Tumor necrosis factor (TNF)-${\alpha}$ is chronically elevated in adipose tissues of obese rodents and humans. Increased levels of TNF-${\alpha}$ have been implicated in both the induction of atherogenic adipokines, such as plasminogen activator inhibitor (PAI)-1, and the inhibition of the anti-atherogenic adipokine, adiponectin. In this study, we investigated the effects of trans-stilbene, piceatannol, rhaponticin, and piceid on the TNF-${\alpha}$-induced atherogenic changes of adipokines in 3T3-L1 cells. Exposure to TNF-${\alpha}$ for 24 hr increased PAI-1 secretion and decreased adiponectin secretion. Among stilbenoids, piceatannol significantly inhibited the increased secretion of PAI-1 induced by TNF-${\alpha}$. Adiponectin secretion decreased by TNF-${\alpha}$ was recovered after trans-stilbene and rhaponticin treatments. Our results showed that stilbenoids exerted different effects on TNF-${\alpha}$-induced changes in adipokines secretion in 3T3-L1 adipocytes according to their structural characteristics.
Antioxidative and protective effects of Ulmus davidiana var. japonica against oxidative damages induced by glutamate in PC 12 cells were investigated. Inhibitory activity against $FeSO_{4}-H_{2}O_{2}$-induced oxidative stress and DPPH radical-scavenging activity were detected in ethyl acetate and butanol fractions of ethanol extracts from stems and roots. Ethyl acetate and butanol fractions of ethanol extracts from roots significantly inhibited glutamate-induced cytotoxicity and reactive oxygen species in PC 12 cells. These results demonstrate ethyl acetate and butanol fractions of ethanol extracts of U. davidiana var. japonica have potent protective effect against glutamate-induced oxidative stress.
Effects of defatted sesame and perilla methanol extracts on cognitive function and antioxidant activity of learning- and memory-impaired animal model SAMP8 mice. Animals were divided into 4 groups and fed with diets containing 0.3%(w/w) defatted sesame (S) or defatted perilla methanol extracts (P) for 12 weeks. Step through latency of SAMP8 control group was significantly higher than that of SAM R1 normal group, whereas significantly increased in S and P groups compared with SAMP8 control on passive avoidance test (p<0.001). Acetylcholinesterase activity of brain in SAMP8 increased compared with SAMR1 but no difference between SAMP8 control group and sample-treated group. Brain TBARS contents of SAMP8 control significantly increased compared with SAMR1 and were lowered significantly by supplementation of defatted sesame and perilla methanol extracts. Defatted sesame and perilla methanol extracts attenuated increased brain superoxide dismutase and glutathione peroxidase activities in SAMP8. These results suggest defatted sesame and perilla methanol extracts could attenuate cognitive deficits induced by aging possibly through activation of antioxidant activity of defatted sesame and perilla methanol extracts.
The effects of cell lytic enzyme treatment on total phenolic content, antioxidant and antityrosinase activities of lotus leaf were investigated. The dried lotus leaves were hydroyzed by cell lytic enzymes such as Promozyme, Ceremix, Pectinex, Ultraflo, Celluclast, Pentopan, Tunicase, Viscozyme at their optimum pHs (pH 5-8) at $50^{\circ}C$ for 4 hrs. Depending on the enzymes used, total phenolic compounds content was measured as $1,079-1,476{\mu}g$/mL, and antioxidant activities and whitening activities were increased by 5~10% and 20%, respectively Among the tested hydrolytic enzymes, Promozyme (pullulanase) was selected as the most suitable enzyme for the extraction of total polyphenol from lotus leaf. The optimal dosage of Promozyme were found to be 1-2% (w/w). By Promozyme treatment, total phenolic compounds content of the lotus extract significantly increased compared to the extraction without enzyme treatment.

In this study, bioactive components such as polyohenols, flavonoids and tocopherols were determined in cultured polar microalgae (Fragilariopsis pseudonana, Chaetoceros neogracile, Stellarima microtrias, Porosiara pseudodenticular). Antioxidant activity of methanol extracts of polar microalgae was also investigated. ${\alpha}-Tocopherol$ contents in Fragilariopsis pseudonana were almost two times higher than those of Chaetoceros neogracile in. The antioxidant activity of methanol extracts of Fragilariopsis pseudonana methanol extracts determined by ABTS assay was higher than other algae. Total polyphenol contents of methanol extracts also showed a similar trend as antioxidant activity. The protective activity against oxidative damages induced by glutamate in PC 12 cells was shown in only Chaetoceros neogracile.
