Development of a single-nucleotide-polymorphism marker for specific authentication of Korean ginseng (Panax ginseng Meyer) new cultivar "G-1"

  • Background: Korean ginseng (Panax ginseng) is a well-known medicinal plant of Oriental medicine that is still in practice today. Until now, a total of 11 Korean ginseng cultivars with unique features to Korean ginseng have been developed based on the pure-line-selection method. Among them, a new cultivar namely G-1 with different agricultural traits related to yield and content of ginsenosides, was developed in 2012. Methods: The aim of this study was to distinguish the new ginseng cultivar G-1 by identifying the unique single-nucleotide polymorphism (SNP) at its 45S ribosomal DNA and Panax quinquefolius region than other Korean ginseng cultivars using multiplex amplification-refractory mutation system-polymerase chain reaction (ARMS-PCR). Results: A SNP at position of 45S ribosomal DNA region between G-1, P. quinquefolius, and the other Korean ginseng cultivars was identified. By designing modified allele-specific primers based on this site, we could specifically identified G-1 and P. quinquefolius via multiplex PCR. The unique primer for the SNP yielded an amplicon of size 449 bp in G-1 cultivar and P. quinquefolius. This study presents an effective method for the genetic identification of the G-1 cultivar and P. quinquefolius. Conclusion: The results from our study shows that this SNP-based approach to identify the G-1 cultivar will be a good way to distinguish accurately the G-1 cultivar and P. quinquefolius from other Korean ginseng cultivars using a SNP at 45S ribosomal DNA region.

Physiochemical and Antimicrobial Activity of Garlic Cultivar (품종별 마늘의 이화학적 특성 및 항균활성)

  • Physicochemical and antimicrobial activities of 12 different garlic cultivars were investigated. Width and weight of California late cultivar (60.44mm, 53.73g) was the biggest and heaviest but Changyoung cultivar (44.04mm, 25.15g) was the smallest and lightest among the variety of garlic. The range of L, a and b color characteristics of garlic surface from different variety were 84.13~90.56, -1.10~0.77 and 18.24~26.61, respectively. Shear force was the lowest in California early, but 94-12-2 cultivar ($4211.35cm/kg^2$) was higher than another cultivars. Soluble solid range was 6.40~11.33 %brix, and Changyoung cultivar was the highest than the others, significantly. pH of garlics from different cultivar were 5.57~6.53. Total thiosulfinate content of California late cultivar (146.05mM/g) was higher, but Italy cultivar (93.23mM/g) was lower than the others. Total pyruvate content was the highest in Yugo cultivar ($162.50{\mu}M/g$) and the lowest in California early cultivar ($147.41{\mu}M/g$).

Antioxidant Activity of Several Cabbage (Brassica oleracea L.) Cultivars

  • Total phenol, flavonoid and antioxidant components of cabbage leaf samples derived from different cultivar were determined. Total phenol compound content showed the highest amount in methanol extracts from ‘YR Howol’ cultivar (11.72 ㎎/g), followed by ‘Harutame’ (10.66㎎/g), ‘Winstar’ (10.34 ㎎/g) and YR Hero (10.20 ㎎/g). The highest amount of total flavonoid content was observed from the methanol extracts of Harutame (5.39 ㎎/g), followed by Winstar (4.28 ㎎/g), Wialhowol (4.10 ㎎/g). The SOD enzyme activity showed a high activity of ‘YR Hogeo’ cultivar, and the cultivar of ‘YR Howol’ cultivar showed the lowest activity of SOD. The activity of CAT and APX showed higher values ‘Ogane’ and ‘YR Hogeol’ cultivars than the other cultivars. The POD activities showed relatively high values ‘Ogane’ and ‘YR Howol’ cultivars compared with other cultivars. The free radical (DPPH) scavenging activity showed lower IC50 values of ‘Harutame’ (15.71) and ‘YR Howol’ cultivar (16.88), however methanol extract of ‘YR Hero’ cultivar (22.49) being the highest. The extracts of all cabbage cultivars in the reaction solution of pH 1.2 could be decomposed nitrite more than 50%. Especially, the cultivar ‘YR Hogeol’ and ‘Ogane’ showed a relatively high nitrite scavenging activity for each 60.13% and 57.20% respectively. The IC50 values of antioxidant activity determined by ABTS were lower in ‘Harutame’ (17.04) and ‘YR Howol’ cultivar (17.97), and its results observed similar with values obtained from the same extracts by DPPH method. The result of this study suggests that the methanol extract of Brassica oleracea L. contains the high amount of phenolic and higher radical scavenging activities.

