Frequency, Distribution of Stomata and Relationship Between Stomatal Frequency and Other Characters of Wheat Cultivars (소맥엽신의 기공분포 및 기공수와 타형질과의 관계)

  • One hundred winter wheat cultivars obtained from the world collections in Korea were planted in 1982. Stomatal frequency of flag leaf of these cultivars were measured, and among these 21 cultivars which showed different stomatal frequency were selected to investigate the relationship of their stomatal frequencies and other characters such as culm length, heading date and characters related to leaf. Stomatal frequency of flag leaf of 100 cultivars different significantly, ranging from 51 to 90 stomata mm$^{-2}$ for the adaxial and 35 to 65 for the abaxial surfaces of leaf. Stomatal frequency was always greater on the adaxial than on the abaxial surface. Mean ratios (adaxial/abaxial) were 1.5 for the flag leaf and 1.3 for the from second to fouth leaves. Among 21 cultivars examined, stomatal size which measured guard cell length varied from 38.5 to 46.6$\mu$ for adaxial surface and from 39.4 to 49.4$\mu$ for abaxial. A negative correlation was obtained between stomatal frequency and its size. Stomatal frequency decreased from flag (L$_1$) to lower leaves, progresively. Stomatal frequency on L$_1$ sheath was compared to those on L$_3$ and L$_4$ abaxial surface of leaf. More stomatal frequency on top and center of adaxial surface showed than that on basal part of a leaf but the abaxial surface resulted in reverse of adaxial one. Cultivars with high stomatal frequency on flag leaf showed higher stomatal frequency on the other leaves and sheaths. Positive correlation between stomatal frequency and other characters such as, culm length, heading date and chlorophyll content showed statistical significance, whearas it revealed the negative correlation of stomatal frequency with leaf weight, leaf area and stomatal aperture. However, no relation appeared between stomatal frequency and leaf vein.

Chromosome Redundancy and Tree Phenotype Variation in Autotetraploid Trifoliate Orange (동질 사배체 탱자에서 염색체 배가와 수체 표현형의 변이)

  • The study was conducted to investigate the possibility that epigenetic DNA methylation causes tree phenotypic variation in autotetraploids through evaluating the phenotypic variation and DNA methylation in autotetraploids occurred spontaneously from diploid trifoliate orange. Chromosome analysis confirmed that fourteen trifoliate orange trees of selected by flow cytometry were tetraploids (2n = 4X = 36) without any aneuploids. Chromomycin A3 staining determined that these trees were all autotetraploid with doubled chromosome set. Tree phenotypes, such as tree height and width, branching number, length, and angle, internode length, and leaf characteristics, varied in the autotetraploids. Chlorophyll indices were diverse in the autotetraploids, but photosynthetic rates were not significantly different. In addition, a wide range of variation was observed in stomatal density and guard cell length. Analysis of global cytosine DNA methylation showed that there was a variation of the methylation level in autotetraploids. More than half of 14 autotetraploids had at least 2 times higher methylation level than diploid trifoliate orange. The results indicate that tree phenotypic variation in autotetraploids might be related to global DNA methylation for reducing gene redundancy.

A Combined Call Control Algorithm based on Moving Speed and Bandwidth in Hierarchical Cellular Systems (계층셀 구조에서 이동 속도와 대역폭을 고려한 복합 호 처리 알고리즘)

  • Recently, the demand for mobile communications and multimedia services has rapidly increased so that conventional cellular system cannot fulfill the requirement of users (capacity and QoS) any more. Therefore, the hierarchical cellular system has been suggested in order to guarantee the QoS and to admit large population of users. IMT-2000 adopts the hierarchical cellular structure, which requires a call control algorithm capable of manipulating and utilizing the complicated structure of hierarchical cellular structure with handiness and efficiency. In this thesis, as an improvement of conventional combined algorithm, a new call control algorithm considering the moving speed of terminal and bandwidth is suggested. This algorithm employs buffers and guard channels to reduce the failure rate. Also, this algorithm considers the moving speed of terminal and bandwidth to elevate the efficiency. Furthermore, calls are handled separately according to the moving speeds of terminal and bandwidths to improve the QoS and reduce the handover rate. As an evaluation of the suggested algorithm, a model hierarchical cellular system is constructed and simulations are conducted with various types of traffic. As the result of the simulations, such indices as block rate, drop rate, channel utilization, and the number of inter layer handovers are examined to demonstrate the excellency fo the suggested algorithm.

The Nuclear DNA Content and Histological Characteristics of Triploid Poplars Grown In Vitro (현사시나무 3배체의 핵 DNA 함량 및 조직학적 특성)

  • Herein we analyzed the nuclear DNA content and the histological characteristics of the triploid of the 'Hyunsasi' (Populus alba ${\times}$ P. glandulosa $F_1$) which were developed for biomass production and molecular breeding research. The flow cytometric analysis showed that the nuclear DNA content of the 3 triploids were 1.6 times greater than those of the diploid. In terms of histological characteristics, the cross-section area of the stem of 'Line-18' was 1.6 times larger than that of the diploid. The area of pith, and cortex and phloem of the stem of 'Line-18' was also 1.6 and 2.0 times larger than that of the diploid, respectively. Moreover, the length and area of guard cell of 'Line-18' was 1.2 times larger than that of the diploid. These results helps to understand the cytological characteristics of the triploid poplar clones, and further investigations in the growth rate and wood properties of the triploids growing in the field will determine whether the triploid poplars are good candidates for molecular breeding programs and for the improvement of industrial biomass productivity.

