Identification of Defect Type by Analysis of a Single PD Pulse in Gas Insulated Structure (가스절연 구조에서 단일 부분방전펄스 분석에 의한 결함 판별)

  • This paper dealt with a defect identification algorithm which is based on single partial discharge (PD) pulse analysis in gas insulated structure. Four types of electrode systems such as a needle-plane, a plane-needle, a free particle and a crack inside spacer were fabricated to simulate defects in gas insulated switchgear (GIS). We measured single PD pulse by an oscilloscope with a sampling rate of 5 GS/s and a frequency bandwidth of 1 GHz. Data aquisition and signal processing were controlled by a LabVIEW program. Physical shapes of PD pulses were compared with kurtosis, skewness and time-based parameters as rising time, falling time and pulse-width. These parameters were analysed by an algorithm with a back propagation algorithm (BPA). By applying the algorithm, the identification rate was 97% for the needle-plane electrode, 96% for the plane-needle electrode, 91% for the free particle and 93% for the crack inside spacer. The results verified that the algorithm could identify the type of defects in GIS.

Production of a Recombinant Laccase from Pichia pastoris and Biodegradation of Chlorpyrifos in a Laccase/Vanillin System

  • The recombinant strain P. pastoris GS115-lccC was used to produce laccase with high activity. Factors influencing laccase expression, such as pH, methanol concentration, copper concentration, peptone concentration, shaker rotate speed, and medium volume were investigated. Under the optimal conditions, laccase activity reached 12,344 U/L on day 15. The recombinant enzyme was purified by precipitating and dialyzing to electrophoretic homogeneity, and was estimated to have a molecular mass of about 58 kDa. When guaiacol was the substrate, the laccase showed the highest activity at pH 5.0 and was stable when the pH was 4.5~6.0. The optimal temperature for the laccase to oxidize guaiacol was $60^{\circ}C$, but it was not stable at high temperature. The enzyme could remain stable at $30^{\circ}C$ for 5 days. The recombinant laccase was used to degrade chlorpyrifos in several laccase/mediator systems. Among three synthetic mediators (ABTS, HBT, VA) and three natural mediators (vanillin, 2,6-DMP, and guaiacol), vanillin showed the most enhancement on degradation of chlorpyrifos. Both laccase and vanillin were responsible for the degradation of chlorpyrifos. A higher dosage of vanillin may promote a higher level of degradation of chlorpyrifos, and the 2-step addition of vanillin led to 98% chlorpyrifos degradation. The degradation of chlorpyrifos was faster in the L/V system ($k_{obs}$ = 0.151) than that in the buffer solution ($k_{obs}$ = 0.028).

The Importance of Filter Integrity Test to Ensure Sterility of Radiophamaceuticals for Using PET Image

  • The radiopharmaceuticals are routinely injected to blood vessel for acquiring PET image. For this reason, It is imperative that they undergo strict quality control measures. Especially, Sterility test is more important than any other quality control procedures. According to the FDA guideline, It requires filter integrity test used in the processing of sterile solutions. Among several methods, we can decide to use bubble point test. We usually use vented GS-filters (Millipore co., USA) which are sterilizinggrade (0.22 um pore size) and are placed upper site on product vial. After the synthesis of $^{18}F$-FDG, solutions wet the membrane in filter and then go into the product vial. By all synthesis steps have finished, we can observe the presence of the bubbles in the product vial. Since we have started this study, we have never found any bubbles in the product vial. Because the maximum pressure intensity of the filter which has set by manufacturer is up to 5 bars, but helium gas pressure is up to 1 bar in our module system. So, we can make 5 bars pressure using helium gas bombe and increase pressure up to 5 bars step by step. However, it does not happen to anything in vial.

Relation of personality preference, and achievement goal orientation, academic self-efficacy, test anxiety in selecting dental hygiene students (일부 치위생과 학생의 성격선호지표와 성취목표, 학업적 자기효능감, 시험불안의 관계)

  • Objectives : The purpose of this study was to identify correlation of personality preference, and achievement goal orientation, academic self-efficacy, test anxiety in selecting college students who majored in dental hygiene. This study also provides a basic data regarding personality for education of dental hygiene students. Methods : 192 students of S college who majored in dental hygiene were asked to answer the MBTI GS form, academic self-efficacy scale, achievement goal orientation scale and test anxiety scale during the month of May, 2009. The total of 160 copies were analyzed. MANOVA, independent-sample t-test were conducted. Results : 1. Looking at distribution of personality preference type of the students, extroversion type (63.1%) was more than introversion type(36.9%), sensing type(87.5%) was more than intuition type(12.5%), thinking type(54.4%) was more than feeling type(45.6%), judging type(59.4%) was more than perceiving type (40.6%) 2. In subscale of achievement goal orientation, performance approach level was higher in extroversion type than in introversion type. Performance avoidance level was lower in extroversion type than in introversion type. However, other preference personality type didn't show any significant difference in subscale of achievement goal orientation. 3. Academic self-efficacy was higher in extroversion type and thinking type than in introversion type and feeling type. There wasn't any significant difference between judging type and perceiving type, sensing type and intuition type. 4. In subscale of academic self-efficacy, confidence was higher in extroversion type than in introversion type. Task difficulty preference and self-regulated efficacy were higher in thinking type than in feeling type. 5. There wasn't any significant difference between personality preference type and test anxiety. Conclusions : Professor should find out difference between students through using information of preference personality and develop a teaching strategy that can encourage strength and make up weakness of each students.

