These results point out the need for continuous surveillance of GAS and non-GAS carriage as well as of antimicrobial resistance in highly susceptible populations, such as school-aged rural children. An extended surveillance program including school-aged children from different cities should be considered to estimate the prevalence of GAS carriage in Argentina.

Increased Expression of P2RY2, CD248 and EphB1 in Gastric Cancers from Chilean Patients

  • Background: Gastric cancer (GC) ranks as one of the major causes of mortality due to cancer worldwide. In Chile, it is currently the leading cause of cancer death. Identification of novel molecular markers that may help to improve disease diagnosis at early stages is imperative. Materials and Methods: Using whole-genome DNA microarrays we determined differential mRNA levels in fresh human GC samples compared to adjacent healthy mucosa from the same patients. Genes significantly overexpressed in GC were validated by RT-PCR in a group of 14 GC cases. Results: The genes CD248, NSD1, RAB17, ABCG8, Ephb1 and P2RY2 were detected as the top overexpressed in GC biopsies. P2RY2, Ephb1 and CD248 showed the best sensitivity for GC detection with values of 92.9%, 85.7% and 64.3% (p<0.05), respectively. Specificity was 85.7%, 71.4% and 71.4% (p<0.05), for each respectively.

Growth and Fruit Quality of 'Lapins' Sweet Cherry (Prunus avium) in Forest (체리나무(Prunus avium) '라핀'의 생육 및 과실 품질)

  • 체리나무는 앵두나무아과(Drupaceae) 벚나무속(Prunus) 앵두나무아속에 속하며 유럽 중남부와 소아시아가 원산지이다. 특히 온대지역에 잘 적응하는 체리나무는 재배기간 중 비가 적은 건조한 기후에 적합하다. 최근 국내에서 체리 소비가 급격히 증가함에 따라 체리 수입량[6,454톤('11)→15,855톤('19)]이 꾸준히 증가하고 있는 추세이다. 또한 국내에서 체리나무 재배에 대한 관심이 높아지면서 농경지 및 산지에 식재되어 생산량[289톤('09)→656톤('14)] 및 재배면적[60ha('08)→144ha('14)]도 꾸준히 증가하고 있다. 그러나 체리나무의 산지재배관련 연구는 전무한 실정으로 고품질 체리의 안정적 생산이 가능한 재배기술 개발이 시급히 요구되고 있다. 따라서 본 연구는 산지재배 체리나무의 생육, 결실 및 과실품질에 대한 기초자료를 확보하기 위하여 실시하였다. 공시품종은 '라핀(Lapins)'으로 대목은 '기셀라 6(Gisela 6)'을 사용한 3년과 4년생 체리나무를 조사하였다. 체리나무의 생육특성 조사 결과, 수고 및 근원경은 3년생 체리나무 234.0±23.0 cm, 5.1±0.7 cm로 나타났고 4년생 체리나무는 346.0±27.0 cm, 7.3±0.9 cm으로 조사되었다. 본당 착과수는 3년생 체리나무의 경우 과실이 착과되지 않았고 4년생 체리나무는 563.6±42.9개로 조사되었다. 4년생 체리나무의 과실특성을 조사한 결과, 입중 8.1±0.9 g, 당도 16.3 °Brix, 경도 9.8 N, 산도 1.3%로 조사되었으며, 본당 수확량은 4.6±0.3 kg으로 예측할 수 있었다.
