Molecular identification and Phylogenetic relationship of the rook (Corvus frugilegus) population in Jeju-do Province, South Korea (제주도에 도래하는 떼까마귀 집단에 대한 분자 종 동정 및 계통 유연관계)

  • In order to identify the species and to reveal the phylogenetic relationship of rook populations found in Jeju-do Province in winter seasons, we determined the sequences of mitochondrial cytochrome c oxidase I (COI) gene and analyzed the genetic structure of maternal lineages and phylogenetic relationship. The rook DNAs were isolated from the post-mortem specimens and plumages collected from agricultural farms in Jeju-do Province including U-do Island. The obtained COI sequences (n=41) showed over 97.0% identities with those previously reported from Corvus frugeligus. Three COI haplotypes (J01-J03) were detected from COI sequences of the rooks obtained in Jeju-do Province but those did not show the site-specific patterns, showing that they might be derived from a common maternal origin. Eight maternal haplotypes were detected from all COI sequences obtained. Among those three haplotypes contained the COI sequences from Northeast Asia including eastern Russia, Mongolia and South Korea. On the other hand, the other five haplotypes contained the COI sequences reported from Central Asia, Middle East, western Russia and European countries. The COI sequences from Jeju-do Province were located on three haplotypes (CF01-CF03) belonging to Northeast Asian rook lineages. The NJ tree showed the distinct branch patterns suggesting two different maternal lineages of C. frugilegus, which proposed as two parapatric subspecies, C. f. frugilegus (Western) and C. f. pastinator (Eastern). These findings using DNA barcoding approaches will be contributed to provide the information about avian fauna for understanding the genetic structure of maternal lineage, phylogenetic relationship and their molecular ecology.

Preliminary Study on the Ultramafic Rocks from the Chungnam Province, Korea (충남지역에 분포하는 초염기성암의 기원규명을 위한 기초연구)

  • Several ultramafic bodies and ultramafic origin talc deposits are distributed in Chungnam province near the contact zone with Ogchun fold belt They occur as discontinued belt form with northeast trending, and most of them are more or less sepentinized. Major, trace, and rare earth elements analyses were made of the ultramafics from the study area to constrain their origin and genetic relationships. Compared to the primitive mantle estimates of privious workers, the correlations defined by the studied rock samples indicate similar Ni but very lower $Al_{2}O_{3}$, CaO and $TiO_{2}$ contents. It is inferred that source material of the studied rocks might be residual mantle which had undergone a large degree of partial melting event. The REE patterns show relatively flat to enriched in LREE (chondrite normalized La/Yb and Sm/Yb ratios are 1.1-5.2 and 1.2-1.6). Several alternative explaination are possible for LREE enrichment patterns in the studied ultramafic rocks such as 1) enrichment due to late stage alteration, 2) enriched pre-melting composition, and 3) mixing of two components. Based on the result, the LREE enrichment characteristic of the studied rocks might be result from the mixture of two geochemically distinct components; one is depleted residual mantle and the other component which determine the abundances of incompatible elements and responsible for the LREE enrichment.

  • The condition, characteristics, and stability of slopes, as well as the consequences of slope failure, need to be understood for the proper stabilization of slopes and preclusion of potential disasters arising from slope failure. Here, a slope code system (SCS) that succinctly and accurately reflects the various conditions of a slope is proposed. The SCS represents the condition, characteristics, and geotechnical stability of slopes, as well as the consequences of slope failure, and the method is quickly and easily applied to a given slope. The SCS comprises five elements: 1) the slope material; 2) the genetic origin (rock type) and geological structure of the slope; 3) the geotechnical stability of the slope; 4) the probability of failure and remedial works made upon the slope; and 5) the consequences of failure. A letter code is selected from each element, and the result of the evaluation and classification of the slope is given as a five-letter code. Because the condition, characteristics, and geotechnical stability of a slope, as well as the consequences of slope failure, are provided by the SCS, this system will provide an effective mechanism for the maintenance and management of slopes, and will also allow more informed decision-making for determining which slopes should be prioritized for remedial measures.

Genetic Environment of the Samsung Gold-Silver Deposit, Republic of Korea: Ore Minerals, Fluid Inclusion and Stable Isotope Studies (삼성 금-은광상의 생성환경: 광석광물, 유체포유물 및 안정동위원소 연구)

