  • 유전자총(Particle bombardment) 방법에 의해 snowdrop lectin (Galanthus nivalis agglutinin; GNA) 유전자가 도입된 해충저항성 형질전환 벼를 개발하였다. GNA 유전자가 도입된 많은 형질전환 식물체가 재분화되었으며, 형질전환 벼의 GNA 유전자의 integration, expression 및 inheritance는 Southern 및 western analysis분석 방법에 의해 증명되었다. 벼 genome 내 도입 유전자 수는 one 혹은 five copies이었다. 형질전환 $R_1$$R_2$ 식물체의 GNA 단백질 발현 분석 결과, 함량은 총 단백질 중 0.01%부터 2.0%까지 포함하고 있었다. 형질전환 식물체 중 GNA 과발현 형질전환 벼를 이용한 생물 검정 결과, 대조구와 비교하여 벼의 주요 해충인 벼멸구(Nilaparvata lugens St${\aa}$l)에 저항성을 나타내었다. 위의 실험 결과를 통해, 해충저항성 유전자인 GNA가 도입된 형질전환 벼는 해충의 증식을 억제하는데 이용될 수 있을 것이다.

추출용매에 따른 이슬송이버섯(Lentinula edodes GNA01) 추출물의 항산화 활성 (Effect of Extraction Solvent on the Antioxidant Activity of Lentinula edodes GNA01 Extract)

  • Antioxidant activities of 80% methanol, water, and 70% ethanol extracts of Lentinula edodes GNA01 were compared and estimated. The yield of Lentinula edodes GNA01 was identified to be in the following order: water>70% ethanol>80% methanol, but there was no significant difference between 80% methanol and 70% ethanol extracts. The highest total phenolic content (TPC) and total flavonoid content (TFC) were found in water extract, and TPC of 80% methanol extract was higher than that of 70% ethanol extract and TFC of 70% ethanol extract was higher than that of 80% methanol extract. Water extract exhibited the strongest DPPH, ABTS radicals, and nitrite scavenging activities, $Fe^{2+}$ chelating ability, and FRAP among the three extracts. In addition, antioxidant activity of 80% methanol extract was higher than that of 70% ethanol extract in most of the experiments. As a result, antioxidant activity of Lentinula edodes GNA01 showed a difference according to extraction solvent and concentration; nevertheless, water extract exhibiting high polarity had the strongest antioxidant effect. Consequently, water extract from Lentinula edodes GNA01 is anticipated to be useful for the development of a high value-added functional product.

표고버섯(Lentinula edodes) 및 표고버섯 신품종 이슬송이버섯(Lentinula edodes GNA01)의 이화학적 특성 및 항산화 활성 비교 (Comparison of Physicochemical Properties and Antioxidant Activities between Lentinula edodes and New Cultivar Lentinula edodes GNA01)

  • 본 연구에서는 관련 연구가 미흡하고 널리 알려져 있지 않아 수요가 부족한 이슬송이버섯의 보급 확대 및 수요 창출을 위한 기초자료를 제공하고자 이슬송이버섯의 이화학적 특성 및 항산화 활성을 조사하였다. 이슬송이버섯은 중국 복건성 삼명진연구소 L26과 경원9015를 모균주로 하여 교배된 신품종 표고버섯으로 표고버섯과 달리 갓과 자루의 구별이 없는 구형을 띠고 있다. 이슬송이버섯의 일반성분을 분석한 결과 수분 및 조단백 함량은 표고버섯보다 낮았으나 조회분, 조지방, 조섬유 및 탄수화물 함량은 표고버섯보다 높았다. 이슬송이버섯의 베타카로틴 함량은 $19.05{\mu}g/100g$으로 표고버섯의 베타카로틴 함량보다 약 3배 높았으며, 비타민 D 함량 또한 $118.53{\mu}g/100g$으로 표고버섯의 약 2배로 확인되었다. 주요 무기질은 K으로 측정되었으며, Mg 함량이 표고버섯의 약 2배로 측정되어 유의적인 차이를 보였다. 이슬송이버섯 및 표고버섯의 지방산 함량과 조성은 모두 불포화지방산이 대부분을 차지하였으며, 특히 linoleic acid가 가장 큰 비중을 차지하고 있었다. 이슬송이버섯의 총 페놀 함량은 표고버섯보다 낮았으며, 1,1-diphenyl-2-picrylhydrazyl picrylhydrazyl(DPPH) radical 소거 활성과 ferric reducing antioxidant power(FRAP) 또한 모든 농도에서 표고버섯보다 낮았다. 그러나 이슬송이버섯은 10 mg/mL 농도에서 80% 이상의 DPPH radical 소거 활성을 보였으며, FRAP 또한 1에 가까운 환원력을 나타내었다. 이상의 결과로 이슬송이버섯은 비록 항산화 활성은 표고버섯에 비해 낮게 나타났으나 베타카로틴 및 비타민 D 함량은 표고버섯보다 높은 것으로 확인되었다. 이에 따라 항산화 활성을 높일 수 있는 방안에 대해 추후 연구가 이루어진다면 더욱 훌륭한 식품소재가 될 것으로 판단된다.

