Histological and Histochemical Studies on the Cutaneous Mucous Glands According to the Development of Frog, Rana nigromaculata (개구리(Rana nigromaculata)발생에 따른 피부점액선의 조직학적 및 조직화학적 연구)

  • The present study was performed histologically and histochemically to observe the cutaneous mucous glands in the frog, Rana nigromaculata during metamorphosis. The cutaneous thssues including dermal plicae in the dorsal portions of the frog tadpoles at each metamorphosis stage were fixed in 10% buffered formalin at$4^{\circ}C$, embedded in paraffin wax, sectioned 4 $\mu$m thickness and stained with periodic acid-Schiff(PAS) and alcian blue (AB) at both pH 2.5 and pH 1.0. The results of observation were as follows: 1. The developments of cutaneous mucous glands of the frog tadpole were begun with appearance of gland cell nest in the dermis at metamorphosis XV stage and significant numerical increases could be seen at metamorphosis XX, XXIII and XXIV stages. 2. This cutaneous mucous gland of the frog tadpole could be divided into two types; A-type glands showed strong positivities to both PAS and AB at pH 2.5 in the gland body cells and to PAS in the gland neck cells, and B-type glands at AB pH 2.5 in the gland body cells. 3. In the A-type mucous glands, the reactivities of the glandular epithelial cells to both PAS and AB stain could be first seen at the metamorphosis XIX stage of frog tadpole. The reactivities of the glandular epithelial cells to both PAS and AB pH 2.5 were gradually increased according to the process of metamorphosis after XX stage of metamorphosis. 4. The B-type mucous glands were first seen at the XX stage and the reactivity of the glandular epithelial cells to AB at pH 2.5 was gradually increased according to the process of metamorphosis after XX stage. 5. The A-type and the B-type mucous glands were in the ratio of 99 : 1, 7 : 3 and 5.5 : 4.5 for each of metamorphosis XX, XXI-XXII and XXIII-XXV stages. 6. The remarkable development of the cutaneous mucous glands of the frog tadpoles might be needed to maintain water and electrolyte balances according to the change of way from aquatic life to amphibious.

  • The cobalt capture method of Gomori's modified technique(Gomori, 1952) was employed to study the histochemistry of the developing frog kidney. Alkaline phosphatase activity was observed in association with the brush borders of pronephros and mesonephros. By the stage of transition from larva to tadpole alkaline phosphatase activity was gradually increased in the brush border of pronephros, and as the pronephros begun to degenerate the enzyme activity was decreased and disappeared. By the time of maximum development of the pronephros the mesonephros began to develop and alkaline phosphatase activity of the mesonephric tubules showed highly positive throughout the stage of metamorphosis. No activity was observed in association with the collecting tubules and ductal elements.

Fine Structure of the Epithelial Apoptosis in the Anuran Tadpole Rana nigromaculata (참개구리(Rana nigromaculata) 유생기 상피 세포사의 미세구조)

  • The fine structural characteristics of the apoptotic cells in the cutaneous epithelium of the anuran tadpole of the black-spotted frog, Rana nigromaculata was examined using the TUNEL (Terminal deoxynucleotidyl transferase-mediated biotinylated d-Uridine triphosphate Nick End Labeling) staining technique and TEM (transmission electron microscopy) observations. The cutaneous epithelium of the tadpole was composed of stratified cuboidal cells and the apoptotic cell death was observed continuously during the tail degeneration stages from the Shumway stage number 31 to 33. The early apoptotic cells shown in the epithelium demonstrated condensation and margination of the chromatin material at the nuclear periphery, and nuclear breakdown and cytoplasmic condensation were followed. Subsequent cytoplasmic degeneration of the apoptotic cell were produced by membrane-bounded cell fragments with relatively well preserved organelles. Following the processes of autophagic degradation, the late apoptotic cells being phagocytosed by other surrounding cells. These nearby cells, presumptive intraepithelial macrophages, contain a variety of lysosomal residual bodies which fuses with other cell organelles or other cytoplasmatic material to form secondary lysosomes. They are soon transformed into lamellar shaped vesicles and finally disappeared during the process of degradation.

