Studies on the Selectivity of Herbicide Alachlor;I. Phytotoxicity and Glutathione Conjugation (제초제 Alachlor의 선택성에 관한 연구;I. 약해와 글루타치온 Conjugation 반응)

  • Present work has been initiated to see if inherent biochemical difference among plants is, in any way, related to the observed selectivity characteristics of preemergence herbicide, alachlor. Application of aqueous solution of alachlor onto three intact plants, soybean, chinese cabbage and barnyard grass resulted in phytotoxicity responses in the testt plants in varying degree. Examination of glutathione (and homoglutathione) contents of the test plants indicated that the phytotoxicity is inversely proportional to the peptide contents of the test plants. It was also noted that four to five water soluble metabolites are readily formed in intact seedling treated with labelled alachlor and glutathionealachlor and homoglutathionealachlor conjugates were tentatatively identified as major metabolites. It is concluded that conjugation reaction involving glutathiones and xenobiotic alachlor, a typical phase II reaction, acts as detoxification reaction in the three test plants and this would, in turn, contribute to observed selectivity of alachlor.

Pathotype Classification of Plasmodiophora brassicae Isolates Using Clubroot-Resistant Cultivars of Chinese Cabbage

  • Clubroot disease caused by Plasmodiophora brassicae is one of the most serious diseases in Brassica crops worldwide. In this study, the pathotypes of 12 Korean P. brassicae field isolates were determined using various Chinese cabbage including 22 commercial cultivars from Korea, China, and Japan, and 15 inbred lines. All P. brassicae isolates exhibited the typical clubroot disease on non-clubroot resistant cultivar, indicating that the isolates were highly pathogenic. According to the reactions on the Williams' hosts, the 12 field isolates were initially classified into five races. However, when these isolates were inoculated onto clubroot-resistant (CR) cultivars of Chinese cabbage, several isolates led to different disease responses even though the isolates have been assigned to the same race by the Williams' host responses. Based on the pathogenicity results, the 12 field isolates were reclassified into four different groups: pathotype 1 (GN1, GN2, GS, JS, and HS), 2 (DJ and KS), 3 (HN1, PC, and YC), and 4 (HN2 and SS). In addition, the CR cultivars from Korea, China, and Japan exhibited distinguishable disease responses to the P. brassicae isolates, suggesting that the 22 cultivars used in this study, including the non-CR cultivars, are classified into four different host groups based on their disease resistance. Combining these findings, the four differential hosts of Chinese cabbage and four pathotype groups of P. brassicae might provide an efficient screening system for resistant cultivars and a new foundation of breeding strategies for CR Chinese cabbage.

Prognostic Role of Methylation Status of the MGMT Promoter Determined Quantitatively by Pyrosequencing in Glioblastoma Patients

  • Objective : This study investigated whether pyrosequencing can be used to determine the methylation status of the MGMT promoter as a clinical biomarker using relatively old archival tissue samples of glioblastoma. We also examined other prognostic factors for survival of glioblastoma patients. Methods : The available study set included formalin-fixed paraffin-embedded (FFPE) tissue from 104 patients at two institutes from 1997 to 2012, all of which were diagnosed histopathologically as glioblastoma. Clinicopathologic data were collected by review of medical records. For pyrosequencing analysis, the PyroMark Q96 CpG MGMT kit (Qiagen, Hilden, Germany) was used to detect the level of methylation at exon 1 positions 17-39 of the MGMT gene, which contains 5 CpGs. Results : Methylation of the MGMT promoter was detected in 43 (41.3%) of 104 samples. The average percentage methylation was $14.0{\pm}16.8%$ overall and $39.0{\pm}14.7%$ for methylated cases. There was no significant pattern of linear increase or decrease according to the age of the FFPE block (p=0.687). In multivariate analysis, age, performance status, extent of surgery, method of adjuvant therapy, and methylation status estimated by pyrosequencing were independently associated with overall survival. Additionally, patients with a high level of methylation survived longer than those with low methylation (p=0.016). Conclusion : In this study, the status and extent of methylation of the MGMT promoter analyzed by pyrosequencing were associated with overall survival in glioblastoma patients. Pyrosequencing is a quantitative method that overcomes the problems of MSP and a simple technique for accurate analysis of DNA sequences.

