Studies on Embryo Cryopreservation and Twinning by Embryo Transfer of Korean Native Cattle I. Transfer of Frozen-thawed Embryos and Production of Calves (한우 수정란의 동결보존 및 쌍자생산에 관한 연구 I. 동결 수정란의 이식과 자우 생산)

  • This study was carried out to establish the techniques for producing the calves of genetically superior Korean Native cattle by transfer of frozen-thawed embryos. The effects of some factors related to embryo recovery following superovulation and pregnancy rate following transfer of frozen-thawed embryos were evaluated. Also calving state was investigated. The results obtained were as follows ; The mean number of total and transferrable embryos recovered per superovulated cow was 8.72 and 4.90, respectively, from a total of 72 superovulations using 34 donor cows. There were no significant differences in the number of total or transferrable embryos recovered per superovulated cow between products of follicle stimulating hormone (FSH), years, seasons, and collection numbers. The pregnancy rate was found 44.44% following transfer of frozen-thawed embryos of Korean Native cattle to a total of 180 recipient cows including 82 Angus, 27 Charolais, 62 Hereford and 9 Korean Native cows. The pregnancy rate was significantly (P<0.05) higher in the transfer of excellent (42.99) and good embryos (40.17%), compared with fair (5.90%) grade embryos. And the pregnancy rate was significantly (P<0.05) higher in the transfer of embryos of morula stage (43.86%) than blastocyst stage (15.51%). But there were no significant differences in pregnancy rates between natural and induced estrus estrus asynchrony of 1 days, breeds, and parities of recipient cows. The normal calving rate of 80 pregnant cows following transfer of frozen4hawed em-bryos was 87.5% and the other 10 pregnant cows showed abortion during the period from pregnancy diagnosis at 50~60 days to calving. The average gestation length of normally delivered recipients was 288.50 days and the average birth weight of 70 calves born was 24.22 kg. The gestation length was significantly (P<0.05) shorter in the recipients delivering female calves (286.70 days) than males (289.39 days). But there were no significant differences in gestation tength and birth weight of calves born between the recipient breeds.

  • Offspring have been produced from somatic cells in a number of species. This biotechnology introduced a new phenomenon in reprogramming and differentiation of somatic cell, namely totipotency. However, birth of oversized calves and perinatal abnormalities such as increased gestation length, lack of spontaneous parturition, higher incidence of dystocia, and reduced perinatal viability of offspring are frequently observed in pregnancies of cloned bovine fetuses. Disturbance of feto-placenta has been proposed as likely causes for abnomal growth. However. Little is known the mechanism responsible for the perinatal problems. Therefore, we focused on gestation length in somatic cell nuclear recipient cows. To solve this issues, placental tissues of control and cloned bovine were obtained by a cesarean section (C-section) before 5 days of paturition. Total RNA from control and cloned bovine placenta was extractd by TRIzol reagent. GeneFishing DEG kits (Seegene) were used to identify differentially expression genes. Total RNA (3 ug) were synthesized by M-MLV reverse transcriptase (200 u/ul) with 10 uM dT-annealing control primer (ACP1) at 42C for 90 min. Then, first-strand cDNA (50 ng) was amplified using the 5 uM arbitary ACP (1-20) and 10 uM dT-ACP2 primers. Some specific expression genes were amplified, Now, we are cloning and sequencing. These finding strongly can be support to solve the problems for parturition delay in nuclear transfer cows, suggest that placenta specific proteins are key indicators for the aberration of gestation and placental function in cows.

Genetic and Environmental Effects on the Performance of Reproduction and Lactation in Holstein Cows (Holstein종 유우의 번식 및 산유능력에 미치는 유전과 환경의 효과)

  • This study was conducted to determine the main effects of year, month, sire and parity on certain reproduction and lactation traits on the basis of the data obtained from 1,510 head of Holstein cows at National Animal Breeding Institute from 1971 to 1981. The results obtained in this study are summarized as follows; 1. The conception interval and number of services per concetption were 124.10 days and 2.19 times, respectively. The effects of year, month and sire on the above traits were significant. 2. The birth weight and gestation length were 42.20kg and 281.52 days, respectively. The effects of year, month and parity on the birth weight, and year, sire and parity on gestation length were significant. 3. The yields of milk and milk fat in 305 days, and the fat percent were 4937.05kg, 174.43kg and 3.56%, respectively. The effects of year, month, sire and parity on the above traits were significant. 4. The peak yield and days reaching the peak yield were 26.46kg and 49.17 days, respectively. The effects of the sire and parity on above traits were significant.

