Studies on the Triglyceride Composition of Job's Tears (율무 지질의 Triglyceride 조성에 관한 연구)

  • Lipids of Job's tears (Yullmoo) were extracted by the mixture of chloroform: methanol (2:1, v/v) and from these lipids, triglycerides were separated by column chromatography (Sephadex G-25 column) and then confirmed by thin layer chromatography (TLC) and reconfirmed by gas chromatography (GC). The triglycerides were fractionated into 6 groups on the basis of their partition numbers (PN) by reverse phase high performance liquid chromatography (HPLC) on a column Shimpack CLC-ODS using tetrahydrofuran-acetonitril (25:75) mixture as a solvent. Each of these collected fractions gave one to two peaks in the GC chromatograms according to the acyl carbon number (CN) of the triglyceride and fatty acid composition of the triglycerides were also analyzed by GC. From the results, the major triglycerides of Job's tears lipids were estimated to be C18:1 C18:2 C18:2 (OLL, 38.2%), C16:0 C18:2 C18:3 (PLLn, 15.7%), C18:1 C18:1 C18:2 (OOL, 12.6%).

Visualization of Artificially Deposited Submicron-sized Aerosol Particles on the Surfaces of Leaves and Needles in Trees

  • To understand the effect of aerosols on the growth and physiological conditions of trees in forests, it is important to know the state of aerosols that are deposited on the surface of the leaves or needles. In this study, we developed methods of visualization of submicron-sized aerosols that were artificially deposited from the gas-phase or liquid phase onto tree leaves or needles in trees. Firstly, we used field-emission scanning electron microscopy (FE-SEM) to observe black carbon (BC) particles that were artificially sprayed onto the leaves or needles. The distribution of BC particles deposited on the leaves and needles were distinguished based on the size and morphological features of the particles. The distribution and agglomerates size of BC particles differed between two spraying methods of BC particles employed. Secondly, we tried to visualize gold (Au) particles that were artificially sprayed onto the leaves using energy dispersive X-ray spectrometry (EDX) coupled to FE-SEM. We detected the Au particles based on the characteristic X-ray spectrum, which was secondarily generated from the Au particles. In contrast to the case of BC particles, the Au particles did not form agglomerates and were uniformly distributed on the leaf surfaces. The present results show that our methods provide useful information of adsorption and/or behavior of fine particles at the submicron level on the surface of the leaves.

  • After recycle of spent materials has been optimised, there remains a proportion of waste which must be dealt with in the most environmentally friendly manner available. For materials such as municipal waste, clinical waste, toxic waste and special wastes such as tyres, incineration is often the most appropriate technology. The study of incineration must take a process system approach covering the following aspects: ${\bullet}$ Collection and blending of waste, ${\bullet}$ The two stage combustion process, ${\bullet}$ Quenching, scrubbing and polishing of the flue gases, ${\bullet}$ Dispersion of the flue gases and disposal of any solid or liquid effluent. The design of furnaces for the burning of a bed of material is being hampered by lack of an accurate mathematical model of the process and some semi-empirical correlations have to be used at present. The prediction of the incinerator gas phase flow is in a more advanced stage of development using computational fluid dynamics (CFD) analysis, although further validation data is still required. Unfortunately, it is not possible to scale down many aspects of waste incineration and tests on full scale incinerators are essencial. Thanks to a close relationship between SUWIC and Sheffield Heat&Power Ltd., an extended research programme has been carried out ar the Bernard Road Incinerator plant in Sheffield. This plant consists of two Municipal(35 MW) and two Clinical (5MW) Waste Incinerators which provide district heating for a large part of city. The heat is distributed as hot water to commercial, domestic ( >5000 dwelling) and industrial buildings through 30km of 14" pipes plus a smaller pipe distribution system. To improve the economics, a 6 MW generator is now being added to the system.

