To develop the methodology of the quantitative analysis of saponin in ginseng milk, conditions of the saponin extraction were optimized using the fractional factorial design with 3 variables and 3 levels by a RSM computer program. The extraction of saponin increased with an increase in extraction temperature up to $90^{\circ}C$ and then decreased significantly at $100^{\circ}C$. Extraction time affected the saponin yield in a similar trend. On the other hand, decreasing cooling temperature increased the amount of the saponin extracted. Recovery yield of the saponin from ginseng milk varied from 70.0% to 92.9%. The optimum extraction temperature, time and cooling temperature determined by partial differentiation of the model equation were $86^{\circ}C$, 2.83 hrs and $4^{\circ}C$, respectively. Red ginseng products manufactured by the Korea Ginseng and Tobacco Corporation were analyzed to determine the crude saponin , total saponin and ginsenoside contentents by gravimetry, spertrometry and HPLC, respectively, to see if effective quality control of the components in the products can be achieved. Medicinal powders, powders, tablets and capsules which were made from ginseng powder showed similarity in saponin content, the ratio of PD to PT saponin, and the ginsenoside content and composition, while extract powder, extract, extract tea, extract pills and tea, which were made of ginseng extract, showed difference in saponin content, ratio of PD to PT saponin, and the content and composition of ginsengside. It is, accordingly, believed that ginseng products which are uniform in contents and saponin composition can be produced by carrying out strict quality control throughout the processes of making raw red ginseng into final products.
The effect of ginseng saponin on acid phosphatase (AP) activity in liver Iysosomes was investigated and the mechanism by which ginseng saponin may function on the integrity of Iysosomes was discussed. The experimental results obtained are summarized as follows; 1, A very marked increase in the AP activity was observed in the supernatant of hypotonic medium, as compared with that of isotonic medium, indicating that the hypoosmotic shock per so results in activation through osmotic Iysis of particles. 2. Ginseng saponin had no effect on the activity of AP if once released from Iysosomes when Iysed in the hypotonic medium, suggesting that ginseng saponin has no effect on the enzyme molecules per se. 3. The AP activity in isotonic medium suspensions was decreased at the concentrations of 10-6, 10-5 and 10-4% of ginseng saponin, but increased at 10-2 and 10-1%. It's suggested that ginseng saponin enhances the integrity of Iysosomes at 10-6, 10-5 and 10-4%, but decreases it at 10-2 and 10-1%. 4. Suspending particles in distilled water resulted in no correlation of AP activity with treatment with ginseng saponin. 5, The AP activity was decreased in the presence of ATP, showing the possible significance of ATP as a Iysosomal stabilizer and the possibility that ginseng saponin may affect a membrane bound ATPase system by which Iysosomal AP release may be controlled.
Ginsenosides, one of the most well-known traditional herbal medicines, are used frequently in Korea for the treatment of cardiovascular symptoms. The effects of ginseng saponin on ischemia-induced isolated rat heart were investigated through analyses of hemodynamic changes including perfusion pressure, aortic flow, coronary flow, and cardiac output. Isolated rat hearts were perfused and then subjected to 30 min of global ischemia followed by 60 min of reperfusion with modified Kreb's Henseleit solution. Myocardial contractile function was continuously recorded. Ginseng saponin administered before inducing ischemia significantly prevented decreases in perfusion pressure, aortic flow, coronary flow, and cardiac output. The ginseng saponin administered group significantly recovered all of the hemodynamic parameters, except heart rate, after ischemia-reperfusion (I/R) compared with ischemia control. The intracellular calcium ($[Ca^{2+}]_i$) content in rat neonatal cardiomyocytes was quantitatively determined. Administration of ginseng saponin significantly prevented $[Ca^{2+}]_i$ increase that had been induced by simulated I/R in vitro (p<0.01) in a dose-dependent manner, suggesting that the cardioprotection of ginseng saponin is mediated by the inhibition of $[Ca^{2+}]_i$ increase. Overall, we found that the administration of ginseng saponin has cardioprotective effects on the isolated rat heart after I/R injury. These results indicate that ginseng saponin has distinct cardioprotective effects in an I/R-induced rat heart.

To evaluate the effect of the saponin of heat-processed ginseng (Sun ginseng, SG), we investigated the protective effect of SG total saponin fraction against adenine-induced chronic renal failure in rats. SG saponin significantly decreased the levels of urea nitrogen and creatinine in the serum, but increased the urinary excretion of urea nitrogen and creatinine, indicating an improvement of renal function. SG saponin also inhibited adenine-induced kidney hypertrophy and edema. SG saponin reduced serum glutamic oxaloacetic transaminase, glutamic pyruvic transaminase, and lactate dehydrogenase activities increased by adenine. Based on these findings, the ameliorating effect of SG on chronic renal failure may result from its saponin.