  • The gestation period and parturition days were accurately predicted by measuring progesterone concentration in the plasma from 40 pregnant companion bitches. The mean length of the estrous cycle based on plasma progesterone concentrations were 8.14 ± 1.39 (Mean ± SD) days for proestrus, 9.19 ± 2.01 days for estrus, and 55.38 ± 1.96 days for diestrus phase, respectively. The gestation length from each based on the days was 65.61 ± 2.47 days from the first day of estrus after the first vaginal discharge, 63.21 ± 0.99 days from the day when plasma progesterone concentrations increase above 4.0 ng/ml, and 54.51 ± 3.51 days from the first day of diestrus, respectively. Therefore, the parturition day was estimated 65 days from the first day of estrus after the first vaginal discharge, 63 days from the day when plasma progesterone concentrations increase above 4.0 ng/ml, and 54 days from the first day of diestrus, respectively.

Comparison of Reproductive Parameters of Sow at the Rural Areas of Rangamati District of Bangladesh

  • It is imperative to know the reproductive parameters of the existing swine breed for profitable farming in Bangladesh. This study was designed to determine the reproductive parameters of sows at hilly areas. A total of 245 pigs with 47 breedable sows were included. The data on age at puberty, oestrous cycle length, oestrus duration, gestation length, interval between furrowing and onset of oestrus, first service pregnancy rate, service per pregnancy and number of piglets born per sow in local and cross bred sows were determined. The present study revealed that age at puberty of local and cross breed sows was $232.5{\pm}8.4$, $221.3{\pm}6.9$ days, respectively. Oestrous duration was $41.1{\pm}3.1$ hours. The interval between furrowing and onset of oestrus was shorter in cross bred sows. There was no variation in values of oestrus cycle length and gestation length. The first service pregnancy rate was higher in both the local and cross bred sow. Number of piglets per sow per farrowing was $6.1{\pm}2.2$ and 60.9% local sows gave birth of 3 to 5 piglets per farrowing whereas 75.0% cross bred sows gave birth of 6 to 9 piglets. These results suggest that reproductive parameters of local sow need to be improved for better production, and cross bred sows should be reared for obtaining expected productivity.

  • prenatal and postanatal study on pranoprofen, as an antiinflammatory agent, was conducted by oral administration in Sprague-Dawley pregnancy rats from day 17 of gestation to day 21 of after delivery. Pranoprofen was intubated doses of 1.0, 2.5 and 5.0 mg/kg/day and dose of 5.0mg/kg/day of Indomethacin was used as positive control. After delivery, several study indexes such as length of gestation, No. of implantations, No. of live pups, No. of perinatal deaths, sex ratio and No. of malformation were checked and then all the newborns were feeded and investigated physical and behavioral changes.

Teratogenic and Embryotoxic Effects of Clomiphene Citrate in Developing Mice

  • The objective of this study was to assess the teratogenic and embryotoxic effects of clomiphene citrate in mice. The pregnant mice were administered a single dose of clomiphene citrate at different concentrations i.e 1.0, 2.0, 4.0 and 6.0 ${\mu}g/g$ BW on day 8 of gestation. Fetuses recovered on day 18 of gestation were analyzed on morphological, morphometric and histological basis. Morphological observations showed defects like open eyelids, anophthalmia, fore and hindlimb micromelia, meromelia, amelia, sacral hygroma, hydrocephaly, hemorrhagic spots, kyphosis and clubbed feet. Morphometric analysis indicated a significant (p<0.001) reduction in fetal body weight, crown rump length, head circumference, eye circumference, forelimb and hindlimb lengths and tail size against controls. The histological observations showed brain defects like hydrocephaly, enlarged ventricles and undifferentiated neuroglial cells in cerebellum. Cleft palate, underdeveloped pharynx and atrophy of jaw muscles were the common anatomical defects of pharyngeal region. It is concluded that the concentrations of clomiphene citrate used during the present study proved teratogenic in mice fetuses.

