Glutathione Contents in Various Plant Cell Lines (다양한 식물배양세포주의 Glutathione 함량)

  • We investigated the levels of glutathione (GSH) and its oxidized form (GSSG) in 24 cell lines derived from various plant species to understand the antioxidative mechanism in plant cell cultures. The total glutathione content was 98$\pm$27 $\mu$g/g cell fresh wt, showing a slight difference in plant species. The average contort of GSH and GSSG was 72$\pm$20 and 26$\pm$10 $\mu$g/g cell fresh wt, respectively. The average GSH content in plant cell lines occupies approximately 73% in total glutathione. During the suspension cultures of Scutellaria baicalensis, one of the plant species we tested, the GSH content decreased in proportion to the cell growth during the exponential growth stage, showing the low level at the stationary growth stage (84 $\mu$g/g cell fresh wt), whereas the GSSG content increased to the stationary growth stage (31 $\mu$g/g cell fresh wt). The results suggested that the ratio of GSH and GSSG should be involved in the cell growth and antioxidative mechanism in cultured cells.

  • The radioprotective effects of white ginseng and Entomopathogenic Fugi Extract on liver damage induced by X-ray were investigated. To one group of ICR male mice were given white ginseng(150 mg/kg/day for 7days, orally) and Entomopathogenic Fugi (200 mg/kg/day for 7days, orally) before X-ray irradiation. To another group were irradiated by 5 Gy(1.01 Gy/min) dose of X-ray. Contrast group were given with saline(0.1 ml). The levels of reduced(GSH) and oxidized(GSSG) glutathione in liver tissue were measured. The ratio of GSSG/total GSH was significantly decreased in the white ginseng and Entomopathogenic Fugi (200 mg/kg/day)(150 mg/kg/day) groups than irradiation group.

  • Purpose: The radioprotective effects of white and fermented ginseng on liver damage induced by $^{60}Co\;{\gamma}$-ray were investigated. Materials and Methods: To one group of ICR male mice were given white(150 mg/kg/day for 7 days, orally) and fermented ginseng(150 mg/kg/day for 7 days, orally) before $^{60}Co\;{\gamma}$-ray irradiation. To another group were irradiated by 5 Gy(1.01 Gy/min) dose of $^{60}Co\;{\gamma}$-ray. Contrast group were given with saline(0.1 mL). The levels of reduced(GSH) and oxidized(GSSG) glutathione in liver tissue were measured. Results: In the fermented(150 mg/kg) and white ginseng(150 mg/kg) groups than irradiation group, the GSH levels were significantly increased, but the GSSG levels were significantly decreased. The ratio of GSSG/total GSH was significantly decreased in the fermented(150 mg/kg) and white ginseng(150 mg/kg) groups than irradiation group. Conclusion: In the fermented(150 mg/kg) groups than white ginseng(150 mg/kg) groups the GSH levels were significantly increased. The radioprotective effects of fermented(150 mg/kg) groups than white ginseng(150 mg/kg) groups were increased.

  • The effects of red ginseng component (water extracts, alcohol extracts, lipophilic extracts, total saponins, panaxadiol and panaxatriol) administration on glutathione (GSH) and lipid peroxidation levels in mice were investigated. 20~25 g ICR mice which were pretreated with water extracts (50 mg/kg), alcohol extracts (50 mg/kg), lipophilic extracts (50 mg/kg), total saponins (50 mg/kg), panaxadiol (50 mg/kg) and panaxatriol (50 mg/kg) for 15 days. The ability of red ginseng component to protect against oxidative damage to the mouse liver was examined by determining the level of lipid peroxidation (MDA), glutathione, and the activities of glutathione peroxidase (GPX). The GSH level was raised by all the ginseng component, but the GSSG level was lowered ]argely by all the ginseng component. The ratio of GSSG/total GSH was decreased because the level of GSSG was decreased more than that of GSH. Finally, the lipid peroxidation (MDA) level was the lowest in lipophilic extracts and panaxadiol nest. In conclusion, the order of effectiveness of anti-oxidants was to be lipophilic extracts>panaxadiol>total saponins.

