Genetic Analysis on Germination Rate and Fat Acidity of Japonica Rice Seeds after High Temperature Storage

  • This study was carried out to get the genetic information on the germination rate and fat acidity after 12 weeks' storage at $35^{\circ}C$. Germination rate decreased with longer storage period, while fat acidity increased. Germination rate was higher in the order of Koshihikari, Hanmaeum and Unkwangbyeo after 12 weeks' storage at $35^{\circ}C$. Fat acidity of Koshihikari and Hanmaeum was gradually increased, but that of Unkwangbyeo was dramatically increased after 4 weeks' storage at $35^{\circ}C$. The genetic mode on germination rate and fat acidity using the joint scaling test was shown to be additive-dominance gene effects. Additive([d]) effect was higher than dominance effect([h]) in all crosses. The average degree of dominance, [h]/[d], was less than 1, indicating that germination rate and fat acidity was partially dominant. Germination rate and fat acidity based on frequency distribution of 2 crosses were quantitative characters expressed by polygenes.

  • The effects of some chemicals for the germinative stimulation of Clostridium chauvoei spore studied. The results obtained were as follows: 1. Cooked meat medium (CMM) was superior to Liver-liver bouillon (LLB) for cultivation of the organisms. 2. Heating the organisms at $70^{\circ}C$ for 30 minutes prior to cultivation in CMM stimulated the germination rate. 3. The addition of 0.5mM L-alanine to CMM was found to be most effective for the rapid germination of the spores. 4. The addition of 0.2 mM D-alanine to CMM inhibited the germination of the spores even if the spores were heat treated. 5. The addition of 0.1mM manganese retarded the germination time, but the final germination rate was greater with 0.1mM manganese than with 0.5mM L-alanine. 6. The addition of both 0.5mM L-alanine and 0.1mM manganese stimulated the germination rate to 100 per cent. 7. The germination rate was greater with the addition of germination stimulants than without any stimulants. The germination rate was approximately 6 per cent greater with prior heat treatment than without heat treatment. 8. The optimum time for the harvest of vegetative forms of the organisms was 15 hours after cultivation in the media which contain suitable germination stimulants.

A Study of Germination Characteristics of Native Plants to be Utilized in DMZ Barren Land (불모지 내 활용 가능한 자생식물의 발아특성 연구)

  • This study suggested suitable soil textures that is proper to propagate native plants to manage and restore barren land in DMZ. Germination tests were conducted for 16 native herbaceous plants growing in the DMZ border area in accordance with FAO-BI (Biodiversity International) standards, and the germination rate and T50 in vitro were investigated. In order to examine the germination characteristics according to the soil textures, we used gravel, bed and mixed soil and investigated the germination characteristics under ordinary room temperature conditions in the greenhouse. As a result, it was observed that the germination rate in the greenhouse was significantly decreased compared to the germination rate in vitro of the species advertised due to soil textures. T50 between the in vitro and each soil texture showed significant differences whereas T50 between soil textures alone did not in all species advertised. The germination rate in vitro of Aster koraiensis, Dendranthema zawadskii var. latilobum, Hosta clausa, and Hosta minor there was no significant difference compared to ordinary room temperature conditions. In addition, as the germination rate is demonstrated more than 70%, which is relatively higher than other species advertised, it is considered to have strong environmentally adaptable. On the other hand, considering that the 6 species of Leontopodium coreanum, Plantago major, Potentilla chinensis, Sedum kamtschaticum, Sedum latiovalifolium, and Veronica kiusiana demonstrated less than 50% of germination rate in vitro, it is expected to be difficult to propagate without pre-treatment. In order to use these 6 species as restoration material plants, it needs to be considered to pre-treat to improve germination rate, or to enhance the vitality of seeds by improving the seed gathering period and storage method.

  • Pollen germination and its' viability in bellflower hybrid system are of great importance. The present study was conducted to investigate the several factors underlying the pollen germination of Platycodon grandiflorum and obtain the basic data for effective artificial pollination for the production of sound specifies. The pollens of Platycodon grandiflorum started germination from one hour after planting, and the germination was actively progressed as time dependent manner. For lighting conditions, the germination of pollens under the light was faster by more than twice than that without the light. Furthermore, the germination was better in the high temperature rather than in the low temperature. The germination rate was higher in the $30^{\circ}C$. For the carbon source, the germination rate was better at the concentration of 15% regardless of the kinds. In particular, the highest value was observed with glucose. The germination rate was decreased substantially as the increasing with the higher pH. The dynamic germination of pollens was observed at the pH 5. With respect to the growth regulator, the higher concentration of NAA induced the higher the germination rate. $GA_3$ showed a good germination rate in $0.05mg{\cdot}L^{-1}$. Meanwhile, for kinetin, lower concentration increased the germination rate, unlike NAA. The higher concentrations of boric acid degraded the germination rate, and the addition of boric acid of $10mg{\cdot}L^{-1}$ demonstrated higher germination rate than the addition of other growth regulators. Notably, the addition of asparaginic acid exhibited the similar results in all test sectors regardless of concentration, whereas a little higher result was observed in the high concentration sector. Taken together, the results concluded that the boric acid was considered as one of the essential minerals that played an important role on the germination of pollens.

