Transfer of the isolated nuclei from Lentinus edodes into protoplasts of Pleurotus florida was induced with polyethlene glycol (PEG) and $CaCl_2$. The intergeneric transfer products were classified into nuclear hybrid, heterokaryon or synkaryon, and reconstituted cell. These progenies except nuclear hybrids formed mature fruiting bodies on sawdust rice bran medium. Formation of fruit bodies was influenced by several factors such as light, temperature, nutrition and physic state of the culture media. Most of fruiting body characters were similar to those of P. florida in synkaryon and L. edodes in reconstituted cell, respectively. All these basidiocarps had clamp connections though initial heterokaryon colonies were lacking. Isozyme patterns of intergeneric progenies were quite different from those of parents. DNA polymorphisms of transfer products were also compared by random amplified polymorphic DNAs (RAPD) analysis based on polymerase chain reaction. The RAPD patterns were different from those of donor and recipient. DNA fingerprints ranged in size from 0.25 to 4.0 Kb. On the basis of RAPD, the transfer products were classified into five groups. Two synkaryon were analysed with distribution of progenies and segregation of genetic markers by random spore analyses. The genetic markers were segregated into wild type and riboflavine requiring auxotrophs.
This study was carried out to understand the flowering and pollination characteristics in Bupleurum falcatum. Estimated total node numbers of primary branches were 106.6, and the total numbers of florets were theoretically 9,622 until secondary branches flower. In the sizes of floral organs according to the branch order, all of ray, pedicel, petal, pistil, stamen, ovary surface, and sepals have a tendancy to be smaller as this order: main stem> 1st branch> 2nd branch. As the branch development proceeds, the size of florets was getting smaller. Each flowering periods, especially stamen emergence, in different branch positons and in intra-branch were clearly divided. In case of florets, umbellets, and compound umbels enveloped in the gauze and paper, they showed 0% fertilization rate. However, the plants which were entirely enveloped in gauze led to the partial fertilization. Consequently, it was admitted that B. falcatum was a partly allogamous plant, fertilized in the way of entomophily. In respect of pollinatability according to the steps of floral organs maturation, this plant was able to bear fruit after 8 days (Pistil maturation). It was at maturity stages, especially 11 through 13 days, that the maximum fertilization rate appeared.
Purpose - Given an increase in the consumption of plums, prices have fluctuated in an unstable manner, making it difficult for farmhouses to sell the product. This study intends to provide information on the cultivation and sale of plums to consumers, thus enabling producers to utilize relevant information to analyze the types of plums that are preferred and consumed by users. Research design, data, and methodology - In this study, a survey was conducted on plum consumption by a consumer panel established and operated by the Rural Development Administration in December 2009. The objective was to identify the purchasing awareness of plums and to analyze panel data from 2010 to 2013 using a linear regression model, a Tobit model, and a panel regression model to derive the purchase characteristics. Results - The outcome of the survey on plums is as follows. Plums are purchased because they are good for the health (90.6%), which means that most customers purchase plums for their health benefits. When plums are in season, the purchase rate is 94.8%, indicating that most plums are purchased when they are in season and that selling plums when they are out of season is difficult. Therefore, we sell most plums in the correct season, and the rest of the plums need to be processed and then sent to markets. The strongest reason for not purchasing plums is that they are difficult to process for consumption (63.1%), followed by the reason that the fruit is unfamiliar (15.5%). Regarding solutions for increasing the consumption of plums, the answers were as follows: distribute a recipe for plums (36.9%), advertise its effect through TV or the press (31.1%), and develop various processed products (15.6%). When customers decide to pick out plums, the major considerations were freshness (4.43), safe to eat (4.16), price (3.96), size (3.87), brand (3.28), and discount event (2.62). Freshness is important for decision making and safe to eat was more important than price because plums are washed and processed into plum jam. According to the results of the linear regression model, a higher family income results in a higher purchasing amount. However, the amount of plums purchased by a person was reduced if his or her income increased. Compared with individuals who used other purchasing agents on weekdays, those who used the traditional market turned out to purchase a higher amount of plums on the weekdays. Conclusions - Considering that numerous people purchase plums for their health benefits, promoting the consumption of plums is anticipated as being successful if they can be produced safely for consumption and for inclusion in recipes and various processed foods, and to promote eco-friendly agricultural practices.
