종류별 감초의 라디칼 소거능 및 H2O2에 의한 C6 glial 세포의 산화적 스트레스 개선 효과 (Free radical scavenging activity and protective effect of three glycyrrhiza varieties against hydrogen peroxide-induced oxidative stress in C6 glial cells)

  • 산화적 스트레스는 신경퇴행성 질환 발병의 원인으로 알려져 있다. 본 연구는 대표적인 감초 종류인 Glycyrrhiza glabra, G. uralensis와 신품종 감초인 신원감(SW)의 in vitro free radical 소거능을 통한 항산화 활성과 H2O2 유도 산화적 스트레스에 대한 C6 glial cell 보호 효능을 확인하고자 하였다. In vitro assay에서 G. uralensis, G. glabra, SW 추출물은 농도유의적으로 2,2-diphenyl-1-picrylhydrazyl, ·OH, O2- radical 소거능이 증가하여 in vitro 항산화 활성을 확인하였다. 또한, SW 추출물은 G. uralensis, G. glabra 추출물에 비해 총 페놀 및 플라보노이드 함량이 가장 우수하였다. H2O2로 산화적 스트레스를 유도한 C6 glial cell에 3가지 감초 추출물을 각각 처리 시, 농도의존적으로 세포 생존율이 증가와 reactive oxygen species 소거능이 증가하여 3가지 감초 추출물의 산화적 손상에 대한 신경교세포 보호 효과를 확인하였다. 특히, SW 추출물은 G. uralensis, G. glabra 추출물에 비해 우수하게 C6 glial cell 보호 효과를 나타내었다. 또한, 3가지 감초 추출물의 신경교세포 보호 메커니즘을 확인하기 위해, 염증 관련 단백질 발현을 측정하였다. 3가지 감초 추출물은 H2O2만을 처리한 control군에 비해 inducible nitric oxide synthase 및 cyclooxygenase-2 발현 감소를 통해 염증반응 조절을 통한 신경교세포 보호 작용기전을 확인하였다. 본 연구는 G. uralensis, G. glabra, SW 등 3가지 감초 추출물이 산화적 손상이 유도된 신경교세포 보호에 유용한 소재로써의 가능성이 있는 것으로 사료된다.

An ESR Study of Amino Acid and Protein Free Radicals in Solution Part Ⅴ. an ESR Study of Gamma-Irradiated Lysozyme in Frozen Aqueous Solutions

  • An electron spin resonance study has been made on lysozyme in. frozen aqueous solutions irradiated with $_{60}Co$ r-rays in air at $77^{circ}K.$ Water resonances are dominant when the concentration and the temperature are both below 20% and $130^{circ}K$ respectively. More solute radicals are produced in the solution of higher concentration. Majority of the solute radicals results from direct hit of the radiation. The same types of radicals are induced at $77^{circ}K$ whether the substances are irradiated in the dry state or in frozen aqueous solution. Based on these results, it is assumed that the number of ESR centers produced by the secondary intermolecular radical reacions and stabilized in aqueous solutions may depend on the concentration of the solution, and the presence of water may facilitate the secondary radical reactions occuring in the solute molecules after heat treatment. Majority of the solute radicals above around $193^{circ}K$ are believed to react with oxygen to form peroxytype radicals. However, when the solution is subiected to heat-treatment at $265^{circ}K$ after irradiation at $195^{circ}K$ the peroxy-type resonance was not observed, suggestin that an appreciable amount of oxygen is condensed into the ice, at $77^{circ}K.$ in addition to the oxygen that has already been dissolved in solution and react with solute free radicals during the process of heat-treatment. When the solution contains $H_2O_2$, no water resonance but $HO_2$, type resonance was observed probably indicating that the radiation-induced OH radicals are trapped in $H_2O_2$ aggregates and react readily with $H_2O_2$ molecules to poroduce $HO_2$ type radicals even at $77^{circ}K.$.