Cultural characteristics of a new spore-less cultivar 'chungheung 1ho' for Lentinula edodes sawdust cultivation (톱밥 재배용 무포자 신품종 '청흥1호'의 생육특성)

  • The competitiveness of the mushroom industry can be increased by diversifying the domestic Lentinula edodes cultivar. Therefore, 'Chungheung 1ho' was cultivated by hybrid breeding method using strain 'E140025,' which was collected by the Chungheung Mushroom Farming Association. Regarding the morphological characteristics of the fruit, the cap of 'Chungheung 1ho' was smaller than that of the control cultivar, whereas, its stem was longer and thicker than that of the control cultivar. In addition, the weight of the individual fruits of the new cultivar was heavier than that of the control cultivar. The period for primordia formation was 24 days for the new cultivar, which was 16-20 days later than that of the control cultivar. Furthermore, it was found that the patterns of the PCR amplification band using six URP primers were different for 'Chungheung 1ho' than those of the mating strains.

Antioxidant activities of brown beech mushroom (Hypsizygus marmoreus) pileus and stipe (갈색 느티만가닥버섯 추출물의 부위별 항산화 활성)

  • This study was carried out to evaluate potential of Hypsizygus marmoreus (brown cultivar) as a functional food and drug materials. H. marmoreus were divided into pileus and stipe and extracted in hot water and 80% ethanol. The total polyphenol content was highest in the hot water extracts (pileus 17.15±0.19 mg of GAE g, stipe 7.37±0.16 mg of GAE/g) and pileus compared to the ethanol extracts (pileus 10.23±0.14 mg of GAE/g, stipe 3.76.±0.19 mg of GAE/g) and stipe. Also, hot water extracts of pileus from H. marmoreus (brown cultivar) was more effective DPPH, ABTS, ORAC value, reducing power than ethanol extracts and stipe extracts. The pileus and stipe extracts were confirmed to be non-cytotoxic in the mouse macrophage cell line RAW 264.7 determined by WST-1 (4-[3-(4-iodophenyl)-2-(4-nitrophenyl)-2H-5-tetrazolio]-1,3-benzene disulphonate) assay. Overall, extracts of H. marmoreus (brown cultivar) was higher antioxidant activity than other mushrooms, and no cytotoxicity. Therefore, H. marmoreus (brown cultivar) showed potential as a functional food and drug materials. The brown cultivar of H. marmoreus have higher antioxidant activity than white cultivar, H. marmoreus seem to have different antioxidant activity depending on the cultivar.

A New High Quality and Yield Mungbean Cultivar "Sohyeon" (고품질 다수성 녹두 신품종 "소현")

  • A new mungbean cultivar "Sohyeon" (Vigna radiata (L.) Wilczek) was bred from the cross between "Suwon28" and "KM94004" at the Jeollanamdo Agricultural Research & Extension Services (JARES) in 2006. "Sohyeon" has erect type, heart shape leaflet, green hypocotyl, black and straight pod when matured, and dull green seed surface. This new cultivar shows 42 g per a thousand seed weight. And sprout yield was up to 9% more than that of comparison cultivar "Owool". It contains 12.0, 12.4 mg/g of vitexin and isovitexin, respectively, which was shown about 38, 31% more than comparison cultivar. It shows resistance to mungbean mottle virus, cercospora leaf spot, and powdery mildew in the normal cultivate condition. The average yield of this cultivar shows 1.62 ton/ha, which was 7% more than comparison cultivar.