Continuous Process for the Etching, Rinsing and Drying of MEMS Using Supercritical Carbon Dioxide (초임계 이산화탄소를 이용한 미세전자기계시스템의 식각, 세정, 건조 연속 공정)

  • The previous etching, rinsing and drying processes of wafers for MEMS (microelectromechanical system) using SC-$CO_2$ (supercritical-$CO_2$) consists of two steps. Firstly, MEMS-wafers are etched by organic solvent in a separate etching equipment from the high pressure dryer and then moved to the high pressure dryer to rinse and dry them using SC-$CO_2$. We found that the previous two step process could be applied to etch and dry wafers for MEMS but could not confirm the reproducibility through several experiments. We thought the cause of that was the stiction of structures occurring due to vaporization of the etching solvent during moving MEMS wafer to high pressure dryer after etching it outside. In order to improve the structure stiction problem, we designed a continuous process for etching, rinsing and drying MEMS-wafers using SC-$CO_2$ without moving them. And we also wanted to know relations of states of carbon dioxide (gas, liquid, supercritical fluid) to the structure stiction problem. In the case of using gas carbon dioxide (3 MPa, $25^{\circ}C$) as an etching solvent, we could obtain well-treated MEMS-wafers without stiction and confirm the reproducibility of experimental results. The quantity of rinsing solvent used could be also reduced compared with the previous technology. In the case of using liquid carbon dioxide (3 MPa, $5^{\circ}C$, we could not obtain well-treated MEMS-wafers without stiction due to the phase separation of between liquid carbon dioxide and etching co-solvent(acetone). In the case of using SC-$CO_2$ (7.5 Mpa, $40^{\circ}C$), we had as good results as those of the case using gas-$CO_2$. Besides the processing time was shortened compared with that of the case of using gas-$CO_2$.

High-Performance Liquid Chromatographic Determination of Phenothrin and Silafluofen Residues in Crops with Mass Spectrometric Confirmation (고성능 액체크로마토그래피와 질량분석법을 이용한 농산물 중 Phenothrin 및 Silafuofen의 정밀 잔류분석법 개발)

  • A high-performance liquid chromatographic (HPLC) method was developed to determine residues of phenothrin and silafluofen, known as synthetic pyrethroids, in agricultural commodities. Insecticide residues were extracted with acetone from representative samples of four crops which comprised rice, apple, pepper and cabbage. The extract was purified serially by liquid-liquid partition and Florisil column chromatography. For rice and pepper samples, acetonitrile/n-hexane partition was additionally adopted to remove nonpolar interferences. Reversed phase HPLC using an octadecylsilyl column was successfully applied to separate two phenothrin isomers and silafluofen from sample co-extractives. Intact parent compounds were sensitively detected by ultraviolet absorption at 226 nm. Recovery experiment at the quantitation limit validated that the proposed method could apparently determine phenothrin and silafluofen residues at 0.02 and 0.01 mg/kg, respectively. Mean recoveries of phenothrin and silafluofen from four crop samples fortified at three levels in triplicate were in the range of 82.4~109.8% and 83.7~109.8%, respectively. Relative standard deviations of the analytical method were all less than 10%, irrespective of crop types and spiking levels. A selected-ion monitoring (SIM) LC/mass spectrometry (MS) with electrospray ionization was provided to confirm the suspected residue of phenothrin, even though no sufficient ionization of silafluofen was obtained. Both phenothrin and silafluofen could be successfully confirmed by gas chromatography/MS SIM with electron impact at 70 eV. The proposed method is sensitive, repeatable and rapid enough to apply to officially routine inspection of agricultural products.

Absorption Characteristics of Carbon Dioxide by Water-lean Diethylenetriamine Absorbents Mixed with Physical Solvents (물리 흡수제를 포함한 디에틸렌트리아민(Diethylenetriamine) 저수계 흡수제에서의 이산화탄소 흡수 특성)

  • In this work, N-methyl-2-pyrrolidone (NMP) was added into diethylenetriamine (DETA) aqueous solution for high $CO_2$ loading via phase splitting of absorbents during $CO_2$ absorption. Immiscible two phases were formed in the range of more than 30 wt% of NMP in 2 M DETA + NMP + water absorbents because of low solubility of DETA-carbamate in NMP solution. As the composition of NMP in the absorbents increased, the difference of $CO_2$ loading between each phase increased and the volume of bottom phase decreased. In $CO_2$ absorption in packed column by 2 M DETA + NMP + water absorbents, the absorption rate decreased in the range of more than 40 wt% of NMP. It is due to the increasing of mass transfer resistance in liquid film of absorbents at the high concentration of NMP. DETA + NMP + water absorbent is expected as the promising one for reducing the regeneration energy of absorbents according to volume reduction of $CO_2-rich$ phase.