Possible Protective Effects of Quercetin and Sodium Gluconate Against Colon Cancer Induction by Dimethylhydrazine in Mice

  • Micronutrients in food have been found to have chemopreventive effects, supporting the conclusions from epidemiologie studies that consumption of fresh fruits and vegetables reduces cancer risk. The present study was carried out to evaluate the role of querctin (Q) and sodium gluconate (GNA) supplementation separately or in combination in ameliorating promotion of colon tumor development by dimethyl-hydrazine (DMH) in mice. Histopathological observation of colons in mice treated with DMH showed goblet cell dysplasia with inflammatory cell infiltration. This pathological finding was associated with significant alteration in oxidative stress markers in colon tissues and carcinoembryonic antigen (CEA) levels in plasma. Mice co-treated with GNA and Q showed mild changes of absorptive and goblet cells and inflammatory cell infiltration in lamina properia, with improvement in oxidative stress markers. In conclusion, findings of the present study indicate significant roles for reactive oxygen species (ROS) in pathogenesis of DMH-induced colon toxicity and initiation of colon cancer. Also, they suggest that Q, GNA or the combination of both have a positive beneficial effect against DMH induced colonic cancer induction in mice.

신품종 이슬송이 버섯(Lentinula edodes GNA01)의 온도별 저장에 따른 품질 변화 (Changes in the Quality of New Cultivar Dewdrop Pine Mushroom (Lentinula edodes GNA01) Depending on the Storage Temperature)

  • Purpose: The purpose of this study was to investigate the quality characteristics and antioxidant activity on consequent storage based on the temperature of new cultivar dew-drop pine mushroom (Lentinula edodes GNA01). Methods: Dewdrop Pine Mushroom were prepared under different storeage temperature (4, 10, $20^{\circ}C$). Results: Weight loss and hardness showed the least reduction rate when it was stored at $4^{\circ}C$ (p<0.05). The color value changed to dark brown at all storage temperatures during the storage period. When it was stored at $4^{\circ}C$, it maintained its initial color for a longer period than at different storage temperatures. With respect to the change in its viable cell count, the cells proliferated to less than 3.0 log CFU/g up to Ed: Please review the change. The earlier part was difficult to understand 2.83 log CFU/g until 15 days of storage at $4^{\circ}C$. On measuring the antioxidant activity of this mushroom, the polyphenol content was maintained without a large change until 9 days of storage at $4^{\circ}C$. The electron-donating action maintained high antioxidant activity, accounting for 81.99% until 12 days of storage from 83.08% during the initial storage at $4^{\circ}C$. When it was stored at $4^{\circ}C$, the sensory characteristics received the highest score among all items, such as appeaance, color, flavor and general preference, etc. Concolusion: In summary, new Cultivar Dewdrop Pine Mushroom (Lentinula edodes GNA01) can maintain its commercial value until the 12th day of $4^{\circ}C$ storage.

N-아세틸글루코사민 생산을 위한 코리네박테리움 글루타미컴의 대사공학 (Metabolic Engineering of Corynebacterium glutamicum for N-acetylglucosamine Production)

  • 대사공학을 이용하여 N-아세틸글루코사민(GlcNAc)을 생산하는 재조합 Corynebacterium glutamicum을 개발하였다. 먼저 GlcNAc를 생산하는 기반균주를 제작하기 위하여, N-acetylglucosamine-6-phosphate deacetylase와 glucosamine-6-phosphate deaminase를 암호화하는 nagAB와 N-acetylmannosamine-6-phosphate epimerase를 암호화하는 nanE를 C. glutamicum ATCC 13032에서 순차적으로 결손하여, 최종적으로 KG208 균주를 제작하였다. 또한, glucosamine-6-phosphate synthase를 암호화하는 C. glutamicum 유래의 glmS와 glucosamine-6-phosphate N-acetyltransferase를 암호화하는 Saccharomyces cerevisiae 유래의 gna1을 각각 여러 발현벡터에 클로닝하였다. 여러 발현 조합의 플라스미드들 중에서 pCXI40-glmS와 pCEI40-gna1을 함유한 제조합균주 KG440은 삼각플라스크 발효에서 1.77 g/l의 GlcNAc와 0.63 g/l의 글루코사민을 생산하였다.