Breeding of 'Daewang' Strawberry for Forcing Culture with Good Taste and Fragrance (맛과 향이 좋은 촉성재배용 딸기 '대왕' 품종의 육성)

  • A new cultivar 'Daewang' strawberry was developed by the National Institute of Horticultural & Herbal Science for forcing culture in 2010. The cultivar 'Daewang' was originated from the cross between 'Maehyang', a high firmness cultivar and 'Wongyo 3111', a high sugar content in 2006. The cultivar shows erect plant type, vigorous growth habit, early flower bud differentiation with 12-15 flowers per cluster from planting on healthy nursery. Fruits of 'Daewang' are conical type having a bright red skin color, and 16-17 g in an average weight. 'Daewang' is suitable for forcing culture as time required for dormancy breaking ranged between 50 and 100 hours. This cultivar has excellent taste for high sugar/acid ratio as sugar content of $11.1^{\circ}Brix$, acidity of 0.39% with abundant texture and can be harvested by late spring because the fruit firmness of 'Daewang' was $18.2g{\cdot}mm^{-2}$ that was about $7.9g{\cdot}mm^{-2}$ higher than $10.3g{\cdot}mm^{-2}$ of 'Akihime' cultivar. But although total yield is not significantly different from 'Akihime' cultivar, its marketable yield is remarkably higher than that of 'Akihime' cultivar. Disease and pest resistance of 'Daewang' have a tendency to sensitive powdery mildew, anthracnose and spotted spider.

Mungbean Cultivar, 'Munpyeong', with a Short Stem and High Yield (단경 다수성 녹두 품종 '문평')

  • A mungbean cultivar "Munpyeong" (Vigna radiata (L.) Wilczek) was derived as a cross between "Keumseong" and "IT163157" at the Jeollanamdo Agricultural Research and Extension Services (JARES) in 2013. "Munpyeong" has an erect growth habit, heart leaflet, green hypocotyls, black and straight pods when matured, and a dull green seed surface. The stem length of "Munpyeong" was 60 cm, which was 6 cm shorter than that of the control cultivar "Owool." The pod number per plant was 25, which was 3 more than that of the control cultivar. The 1000 seed weight of "Munpyeong" was 45 g, which was 4 g lighter than that of the control cultivar "Owool." Its field resistance to mungbean mottle virus, cercospora leaf spot, and powdery mildew are similar to the control cultivar, whereas its field resistance to lodging was slightly stronger. The hard seed rate of "Munpyeong" was 3.2%, which was similar to that of the control cultivar. The sprout yield ratio was 7.54 times, which was 9% more than that of the control cultivar. The average yield of "Munpyeong" was 1.93 ton/㏊, which was 21% more than that of the control cultivar "Owool" (Cultivar registration number: 5878).

Identification of Soybean Mosaic Virus Strains and a Consideration on Genetics of Soybean for Resistance to SMV Strains (콩 모자이크 바이러스의 계통분류와 콩 품종의 저항성 유전에 관한 고찰)

  • The soybean necrotic disease has been shown to be caused by a virulent strain or strains of soybean mosaic virus (SMV) in soybean cultivar Kwnaggyo. However, the disease was found in soybean cultivar Hwanggeum which was released as a leading and mosaic resistant soybean cultivar in Korea. The strain SMV-G5H appeared to an isolate showing similar characteristics with the strain SMV-G7, although there were some variations in reactions of soybean differentials used.

A New Mungbean Cultivar, "Daseon" with Greenish Purple Hypocotyl and High Yielding (녹자색배축 다수성 녹두 신품종 "다선")

  • A new mungbean cultivar Daseon (Vigna radiata (L.) Wilczek) was derived from the cross between KM9003-2B-783 and Keumseong at the Jeollanamdo Agricultural Research & Extension Services (JARES) in 2006. Daseon has erect growth habit, heart leaflet, greenish purple hypocotyl, black and straight pod when matured, and dull green seed surface. A thousand seed weight was 52 g, sprout yield ratio was 630%, and the anthocyanin (delphinidin 3-glucoside) of fresh sprouts contained 1.7 ug/g. The average plant height was 2 cm shorter than the check cultivar (Owool). It is field resistance to mungbean mottle virus, cercospora leaf spot, and powdery mildew. The average yield was 1.76 ton/ha, which was 21% more than check cultivar.