  • 목 적 : 사람의 미세변화 신증후군과 유사한 신증을 일으키는 것으로 알려진 puromycin aminonucleoside (PAN)의 1회 투여로 발생하는 신세포의 변화는 여러 기전이 작용하는 것으로 알려져 있으며, 최근에는 oxygen free radical이 사구체 손상의 중요한 한 가지 원인이라고 한다. 백서에 PAN을 투여하여 신증을 발생시킨 후, $\alpha$-tocopherol을 투여하였을 때 PAN신증에 미치는 영향에 대해 연구하고자 한다. 방 법 : 건강한 수컷 Sprague-Dawley 흰 쥐를 3군으로 나누어, 1군은 생리 식염수를 1회 투여한 대조군(N=6), II군은 PAN 1회 단독 투여군 (N=8), III군은 PAN투여 후 $\alpha$-tocopherol을 투여한 군 (N=7)으로 하였다. PAN 투여 직전과 투여 후 5일, 11일, 18일에 24시간 뇨의 protein, creatinine을 측정하였고, 18일째에는 혈청 protein, albumin, cholesterol도 함께 측정하였다. 또한 Oxygen free radical에 의한 신장 조직 손상을 평가하기 위해 신장 피질 미세균질액의 lipid peroxide와 reduced glutathione을 측정하였다. 결 과 : PAN투여 5일 후 24시간 뇨 protein/creatinine비는 II군과 III군에서 대조군에 비해 유의하게 증가하였다(P<0.01), 그러나 18일째에는 $\alpha$-tocopherol를 투여한 III군이 PAN을 단독 투여한 II군에 비해 유의하게 단백뇨가 적었다 (P<0.05). 신장 피질의 lipid peroxide과 reduced glutathione은 III군에 비해 II군에서 유의하게 증가하였다(P<0.05). 18일째의 신조직의 형태학적 변화는 PAN단독으로 투여한 II군에서는 사구체 상피세포의 족돌기가 심하게 융합되어 있으나, $\alpha$-tocopherol을 투여한 III군에서는 족돌기가 잘 보존되고 있었다. 결 론 : PAN투여로 초래된 백서 신증에서 $\alpha$-tocopherol의 투여는 lipid peroxidation의 진행을 차단하여 족돌기의 손상을 회복시키며 단백뇨의 배설도 감소시키는 것으로 사료된다.

일산화탄소 중독시 고압산소투여가 8-hydroxydeoxyguanosine 생성에 미치는 영향 (Effects of Hyperoxia on 8-Hydroxydeoxyguanosine Formation in Carbon Monoxide Exposed Rats)

  • Hyperbaric oxygen (HBO) therapy for carbon monoxide (CO) poisoning eventually inducing the hypoxia-reoxygenation condition, may produce oxygen free radicals, which forms 8-hydroxydeoxyguanosine (8-OH-dG) by attacking C-8 position of deoxyguanosine (dG) in DNA. Effects of oxygen partial pressure or duration of HBO therapy with or without CO poisoning on the tissue 8-OH-dG formation were investigated. Male Sprague-Dawley rats were grouped and exposed to air (control group), 4000 ppm of CO for 10 to 30 minutes (CO only group), air for 30 minutes after 30 minute exposure to 4000 ppm of CO(CO-air exposure group), HBO after 30 minute exposure to 4000 ppm of CO(CO-HBO group), or HBO therapy fo. $10{\sim}120$ minutes(HBO only group). The 8-OH-4G concentrations in the brain and the lung tissues were measured with high performance liquid chromatography and electrochemical detector (ECD). Average concentrations of the 8-OH-dG of each group were statistically compared. In the brain tissues, 8-OH-dG concentrations of the CO only group, the CO-air exposure group, and the CO-HBO group did not significantly differ from those of the control group. Similar insignificance was also found between the CO-HBO group and the HBO only groups. No appreciable dose-response relationship was observed between the 8-OH-dG concentration and the oxygen partial pressure or the duration of HBO. However, the 8-OH-dG concentrations of the 30 minute CO only group were higher than those of the CO-air exposure group (p-value<0.05). In the lung tissues, there were no significant differences between the 8-OH-dG concentrations of the control group and those of the CO only group, the CO-air exposure group, and the CO-HBO group. However, mean 8-OH-dG concentration of the CO-air exposure group was significantly higher than that of the CO only group under the same CO exposure condition(p-value<0.05). With the duration of CO exposure, the 8-OH-dG concentrations of the lung tissues decreased significantly (p-value<0.05). The concentrations of 8-OH-dG in the lung tissues proportionally increased with the duration of HBO, but no such relation was observed with the oxygen partial pressure. These results suggest that the brain may be more resistant to oxygen free radicals as compared with the lungs, and that oxygen toxicity following HBO may be affected by factors other than oxygen free radicals.

