  • To investigate germination characteristics of soils on Fusarium species under different soil conditions, this study was carried out to test spore germination of 4 Fusarium species. Among 102 soil samples, spore germinations of Fusarium solani f. sp. pisi, F. oxysporum f. sp. cucumerium, F. oxysporum f. sp. raphani and F. coccophilum were under 50% in 98, 85, 82 and 83 soil samples, respectively. The highest spore germination of F. solani f. sp. pisi, F. oxysporum f. sp. cucumerium and F. coccophilum was obtained in 21 soil samples of pH 5.1-6.0, whereas F. oxysporum f. sp. raphani was in 33 soil samples of pH 4.1-5.0. The highest spore germination of F. solani f. sp. pisi, F. coccophilum and F. oxysporum f. sp. cucumerium was obtained in 4 soil samples holding soil moisture content of 41-60%, while F. oxysporum f. sp. raphani was in 22 soil samples of 21-30%. F. oxysporum f. sp. raphani only showed a direct correlation(r=0.29) between spore germination and soil moisture content. On the basis of each spore germination in cultivated and non-cultivated soils, there was significant difference(p=0.01) in 4 Fusarium species. F. solani f. sp. pisi only indicated significant difference(p=0.0l) between two contrasting-soils, cultivated and non-cultivated soils. The numbers of microbial population were higher in soils suppressive to spore germination than soils conducive to spore germination.

  • This study was carried out in order to secure basic data of seedling mass propagation technique of Rhododendron mucronulatum which is the native tree species of Korea by surveying the characteristics of its fruit and seed. The fruits were collected at Mt. Goryeo in Ganghwa-gun on different dates in 2013; August 26th, September 5th, September 12th, October 4th. The seed germination test was carried out at 5, 10, 15, 20, 25 and $30^{\circ}C$. Moisture content of the fruit was highest (54.5%) in the fruit collected on September 5th. Number of the seeds in a fruit was 91.3 to 116.3, regardless of the collection date. Seed length was highest ($1947.4{\mu}m$) in the seeds collected on October 4th and seed width was highest ($727.3{\mu}m$) in the seeds collected on September 12th. Germination rate of the seeds was highest at $25^{\circ}C$ regardless of the seed collection date, which showed the highest value(27.3%) in the seeds collected September 12th. Meanwhile, the seeds were not germinated not at all at 5, 10 and $30^{\circ}C$. $T_{50}$ and mean germination time of the seeds got shorter at the higher temperature. Germination uniformity got lower when the collection date got later. Germination speed of the seeds was fastest at $25^{\circ}C$. According to the results of this study, it seems that the appropriate time to collect fruit and seed is between September 12th and October 4th, and the appropriate temperature for the seed germination is $25^{\circ}C$.

  • From soil samples, 380 antagonistic microorgnisms were isolated. Among the isolates, 42 strains had mycelia growing inhibition ability against Fusariun solani, ginseng root rot causing pathogen. Isolates CHA 1 and S-PFHR 6 were proposed as antagonists for this study and they were identified as Promicromonospora sp. and Pseudomonas pseudoalcaligenes respectively. As an antagonism against hyphae of F. solani in dual culture test, CHA 1 and S-PFHR 6 inhibited linear growing, caused abnormal branching, and the membrane projection which formed by cell wall destruction. The secondary metabolites contained in the culture filtrates which prepared from PD broth and Nutrient broth inhibited the spore germination to 14.3%. The culture filtrate of S-PFHR 6 which prepared by a little amount of soil extract addition to nutrient rich medium had more strongly. inhibited the spore germination and spore germination decreased to less than 4.0% in it. The soil used in this study had fungistasis and the germination rate of macroconidia and chlamydospore of F.solani was 19.4% and 17.7% respectively. The steam sterilized soil lost fungistasis and germination rate of conidia increased to more than 97.9%. The soils amended with the propagule of CHA 1 and S-PFHR 6 increased fungistasis and the germination rate of macroconidia decreased to 14.7% and 11.7% respectively in each treatments. But the soil ammended with glucose and asparagine annulled fungistatic ability and the germination rate of macroconidia increased to more than 48.0%. As an antagonistic activity of the secondary metabolites of two antagonistic isolates in soil, the germination rate of macroconidia of F. solani was 9.3% in the soil amended with the culture filtrate of CHA 1 but the culture filtrate of S-PFHR 6 had no such activity. In the soil which treated with antagonist propagule or culture filtrate, the chlamydospore germination rate was lower than that in natural soil. The addition of glucose and asparagine to antagonist propagule treated soil did not enhanced the chlamydospore germination.

