Involvement of Ca2+/Calmodulin Kinase II (CaMK II) in Genistein-Induced Potentiation of Leucine/Glutamine-Stimulated Insulin Secretion

  • Genistein has been reported to potentiate glucose-stimulated insulin secretion (GSIS). Inhibitory activity on tyrosine kinase or activation of protein kinase A (PKA) was shown to play a role in the genistein-induced potentiation effect on GSIS. The aim of the present study was to elucidate the mechanism of genistein-induced potentiation of insulin secretion. Genistein augmented insulin secretion in INS-1 cells stimulated by various energygenerating nutrients such as glucose, pyruvate, or leucine/glutamine (Leu/Gln), but not the secretion stimulated by depolarizing agents such as KCl and tolbutamide, or $Ca^{2+}$ channel opener Bay K8644. Genistein at a concentration of $50{\mu}M$ showed a maximum potentiation effect on Leu/Gln-stimulated insulin secretion, but this was not sufficient to inhibit the activity of tyrosine kinase. Inhibitor studies as well as immunoblotting analysis demonstrated that activation of PKA was little involved in genistein-induced potentiation of Leu/Gln-stimulated insulin secretion. On the other hand, all the inhibitors of $Ca^{2+}$/calmodulin kinase II tested, significantly diminished genistein-induced potentiation. Genistein also elevated the levels of $[Ca^{2+}]_i$ and phospho-CaMK II. Furthermore, genistein augmented Leu/Gln-stimulated insulin secretion in CaMK II-overexpressing INS-1 cells. These data suggest that the activation of CaMK II played a role in genistein-induced potentiation of insulin secretion.

  • Since beginning of subway construction in 1974, for supplying the demand for increasing urban traffic and solving the traffic jam due to expansion of society and elevation of an individual income, the $1^{st}\;and\;2^{nd}$ construction have been almost done and $3^{rd}$ has been driving forward. However, according to an increment of com-mon usage with the passage of time, structures are superannuated and the amount of improvement and repairs keeps on increasing continuous. And many problems are taking place at underground space because of new constructions for underground facilities such as urban gas, electric power, waterworks, and sewerage. In consequence. The consideration of a counterplan for safety is required deeply This study analyzed problems on maintenance of structure about $1^{st}$ class subway, and will present means that a manage civil structures scientifically and systematically by using GSIS concept that imported into several organizations for facility management recently as making digital graphic elements and historic data into a database rather than using an old method - documents and paper drawings.

  • Recently, the rapid economic growth has caused urban problems due to the increase of urban population and the rapid expansion as a result of exodus phenomenon in rural territories. In order to solve the problems, we need to perform rational and effective plans for national land development. Generally, we need speedy and accurate topographical information of the object area to go ahead with balanced national land development. Acquisition method for topographical data include utilization of existing geographic information such as national base maps and related thematic maps and grasping of accurate present condition of object area using aerial photographies or satellite images. When the object area is small, existing methods for acquiring topographical information reveals imitations in accuracy, speed and economic aspect. Therefore, development of a new method is in order. As a result of this study, we recogized that Photo Balloon System to be manufactured and digital ortho-map to be generated could be used to obtain a topographical information for small areas.

  • Recently, the view of value and the national consciousness are expressed in various fields because of a rapid urban expanding, enforcement of local self-government system and tendency of democratization over the whole fields of society. It is difficult to satisfy this tendency of social environment change with only existing data management system. Therefore, we would use GSIS mainly used in facility management to develop the integrated urban management system which is connected the related information to related Agency. As a result, we can expect the efficient development, maintenance-management of urban and much civil affairs. On this study, firstly, we select a study area and present a basic direction of various urban facility management system. secondly, we develop the integrated urban information system with related Agency that combined the various urban facility management. Finally, we intended of its expression. It aims to manage as far as the step which is presented a model of urban integrated management system at the study area

  • Today Geo-Spatial Information System(GSIS) has developed and used more widely, and especially it has been more greatly asked for Land Information System as a part of GSIS. Now it has been planning and monitoring for remaking of cadastral maps by the Ministry of Government Administration and Home Affairs. But this is long term project because of the importance of itself. And then it has been needed to computerize cadastral maps for implementation of relative tasks and services. Cadastral maps have some problems; for example, map join in different scales, different administration boundary and so on. In this study General topology algorithm, Polygonization algorithm and Pseudo-topology algorithm was adapted in computerization of cadastral maps. This is called digital map. The area and shape of parcels do not change. As a result it was found that Polygonization algorithm is the best of 3 algorithm and General topology algorithm is the worst.