Development and Performance Tests of the Waste Water Diffusers using Acoustic Resonance and Oscillatory Pulsation (음향공진과 맥진동 현상을 이용한 폐수처리용 산기관 개발 및 성능시험)

  • Using the acoustic resonances and oscillatory pulsations considered as the branch of wave technologies, the concept of the acoustic resonance diffusers for waste water treatment which maximize the oxygen transfer efficiency in gas-liquid two phase medium have been proposed, and studies for the principles and performance tests were accomplished. Besides, the design concepts for the low pressure Helmholtz resonator, cylinder and annular type reflection resonator and combined type resonance system have been implemented. The acoustic resonance energy which can speed up the mass transfer process increase the oxygen transfer efficiency, and periodic pulsations generated from the instability of air jet from nozzle make very small air bubbles. Then, the annular type jet resonator(AJR) applying these two principles successfully was evalulated as the most promising device and also the efficiency showing $20{\sim}30%$ better than conventional diffusers has been verified experimentally.

Verification of SPACE Code with MSGTR-PAFS Accident Experiment (증기발생기 전열관 다중파단-피동보조급수냉각계통 사고 실험 기반 안전해석코드 SPACE 검증)

  • The Korean nuclear industry developed the SPACE (Safety and Performance Analysis Code for nuclear power plants) code and this code adpots two-phase flows, two-fluid, three-field models which are comprised of gas, continuous liquid and droplet fields and has a capability to simulate three-dimensional model. According to the revised law by the Nuclear Safety and Security Commission (NSSC) in Korea, the multiple failure accidents that must be considered for accident management plan of nuclear power plant was determined based on the lessons learned from the Fukushima accident. Generally, to improve the reliability of the calculation results of a safety analysis code, verification work for separate and integral effect experiments is required. In this reason, the goal of this work is to verify calculation capability of SPACE code for multiple failure accident. For this purpose, it was selected the experiment which was conducted to simulate a Multiple Steam Generator Tube Rupture(MSGTR) accident with Passive Auxiliary Feedwater System(PAFS) operation by Korea Atomic Energy Research Institute (KAERI) and focused that the comparison between the experiment results and code calculation results to verify the performance of the SPACE code. The MSGR accident has a unique feature of the penetration of the barrier between the Reactor Coolant System (RCS) and the secondary system resulting from multiple failure of steam generator U-tubes. The PAFS is one of the advanced safety features with passive cooling system to replace a conventional active auxiliary feedwater system. This system is passively capable of condensing steam generated in steam generator and feeding the condensed water to the steam generator by gravity. As the results of overall system transient response using SPACE code showed similar trends with the experimental results such as the system pressure, mass flow rate, and collapsed water level in component. In conclusion, it could be concluded that the SPACE code has sufficient capability to simulate a MSGTR accident.

Fluid Inclusion Studies on the Wolak Tungsten-Molybdenum Deposits, Korea (월악 중석-몰리브덴 광상의 유체포유물 연구)