Spatial Variability of Soil Moisture and Irrigation Scheduling for Upland Farming (노지 작물의 적정 관개계획을 위한 토양수분의 공간변이성 분석)

  • Due to droughts and water shortages causing severe damage to crops and other vegetations, much attention has been given to efficient irrigation for upland farming. However, little information has been known to measure soil moisture levels in a field scale and apply their spatial variability for proper irrigation scheduling. This study aimed to characterize the spatial variability and temporal stability of soil water contents at depths of 10 cm, 20 cm and 30 cm on flat (loamy soil) and hill-slope fields (silt-loamy soil). Field monitoring of soil moisture contents was used for variogram analysis using GS+ software. Kriging produced from the structural parameters of variogram was applied for the means of spatial prediction. The overall results showed that the surface soil moisture presented a strong spatial dependence at the sampling time and space in the field scale. The coefficient variation (CV) of soil moisture was within 7.0~31.3 % in a flat field and 8.3~39.4 % in a hill-slope field, which was noticeable in the dry season rather than the rainy season. The drought assessment analysis showed that only one day (Dec. 21st) was determined as dry (20.4 % and 24.5 % for flat and hill-slope fields, respectively). In contrary to a hill-slope field where the full irrigation was necessary, the centralized irrigation scheme was appeared to be more effective for a flat field based on the spatial variability of soil moisture contents. The findings of this study clearly showed that the geostatistical analysis of soil moisture contents greatly contributes to proper irrigation scheduling for water-efficient irrigation with maximal crop productivity and environmental benefits.

  • Spatial decision support systems(SDffi), a new class of decision support system(DSS), result from the melding together of GIS and DSS, Planning support systems(PS5) add more advanced spatial analysis functions than GIS and intertemporal functions to the functions of SDSS. This paper reports the development of a planning support system providing a framework that facilitates urban planners and civil engineers in conducting coherent deliberations about the generation of satisficing sewers. 1he planning support system for the generation of satisficing sewers(PS5/GSS) was designed from the understanding that land use and development drive the demand for storm and sanitary sewers. Through four stages of supply, demand, alternative generation, and evaluation, PSS/GSS integrates basic planning, preliminary design, and engineering design of sewer. GIS and graphic user interface are excellent toolboxes for designing sewer networks, estimating the quantity of wastewater, and showing generated alternative sewers. A sewer model using simulation tedmique can generate an initial sewer. Users can define alternative sewers by the direct manipulation of sewer networks or by the manipulation of parameters in the sewer model. The sewer model evaluates the performance of the user defined alternatives.

A Case Study on Behavior of High-Raised Reinforced Soil Wall (고성토 보강토옹벽의 거동에 관한 사례연구)

  • This paper describes a field experience on geogrid-reinforced soil walls rising up to 29.5m in height. Since experiences of design and construction on very high-raised geogrid reinforced soil wall were limited, thorough design and construction management was performed for safe construction of the wall. Regarding design of the wall, both internal and external stabilities were examined based on the design guideline specified by FHWA and overall slope stability analyses were performed by using Bishop simplified method. Moreover, a series of instrumentations were performed. The results of instrumentation for two tiered reinforced soil wall showed that not only the deformations of both the wall face and the reinforcement but also the horizontal earth pressures acting on the wall facing were very small. These results indicate that the reinforced soil wall technology can be applied successfully for high-raised tiered wall more than 20m heights and FHWA design guideline is very conservative for that large wall.

Effect of Mobile Crane Load on Excavated Slope Stability (이동식 크레인 하중이 굴착사면 안정성에 미치는 영향 분석)

  • The effect of heavy construction equipment on the excavated slope is investigated by slope stability analysis. A mobile crane with 500 kN capacity is applied as a working load to the background surface of the excavated slope, in both sandy soil and clay, designed to guarantee the safety of slope stability. Major parameters such as the distance between the edge of the slope and the mobile crane, groundwater level, and ground plate size of the mobile crane are considered. Only 23.8% and 14.3% of the analysis models with sandy soil and clay excavated slope, respectively, satisfied the slope stability. By changing the slope of the sandy soil from 1:1.0 to 1:1.2, the number of analysis models securing slope stability increased from 23.8% to 40.5%. For the clay excavated slope, the analysis models securing slope stability increased from 14.3% to 42.9% by changing slope inclination from 1:0.8 to 1:1.2. In addition, it is found that the increase in the size of the ground plate of the mobile crane increases the analysis models that secure slope stability. Therefore, it is an effective way to relax the excavated slope's inclination angle and simultaneously increase the ground plate size to guarantee stability.