Comparison of carbohydrate moieties of sparganum proteins of the snake, mouse and those of adult worm

  • The carbohydrate moieties of larval sparganum proteins in two different species, the snakes, Elaphe rufodorsata, the Balb/c mouse and those of the adult worm, Spirometra erinacei, were compared using five different lectins including GNA, SNA, MAA, PNA and DSA. The GNA positive 53 kDa molecule, which is excretory-secretory protease in the sparganum from the snake showed a stage specific and developmental regulation. We also suggested that sparganum glycosylation may be involved in immune evasion and differentiation into an adult worm.

  • Purpose: The purpose of this study was to investigate quality characteristics and antioxidant effects on storage by temperature between new cultivar Dewdrop Pine Mushroom (Lentinula edodes GNA01) and button mushroom (Agaricus bisporus). Methods: Dewdrop Pine Mushroom and button mushroom were prepared at low storage temperature. Results: Weight loss increased with time, whereas the new cultivar Dewdrop Pine Mushroom showed a decrease of less than 4% until 15days. Hardness of new cultivar Dewdrop Pine Mushroom was reduces less than button mushroom during storage. Color change of new cultivar Dewdrop Pine Mushroom was not altered during storage. For viable cell count, new cultivar Dewdrop Pine Mushroom proliferated less than button mushroom. For antioxidant activity, polyphenol content increased with storage period of both mushrooms. The electron-donating action of new cultivar Dewdrop Pine Mushroom maintained high antioxidant activity accounting for 80% until 12 days of storage. New cultivar Dewdrop Pine Mushroom was evaluated as better than pine mushroom as it exceeded the middle of storage in sensory characteristics; such as appearance, color, aroma and overall acceptability, etc. Conclusion: In summary, new cultivar Dewdrop Pine Mushroom was stored for 12 days while button mushroom was stored for 9 days.

신선편이 엽채류의 병원성 E. coli 검출을 위한 생화학적동정법 비교 분석 (Comparison of Biochemical Identification to Detect Pathogenic Escherichia coli in Fresh Vegetables)

  • 본 연구에서는 선택배지와 건조필름을 이용하여 신선편이 엽채류 3종(양상추, 양배추, 어린잎채소)에서 병원성 E. coli를 분리하였고, 의심집락 동정을 위해 생화학적 분석법을 사용하여 동정한 후 결과를 비교 분석하였다. 양상추 20 g, 양배추 20 g, 어린잎채소 10 g에 병원성 E. coli 혼합 균액(Enterohemorrhagic E. coli NCCP11142, Enterotoxigenic E. coli NCCP14037, Enteropathogenic E. coli NCCP14038, Enteroaggregative E. coli NCCP14039, Enteropathogenic E. coli NCCP15661)을 최종농도가 1, 2, 3 log CFU/g이 되도록 접종하였고, BPW (80~90 ml)을 넣은 후 60초 동안 균질화하여 분석하였다. 연구결과, 모든 시료에서 건조필름 시험 양성, 양상추 시료 일부(최종농도 3 log CFU/g 접종시료)를 제외한 모든 시료에서 증균배양법을 이용한 정성시험결과가 음성으로 나타났다. 증균배양과 건조필름 시험을 통해 분리한 병원성 E. coli를 이용하여 생화학적 분석을 실시한 결과, 양상추에서 분리한 병원성 E. coli의 경우, API 20E 100% (44/44), Microgen GNA 100% (44/44), Food System 66.7% (10/15)의 동정률이 나타났다. 양배추의 경우, API 20E 64.7% (22/34), Microgen GNA 50% (16/32), Food System 60% (9/15), 어린잎채소의 경우, API 20E 65.1% (28/43), Microgen GNA 62.3% (27/43), Food System 53.3% (8/15)가 병원성 E. coli로 동정되었다. 본 연구의 결과는 신선편이 엽채류에 대한 병원성 E. coli 검출법 선택에 유용하게 이용될 것으로 판단된다.