Removal Effect of Biostone and Green Tea on the Heavy Metal Toxicity during Seed Germination of Arabidopsis thaliana (애기장대의 종자 발아에 미치는 맥반석과 녹차의 중금속 제거 효과)

  • This experiment was carried out to investigate the effects of heavy metals (cadmium, chromium, copper and lead) on the seed germination of Arabidopsis thaliana, and examinated the removal effects of biostone and green tea on the heavy metal toxicity. Cadmium and chromium among the four heavy metals had no effect on the seed germination even in the concentration fifty times higher than in the official standard concentration of pollutant exhaust notified by the Ministry of Environment. However, seeds were not germinated in the concentration of copper ten times higher and in the concentration of lead fifty times higher than the official standard concentration. When seeds were sown in the solutions of lead (15, 20, 25 and 30 mg/L) and copper(15 and 20 mg/L), the seed germination rates were 0% and less than 10%, respectively. However, when biostone(3 g/30 $m\ell$) was added, the seed germination rate was 100% in all the concentrations. The germination rate was 100% in distilled water and copper solution (5 mg/L). However, green tea (0.2 g/30 $m\ell$) was added, the seed germination rate was 0% in both. The results show that cadmiun and chromium had no effect on the seed germination, but lead and copper decreased the rate of seed germination of Arabidopsis thaliana, Biostone removed heavy metal toxicity, but green tea did not removed heavy metal toxicity during germination.

Effect of Temperature and Sulfuric acid treatment on the Germination of Hovenia dulcis Thunb (온도 및 황산처리가 헛개나무 종자의 발아에 미치는 영향)

  • This study was conducted to investigate the optimum condition of breaking dormancy and to increase the germination rate of Hovenia dulcis Thunb. seeds. Germination of seeds which did not germinate at the various constant temperature was induced by the artificial breaking of seed coat, and the germination rate of seeds which the seed coats were broken was the highest(94%) at $15^{\circ}C$. The germination rate of seed was highest when seeds were treated with concentrated $H_2SO_4$ for 40 minutes. When seeds were stored in dry condition, the germination capability was maintained up to 5 years after harvest. However, their germination rate become lowered and average germinating duration was extended. Germination rate of seeds which were stored for 5 years significantly increased by the low temperature treatment more than 30 days.

  • The germination responses of Cassia mimosoides var. nomame seeds to temperature were examined under various conditions. The temperature range allowing germination was $20~40^{\circ}C$, and the linear relationship between the germination rate and temperature appeared to exist between $28~38^{\circ}C$ The total thermal time required for germination (10~80%) of C. mimosoides seeds ranged from 259 Kh (degree Kelvin$\times$hours) to 421 Kh and base temperature range was relatively constant, i.e., $23.39~26.68^{\circ}C$. In the increasing temperature (IT) regime, C. mimosoides seeds started to germinate at $16^{\circ}C$ and showed greater germination rate with increasing temperatures. The final germination rate was 64% at $36^{\circ}C$. On the other hand, in the decreasing temperature (DT) regime, the seeds began to germinate at $36^{\circ}C$, and the final germination rate was 52% at $20^{\circ}C$. An induced dormancy occurred at $16^{\circ}C$ in the DT regime.

  • This study was conducted to investigate the seed storage method and seed germination characteristics of Synurus deltoides (Ait.) Nakai, a kind of wild vegetables. Several parameters affecting seed germination, such as germination temperature, chilling duration, imbibition duration, and storage method were evaluated. The seed germination of Synurus deltoides showed the highest germination rate (GR), promptness index (PI), germination performance index (GPI), mean daily germination (MDG) at 25℃. This implies that the optimum germination temperature was 25℃. And also, Seeds stored at -4℃, after 150 days showed relatively high germination rate about 98%. Therefore, The optimum germination temperature of Synurus deltoides was 25℃ and to increase the germination rate over 98% of Synurus deltoides, cold-wet storage 20 days treatments at 4℃ were effective in the improvement of seed germination.

  • The germination rate and longevity of seeds of Gastrodia elata Blume have been observed for 48 weeks using Mycena osmundicola strain H-21, one of fungi stimulating seed germination. Storage condition of post-harvest seeds was observed in the different temperature ranges of $-30^{\circ}C,\;-5^{\circ}C,\;5^{\circ}C\;and\;30^{\circ}C$ for 48 weeks. After storage period of 48 weeks, the germination rate of G elata was 65.7% at $5^{\circ}C$ and 71.6% at $-5^{\circ}C$, respectively. Although the germination rate of G. elata was 77.3% for 11 weeks at $25^{\circ}C$, the germination rate had been decreased gradually to 49.3% at 13 weeks, 0.3% at 23 weeks and then 0% at 25 weeks. The germination rate was reached to the level of 10% for 2 weeks at $-30^{\circ}C$ and then decreased to 0%.

Effects of Temperature, Pelleting Materials and Size on Germination of Rehmannia glutinosa $L_{IBOSCH}$ (Pelleting 소재와 크기가 지황종자의 발아에 미치는 영향)

  • This experiment was conducted to evaluate the germination rate according to temperature, pelleting materials, pelleted size, seed storage periods under light and dark conditions in Rehmannia glutinosa. The highest germination rate was obtained from $25^{\circ}C$ in both light and dark condition. Two-year old seeds showed higher germination rate compared to one-year old seed. Talc material showed the highest germination rate among the pelleting materials by 82.5%. Among the different pelleting sizes, the highest germination rate was shown in 2.0 mm, and especially Talc pelleting showed 100% germination rate on 17days after seeding. In case of combination of pelleting materials, both of talc + vermiculite, zeolite + vermiculite showed lower germination rate than the control and 0.7 mm pelleted seed showed the highest germination rate.