Antioxidative Effect of Rhus javanica Linne Extract Against Hydrogen Peroxide or Menadione Induced Oxidative Stress and DNA Damage in HepG2 Cells

  • The free radical scavenging activities and the protective effects of Rhus javanica extracts against oxidative damage induced by reactive oxygen species (ROS) were investigated. n-Hexane, ethyl acetate and water fractions were prepared from a methanol extract. DPPH radical, superoxide anion and hydroxyl radical scavenging activities were estimated. Intracellular ROS formation was quantified using fluorescent probes, 2', 7'-dichlorofluorescin diacetate (DCFH-DA) for hydroxyl radical and dihydroethidium (DHE) for superoxide anion. The oxidative DNA damage was investigated by the comet assay in HepG$_2$ cells exposed either to $H_2O$$_2$ or to menadione. The highest $IC_{50}$/ values for DPPH radical scavenging activity was found in the ethyl acetate fraction with a value of 5.38 $\mu\textrm{g}$/mL. Cells pretreated with $\geq$ 1 $\mu\textrm{g}$/mL of the ethyl acetate extract had significantly increased cell viability compared to control cells, which were not pretreated with the extract. Intracellular ROS formation and DNA damage in HepG$_2$ cells, which were pretreated with the various concentrations of Rhus javanica ethyl acetate extract and then incubated either with $H_2O$$_2$ or with menadione, reduced in a dose-dependent manner. These findings suggest that Rhus javanica might have biologically active components which have strong protective effects against ROS induced oxidative damages to the biomolecules, such as cell membranes and DNA.

  • 비록 스트레스의 종류는 다를지라도, 환경 스트레스들은 공통적으로 식물채 내의 활성 산소의 양을 증가시킨다. 활성산소에 의하여 스트레스-에틸렌의 생성량은 증가하고 결국 식물체의 노화는 촉진된다. 반면에, 활성산소 분해 효소들과 antioxidiant들에 의하여 활성산소에 의한 스트레스-에틸렌 생성이 제한될 경우 한정된 양의 에틸렌에 의하여 스트레스에 대한 저항이 유도될 가능성이 있다.

The Isolation and Antioxidative Effects of Vitexin from Acer palmatum

  • Free radicals and reactive oxygen species (ROS) caused by UV exposure or other environmental factors are critical players in cellular damage and aging. In order to develop a new antiphotoaging agent, this work focused on the antioxidant effects of the extract of tinged autumnal leaves of Acer palmatum. One compound was isolated from an ethyl acetate soluble fraction of the A. palmatum extract using silica gel column chromatography. The chemical structure was identified as apigenin-8-C-beta-D-glucopyranoside, more commonly known as vitexin, by spectral analysis including LC-MS, FT-IR, UV, $^{1}H-$, and $^{13}C-NMR$. The biological activities of vitexin were investigated for the potential application of its anti-aging effects in the cosmetic field. Vitexin inhibited superoxide radicals by about $70\%$ at a concentration of $100\;{\mu}g/mL$ and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals by about $60\%$ at a concentration of $100\;{\mu}g/mL$. Intracellular ROS scavenging activity was indicated by increases in dichlorofluorescein (DCF) fluorescence upon exposure to UVB $20\;mJ/cm^2$ in cultured human dermal fibroblasts (HDFs) after the treatment of vitexin. The results show that oxidation of 5-(6-)chloromethyl-2',7'-dichlo-rodihydrofluorescein diacetate ($CM-H_{2}DCFDA$) is inhibited by vitexin effectively and that vitexin has a potent free radical scavenging activity in UVB-irradiated HDFs. In ROS imaging using a confocal microscope we visualized DCF fluorescence in HDFs directly. In conclusion, our findings suggest that vitexin can be effectively used for the prevention of UV-induced adverse skin reactions such as free radical production and skin cell damage.

