녹화용 벼과식물 종자의 채종시기 및 온도별 발아특성 (Germination Characteristics by Temperature and Production Time to Poaceae Plant Seed)

  • This text was experimented and investigated the optimum production time and germination characteristics of seed that collect in Korean Chungnam, in order to offer the basic informations for slope restoration and revegetation using Poaceae plant. Optimum time of seed production was mid-late October of Miscanthus sinensis, Pennisetum alopecuroides, early November to late October of Themeda triandra var. japonica and M. sacchariflorus, and mid November of Phragmites communis. Epiphytic amount of seed full ripe was the most times in mid October of M. sinensis, P. alopecuroides and M. sacchariflorus, early November of T. triandra var. japonica, and mid November of P. communis. Seed rate of maturity was the highest times in early November of M. sinensis, P. alopecuroides and M. sacchariflorus, and mid November of T. triandra var. japonica and P. communis. Germination rate by seed collection time was the highest times in early October of M. sacchariflorus, mid October of T. triandra var. japonica, early November of M. sinensis and mid November of P. communis. Germination days by seed production time was the shortest times in early November of M. sinensis and M. sacchariflorus, mid November of T. triandra var. japonica and P. communis. Optimum temperature of germination was $20{\sim}25^{\circ}C$ in M. sinensis and P. alopecuroides, $25^{\circ}C$ in T. triandra var. japonica, and $25{\sim}30^{\circ}C$ in P. communis.

Resistance of Cucumber Grafting Rootstock Pumpkin Cultivars to Chilling and Salinity Stresses

  • Grafting using a pumpkin (Cucurbita sp.) rootstock is an effective way to improve cucumber (Cucumis sativus) resistance to a combination of chilling and salinity stresses. We evaluated the tolerance of 15 pumpkin cultivars to chilling, salinity, and combined stresses at the germination and seedling stages. Selected plant characteristics, including germination rate, germination potential, germination index, plant height, stem thickness, fresh weight, and dry weight, were analyzed. We used the unweighted pair group method with arithmetic mean for cluster analyses to determine the stress tolerance levels of the pumpkin cultivars. The 15 cultivars were divided into three clusters: tolerant, moderately tolerant, and susceptible to stress treatments. The stress tolerances of all cultivars were variable in the germination and seedling stages, and most cultivars were not tolerant to individual treatments of chilling or salinity stresses at both stages. These results suggest that identifying suitable cultivars for use as rootstock during cucumber grafting should involve the evaluation of stress tolerance during different growth stages. Additionally, cultivars tolerant to chilling stress may not be tolerant to salinity stress; therefore, the choice of pumpkin rootstock should depend on where the grafted plant will be grown. Cultivars tolerant to a combination of chilling and salinity stresses may be useful as rootstock for cucumber grafting. Our findings may serve as reference material for choosing appropriate pumpkin rootstocks for cucumber grafting.

비탈면 녹화용 큰낭아초 종자의 형태 및 발아특성 (Germination Characteristics and Shape of Indigofera amblyantha Seed for Slope Revegetation)

  • This text was analyzed and investigated the morphological character, production time and germination characteristic of seed that collect in Chungnam, in order to offer the basic informations for slope restoration and revegetation using Indigofera amblyantha of wild shrub. Legume was cylindrical type and length 31.5mm, width 2.5mm, and seed number of per legume was full ripe 5.5, unripe 0.9. Seed was elliptical type and length 2.4mm, width 1.7mm, and weight of 1,000 seeds was 5.9g. Because seed matured within December and next year January, optimum time of seed production was in mid of January. In the treatment $H_2SO_4$ of 15 minutes, seed germination rate was the highest as 91.0%, and seed germination time (days) was the shortest as 6.7 days. Seed germination time by temperature was the shortest from $25^{\circ}C$. Therefore, seed germination of Indigofera amblyantha was very useful in the treatment $H_2SO_4$ of 15 minutes from $25^{\circ}C$ temperature.