  • The Wolak tungsten-molybdenum deposits are tungsten-molybdenum bearing quartz veins which filled the fractures in Pre-Cambrian pebble-bearing calcareous hornfels, hornfels and Cretaceous granite. There are two vein groups in this mine, Dongsan vein group in the west and Kwangcheon vein group in the east. The ore minerals are wolframite, scheelite, molybdenite, native bismuth, bismuthinite, pyrite, arsenopyrite, chalcopyrite, cubanite, stannite, pyrrhotite, sphalerite, galena, marcasite, Pb-Bi sulfosalt and ilmenite. Quartz, calcite, beryl, fluorite, muscovite, rhodochrosite and siderite are gangue minerals. Fluid inclusion studies were carried out for the quartz, beryl, scheelite, early and late fluorite. Fluid inclusion studies reveal that liquid-gas inclusions are most common and occur in all of the minerals examined. Filling degree of the inclusions in the late fluorite is much higher than that of the inclusions in quartz and early fluorite. Liquid $CO_2$ bearing liquid-gas inclusions occur in quartz and early fluorite. Liquid, gas and solid phase inclusions occur in quartz, beryl and scheelite. Salinities of inclusions in quartz and beryl from Dongsan vein group range from 3.9 to 8.0, from 5.3 to 7.7 wt.% NaCl equivalent respectively. Salinities in the late fluorite range from 1.5 to 3.2 wt.% NaCl equivalent. In Kwangcheon vein group salinities range from 3.9 to 9.6 wt.% NaCl equivalent in quartz, from 2.8 to 7.3 wt.% NaCl equivalent in early fluorite, from 1.3 to 1.5 wt.% NaCl equivalent in late fluorite. Homogenization temperatures of inclusions range from $239^{\circ}$ to higher than $360^{\circ}C$ in quartz, over $360^{\circ}C$ in scheelite, from $288^{\circ}C$ to higher than $360^{\circ}C$ in beryl, and from $159^{\circ}$ to $202^{\circ}C$ in late fluorite of the Dongsan vein group. In Kwangcheon vein group, homo genization temperatures of inclusions range from $240^{\circ}C$ to higher than $360^{\circ}C$ in quartz and from $240^{\circ}$ to $328^{\circ}C$ in early fluorite. As a whole, in Dongsan and Kwangcheon vein groups it seems that there are no distinct differences in mineralogy, salinities and homogenization temperatures. No distinct variations in homogenization temperatures are revealed through about 300 m vertically in both district. The faint trend of increase in salinities in the lower level can be detected. The salinity, $CO_2$ content and the temperature of ore fluid were much higher in the early vein stage and then dropped off in the late stage of mineralization as represented by the quartz and fluorite inclusion data.

Influence of Temperature and pH on Fermentation Pattern and Methane Production in the Rumen Simulating Fermenter (RUSITEC)

  • An experiment was conducted to study the effect of temperature and pH on in vitro nutrient degradability, volatile fatty acid profile and methane production. The fermenter used was the semi-continuous system, known as the rumen simulation technique (RUSITEC). Sixteen cylinders were used at one time with a volume of 800 ml, the dilution rate was set at 3.5%/hour, the infused buffer being McDougall's artificial saliva. Basal diet (9.6 g DM) used in RUSITEC consisted of (DM) 6.40 g Timothy hay, 1.86 g crushed corn and 1.34 g soybean meal. The food for the fermentation vessel was provided in nylon bags, which were gently agitated in the liquid phase. The experiment lasted for 17 d with all the samples taken during the last 5 d. Treatments were allocated at random to four vessels each and were (1) two temperature levels of $39^{\circ}C$ and $41^{\circ}C$ (2) two pH levels of 6.0 and 7.0. The total diet contained ($g\;kg^{-1}$ DM) 957 OM, 115 CP and $167MJ\;kg^{-1}$ (DM) GE. Although increase in temperature from $39^{\circ}C$ to $41^{\circ}C$ reduced degradation of major nutrients in vitro, it was non-significant. Interaction effect of temperature with pH also reflected a similar trend. However, pH showed a significant (p<0.05) negative effect on the degradability of all the nutrients in vitro. Altering the in vitro pH from 7 to 6 caused marked reduction in DMD from 60.2 to 41.8, CPD from 76.3 to 55.3 and GED from 55.3 to 35.1, respectively. Low pH (6) depressed total VFA production (61.9 vs. 34.9 mM) as well as acetate to propionate ratio in vitro (from 2.0 to 1.5) when compared to pH 7. Compared to pH 7, total gas production decreased from 1,841 ml to 1,148 ml at pH 6, $CO_2$ and $CH_4$ production also reduced from 639 to 260 ml and 138 to 45 ml, respectively. This study supported the premise that pH is one of the principal factors affecting the microbial production of volatile fatty acids and gas. Regulating the ruminal pH to increase bacterial activity may be one of the methods to optimize VFA production, reduce methane and, possibly, improve animal performance.