  • The Samsung gold-silver deposit consists of quartz veins that fill along the fault zone within Cretaceous shale and sandstone. Mineralization is occurred within fault-breccia zones and can be divided into two stages. Stage I is main ore mineralization and stage II is barren. Stage I is associated with wall-rock alteration minerals(sericite, pyrite, chlorite, quartz), rutile, base-metal sulfides(pyrrhotite, pyrite, sphalerite, chalcopyrite, galena), and electrum. Stage II occur quartz, calcite and pyrite. Fluid inclusion data indicate that homogenization temperatures and salinities of stage I range from 145 to $309^{\circ}C$ and from 0.4 to 12.4 wt.% NaCl, respectively. It suggests that hydrothermal fluids were cooled and diluted with the mixing of meteoric water. The main deposition of base-metal sulfides and electrum occurred as a result of cooling and dilution at temperature between $200^{\circ}C$ and $300^{\circ}C$. Sulfur(9.3~10.8‰) isotope composition indicates that ore sulfur was mainly derived from a magmatic source as well as the host rocks. The calculated oxygen[-2.3~0.9‰(quartz: 0.3‰, 0.9‰, calcite: -2.3‰)] and hydrogen[-86~-76‰(quartz: -86‰, -82‰, calcite: -76‰)] isotope compositions indicate that hydrothermal fluids may be meteoric origin with some degree of mixing of another meteoric water for paragenetic time.

Genetic Consideration of Sericite Deposits Derived from Granitic Rocks in the Taebaegsan Region (태백산지역에 분포하는 화강암체 기원 견운모광상의 성인적 고찰)

  • Yeongweol sericite deposit of Gangwon Province is regarded as one of the sericite deposits derived from granitic rocks due to post-magmatic alkali metasomatism, and the other sericite deposit of the same origin is the Daehyun mine of Gyungbug Province. Sericite ores were originated from leucocratic granitic stocks of Cambrian-Triassic age which intruded the pegmatitic migmatite of the unknown age and granite of the Pre-cambrian age, respectivcly. Jangsan quartzite of the lowermost formations of the Paleozoic era, which played as the capping rock protected from the leakage of the hydrothermal solution. It is well known that those sericite deposits arc formed during formation of the geosyncline, and they are also situated in the margins of the Hambaeg Syncline. Leucocratic granites commonly contain pegmatites with tourmaline crystals, and are rich in potassium feldspars, and sodium plagioclase as well. Sericitized ores are mainly found as we go up to the higher elevations or to the margins of the stocks. And some of the Highest grade sericite ores show the monominerallic character composed of nearly pure sericite probably doc to the ultra greisenization. Chemical analysis shows higher $Na_{2}O$ and $K_{2}O$ contents $(2.00\sim7.03wt%)$ as the sericitizations arc preceded and they represent obvious greisenization. But low CaO contents $(0.05\sim4.51wt%)$ indicate that albitizations are so weak. Pyrophyllite of the Youngweol area is often accompanied by the sericite, indicating rather stronger thermal effect than the Daehyun mine. It is known that there are several Sn deposits originated from greisenization in the Taebaegsan region. And greisens are inclined to contain W, Mo and several REE's such as Be, Nb and Li, and so Taebaegsan region interbedded with lots of carbonate formations are still worthwhile to survey for those metallic deposits.

  • Purpose : We examined the relationships between the root curvatures of the mandibular third molars and their locations on the mandible, so that it might be contributed to elucidate the origin of the formation and development of the Korean mandibular third molar and help many clinicians to treat the mandibular third molars. Materials and Methods : Panoramic radiographs of 237 patients who had been treated at Korea University Ansan Hospital from March, 2000 to April, 2001 and had bilateral mandibular third molars were examined and analyzed. Results : There was no statistically significant difference between men and women (p<0.05). But there were significantly fewer subjects with bilateral curved roots in bilateral third molars regardless of their status (p<0.01) and were statistically significant differences between the groups with bilateral curved roots in the mandibular third molars with same status and with unilateral curved roots in the mandibular third molars with different status and between the groups with unilateral curved roots in the mandibular third molars with same status and with bilateral curved roots in the mandibular third molars with different status(p<0.05). Subjects with bilateral curved roots in the mandibular third molars with bilateral partial impacted status and with bilateral curved roots in the mandibular third molars with bilateral complete eruption status were significantly fewer (p<0.01) and subjects with the mandibular third molars with partial impacted and complete impacted status and with the mandibular third molars with complete erupted and partial impacted status had significantly fewer bilateral curved roots (p<0.05). Conclusions : When we postulate that it is genetic and environmental factors that affect the root formation, it seems that environmental factors have more effects on the formation of the mandibular third molars than on that of the mandibular first and second molars

  • Molecular cytogenetics allows the identification of unknown chromosome rearrangements, which is clinically useful in patients with mental retardation and/or development delay. We report on a 31-year-old woman with severe mental retardation, behavior development delay, and verbal performance delay. Conventional cytogenetic analysis showed a 46,XX,add(8)(p23.3) karyotype. To determine the origin of this unbalanced translocation, we performed array CGH and subtelomeric FISH. The results showed that the distal region of chromosome 8p was added to the terminal of chromosome 13q. This was confirmed the final result of 46,XX,der(8)t(8:13)(p23.3;q32.1)dn.