쇠뜨기 추출물의 항산화 작용에 관한 연구(I) (Evaluation of Antioxidant Potential of Extract/Fractions of Equisetum arvense (I))

  • 본 연구에서는 쇠뜨기 추출물의 항산화능을 조사하였다. 추출물의 free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) 소거활성($FSC_{50}$)은 50% 에탄올 추출물($182.04{\mu}g/mL$) < ethylacetate 분획(54.50) < 당을 제거시킨 플라보노이드 aglycone 분획(14.13) 순으로 증가하였다. Luminol-의존성 화학발광법을 이용한 $Fe^{3+}-EDTA/H_2O_2$ 계에서 생성된 활성산소종(reactive oxygen species, ROS)에 대한 총항산화능은 당을 제거시킨 플라보노이드 aglycone 분획($OSC_{50}$, $3.54{\mu}g/mL$) < 50 % 에탄올 추출물(0.80) < ethylacetate 분획(0.006) 순으로 ethylacetate 분획에서 가장 큰 활성을 나타내었다. 쇠뜨기 추출물에 대하여 rose-bengal로 증감된 사람 적혈구의 광용혈에 대한 보호효과를 조사하였다. 50% 에탄올 추출물의 경우 농도 의존적($1{\sim}100{\mu}g/mL$)으로 광용혈을 억제하였다. 특히 ethylacetate 분획에서 당을 제거시킨 aglycone 추출물은 $10{\mu}g/mL$ 농도에서 $\tau_{50}$이 161.10min으로 매우 큰 세포보호 활성을 나타내었다. 이상의 결과들은 쇠뜨기 추출물이 $^1O_2$ 혹은 다른 ROS를 소광시키거나 소거함으로써 그리고 ROS에 대항하여 세포막을 보호함으로써 생체계, 특히 태양 자외선에 노출된 피부에서 항산화제로서 작용할 수 있음을 가리킨다.

Comparison of Hydroxyl Radical, Peroxyl Radical, and Peroxynitrite Scavenging Capacity of Extracts and Active Components from Selected Medicinal Plants

  • The ability of 80% ethanol extracts from five medicinal plants, Aralia continentalis, Paeonia suffruticosa, Magnolia denudata, Anemarrhena asphodeloides, and Schizonepeta tenuifolia, to neutralize hydroxyl radical, peroxyl radical and peroxynitrite was examined using the total oxyradical scavenging capacity (TOSC) assay. Peroxyl radical was generated from thermal homolysis of 2,2'-azobis(2-methylpropionamidine) dihydrochloride (ABAP); hydroxyl radical by an iron-ascorbate Fenton reaction; peroxynitrite by spontaneous decomposition of 3-morpholinosydnonimine N-ethylcarbamide (SIN-1). The oxidants generated react with $\alpha$-keto-$\gamma$-methiolbutyric acid (KMBA) to yield ethylene, and the TOSC of the substances tested is quantified from their ability to inhibit ethylene formation. Extracts from P. suffruticosa, M. denudata, and S. tenuifolia were determined to be potent peroxyl radical scavenging agents with a specific TOSC (sTOSC) being at least six-fold greater than that of glutathione (GSH). These three plants also showed sTOSCs toward peroxynitrite markedly greater than sTOSC of GSH, however, only P. suffruticosa revealed a significant hydroxyl radical scavenging capacity. Seven major active constituents isolated from P. suffruticosa, quercetin, (+)-catechin, methyl gallate, gallic acid, benzoic acid, benzoyl paeoniflorin and paeoniflorin, were determined for their antioxidant potential toward peroxynitrite, peroxyl and hydroxyl radicals. Quercetin, (+)-catechin, methyl gallate, and gallic acid exhibited sTOSCs 40~85 times greater than sTOSC of GSH. These four components also showed a peroxynitrite scavenging capacity higher than at least 10-fold of GSH. For antioxidant activity against hydroxyl radical, methyl gallate was greatest followed by gallic acid and quercetin. Further studies need to be conducted to substantiate the significance of scavenging a specific oxidant in the prevention of cellular injury and disease states caused by the reactive free radical species.