An Intra Prediction Hardware Design for High Performance HEVC Encoder (고성능 HEVC 부호기를 위한 화면내 예측 하드웨어 설계)

  • In this paper, we propose an intra prediction hardware architecture with less processing time, computations and reduced hardware area for a high performance HEVC encoder. The proposed intra prediction hardware architecture uses common operation units to reduce computational complexity and uses $4{\times}4$ block unit to reduce hardware area. In order to reduce operation time, common operation unit uses one operation unit to generate predicted pixels and filtered pixels in all prediction modes. Intra prediction hardware architecture introduces the $4{\times}4$ PU design processing to reduce the hardware area and uses intemal registers to support $32{\times}32$ PU processmg. The proposed hardware architecture uses ten common operation units which can reduce execution cycles of intra prediction. The proposed Intra prediction hardware architecture is designed using Verilog HDL(Hardware Description Language), and has a total of 41.5k gates in TSMC $0.13{\mu}m$ CMOS standard cell library. At 150MHz, it can support 4K UHD video encoding at 30fps in real time, and operates at a maximum of 200MHz.

  • AL1-gene, necessary for the replication of the genome of a gemini virus TGMV, was inserted in the opposite direction to the promoter CaMV35S resulting in the construction of a plant transformation binary vector pAR35-2. The vector pAR35-2 contains the chimeric gene cassette involving the duplicated promoter CaMV35S, opposite direction of AL1-gene fusioned with hygromycin resistant gene, and the gene cassette of the neomycin phosphotransferase II gene. The plasmid was transferred to tobacco and tomato plants by leaf disk infection via Agrobacterium. The transgenic plants were selected and grown on the MS-agar medium containing kanamycin and hygromycin. The shoots induced from the calli were regenerated to the whole transgenic plants. The antisense AL1-gene was detected in the genomic DNA isolated from the leaves by using the PCR mediated Southern blot analysis. The expression of the antisense AL1-gene was also observed using the RT-PCR mediated Southern blot analysis. The observation of chloroplasts in guard cell pair indicated that the transgenic tomato plants were diploid.

Stomatal Closure due to Water Stress in Plants (수분 스트레스에 의한 식물의 기공 닫힘)

  • The environmental stress that plants are most susceptible to is water stress. Abscisic acid (ABA) is a plant hormone synthesized by plants to counteract environmental stress. The role of stomata in plants is to allow the synthesis of sucrose by absorbing CO2, which greatly affects photosynthetic activity. In addition, stomata are pathways for transpiration, which releases H2O and help establish a water potential gradient that allows plant roots to continuously absorb water and inorganic substances from the soil. Plants have a mechanism to minimize water loss by closing their stomata when exposed to water-stressed environments. The most well-studied hypothesis concerning the mechanism of stomatal closure is the response to water stress. When a plant receives sufficient water, its stomata open during the day and close at night due to its circadian rhythm. In addition, stomatal closure occurs when the concentration of CO2 in the intercellular space increases. However, the mechanism of stomatal closure due to circadian rhythm and increased CO2 concentration in the intercellular space is not well understood. When plants undergo water stress, the increased concentration of ABA in the guard cell cytoplasm induces an increase in Ca2+ concentration, resulting in cytoplasmic depolarization. As a result, the outward K+-channel of the tonoplast and the slow-type anion channels SLAC1 and SLAH3 are activated, releasing K+, Cl-, and malate2-, causing the stomata to close. Therefore, in this paper, the mechanism of stomatal closure caused by water stress was investigated.

Taxonomic significance of the leaf micromorphology in the tribe Sorbarieae (Spiraeoideae: Rosaceae) (쉬땅나무족(조팝나무아과: 장미과) 잎표피 미세형태학적 형질의 분류학적 유용성)