Morphological study on effect of radiation in developing mouse brain after fetal exposure (태아시기의 방사선 노출에 의한 마우스 뇌 발달에 미치는 영향에 관한 형태학적 연구)

  • The present study analyze the morphological aspects of the cerebrum of mice with prenatal exposure to high and low dose (0.5, 1, 2 Gy) of $\gamma$-radiation on gestation day 12 or 16. The animal were allowed to give birth and the offspring were sacrificed at postnatal days 28 for gross and microscopic examination of cerebrum. Their body weight, brain weight, brain length, brain width, cortical thickness and area of cingulum bundle were examined. The histological and planimetric analysis were performed observing coronal sections. The gross malfomation (microcephaly) and abnormality of cortical architecture were prominent after exposure to 2 Gy on day 12 of gestation. significant dose-related reductions in body weight, brain weight, brain size were found in all irradiated groups. A significant change was found in thickness of the cerebral cortex and area of the cingulum bundle in the groups exposed to 0.5 Gy or more. There was no difference a lamina patter of six layers in cerebral cortex between the control and irradiated groups, but cell packing density increased significantly in the group exposed to 1 Gy or more. These results suggested that dose as low as 0.5 Gy could cause a morphologically reduce change in developing mouse cerebrum and exposure on day 12 of gestation to $\gamma$-irradiation is a particularly sensitive phase in causing malformation and abnormality of central nerve system.

Changes of the Osmolarity in Embryonic Body Fluid and the Maternal Ovarian Tissue of the Viviparous Teleost, Ditrema temmincki, during the Gestation Period (태생 경골어류, 망상어(Ditrema temmincki)의 임신기 동안 체내자어의 체액삼투압 및 모체 난소조직상의 변화)

  • Dry weight and body fluid osmolarity of embryo, and maternal ovarian fluid and the ovarian inner tissue of the viviparous teleost, Ditrema temmincki, changed considerably during the gestation period. After the complete absorption of the egg yolk, average dry weight of the embryo increased to 373.76 mg, and the range of total length (TL) was from 6.0 to 60.0 mm. Osmolarity of the embryonic body fluid was 796,8 mOsmol/kg with TL 64.0 mm right before parturition. Ovarian outer membrane started to swelling clearly after fertilization, and maximized in March. The swelling of ovigerous folds was maximized in late April. Expansion of blood vessels and increase of hemocytes reached to their maximum right before parturition. The results of this study indicated that these changes are related to the nutritional and environmental adaptation of both the embryo and the maternal body during the gestation period.

A COMPARATIVE STUDY ON THE PERFORMANCES OF F1 CROSSBRED COWS UNDER RURAL CONDITIONS

  • Data were collected on productive and reproductive performance of different $F_1$ crossbred cows (Deshi cows joined by A.I. to Sindhi, Sahiwal, Jersey or Holstein-Friesian Bull semen) reared by farmers in and around the Bangladesh Agricultural University campus at Mymensingh. The traits considered were birth weight, age at first heat, age at first calving, gestation length, post-partum heat period, milk yield, lactation length, dry period and calving interval. Data were analyzed statistically using a completely randomized design and Duncan's Multiple Range Test to compare and determine the significance of between individual group means. It was observed that the Holstein $\times$ Deshi had the highest birth weight (21.4 kg) followed by Sahiwal (17.6 kg), Jersey (16.7 kg) and Sindhi (16.1 kg). Shortest age at first heat (854 and 920 days) and age at first calving (1002 and 1201 days) were found for the Jersey $\times$ Deshi and Holstein $\times$ Deshi crosses respectively. The post-partum heat period were significantly different between breeds with Jersey and Holstein sired calves having the shortest intervals. There was no difference in the gestation length of the four crosses. Holstein $\times$ Deshi and Jersey $\times$ Deshi cows gave the highest milk yields due to longer lactation periods and higher daily milk production. Similarly, Holstein $\times$ Deshi and Jersey $\times$ Deshi crosses had significantly shorter dry periods and Jersey $\times$ Deshi had a shorter calving interval. From this study it may be concluded that exotic genotypes such as Holstein-Friesian and Jersey can perform very well under rural conditions in Bangladesh.