Influence of Cu(II) on the Growth of Korean Tadpole, Rana nigromaculata (銅이온이 참개구리 幼生의 成長에 미치는 影響)

  • The author hatched the eggs of Korean frog, Rana nigromaculata in natural water, and reared the tadpoles in natural water as control group and in seven copper ion groups contaminated by 0.1, 0.2, 0.3, 0.4, 0.5, 0.7, and 1.0 ppm of copper ion. The influences of copper ion on the growth of the tadpoles were analyzed, and the results were summarized as follows: 1) The copper ion inhibited the growth of the tadpole. 2) The tadpoles were not survived for 20 days in the copper ion of 0.4~0.5ppm, and in the 0.7ppm, the tadpoles were not survived for 1 day after hatch. 3) The critical lethal concentration of Korean tadpole is regarded as 0.6ppm of copper ion. 4) The growth of head with and body length show a convexin increase pattern. 5) In the growth quantity of head width and body length, that of natural water shows the most rapid increase pattern, and that of the copper ion groups 0.1, 0.2, and 0.3ppm follows in that order. 6) The coefficient of relative growth($\alpha$) of control group is the greatest value, and that of the copper ion groups of 0.1, 0.2, and 0.3ppm follows in that order. The natural water shows the positive allometry, the contaminated groups shows the negative allometry in the relative growth of the contaminated groups to the natural water. 7) Body length shows positive allometry in the relative growth to head width.

A Study on the Effect of Chemicals on the Survival Rate and the Metamorphosis Speed in Rana nigromaculata Hallowell. (수종의 약품이 개구리의 생존율과 변태속도에 미치는 영향에 관한 연구)

  • This experiment is to search what some chemicals polluting water have an effect on the Rana nigromaculata Hallowell, in the survival rate and the metamorphosis speed from a tadpole to a young frog. 1. The fatal thickness by some agricultural chemicals and copper sulfate is different from one another. The fatal rate in the contrast group is 15 percent, and particulary Dithane M-45 is the most among the all reagents. 2. The effect on the anamorphosis is very severe. In the case of contrast group, it appears to stage 25, but it is not to appear at the group used agricultural chemicals. 3. The less the thickness of that is, the less the influence of the survival rate and the metamorphosis speed. 4. The density of the agricultural chemical is in inverse proportion to the survival rate and the metamorphosis speed, and the other side, the fatal rate is proportion.

Morphological Characterization and Classification of Anuran Tadpoles in Korea

  • The tadpoles of 12 Korean anuran species, including Bombina orientalis, Bufo gargarizans, B. stejnegeri, Hyla japonica, Kaloula borealis, Rana dybowskii, R. huanrenensis, R. coreana, R. nigromaculata, R. chosenica, R. rugosa, and R. catesbeiana, were classified based on their morphological characteristics. We collected eggs or tadpoles of the 12 Korean anuran species from Gangwon, Incheon, Chungcheong, and Gyeonggi districts in 2005 and 2006 breeding seasons. When the tadpoles reached at $27{\sim}37$ Gosner's developmental stages, we described morphological characteristics of the tadpoles of each anuran species and measured their physical parameters such as total length, body length, and body mass. After that, we chose 12 morphological characteristics to identify each species and to use them as classification keys such as eye location, caudal musculature pattern, spiracle location, oral disc morphology, and labial tooth row formula. In this paper, we presented classification keys, morphological characteristics, and drawings for the tadpoles of 12 anuran species.

Histochemical and Fine Structural Visualization of the Epithelial Apoptosis in the Anuran Tadpole during the Tail Regression Stages (무미양서류 꼬리 퇴화과정중 피부 상피조직의 세포사에 관한 조직화학적 및 미세구조적 연구)

  • The programed cell death of the cutaneous epithelial tissue during tail regression stages in anuran tadpoles of the blackspotted frog, Rana nigromaculata were visualized by the histochemical and transmission electron microscopic techniques. Metamorphotic changes in the tail regression during the period of the Shumway stage number 31 to 33 are characterized by the disappearance of mucous layer and formation of compound epithelium through cutaneous thickening. Following the TUNEL (terminal deoxynucleotidyl transferase-mediated biotinylated d-uridine triphosphate nick end labeling) staining technique, the apoptotic cells were detected at the distal region of the tail skin initially, but they can be seen at the proximal region according to their following development. It has been also revealed that the number of the TUNEL-positive cells gradually increased from apical to basal direction of the epithelial layers during the tail regressing stages. Following the TEM observation, the early apoptotic cells shown in the epithelium demonstrated condensation and margination of the chromatin material at the nuclear periphery. Another epithelial apoptotic cells were shown nuclear fragmentation, membrane blebbing and cytoplasmic condensation. Following the process of the apoptotic degradation, well preserved organelles and nuclear fragments can be identified in the cytoplasm of lysosome-rich cells, however they soon reduced to lysosomal residual bodies through the progressive degradation.