Enhancing the Eco-product Consumption as a Climate Change Mitigation Measure: The Case of Recycled Copy Paper (기후변화 완화대책으로서의 재생복사용지 소비 개선 연구)

  • Purpose: The paper aims to: 1) identify the reasons for the low usage of recycled paper in domestic workplaces and the premature state of recycled paper industry; 2) assess the environmental effects of replacing domestically consumed copy papers with recycled papers; and 3) suggest ways in which the recycled paper industry can expand in South Korea. Method: Questionnaire survey with cluster sampling is used to identify the attitudes and behavior with respect to recycled copy paper, the results of which are analyzed using SPSS. The environmental effect of replacing copy papers with recycled paper is assessed through the Life Cycle Assessment approach and Paper Calculator V4.0. Result: While the respondent's experience in using recycled copy paper was relatively low, they tend to acknowledge the need for its use and show relatively high satisfaction with the quality of the recycled paper. The environmental benefits of replacing ordinary copy paper with 40%+ recycled paper under the 10% market share increase scenario amounts to 60,000 tons of CO2 emissions reductions. Conclusion: The results from the attitude survey and market research, five approaches to improving the recycled copy paper market are suggested.

Intercropping of Cow Pea (Vigna unguiculata) as Summer Forage Yield with Grewia tenax in Irrigated Saline Soil of Khartoum State, Sudan

  • Agroforestry in terms of intercropping cow pea as summer forage with Grewia tenax was undertaken under sub -irrigation system in two consecutive seasons of 2017 and 2018 in saline soil of Khartoum State of Sudan. The aims were to find out suitable agro forestry system for saline soils as well as to investigate effect of tree spacing on field summer forage crop under semi -irrigation system. Therefore G. tenax trees that spaced at 4×4 m were used as main factor versus cow pea crop that incorporated at 25×50 cm intervals by using completely randomized block design with 3 replications. Trees and crop parameters were determined in terms of plant growth and yield. In addition to land equivalent ratio and soil chemical and physical properties at different layers were determined. The results revealed that, soil parameters in terms of CaCo3, SAR, ESP, pH paste and EC ds/m were increased with increasing soil depths. Meanwhile tree growth did not show any significant differences in the first season in 2017. Whereas in the second season in 2018 tree growth namely; tree height, tree collar and canopy diameters were higher under intercropping than in sole trees. Cow pea plant height recorded significant differences under sole crop in the first season in 2017. Unlike the forage fresh yield that was significant under the inter cropped plots. Tree fruit yield was higher under sole trees and land equivalent ratio was more advantageous under GS2 (1.5 m) which amounted to 4. Therefore it is possible to introduce this agroforestry system under saline soils to provide summer forage of highly nutritive value to feed animals and to increase farmers' income as far as to halt desertification and to sequester carbon.

Multi - Modal Interface Design for Non - Touch Gesture Based 3D Sculpting Task (비접촉식 제스처 기반 3D 조형 태스크를 위한 다중 모달리티 인터페이스 디자인 연구)

  • This research aims to suggest a multimodal non-touch gesture interface design to improve the usability of 3D sculpting task. The task and procedure of design sculpting of users were analyzed across multiple circumstances from the physical sculpting to computer software. The optimal body posture, design process, work environment, gesture-task relationship, the combination of natural hand gesture and arm movement of designers were defined. The preliminary non-touch 3D S/W were also observed and natural gesture interaction, visual metaphor of UI and affordance for behavior guide were also designed. The prototype of gesture based 3D sculpting system were developed for validation of intuitiveness and learnability in comparison to the current S/W. The suggested gestures were proved with higher performance as a result in terms of understandability, memorability and error rate. Result of the research showed that the gesture interface design for productivity system should reflect the natural experience of users in previous work domain and provide appropriate visual - behavioral metaphor.

Assessment of the Effect of a Disinfectant to Control Vibrio Strain from White Leg Shrimps Cultured in Korea (국내산 양식 흰다리새우 유래의 비브리오균에 대한 소독제의 방제 효과 검증)

  • White leg shrimps which were cultured in a private aquaculture farm showed abnormal swimming behavior and appetite reduction in July 2023. Then, gradual mortality was observed in the aquaculture farm. During the diagnosis, bacterial strain KNUAFVaSHP08147 was isolated from the hepatopancreas of the dead shrimps. Based on the sequence of 16S rRNA gene, KNUAFVa-SHP08147 was proved to be Vibrio alginolyticus, showing 99.71 % nucleotide identity with that of V. alginolyticus MG54 and GS MYPK1. According to the result of the growth of KNUAFVa-SHP08147, the treatment a commercial disinfectant, VirkonTMS was not proved to be effective to prohibit the growth of Vibrio.