Distribution and Frequency of SSR Motifs in the Chrysanthemum SSR-enriched Library through 454 Pyrosequencing Technology (국화 SSR-enriched library에서 SSR 반복염기의 분포 및 빈도)

  • Chrysanthemums, often called mums or chrysanths, belong to the genus Chrysanthemum, which includes about 30 species of perennial flowering plants in the family Asteraceae. We extracted DNA from Dendranthema grandiflorum ('Smileball') to construct a simple sequence repeat (SSR)-enriched library, using a modified biotin-streptavidin capture method. GS FLX (Genome Sequencer FLX System which provides the flexibility to perform the broad range of applications) sequencing (at the 1/8 run specification) resulted in 18.83 mega base pairs (Mbp) with an average read length of 280.06 bp. Sequence analyses of all SSR-containing clones revealed a predominance of di-nucleotide motifs (16,375, 61.5%) followed by tri-nucleotide motifs (6,616, 24.8%), tetra-nucleotide motifs (1,674, 6.3%), penta-nucleotide motifs (1,283, 4.8%), and hexa-nucleotide motifs (693, 2.6%). Among the di-nucleotide motifs, the AC/CA class was the most frequently identified (93.5% of all di-nucleotide types), followed by the GA/AG class (6.1%), the AT/TA class (0.4%), and the CG/GC class (0.03%). When we analyzed the distribution of different repeat motifs and their respective numbers of repeats, regardless of the motif class, of 100 SSR markers, we found a higher number of di-nucleotide motifs with 70 to 80 repeats; we also found two di-nucleotide motifs with 83 and 89 repeats, respectively, but their product lengths were within optimum size (297 and 300 bp). In future work, we will screen for polymorphisms of possible primer pairs. The results will provide a useful tool for assessing molecular diversity and investigating the population structure among and within Chrysanthemum species.

A New Wheat Variety, "Sukang" with Good Noodle Quality, Resistant to Winter Hardiness and Pre-harvest Sprouting (내한 내수발아성 제면용 밀 신품종 "수강밀")

  • "Sukang", a winter wheat (Triticum aestivum L.) cultivar was developed by the National Institute of Crop Science, RDA. It was derived from the cross "Suwon266" / "Asakaze" during 1994. "Sukang" was evaluated as "Iksan312" in Advanced Yield Trial Test in 2005. It was tested in the regional yield trial test between 2006 and 2008. "Sukang" is an awned, semi-dwarf and hard winter wheat, similar to "Keumkang" (check cultivar). The heading and maturing date of "Sukang" were similar to "Keumkang". Culm and spike length of "Sukang" were 90 cm and 8.1 cm, longer culm length and similar spike length compared to "Keumkang" (80 cm and 7.9 cm, respectively). "Sukang" had similar test weight (819 g/L) and lower 1,000-grain weight (40.2 g) than "Keumkang" (813 g/L and 44.9 g, respectively). "Sukang" showed resistance to winter hardiness and pre-harvest sprouting, which lower withering rate on the high ridge (4.5%) and rate of pre-harvest sprouting (0.2%) than "Keumkang" (21.9% and 30.4%, respectively). "Sukang" had lower flour yield (71.1%) and higher ash content (0.45%) than "Keumkang" (74.1% and 0.42%, respectively). "Sukang" showed lower lightness (89.13) and higher yellowness (10.93) in flour color than "Keumkang" (90.02 and 9.28, respectively). It showed higher protein content (12.8%) and gluten content (11.1%) and lower SDS-sedimentation volume (56.8 ml) and mixing time of mixograph (2.6 min) than "Keumkang" (11.9%, 10.2%, 62.3 ml and 4.7 min, respectively). Fermentation properties, amylose content and pasting properties of "Sukang" were similar to "Keumkang". "Sukang" showed different compositions in high molecular weight glutenin subunits (HMW-GS, $2^{\ast}$, 13+16, 2+12) and puroindolines (pina-1b/pinb-1a) compared to "Keumkang" ($2^{\ast}$, 7+8, 5+10 in HMW-GS and Pina-1a/Pinb-1b in puroindolines, respectively). "Sukang" showed lower hardness (4.53 N) and similar springiness and cohesiveness of cooked noodles (0.94 and 0.63) compared to "Keumkang" (4.65 N, 0.93 and 0.64, respectively). Average yield of "Sukang" in the regional adaptation yield trial was 5.34 MT/ha in upland and 4.72 MT/ha in paddy field, which was 4% and 1% lower than those of "Keumkang" (5.55 MT/ha and 4.77 MT/ha, respectively). "Sukang" would be suitable for the area above $-10^{\circ}C$ of daily minimum temperature in January in Korean peninsula.