The Effect of Incubation Temperature at Egg Hatching of the Boreal Digging Frog, Kaloula borealis (부화 온도가 맹꽁이(Kaloula borealis)알의 부화에 미치는 영향)

  • This study aimed to determine the egg-hatching period of boreal digging frogs, Kaloula borealis, and investigate whether the incubation temperature affects the hatching period. In this study, the egg hatching was recorded based on the appearance of the tadpole. The results of this study showed that all the eggs hatched within 48 hours after spawning, with 28.1% (±10.8, n=52) hatching within 24 hours and 99.9% (±0.23, n=49) within 48 hours after spawning. The mean hatching rate of tadpoles showed significant differences depending on the difference in water temperature. The mean hatching rate between 15 and 24 hours after spawning was higher at a water temperature of 21.1 (±0.2) ℃ than at 24.1 (±0.2) ℃. The results suggest rapid hatching occurs at relatively low water temperatures because the spawning habits that spawn eggs in temporary ponds or puddles in the rainy season require rapid hatching before the puddles dry out. The results of this study are helpful for understanding the most suitable temperature conditions for the incubation of eggs of the endangered species, boreal digging frog.

Life History and Morphological Responses of Daphnia similis against Chemical Compounds Exuded by Various Cohabiting Animals and Triops longicaudatus in the Rice Paddy Ecosystem (논 생태계에 서식하는 생물과 긴꼬리투구새우가 분비한 화학물질이 물벼룩의 생활사와 형태에 미치는 영향)

  • The aim of this study was to assess the interaction between Daphnia similis and various organisms related to the rice paddy ecosystem. We selected several organisms that are likely to prey on D. similis and evaluate predation rate as well as responses of D. similis to the chemical compounds exuded by these organisms. As a result of predation experiment, larval dragonfly (Anax parthenope) and Triops longicaudatus were clearly shown decreasing abundances of D. similis. Especially, Triops longicaudatus was observed higher feeding rates on D. similis than larval dragonfly. Chemical compounds from the vertebrates such as fish (Misgurnus anguillicaudatus, Pseudorasbora parva, Micropterus salmoides) and tadpole of frog (Rana nigromaculata) did not affect the life history of Daphnia. However, a potential predatory fish P. parva induced significantly longer tail spine in Daphnia. In addition, among the invertebrates (T. longicaudatus, A. Parthenope, Micronecta spp., Palaemon paucidens), chemical compounds exuded by T. longicaudatus induced shorter body and significantly longer tail spine in D. similis.

Growth and development of Fibricola seoulensis metacercariae in tadpoles (Fibricola seoulensis 피낭유충의 실험감염 올챙이내 성장 및 발육)

  • In order to observe the growth and development of Fibricola seoulensis metacercariae, the tadpoles of Rana nigromaculata were experimentally infected with the cercariae. The meta cercariae of various developmental stages were recovered from the tadpoles after 2 to 65 days of infection. They were prepared for morphological observation, and were given orally to mice to observe their infectivity. The following results were obtained. 1. All of the tadpoles exposed to the cercariae were observed to harbour the larvae in their abdominal cavity. 2. The young metacercariae of 2 days after infection were $121.1{\mu}m$ long and $63.3{\mu}m$ wide. They grew linearly for the first 14 days to be $262.0{\mu}m$ long and $166.4{\mu}m$ wide. Thereafter, no more growth recognized until 65 days. 3. The larvae of 2 days old were similar with cercarial body and had 2 suckers, a pharynx, 2 ceca and a primordium of germ cells but no tribocytic organ. On the 8th day, they had tribocytic organ, and their morphology resembled that of mature metacercariae. 4. The metacercariae younger than 10 days could not infect the mice. Only the metacercariae older than 14 days had infectivity. The recovery rates increased by the age of metacercariae from 19.0% in 14 days old to 70.0% in 40 days old. Above findings indicate that the tadpole is indispensable for metacercarial development and it needs at least 2 weeks for maturation. The tadpole is a pivotal host in the life cycle of F. seoulensis for connection between the snail and the frog.