한국산 가막사리속(Bidens L.)에 포함된 10분류군(8종 2변종)의 수과형태를 연구하기 위하여 입체현미경과 주사전자현미경을 이용하여 관찰하였다. 연구된 가막사리속에서 까끄라기를 제외한 수과의 크기는 $3.0-19.5{\times}0.7-2.6 mm$이다. 관모는2-4개의 까끄라기로 구성되어 있고, 까끄라기의 길이는 2.2-5.6 mm이다. 강모의 배열은 1-3열이었다. 수과의 다형화가 6개의 분류군(B. bipinnata, B. biternata, B. frondosa, B. parviflora, B. pilosa var. pilosa, B. tripartita var. tripartita)에서 확인되었다. 수과의 형태와 까끄라기의 수에 의해 4가지 유형으로 구분되었다. - Type A: 장타원형이고, 까끄라기 수는 2개(B. parviflora); Type B: 장타원형이고, 까끄라기 수는 3개 이상(B. bipinnata, B. biternata, B. pilosa var. pilosa, B. pilosa var. minor) Type C: 도란형이고, 까끄라기의 수는 2개(B. frondosa, B. radiata var. radiata, B. radiata var. pinnatifida, B. tripartita var. tripartita); Type D: 도란형이고, 까끄라기의 수는 3개 이상(B. cernua). 까끄라기의 강모 표면무늬는 3가지 즉 나선상, 평활상, 불규칙한 직선형으로 나타났다. 수과의 표면은 각피층이 발달한 다각형, 평활상 그리고 굴곡형으로 나타났다. 수과형질의 분류학적 적용에 대하여 간략하게 토의하였다. 수과형질을 바탕으로 한국산 가막사리속 분류군에 대한 검색표를 제시하였다.
Life history and control methods of Aphanostigma iakusuiense Kishida were studied during April to November, 1974, in the Naju pear orchard area, Cholla Namdo. The results are summarized as follows: 1. More than $60\%$ of wintering eggs were found at the height of 1.5 to 2.0m from the soil surface, with approximately $10\%$ at 0.5 to 1.5m and 1.5 to 2.5m respectively. 2. There was little difference in the size of egg masses and hatching ratio $(12.7-13.7\%)$ between the varieties of host trees, Mansamkil and Kooksoo. 3. Hatching was not observed until April 6, but hatches of $12.8\%$ on April 13 and $90.6\%$ on May 4 were found. 4. During the hatching season in early spring, 8 chemicals were applied to test effectiveness of control. C8514, Metasystox, Acar 338, and Parathion were more effective than either Sevin or Malix for control of this insect. 5. Highest populations of pear phylloxera ocurred from August 20 to September 20 at which time levels of eggs and larvae were simillar in abundance. 6. In tests for control of this insect after in paper bags, DDVP, Loxon, C8514, Sevin, and Binapacryl were more effective than either EPN or Otran. 7. Predacious mite, Protolaelape pygmaeus Miller, and several species of lady beetles (Coccinellidae) preying on this insect as natural enemies were collected. The predation ratio of mites varied according to region: that is 0.77 to $18.6\%$ in Bia Orchard; $7.88-48.7\%$ in Noan Orchard and almost none in Youngsanoo Orchard. 8. The varietal difference in fruit damage by this insect was in the order of Choseng Chuck, Kooksoo, followed by Sin Ko, Mansamkil, Keumchon Choo, and Changsimrang.
We investigated quality changes in Setoka ([Citrus unshiu$\times$Citrus sinensis]$\times$Citrus reticulate]$\times$[Citrus reticulata$\times$Citrus sinensis]) wrapped in 30-m pore size low-density polyethylene film (LDPE) and stored at different temperatures. LDPE wrapping effectively inhibited weight loss, and fruits seemed fresh even after room temperature storage. The decay rate was higher at room temperature and with LDPE wrapping. The respiration rate of LDPE- wrapped fruits was somewhat higher than that of control samples. Thus, no modified atmosphere packaging (MAP) effect was observed when 'Setoka' citrus was studied. Acidity decreased at a constant but different rate with each storage method used, and these rates were correlated with respiration rates. Soluble solid content (SSC), however, increased during 45 days of storage a subsequent rapid decrease was then observed.
Citrus juice, a concentrate manufactured by the Jeju Provincial Corporation, was converted into vinegar orderly by alcohol and acetate fermentation. The juice with 6 folds dilution by distilled water was used as the sole nutrient source through out experiments. Diluted juice contained $12.96^{\circ}Brix$ of total sugar, $0.632\%$ of total acid and $20.23{\mu}g/m{\ell}$ of hesperidin. Naringin was not detected from the juice. Citrus wine having $5.6\~6.3\%$ alcohol was produced from diluted juice by 3 days of fermentation at $28^{\circ}C$. A kind of malomelo yeast CMY-28 was used for wine fermentation. The wine was succeedingly fermented for 8 days at $30^{\circ}C$ after inoculation of seed vinegar which contained active cells of acid producing bacteria CV1. Inoculum size of seed vinegar was controlled to $10\%$(v/v) of citrus wine. The wine converted into vinegar by the fermentation. Citrus vinegar, the final product of fermentation, was colored with very thin radish-yellow and transparent. It's acidity ranged between $5.8\~6.2\%$ as acetic acid. The vinegar got the best score by sensory test among several natural fruit vinegars. It was clear from the results that citrus vinegar in high quality could be produced from concentrated citrus juice, however fermentation conditions should be improved to reduce the amount of reducing alcohol.