Astaxanthin is an unsaturated compound with a double bond. So it is easily decayed by heat and oxidation (light) during its storage and processing of it. Nanoliposome formulation technology was utilized to improve the stability of astaxanthin. Nanoliposome preparation conditions were established and the stability of astaxanthin encapsulated nanoliposome and free astaxanthin was investigated. Thermal stability and UV-stability of astaxanthin encapsulated nanoliposome increased up to two times and tree times, respectively. Astaxanthin encapsulated nanoliposome could be used as a stable functional material for industrial purposes.
The relationship between cognitive function and dietary patterns in Korean elderly women was investigated in this study. Twenty-four-hour dietary recall and cognitive function tests were performed in 170 elderly women aged over 60 years. Average age of the subjects was 71 years and the average number of family members was 3.3. Most of the subjects had less than seven years of education and their monthly income was much lower than the Korean average. Most of the subjects engaged in regular exercise and did not indulge in drinking and smoking. The average cognitive function score of the subjects was 58.4. Most of the subjects appeared to consume insufficient amount of food ; average intakes of energy, Ca, iron, vitamin A, and riboflavin were lower than RDA for the elderly. Cognitive function score was positively related to education level and regularity of exercise. Also, cognitive function score had a strong relationship with food intake such as total amounts of foods, fish and shellfish, milk and dairy products, total animal food, fruit, bread and sugar. Energy, protein, Ca, p, riboflavin, and niacin were also shown to have positive relationships to cognitive function score, while carbohydrate caloric ratio had a negative relationship with cognitive function. According to the results of multiple regression analysis, factors that most influenced on cognitive function were education level and riboflavin intake. The subjects who had more than seven years of education had significantly higher cognitive function scores than those with no school education. And cognitive function scores f3r those who consumed more than 2/3 of the RDA for most nutrients were significantly higher than the ones fir the subjects who consumed less than 2/3 of the RDA. These results suggest that proper education and adequate nutrient intake in quantity and quality by achieving food diversity are essential in maintaining cognitive function in later life (Korean J Nutrition 31(9) : 1457-1467, 1998)

Purpose: This study was conducted to investigate the effect of freeze-dried Gastrodiae rhizoma powder (GR) on the liver function and alcohol metabolism in alcohol treated rats. Methods: The rats were administered various concentrations of GR (100 mg, 200 mg, 300 mg/kg B.W.) for 3 days 1 hour before 50% (v/v) ethanol (3 g/kg B.W.) administration. Two tests focusing on liver function and alcohol metabolism in acute alcohol treatment were carried out. Results: Glutamic oxaloacetic transaminase activity was significantly increased by alcohol treatment, and was decreased by 100 mg GR administration. Acute ethanol treatment led to significant increase in alcohol and acetaldehyde levels of serum and liver. However, 100 mg GR administration led to a significant reduction in increased alcohol level in the serum with decreased alcohol dehydrogenase (ADH) activity and increased acetaldehyde level in liver was significantly reduced by three levels. Conclusion: These results suggest that GR can be effective in enhancing liver function and alcohol metabolism in the alcohol-treated rats. Studies on the appropriate dosage of GR should further be developed to treat alcohol detoxification and stimulate liver function.
A well-holder type piezoelectric chloramphenicol (CAP) immunosensor which was prepared by binding an antiCAP antibody to the chemisorbed monolayers of various thiol or sulfide compounds over the gold electrode surface of quartz crystals through a carboxyl-amine coupling procedure, using the activation with l-ethyl- 3-(3-dimethylarninopropyl)carbodiimideHCl and N-hydroxysulfosuccinimide, was determined for its responses to CAP, CAP succinate, and water-soluble CAP. The reaction phase used in the well holder was 0.01 M phosphate buffer (pH 7.4), and the solvent for analyte dissolution varied according to the solubility of the individual analyte. The analyte detection which was indicated by a steady-state frequency shift was finished within 10 min, except for CAP dissolved in methanol. The responses of CAP succinate and water-soluble CAP in the reaction phase were very stable, while a minute fluctuation was found with CAP.
The optimization of a batch-type quartz crystal microbalance (QCM)-precipitation sensor measuring acetylcholinesterase (AChE) activity was conducted. To covalently bind AChE onto the gold electrode of a QCM surface, glutaraldehyde cross-linking to a cystamine self-assembled monolayer was tried at different cystamine concentrations. At the optimum conditions of the QCM-precipitation sensor, 0.1 M potassium phosphate buffer (pH 8.0), containing 0.01% Tween 80, was used as the reaction buffer, with the enzyme amount of 5 units for immobilization and the substrate concentration of 50 mg/ml. The current biosensor might find a future applicability to the sum parameter detection on organophosphorus and carbamate pesticides.