Early Growth, Carbohydrate and Phytic Acid Contents of Germinating Rice Seeds under NaCl Stress

  • Germination characteristics and alterations in soluble sugar-starch transition and phytic acid during germination were studied in rice seeds under saline conditions. NaCl significantly reduced the speed of germination. Also, the radicle growth out of seeds was severely inhibited by the exposure to NaCl solution, thus, seeds were almost impossible to grow to seedlings. Soluble sugar was remarkably accumulated, whereas starch was decomposed stepwise during seed germination. The metabolism of soluble sugar and starch in germinating seeds showed a distinct difference. The level of phytic acid in seeds decreased in all NaCl treatments during germination, but the level was affected differently by NaCl concentration in the two varieties. Overall, our results suggest that salt stress retard the radicle growth of rice seeds, and affect the starch-to-sugar conversion and the decomposition of phytic acid differently in two varieties.

  • Plasma technology has been widely used for decontamination, differentiation, and disease treatment. Recently, studies show that plasma has effects on increasing seed germination and plant growth. In spite of increasing number of studies about plasma effects, the interaction between plasma and plants has been rarely informed. In this study, we have analyzed the effects of nonthermal atmospheric pressure plasma on seed germination and growth of coriander (Coriandum sativum), a medicinal plant. We used to Ar, air, and N2 plasma on seed as feeding gases. Plasma was discharged at 0.62 kV, 200 mA, 9.2 W. Seed germination was increased over time when treated with N2 based DBD plasma for exposure times of 30 seconds and 1 minute, everyday. After 7 days, about 80~100% of seeds were germinated in the treatment with N2 based DBD plasma, compared to control (about 40%, only gas treated seeds). In order to elucidate the mechanism of increased germination, we have analyzed characteristics of changes in plant hormones and seed surface structure by SEM.

Culturability of Clostridium botulinum Spores under Different Germination Conditions, Sublethal Heat Treatments, and in the Presence of Nisin

  • Thermal resistance and heat activation characteristics of Clostridium botulinum ATCC 25763 spores were evaluated. The effects of nisin and pH on the activation and subsequent germination were also investigated. Spores of C. botulinum were not inactivated by heat treatments up to $92^{\circ}C$ for 2 hr. Heat treatment at $85^{\circ}C$ for 90 min was selected as the optimal activation condition based on monitoring subsequent germination. L-alanine alone or in combination with L-cysteine was not sufficient to germinate the spores of this strain. Tryptone-Peptone- Glucose-Yeast extract (TPGY) broth supplemented with L-alanine was used as a suitable germination medium. Decreasing pH of activation suspension increased the degree of phase darkening, i.e., germination. In addition, the presence of nisin during activation increased the degree of phase darkening. The majority of spore populations were dormant at a pH of less than 2.8, and these populations required heat activation to increase the culturability on TPGY agar medium. However, extended heating in the presence of nisin at pH 2.8 decreased the spore count; however, heat activation was less necessary at pH 3.4, compared at pH 2.8.

Seed Dormancy and Germination Characteristics of Endemic Elder Species (Sambucus racemosa subsp. pendula) and Common Elder Species (S. williamsii) in Korea

  • This study was conducted to determine the seed dormancy types of Sambucus racemosa subsp. pendula Nakai and S. williamsii Hance. Low temperature stratification (1, 2, 3, 4 months) and GA3 treatment (1,000 mg/L) were performed on seeds to determine the type of seed dormancy. After the treatment, seeds were placed on a petri dish at 25℃ under light conditions. The germination rate and mean germination time were investigated. Results showed that cold stratification was effective in breaking the dormant state of the seed in both species. In the low temperature stratification treatment, the seed germination performance was improved as the treatment period was prolonged. Gibberellin treatment was effective in breaking the dormant state of S. racemosa subsp. pendula without low temperature stratification. However, S. williamsii did not break the dormant state of the seed by gibberellin treatment without low temperature stratification treatment. In the gibberellin treatment, germination performance was improved according to the low temperature stratification treatment period. As a result of this study, the seeds of S. williamsii have both an intermediate complex and a deep complex morphophysiological dormancy (MPD). In comparison, it was found that the S. racemosa subsp. pendula had intermediate composite MPD.