Effect of Ischemic Preconditioning on the Oxygen Free Radical Production in the Post-ischemic Reperfused Heart

  • 허혈전처치(ischemic preconditioning)의 재관류심근손상 보호작용과 그 기전을 규명하기 위한 연구의 일환으로 허혈전처치가 심근세포의 산소라디칼 생성능력에 미치는 영향을 검토하였다. 흰쥐 적출심장의 Langendorff 관류표본에서 실험적인 허혈(30분)-재관류(20분)손상을 유도하였고, 허혈전처치는 재관류손상 유도전에 5분 허혈-5분 재관류를 3회 반복하여 시행하였다. 허혈심근 재관류손상의 지표로 심수축기능, 세포질효소 유출, 칼슘 유입 및 미세형태학적 변화를, 그리고 심근세포의 산소라디칼 생성기전으로 xanthine oxidase system의 변동을 허혈전처치와 비전처치 재관류 심장들에서 비교검토하였다. 연구 성적은 다음과 같다. 1. 허혈전처치는 허혈-재관류 심장의 관상혈류량, 심박수, 좌심실압 등 심기능의 저하를 현저히 회복시켰다(회복률; 91%) 2. 허혈-재관류 심장에서 lactate dehydrogenase 유출증가는 허혈전처치에 의해 현저히 저하되었다. 3. 허혈-재관류 심근세포의 전자현미경상 미세구조는 허혈전처치시 비교적 잘 보존되었으며, 특히 myofibril 구조의 보존이 매우 뚜렷하였다. 4. 허혈-재관류시 산화성 조직손상 척도의 하나인 malondialdehyde 생성이 허혈전처치에 의하여 현저히 감소되었다. 5. 허혈전처치 심장에서 xanthine oxidase(D,O 및 D/O형)활성은 변화되지 않았으나 그 기질인 hypoxanthine의 조직함량은 현저히 감소되었다. 이상의 결과들로 부터 허혈전처치는 세포수준에서 허혈심근의 재관류손상을 방지하며, 허혈전처치에 따른 산소라디칼 생성 감소가 재관류손상 방지에 일부 기여할 수 있으리라 사료된다.an을 2주간 처치한 경우에도 영향을 받지 않았다. 이상의 결과로 미루어 아마도 losartan의 내피세포에 대한 작용은 constitutive NO 생성을 증가시키나 inducible NO 생성에는 영향을 미치지 않을 것으로 여겨진다.cium ion이 상당히 중요한 역할을 하는데, 특히 소뇌에서의 NO생성의 감소는 이들 약물들의 치명적 부작용인 tardive dyskinesia와 매우 깊은 관련을 추측할 수 있다. 그러나, 더 많은 약물들의 검색으로 일관적인 기본 결과가 필요 되고 또 각개 약물의 특정적 기전이 연구되기 위하여 현재 실험중이다.(신칭(新稱), Cystostereum subabruptum), 털융단버섯(신칭(新稱), Tomentella pilosa), 노란소나무무늬버섯(신칭(新稱), Asterostroma laxum), 붉은소나무비늘버섯(신칭(新稱), Hymenochaete cruenta), 가루소나무비늘버섯(신칭(新稱), Hymenochaete fuliginosa), 소나무비늘버섯(신칭(新稱), Hymenochaete tabacina), 갈색시루삔버섯(신칭(新稱), Inonotus radiatus), 벚나무진흙버섯(신칭(新稱), Phellinus pomaceus), 회주름구멍버섯(신칭(新稱), Antrodia crassa), 층주름구멍버섯(신칭(新稱), Antrodia serialis), 흰그물구멍버섯(신칭(新稱), Ceriporia reticulata), 겹친손등버섯(신칭(新稱), Oligoporus balsameus), 점박이손등버섯(신칭(新稱), Oligoporus guttulatus), 무른흰살버섯(신칭(新稱), Oxyporus cuneatus), 각목버섯(신칭(新稱),