  • A comparative study of leaf epidermal microstructures in the tribe Sorbarieae (Adenostoma: 3 spp., Chamaebatiaria: 1 sp., Sorbaria: 11 spp., Spiraeanthus: 1 sp.) including related genera Gillenia (2 spp.) and Lyonothamnus (2 spp.) was carried out using scanning electron microscopy (SEM) in order to evaluate their significance in taxonomy. The leaves of Adenostoma, Chamaebatiaria, and Spiraeanthus were amphistomatic, whereas Gillenia, Lyonothamnus, and Sorbaria were hypostomatic. The size range of the guard cells is $7.84-48.7{\times}5.86-38.6{\mu}m$; the smallest one was found in Sorbaria tomentosa var. tomentosa ($7.84-11.8{\times}6.84-10.5{\mu}m$), while the largest measured example was Adenostoma fasciculatum var. obtusifolium ($30.3-48.7{\times}18.8-38.6{\mu}m$). Anomocytic stomata complex were the most frequent type (rarely cyclocytic), with usually both anomocytic and actinocytic types occurring in one leaf. On the surfaces, both the adaxial and abaxial anticlinal walls of the subsidiary cells vary (e.g., straight/curved, undulate, sinuate). Four types (unicellular non-glandular trichome, stellate, glandular trichome, pustular glandular trichome) of trichomes are found in the leaves. The epicuticular wax can be divided two types: membraneous platelets (Lyonothamnus) and platelets (Sorbaria arborea var. arborea, S. arborea var. subtomentosa, S. kirilowii, S. tomentosa var. tomentosa, Spiraeanthus schrenkianus). The trichome diversity (in particular, stellate, gland) and the existence of epicuticular wax may have taxonomic significance, although the leaf epidermal micromorphological characteristics do not provide synapomorphy in this tribe. These leaf micromorphological features are most likely better understood in the Sorbarieae when used in conjunction with external morphological characters.

Leaf epidermal microstructure of the genus Scopolia Jacq. s.l. (Solanaceae-Hyoscymeae) and its systematic significance (광의의 미치광이풀속(Scopolia Jacq. s.l., 가지과-Hyoscymeae족)의 잎표피 미세구조와 이의 계통분류학적 중요성)

  • To examine the leaf epidermal microstructure of three genera (Scopolia s.s., Anisodus, AtroPanthe, including Przewalskia as an outgroup) in the genera Scopolia Jacq. s.l., leaves of 10 species (37 specimens) were investigated by the light microscopy (LM) and scanning electron microscopy (SEM). The stomata of studied taxa were 'amphistomatic type' and the size (guard cell) range was $18-64{\times}11-48{\mu}m$. The size of stomata is slightly differed from between the taxa; the smallest size of stomata were found in the monotypic genus, Przewalskia ($24-27{\times}16-17{\mu}m$), on the other hand the largest one was found in Anisodus carniolicoides ($62-64{\times}43-48{\mu}m$). The stomatal complex was mostly anomocytic (in Scopolia s.s., Anisodus taxa : A. luridus, A. carniolicoides, A. acutangulus) and sometimes anisocytic (in Anisodus tanguticus, Przewalskia, Atropanthe). The stomata is mostly crescent in shape, but rarely circular, especially in Przewalskia tangutica. The shapes of epidermal cells are similar in both adaxial and abaxial sides, and mostly undulate/sinuate polygonal anticlinal wall, but rarely arched in Przewalskia tangutica. The epicuticular wax was not well developed in most studied taxa, except Anisodus tanguticus which is well developed cuticular striae around the stomatal complex. The elongate-headed glandular trichomes were found in Scopolia s.s. and Przewalskia. While the taxa of Anisodus and Atropanthe have not any trichomes (i. e., glabrous), except Anisodus luridus, which has simple or sometimes branched (dendritic- type) non-glandular trichome. Finally, the systematic and ecological significance of the leaf micromorphological features (stomata complex, trichome, etc.) in identification and elucidation of Scopolia s.l. including Przewalskia, especially between or within the genera including among the species is also discussed.

Short-term changes of phytoplankton communities after nutrient addition and establishment of stable mass culture condition to prepare the type approval test of USCG Phase-II in mesocosm enclosure (메소코즘에서 USCG phase-II 형식승인 대비 영양염 첨가에 따른 식물플랑크톤 대량 배양조건 확립 및 군집구조의 단주기변화)

  • In order to prepare for the type approval test for the United States Coast Guard (USCG) Phase-II of Ballast Water Treatment System (BWTS), a phytoplankton mass culture was conducted in a mesocosm enclosure. We evaluated the response of the phytoplankton community after nutrient addition (+N, +P, and +NP) and investigated the development of the species with increasing culture time. After nutrient dosing, the phytoplankton population significantly (p < 0.05) increased from day 1 to day 3, depending on the nutrient treatments In particular, the specific growth rate of the phytoplankton community in the case of +NP treatment and + N treatment were estimated to be $2.47d^{-1}$ and $1.98d^{-1}$, respectively. The phytoplankton population density in the case of + NP treatment was approximately 50 times higher than that of the control group, suggesting that these treatments could be useful for mass culturing phytoplankton (> 75% of natural community) for the approval regulation of USCG Phase-II. In the phytoplankton community of the mesocosm, Pseudo-nitzchia spp. dominated in the logarithmic growth phase. The cell density decreased significantly (p < 0.05) with increasing time, coinciding with the nutrient limitation. At that time, the dominance of Pseudo-nitzchia spp. shifted to that of Cylindrotheca closterium. Therefore, the optimum nutrient concentration ($N:30{\mu}M$, $P:3{\mu}M$) and reasonable harvesting time (after 3 days in summer) found in this study for the mass culturing of phytoplankton may be helpful to meet the USCG Phase-II biological criteria to be used in BWTS.