  • Regardless of the test species, germination was adversely affected by the different concentrations of the acidic and neutral fractions of sunflower root exudates while the basic and aqueous fractions had no effect on germination. In both test species, root lengths were inhibited slightly more than shoot lengths. Significant reduction in fresh weights of the test species was observed when the test species were treated with the acidic and neutral fractions but not with the basic and aqueous fractions. Six compounds, hydroquinone, ${\beta}$-resorcyclic acid, vanillic acid, caffeic acid, salicylic acid, and quercetin, were characterized from the acidic fraction. Seven compounds, hydroquinone, gentisic acid, ${\beta}$-resorcyclic acid, vanillic acid, caffeic acid, ferulic acid, and quercetin, were elucidated from the neutral fraction.

  • The germination test was used to detect biological changes in irradiated rice and glutenous rice at low doses. Grains were irradiated at below 0.5 kGy, husked and placed on distilled water moistend filter paper in a covered petri-dish. A germination test of 20 grains was carried out at room temperature for 5 days. The shoots and roots of non-irradiated rices grew well in comparison with those of irradiated rices above 0.3 kGy. The roots of rices were more sensitive to irradiation than the shoots, and the growth of roots was significantly decreased with the increasing doses. In glutinous rices, the growth of shoots and roots was retarded by irradiation at 0.2 kGy or more after 3rd days. We concluded that if the shoot or root length is 10 mm or longer within 5 day, the rices and glutinous rices are identified as non-irradiated.

  • The purpose of this paper is to elucidate physiologically the cause of the hastening germination of dormant Taxus cuspidata seeds by stratification. During the stratification the exchange of chemical substances such as sugar, protein, starch and fat were observed, and growth promoting and inhibiting substances were extracted and seperated from seeds by the conventional chromatographic method with coleoptile straight-growth test. An intensive investigation was made on the balance between the promoters and inhibitors. consequently, it was confirmed that germination of seeds was accelerated with exchange of chemical substances by stratification. The results obtained may be summarized as follows: 1. During the stratification growth promoters were increased and growth inhibitors were decreased rapidly in the endosperm of seeds. Thus, it was presumed that hastening germination was controlled by balace between the promoters and inhibitors from November to next March after a year's stratification. On the other hand growth promoters were almost constant and growth inhibitors were decreased rapidly in the seed coats, and it was presumed that hastening germination was influenced by exchange of inhibitors more than by that of promoters. 2. As a results of germination test of lettuce seeds, it was generalized that hastening germination was controlled by a decreased amount of growth inhibitors more than by an increased amount of promoters. 3. During the stratification sugar and crude protein contents were increased gradully with moisture content, while starch and crude fat were decreased in endosperm of seeds. So it was assumed that the exchange of these chemical substances was closely related to the germination of seeds.

  • Gibberellin (GA) has been widely used to break seed dormancy for better stand establishment. The experiment was done to clarify the effect of $GA_3$ (concentration; period) and light quality (red; white; dark) after sowing on seed germination and radicle elongation of Campanulaceae (Platycodon grandiflorum; Codonopsis lanceolata; C. pilosula) to get an information on their field emergence. The germination test was carried out with 12 hours irradiation for 9 days after priming treatment. In the darkness, the mean germination rate of all the species was decreased in the order to P. grandiflorum, C. lanceolata, C. pilosula. Their mean germination rates and radicle lengths were increased with increased concentration to 0.1mM of $GA_3$. Earlier germination rate was higher but later one was less in 4-day $GA_3$ treatment than in 1- or 2-day $GA_3$ treatment. Light treatment. especially red light given after $GA_3$ treatment. eliminated the $GA_3$ treatment effect. Red light done after $GA_3$ treatment nearly blocked the germination of P. grandiflorum and C. pilosula but delayed that of C. pilosula compared to the other light quality treatments having the similar rate. In addition. the radicle elongation of all three species affected by light quality treatment showed the same result as the germination rate.