  • Photovoltaic system is the core one among the new and renewable energy of the low Carbon and green growth. Recently, the necessity developing PV is emerging since its of less green hose gas emissions. However, a survey or research on the PV system has been hardly performed. It's not easy to find a appropriate location in consideration of environment. These circumstances encourages a systematic approach for the PV system development. The purpose of this study is to propose a methodology of the location analysis for developing PV system. With this, constraint and location criteria with weights of Analytic Hierarchy Process are established and quantification method of each factor is presented. The location analysis of PV system using the GIS were generalized and the results of analysis for redundant topographic features were presented as 3 criteria of the suitability rank.

  • The highway is greatly changed by the constant economic growth for a long times the traffic situation such as the large volumes and the performance vehicles, the performance enlargement of vehicles, the high speedization, etc., due to growth economic. A study of an optimal route selection model is researched over late 1980s by development of computer and GSIS, and consisted including research about the optimal route that uses digital terrain model in domestic such as the earth volume calculations, the mass curve output and the automation system construction. Lately, the study of the driving simulation of the highway and the virtual reality using VGIS(Virtual Geographic Information System) is researched. This study shows when the alternative highway selection considered surrounding facilities, development plan and according to estimate amount of traffic and the additional possibility of view analysis and environment effect analysis element will study through 3D simulation method.

Human Postural Dynamics in Response to the Horizontal Vibration

  • The dynamic responses of human standing postural control were investigated when subjects were exposed to long-term horizontal vibration. It was hypothesized that the motion of standing posture complexity mainly occurs in the mid-sagittal plane. The motor-driven support platform was designed as a source of vibration. The AC Servo-controlled motors produced anterior/posterior (AP) motion. The platform acceleration and the trunk angular velocity were used as the input and the output of the system, respectively. A method was proposed to identify the complexity of the standing posture dynamics. That is, during AP platform motion, the subject's knee, hip and neck were tightly constrained by fixing assembly, so the lower extremity, trunk and head of the subject's body were individually immovable. Through this method, it was assumed that the ankle joint rotation mainly contributed to maintaining their body balance. Four subjects took part in this study. During the experiment, the random vibration was generated at a magnitude of $0.44m/s^2$, and the duration of each trial was 40 seconds. Measured data were estimated by the coherence function and the frequency response function for analyzing the dynamic behavior of standing control over a frequency range from 0.2 to 3 Hz. Significant coherence values were found above 0.5 Hz. The estimation of frequency response function revealed the dominant resonance frequencies between 0.60 Hz and 0.68 Hz. On the basis of our results illustrated here, the linear model of standing postural control was further concluded.

A Study of Land Suitability Analysis by Integrating GSIS with Artificial Neural Networks (GSIS와 인공신경망의 결합에 의한 토지적합성분석에 관한 연구)

  • This study is tried to organic combination in implementing the suitability analysis of urban landuse between GSIS and ANN(Artificial Neural Network). ANN has merit that can decide rationally connectivity weights among neural network nodes through procedure of learning. It is estimated to be possible that replacing the weight among factors needed in spatial analysis of the connectivity weight on neural network. This study is composed of two kinds of neural networks to be executed. First neural network was used in the suitability analysis of landuse and second one was oriented to analyze of optimum landuse pattern. These neural networks were learned with back-propagation algorithm using the steepest gradient which is embodied by C++ program and used sigmoid function as a active function. Analysis results show landuse suitability map and optimum landuse pattern of study area consisted of residental, commercial. industrial and green zone in present zoning system. Each result map was written by the Grid format of Arc/Info. Also, suitability area presented in the suitability map and optimum landuse pattern show distribution pattern consistent with theroretical concept or urban landuse plan in aspect of location and space structure.