The present study was undertaken to elucidate the characteristics of red ginseng total saponin in behavioral changes on ambulation, forced swim test and convulsion in mice. The ambulation and the duration of immobility on forced swim test were not affected by red ginseng total saponin. On the other hand, the duration of immobility inducted by DMI, PGL but not CIP was significantly decreased, and the onset of convulsion induced by pentylenetetrazole was significantly shortened by preadministration of red ginseng total saponin. These results suggest that red ginseng total saponin component may play an important role in modulating synergism with drugs acting on depression and convulsion, and that the characteristics of synergetic effect induced by red ginseng total saponin potentate the central norepinephrine neuron activity.
The effects of 13-cis-retinoic acid and ginseng saponin iron Korean red ginseng on hyperkeratinization of guinea pig skin were investigated by means of enzymatic analysis and light microscopic observation. To induce hyperkeratinization, hexadevance It was topically applied to the dorsal skin of female guinea Pigs every other day for eight days and 13-cis- retinoic acid or ginseng saponin solution was administered orally or topically applied daily during the experimental period. As a result, both topical application of ginseng saponin and oral administration of 13-cis-retinoic acid showed prepentive effects on hyperkeratinization while topical application of 13-cis-retinoic acid inhibited normal epidermal cell proliferation and reduced epidermal enzyme activities such as LDH. ICD and GSPDH below the levels in a normal epidermis. It is suggested that topical application of ginseng saponin and oral administration of 13-cis-retinoic acid may have beneficial efforts against hyperkeratinization possibly by controlling epidermal proliferation and enzyme activities related to epidermal energy metabolism.
In an endeavour to elucidate effects of ginseng on some characteristics of enzymes, malate dehydrogenase (EC 1.1.1.37) was chosen as a model enzyme and effects of ginseng saponin on the enzyme such as optimum pH, product inhibition, optimum temperature and the activity was investigated. The product inhibition by NADH-a reaction product of the enzyme-was increased 33% by 0.3% ginseng saponin. And the optimum pH of the enzyme was 8.3 but in the presence of 0.3% ginseng saponin it increased to 8.5. The enzyme activity and the optimum temperature was not affected by ginseng saponin in the concentration of 1.0% and 0.3%, respectively. In this work, the possibility of contribution of ginseng saponin to the adaptogen activity is suggested; Potentiation of the regulatory activity of an enzyme may contribute to the normalization of the physiological state and consequently may increase the nonspecific resistance of an organism.
This study was performed to investigate the effect of ginseng saponin from Korean red ginseng on the learning and memory. Total (50, 100 mg/kg, bw) and panaxadiol saponin (15, 30 mg/kg, bw) treated groups did not show the difference of the time score and the number of error in comparison with control group. Panaxatriol saponin (15, 30 mg/kg, bw) significantly decreased both the time score and the number of error in water maze test. These results indicate that panaxatriol saponin from Korean red ginseng may improve the learning ability of rat in water multiple T-maze.
Intravenous administration of saponin from the root of Panax ginseng (red ginseng) lowered the blood pressure in a dose-dependent manner (10~100 mg/kg B.W) in anesthetized rats. Therefore, experiments were designed to study whether this lowering of blood pressure is associated with the release of endothelium-derived relaxing factor. Rings of thoracic aorta with and without endothelium were suspended for the measurement of isometric tension in organ chamber. All experiments were performed in the presence of indomethacin (10-5 M). Ginseng saponin (10-5~3$\times$10-4 g/ml) relaxed contractions induced by phenylephrine (10-5 M) in the aorta with endothelium but not in that without endothelium. Treatment of aortic rings with NG_monomethyl-L-arginine (L-NMMA 10-4 M for 30 min), a competive inhibitor of nitric oxide synthase and methylene blue (M.B., 3$\times$10-7 M for 30 min), an inhibitor of soluble guanylate cyclase, diminished the relaxation induced by ginseng saponin. In thoracic aortic rings from rats treated with ginseng saponin for 2 weeks intraperitoneally, the relaxation to acetylcholine was increased compared with non-ginseng treated rings. These data suggest that red ginseng saponin evokes hypotension and that vascular relaxations induced by red ginseng saponin are inediatpd by release of endothelium-derived relaxing factor.