  • The effects of acute and chronic ethanol administration on hepatic and cerebellar glutathione (GSH) statuses and lipid peroxide levels in rats were investigated. In the liver, chronic ethanol feeding (6.9 g/kg, per day) as 10% (v/v) drinking water for 4 weeks produced a slight decrease of total GSH and an increase in the ratio of GSSG/total GSH without change of GSSG (oxidized GSH). Lipid peroxide level however was not modified. Many other studies have shown the acute ethanol loading effect in the rat liver, that is moderate decrease of total GSH and elevation of lipid peroxide level. Relating to this, it was observed that total GSH in the plasma obtained from post. hepatic inferior vena cava was increased by acute ethanol injection (50 mmol/kg, i.p.). This increased hepatic efflux of GSH into blood, in addition to the promoted antioxidative utilization of GSH, could be suggested as one of the possible reasons for the decrease of hepatic GSH induced by ethanol load. In the cerebellum, acute ethanol load did not change the total GSH and GSSG, but increased the lipid peroxidation rate. In the chronic, neither GSH pattern nor lipid peroxidation rate was changed.

The Effect of Kimchi Intake on Free Radical Production and the Inhibition of Oxidation in Young Adults and the Elderly People (김치가 청장년 및 노인의 유리기 생성 및 산화억제에 미치는 영향)

  • This study was performed to investigate the effect of kimchi intake on free radical and oxidative substance production in young adults and the elderly. Daily kimchi intake by people in their twenties (n = 93, 20 to 29 years old) and over sixty-five (n = 143, over 65 years old) in M city were surveyed and blood was drawn to analyze the free radicals in their plasma. The average amount of kimchi intake by the subjects was 115.8 $\pm$ 91.7 g. The amount of kimchi intake of those in their twenties (106.1 $\pm$ 80.6 g) was significantly lower than that of those over sixty-five (125.5 $\pm$ 102.9 g, p < 0.05). Concentrations of total free radicals and OH radicals were 27 and 33% greater respectively, in those over sixty-five than in those in their twenties, indicating that more free radicals were produced by the older group. The concentration of GSH was not signiacantly different in the two groups, but that of GSSG in the over sixty-five age group was 53% greater than in the twenties group, which resulted in a 35% reduction in GSH/GSSG in the elderly group. TBARS concentration in the over sixty-five group was 26% greater than that of the twenties group. In order to see the effect of kimchi intake on free radical production, subjects in same age group were divided into two sub groups-the mean over and the mean under groups-according to the average amount of kimchi intake, which was 115.8 g. The total free radicals, the OH radicals, the GSH, the GSSG, and the GSH/GSSG in the twenties group were not significantly different in the two kimchi intake groups. However, those in the over sixty-five group were significantly different. The concentration of total free radicals and OH radicals of the mean over group were 21 and 26% lower respectively, than those of the mean ova. group (p < 0.05). The GSH and GSH/GSSG of the mean ova. groups were higher by 8 and 12%, respectively. The correlation coefficient between the kimchi intake and the total free radicals was -0.1862 (p < 0.05) and that for GSH/GSSG was 0.1861 (p < 0.05). In conclusion, the production of free radicals and oxidative substances increased with age, and kimchi seemed to retard this phenomena.

  • To identify the pathogenetic role of reactive oxygen free radical-induced oxidation reaction in endotoxin-induced acute lung injury, we infused endotoxin into 8 domestic pigs; endotoxin only (n=3), pretreatment with dimethylthiourea (DMTU) (n=5). We observed the sequential changes in hemodynamic parameters, the concentration of plasma oxidized glutathione (GSSG) in pulmonary arterial and venous blood, and albumin content in bronchoalveolar lavage fluid (BALF). The results were as follows. 1) While cardiac output decreased, mean pulmonary arterial pressure, pulmonary vascular resistance, and alveolar-arterial oxygen difference increased over phase 1 (0-2 hr) and phase 2 (2-4.5 hr) by endotoxin, DMTU attenuated the above changes only during phase 2. 2) While the concentration of plasma GSSG increased significantly by endotoxin during phase 2, there were no significant differences between pulmonary arterial and venous GSSG contents during both phases. The increase in plasma GSSG content was attenuated by DMTU. 3) The content of BALF albumin was significantly lower in DMTU group than that of endotoxin group. These results suggest that reactive oxygen free radical-induced oxidation reaction may have an important pathogenetic role in endotoxin-induced acute lung injury in pigs, which seems to be greater during phase 2 rather than phase 1.