Gerstmann-Sträussler-Scheinker Disease: A Case Report (Gerstmann-Sträussler-Scheinker병: 증례 보고)

  • Gerstmann-Sträussler-Scheinker (GSS) disease is a rare hereditary prion disease which is clinically characterized by a progressive cerebellar ataxia followed by cognitive impairment. We report a rare case of GSS disease in a 39-year-old male patient who complained of a progressive gait disturbance followed by dysarthria with cognitive impairment, after five months from the onset of initial symptom. His brain MRI scan revealed multifocal symmetric diffusion restricted lesions with T2/FLAIR hyperintensities in bilateral cerebral cortices, basal ganglia, and thalami. His family members also manifested similar symptoms in their 40-50s, suggesting the possibility of a genetic disease. Finally, he was genetically diagnosed with GSS disease by real-time quaking-induced conversion and prion protein (PRNP) gene sequencing test.

Antibacterial Activity of Artemisa Capillaris THUNB on Oral Bacteria (인진쑥의 구강세균에 대한 항균작용)

  • Recently it is very interesting that the plant extracts use to prevent or treat the oral diseases. The present study was performed to observe the antibacterial effect on S. gordonii Challis, S. gordoii G9B, S. mutans GS5, S. sobriuns 6715, E. faecalis ATCC 4083, A. actinomycetem Y4, P. gingivalis A7A1-28, P. gingivalis W83, Pr. intermedia ATCC 25611, F. nucleatum KTCT 2488, C. albicans ATCC 18804 of Artemisa capillaris THUNB employing the viable cell counts. The results were as follows: 1. Minimum inhibitory concentration(MIC) and Minimum bactericidal concentration(MBC) of extracts of Artemisa capillaris THUNB for P. gingivalis A7A1-28, P. gingivalis W83, and Pr. intermedia ATCC 25611, which are the pathologic bacteria of periodontal diseases, was observed under 2%. 2. MIC of extracts of Artemisa capillaris THUNB for P. gingivalis A7A1-28 was determined to be 1.2% and MBC was determined to be 2.0% respectively. 3. MIC of extracts of Artemisa capillaris THUNB for P. gingivalis W83 was determined to be 1.4% and MBC was determined to be 2.0% respectively. 4. MIC of extracts of Artemisa capillaris THUNB for Pr. intermedia ATCC 25611 was determined to be 1.2% and MBC was determined to be 2.0% respectively. The overall results indicate that Artemisa capillaris THUNB used for this study has a strong antibacterial activity against P. gingivalis A7A1-28, P. gingivalis W83, and Pr. intermedia ATCC 25611, which are the periodontopathic bacteria. Therefore, the extracts of Artemisa capillaris THUNB can be used as a candidate for prevention and therapeutic agent against periodontal diseases.

Development of Near Isogenic Lines with Various Seed Sizes and Study on Seed Size-related Characteristics in Watermelon (다양한 종자크기를 가진 Near Isogenic 수박 계통 육성 및 종자관련 특성 분석)

  • We inspected seed sizes of 353 genetic accessions of watermelon to diversify functional utility related to seed size and classified them into six representative groups based on their seed sizes. Each group was named as giant seed (GS), big seed (BS), medium size (NS), small size (SS), micro seed (MS) and tomato seed (TS) from the biggest. As the seed size was getting smaller, decreased seed length and seed width, increased seed number per fruit, and decreased seed weight per fruit were observed, but seed shape did not change significantly. In order to study the effect of seed size on fruit weight and seed germination, we developed three near isogenic lines (NILs) with three different seed sizes, SS, MS and TS, from crossing between two accessions 'NT' and 'TDR', and one NIL with seed size of TS from crossing between two accessions 'S55' and 'TDR'. In the study on the fruit weight of NILs with various seed sizes, NS, SS, MS, and TS NILs produced an average of 6.4, 6.3, 5.9, and 4.2 kg fruits, respectively. The bigger seed types showed the better germination rate. NS type showed the highest germination percentage, while TS showed very low germination percentage. Fermentation treatment for 48 hrs increased the germination percentage on TS type seed, but still remained at a low level. In NS, SS, and MS material, the ratio of embryo/whole seed weight was over 50%, meanwhile that of TS was only 44.4% of which low embryo percentage would be one of the reason of low germination percentage. From this study, we concluded that watermelon has very wide genetic diversity on seed size which is somehow related to fruit sizes and germination rate.