  • We studied the effect of SMP (solid matrix priming) treatment, seed pretreatment for germination enhancement, to tobacco seeds by measurement of germination rate and contents of fatty acid, energy source for tobacco germination. The results were as follows. In result of germination test, germinative ability was the highest in KF109 at nine day treatment and in KB108 at seven day treatment. The composition of fatty acid in tobacco seed confirmed by gas chromatography were palmitic acid, stearic acid, oleic acid+elaidic acid, linoleic acid+linolelaidic acid, and $\alpha-linolenic$ acid. Palmitic acid, stearic acid, oleic acid and elaidic acid, and linoleic acid and linole-laidic acid were highest in KF109 at eight or nine day treatment and in KB108 at six or seven day treatment. Especially, content of oleic acid + elaidic acid, and linoleic acid+linolelaidic acid were changed largely by treatment, so these might be used for index to examine treatment effect.

  • To investigate phytotoxic substances in Artemisia argyi, the donor plant, and their biological activities, seed germination and seedling growth of receptor plants such as Arundinella hirta, Echinochloa crus-galli, Rumex crispus and Lactuca sativa were examined at different concentrations of aqueous extracts of the donor plant. Germination of four receptor species was inhibited by the extracts, while seedling growth was decreased to a lesser degree than in the germintion test. Germination, seedling growth and dry weight growth of Achyranthes japonica grown in pot were proportionally inhibited by the extracts. Volatile substances emitted from A, argi plant caused slight inhibition in the germination and seedling growth of the receptor species. Essential oil of the plant extracted by Karlsruker's apparatus inhibited growth of microorganisms and callus growth of Pinellia ternata and Oryza sativa. The GC /MS method was employed for analysis and identification of allelochemicals from A. argyi leaves. Sixty-one chemical substances such as a-pinene, camphene, 1. 8-cineol, etc. were identified from essential oil of A. argyi. The results of this experiment on seed germination, seedling growth, microorganism culture and tissue culture indicated that naturally occurring chemical substances from A. argyi would be responsible for the growth inhibition of plants studied.

  • Dried top and root extracts of seven different weed species, fresh top and root extracts, and various concentrations of extract (0, 5, 10 and 15%, W/V) and residue rate (0.0, 0.25. 0.5, 0.75 and 1.0%) of velvetleaf (Abutilon theophrasti Medic.) were used to study their allelopathic effects on alfalfa in the laboratory and greenhouse. Top and root aqueous extracts of common lambsquarter (Chenopodium album L.), giant foxtail (Setaria faberii Herrm), redroot pig-weed (Amaranthus retroflexus L), velvetleaf, large crabgrass (Digitaria sanguinalis L), canada thistle (Cirsium arvense L.) and prostrate knotweed (Polygonium aviculare L.) significantly inhibited germination, seedling length, weight, and vigor in alfalfa. Top growth extracts of weeds exhibited greater allelopathic effects than root extracts. Alfalfa test species, WL-320, responded significantly different to the various weed species extracts in terms of allelopathic effect. The regression slopes of various top extracts showed significant variation with respect to germination percentage. Velvetleaf (b=3.69) extracts were the most inhibitory, while large crabgrass (b=2.39) extracts had the least allelopathic effect on alfalfa. When compared the activity of fresh velvetleaf extract to that of dry velvetleaf extract, dry extract was more inhibitory to alfalfa germination and seedling growth. Germination, seedling length and weight of alfalfa were inversely proportional to the concentration of dried velvetleaf extracts. Seedling emergence and survival percentage was inhibited by velvetleaf residue mixture treatment. Also, more of the toxic effects were observed from the dried top extracts, as compared to extracts from fresh top and root. These results demonstrate the allelopathic activity of different weed species extracts and suggest that weeds may affect crop growth and development due to the inhibitory effects of allelochemicals present in weed tissue.