Temperature Effect on Seed Germination and Seedling Growth of Medicinal Umbelliferae Plants

  • Umbelliferae medicinal crops have low germination under natural condition. Special objective of this study is to investigate morphological characteristics of seeds and effects of environmental factors such as light and temperature on their germination and seedling growth in Umbelliferae plants, using them as oriental medicine materials. Fruits of Umbelliferae are bi-partite type and form 2 seeds. Their oil canals and funiculus or carpaphore has apparently specific shape. Seed shape of Buplerum falcatum, Ligusticum acutilobum and Foeniculum vulgare are oblong, Ledeboutiella seseloides, Angelica dahurica, Angelica gigas elliptic, and Ostericum koreanum ovate. Color of seed coats varied from yellow to dark brown. Seed size was 3.2 to 6.9 mm in length, 1.7 to 4.9 mm in width. One thousand-seed weight of Buplerum falcatum was lowest(at 1,660 mg) and Ledebouriella seseloides and Angelica gigas were relatively highest(at 3,970 mg) having larger size than any others. Optimum temperatures for seed germination and seedling growth ranged from 20 to $25^{\circ}C$ and especially temperature at $25^{\circ}C$ improved seed germinations of Ledebouriella seseloides, Angelica dahurica and Ligusticum acutilobum regardless of light condition.

Effect of Cold Stratification and Gibberellin Treatment on Androsace septentrionalis L. Seed Germination

  • Androsace septentrionalis is a grass species restricted to North Korea; however, it is at the brink of extinction due to habitat loss and environmental changes caused by natural disasters and anthropogenic activities. This study was conducted to characterize the dormancy conditions of A. septentrionalis in an effort to conserve this North Korean plant resource. For this purpose, the morphological characteristics and vigor of A. septentrionalis seeds were examined, and its germination characteristics under different temperature conditions (15/6℃ and 25/15℃ ), low-temperature stratification, and gibberellin (GA3) treatment were determined. The results revealed that A. septentrionalis exhibits non-deep type morpho-physiological dormancy, and low-temperature stratification treatment was not effective in breaking the dormancy of A. septentrionalis seeds. Meanwhile, GA3 treatment significantly increased the mean germination time, rate, and speed of the seeds. The optimal conditions for the germination of A. septentrionalis seeds were 25/15℃ fluctuating temperature and 500 mg·L1 GA3 treatment. The results of this study are useful for the mass propagation of A. septentrionalis.

하천 제방 비탈면 녹화용 자생 지피식물의 종자발아 특성에 관한 연구 (A Study on Characteristics of Seed Germination of Native Plants for Revegetation on the Slope of River bank)

  • To investigate the attributes of seed germination of the plants for greening of river bank slope, low-temperature treatment, chemical treatment, plant hormones treatment, and stratification were conducted using the seeds of 6 herbs growing naturally around river banks, such as Setaria glauca and faberii, Imperata cylindrica var. koenigii, Miscanthus sinensis, Pennisetum alopecuroides, Lespedeza cuneata and the seeds of 3 shrubs, such as Lespedeza bicolor., Rosa multiflora, Spiraea prunifolia for. simpliciflora. The findings are as follows. In case of herbs, Setaria glauca and faberii. have stone seeds. In the treatment with $H_2SO_4$, they were germinated through dormancy breaking, even though there were some differences depending on immersion time, and the germination rate of Setaria faberii was a bit higher than that of Setaria glauca. The germination rate of Imperata cylindrica var. koenigii was low in the treatment with $GA_3$, NAA, and the plant hormone, but the seed germination rate has improved and the germination rate of Miscanthus sinensis and Pennisetum alopecuroides showed the improved seed germination rate in low temperature treatment. In case of Lespedeza cuneata and bicolor, the leguminous plants, the treatment with $H_2SO_4$ was most effective for the improvement of seed germination and the decrease in germination time. Only Spiraea prunifolia for. simpliciflora, the rose among 3 woody plants showed the seed germination rate above 50% without any additional treatment. Thus, it was thought to be appropriate for greening. Rosa multiflora showed the attribute of germination in a dark condition at low temperature through stratification, despite its low germination rate. Based on the results as above, with respect to the greening using directly collected seeds, Miscanthus sinensis, Pennisetum alopecuroides, and Spiraea prunifolia for. simpliciflora are suitable for greening, because they do not need any additional treatment, and Lespedeza cuneata and bicolor are also suitable for greening because it is possible to improve the germination rate and reduce the germination time through the treatment on the seeds before sowing.