This study examined how achievement of session goals contributes to outcomes of subjects after participation in a 12-week lifestyle intervention program in men with metabolic syndrome (MetS). Thirty office workers with MetS, aged $47.2{\pm}6.6$ years, participated in this study, from March to July, 2011. The intervention program included face-to-face counseling five times during the 12-week period. Counselors and subjects designed session goals for each round. The average of the goal achievement rate was calculated based on compliance for each round. The subjects were divided into three groups according to their tertiles of achievement rate: Low-compliance group (LC, < 59%), medium-compliance group (MC, 59-70%), and high-compliance group (HC, > 70%). Anthropometry, biochemical index, and nutrient intake were examined at baseline and at the end of the 12-week intervention program. After the intervention, diastolic blood pressure (DBP) showed a significant decrease in the LC group, and waist circumference (WC) showed a significant decrease in the MC group. Systolic blood pressure (SBP), DBP, and low-density lipoprotein cholesterol (LDL) showed a significant decrease in the HI group. Changes in SBP and DBP were significantly lower in the HC group than in the MC group (p < 0.05, p < 0.01). Changes in LDL were significantly lower in the HC group than in the MC group (p < 0.05). Results for intake of total energy, protein, fat, and sodium, as well as rates of carbohydrate and fat intake, showed a significant decrease in all participants (p < 0.05). The change in fiber was significantly higher in the HC group than in the MC group (p < 0.05). The change of fruit serving size showed a significant increase in the HC group (p < 0.01). The number of risk factors for MetS showed a significant decrease in the LC and HC groups (p < 0.05), however, no significant mean differences were observed among the three groups. In conclusion, participation in this intervention program resulted in positive effects on risk factors for MetS, nutrient intake, and dietary habits, especially in the High-compliance group.
This study was conducted to reduce the phenomenon of biased cultivation and to pursue the diversification of mushroom items. We focused on developing Sparassis latifolia varieties with a short cultivation period, stable pinheading, and a high yield. We have collected and determined the characteristics of genetic resources domestically and abroad since 2016. We bred the unique domestic variety, "Hanyeoul" and the results of this study are as follows. The optimal temperatures for mycelial growth and fruit body growth were 22~25℃ and 19~21℃, respectively. The color of the pileus was pale-yellowish white, which was similar to the color of the control variety, "Neoul". The number of days required for cultivation was 125 for bottle cultivation and 98~102 for bag cultivation, which was shorter than the number of days required for the control variety. Bottle cultivation took 39 days shorter for the new variety than the control variety. The fruiting body of the new variety was similar in size or larger than that of the control variety. The pinheading rate was more than 90% in bottle cultivation and bag cultivation, which was more stable than pinheading in the control variety. The yield was 79.1 g/bottle (1,100 cc) for bottle cultivation, which was 22.6% higher than the yield of the control variety, and 641.1~689.5 g/bag (2.6 kg) for bag cultivation, which 5.8~10.4% higher than yield of the control variety.
Purpose: The size of the salad consumption market has expanded since Covid-19, and continuous growth is predicted. Therefore, by extracting influential core purchasers in the salad consumption market and analyzing their purchasing behaviors and consumer types, this study intended to provide basic data for establishing a marketing strategy. Methods: The analysis data is the purchasing data of 576 people who have purchased salads between 2016 and 2020 (panel data of the Rural Development Administration), and in the social network analysis, the centrality structure was analyzed. Results: First, in the results of analyzing the causes of the rapid increase in salad consumption in 2020, it was found that the increase in consumption of new purchasers (n=102) had little effect. The existing consumer type (n = 474), which has been the majority of the salad consumption market so far, were consumers with stable income. However, the results of study indicated that the type of consumers has expanded since low-income class as well as high-income class increased consumption of purchasing salad. Second, in the results of analyzing the types of key purchasers with great influence in the salad consumption market, there was a difference from the results of frequency analysis in age, number of family members, existence/absence of children, and income decile. This suggests that there should be a difference between the type of customers according to the apparent quantitative figure and the actual influential purchasers. Third, in the results of analyzing the salad purchasing behaviors of core purchasers, the purchasing site for existing purchasers was large-scale marts and for new purchasers it was corporate-type supermarkets. Purchases were concentrated on Saturdays for both existing and new purchasers. As for the purchased products, existing purchasers had a high preference for products made of chicken, and new purchasers had a high preference for vegetable/fruit salad. In particular, in the results of purchased products by age group, in the case of 50s and 60s, it was an interesting result that there was a difference between the products purchased by the existing and new purchasers even though they were the same age. Conclusion: When establishing a marketing strategy in the salad consumption market, it is necessary to pay attention to the purchasing behavior of key buyers.