Lack of genetic divergence between Mogera wogura coreana from Korea and M. w. robusta from Northeastern China and adjacent Russia (Soricomorpha: Mammalia), reexamined from 12S rRNA and cytochrome b sequences

  • To reexamine taxonomic status of endemic Mogera wogura coreana from Korea, we first obtained partial 12S rRNA sequences (893 bp) and complete cytochrome b gene sequences (1140 bp) of this subspecies, and these sequences and partial cytochrome b sequences (402 bp) were compared to the corresponding haplotypes of M. wogura from East Asia, obtained from GenBank. The one of three 12S rRNA haplotypes in M. w. coreana was identical to one 12S rRNA haplotype of M. w. robusta from East Asia: 10 complete and 13 partial cytochrome b haplotypes of M. w. coreana formed a single clade with one complete and four partial cytochrome b haplotypes of M. w. robusta, respectively. We considered that M. w. coreana from Korea is an endemic subspecies with only morphological differences, although it is necessary to reexamine the subspecies status of M. w. coreana. Additionally, in the 12S rRNA and complete cytochrome b sequences, M. wogura from Japan was distinct from the two continental subspecies of M. w. coreana and M. w. robusta with average distances of 1.76 and 5.65%, respectively; insular M. wogura, with within-group distances of 2.09 and 4.38%, respectively, was also genetically more divergent than the mainland M. wogura, with within-group distances of 0.08 and 0.57%, respectively. Thus, we considered that insular M. wogura of Japan dispersed into neighboring East Asian continent, which is opposite to the traditional hypothesis on the origin of Japanese M. wogura.

Teratoma Formation in Immunocompetent Mice After Syngeneic and Allogeneic Implantation of Germline Capable Mouse Embryonic Stem Cells

  • Background: Embryonic stem cells (ESCs) have the potential to form teratomas when implanted into immunodeficient mice, but data in immunocompetent mice are limited. We therefore investigated teratoma formation after implantation of three different mouse ESC (mESC) lines into immunocompetent mice. Materials and Methods: BALB/c mice were injected with three highly germline competent mESCs (129Sv, BALB/c and C57BL/6) subcutaneously or under the kidney capsule. After 4 weeks, mice were euthanized and examined histologically for teratoma development. The incidence, size and composition of teratomas were compared using Pearson Chi-square, t-test for dependent variables, one-way analysis of variance and the nonparametric Kruskal-Wallis analysis of variance and median test. Results: Teratomas developed from all three cell lines. The incidence of formation was significantly higher under the kidney capsule compared to subcutaneous site and occurred in both allogeneic and syngeneic mice. Overall, the size of teratoma was largest with the 129Sv cell line and under the kidney capsule. Diverse embryonic stem cell-derived tissues, belonging to the three embryonic germ layers, were encountered, reflecting the pluripotency of embryonic stem cells. Most commonly represented tissues were nervous tissue, keratinizing stratified squamous epithelium (ectoderm), smooth muscle, striated muscle, cartilage, bone (mesoderm), and glandular tissue in the form of gut- and respiratory-like epithelia (endoderm). Conclusions: ESCs can form teratomas in immunocompetent mice and, therefore, removal of undifferentiated ESC is a pre-requisite for a safe use of ESC in cell-based therapies. In addition the genetic relationship of the origin of the cell lines to the ability to transplant plays a major role.

Production of Chimeric Mice Following Transgenesis of Multipotent Spermatogonial Stem Cells (유전자변형 다분화능 정원줄기세포를 이용한 키메라 생쥐의 생산)

  • Multipotent spermatogonial stem cells (mSSCs), derived from uni-potent SSC, are a type of reprogrammed cells with similar characteristics to embryonic stem cells (ESCs). The aim of this study was to evaluate the potential for transgenesis of mSSC derived from outbred mice and the production of transgenic animal by the mSSC-insertion into embryo. mSSCs, established from outbred mice (ICR strain) in the previous study, were maintained and then transfected with a lenti-viral vector expressing green fluorescent protein (GFP), CS-CDF-CG-PRE. Embryonic stem cells (ESCs) were derived from inbred transgenic mice (C57BL/6-Tg (CAG-EGFP)) and were used as an experimental control. Transfected mSSCs were well proliferated in vitro and maintained their characteristics and normal karyotype. Ten to twelve mSSCs and ESCs were collected and inserted into perivitelline space of 8-cell mouse embryos, and then transferred them into uteri of poster mothers after an additional 2-days of culture. Percentage of mSSC-derived offsprings was 4.8% (47/980) and which was lower than those (11.7% (67/572)) of ESC-derived ones (P<0.05). However, even though different genetic background of mSSC and ESC origin, the production efficiency of coat-colored chimeric offspring in mSSC group was not different when compared it with ESC (6.4% (3/47) vs. 7.5% (5/67)). From these results, we confirmed that mSSC derived from outbred mice has a pluripotency and a potential to produce chimeric embryos or mice when reaggregatation with mSSC is performed.