  • The monomer N,N'-bis(2-pyrrol-1-yl-propyl)-4,4'-bipyridine(bpb) was electrochemically polymerized on the glassy carbon electrode surface, which was modified with 1:1 ratio of erichrome black T(EBT) and glutathione(GSSG) to give a type of GC/poly-bpb, EBT, GSSG electrode for depositing Zn(II). The diffusion coefficients of the incorporated ions were 2.43${\times}10^{-15}$ and 9.14${\times}10^{-15} cm^2s^{-1}$ before taking Zn(II) ions and after them respectively. The modified electrodes are stable at the electrode process. The polymerized poly-bpb of 2.83${\times}10^4gmol^{-1}$ can deposit 2.15${\times}10^4gmol^{-1}$ of Zn(II). The number of pumping ions involving in the redox procedure at 0.77 V was 81.7% of the captured 180 ions into the polymer matrix, which was 3 times larger than that of the electrode modified with EBT alone.

Effects of Saururus chinensis Baill on Glutathione and Antioxidative Activity Against TCDD-treated Rats (환경 Hormone에 대한 삼백초의 Glutathione 및 항산화 활성 효과)

  • The effects of Saururus chinensis Baill administration on the biochemical parameters of function in liver of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) treated rats were investigated. Saururus chinensis Baill (200mg/kg) was administered into rats intraperitoneally for four weeks, seven days after the injection of TCDD(1 ${\mu}g/kg$). We examined the antioxidative enzymatic activity by measuring the level of AST and ALT in serum and SOD, Catalase, GPx, GSH and GSSG in liver tissue of rats. STT group showed 70.7% of inhibitive effect in AST acrivity compared to TTA group. ALT level of STT group was decreased to the level of NTT group. SOD and Catalase in TTA group were lower than in NTT group, but SOD and Catalase in STT group were increased by 82% and 55.45% respectively compared to TTA group. GSH contents in STT group were 74.20% increased compared to TTA group. GSSG contents in STT group were 61.08% decreased compared to TTA group. These results suggest that Saururus Chinensis Baill has a potent hepathprotective effect against TCDD intoxicated rats.

The effect of Panax notoginseng saponins on oxidative stress induced by PCV2 infection in immune cells: in vitro and in vivo studies

  • Background: Panax notoginseng saponins (PNS) are bioactive substances extracted from P. notoginseng that are widely used to treat cardiovascular and cerebrovascular diseases and interstitial diseases. PNS have the functions of scavenging free radicals, anti-inflammation, improving blood supply for tissue and so on. Objectives: The aim of this study was to investigate the effects of PNS on the oxidative stress of immune cells induced by porcine circovirus 2 (PCV2) infection in vitro and in vivo. Methods: Using an oxidative stress model of PCV2 infection in a porcine lung cell line (3D4/2 cells) and mice, the levels of nitric oxide (NO), reactive oxygen species (ROS), total glutathione (T-GSH), reduced glutathione (GSH), and oxidized glutathione (GSSG) and the activities of xanthine oxidase (XOD), myeloperoxidase (MPO) and inducible nitric oxide synthetase (iNOS) were determined to evaluate the regulatory effects of PNS on oxidative stress. Results: PNS treatment significantly reduced the levels of NO and ROS, the content of GSSG and the activities of XOD, MPO, and iNOS (p < 0.05), while significantly increasing GSH and the ratio of GSH/GSSG in infected 3D4/2 cells (p < 0.05).Similarly, in the in vivo study, PNS treatment significantly decreased the level of ROS in spleen lymphocytes of infected mice (p < 0.05), increased the levels of GSH and T-GSH (p < 0.05), significantly decreased the GSSG level (p < 0.05), and decreased the activities of XOD, MPO, and iNOS. Conclusions: PNS could regulate the oxidative stress of immune cells induced by PCV2 infection in vitro and in vivo.
