Salmonella enterica serovar Gallinarum is the causative agent of fowl typhoid (FT) and Salmonella enterica serovar Pullorum is pullorum disease (PD), a severe systemic disease of chick and it has the same antigenic fomula, the close relation but distinct pathogen. The traditional bacteriologic and serologic methods routinely used but tedious, time consuming. some of biochemical differences are helpful in differentiating the two organisms, however variation in the characteristics of some strains can be observed. During 2006 to 2008, there was isolated 30 strains. The biochemical characteristics of S. Gallinarum was nonmotile, fermentation of dulcitol, maltose but positive arginine (6.6%), lysine (83.3%) and arabinose (20.0%). The antimicrobial susceptibility test showed 100% sensitive to amikacin, ampicillin, amoxicillin/clavulanic acid and florfenicol, but resistant to penicillin (100%) and erythromycin (60.0%). This PCR method can be applied in the diagnosis between S. Gallinarum and S. Pullorum.
본 조사 연구는 종란을 생산하는 원종계, 종계, 백세미씨알 생산 농장을 대상으로 수직 감염(난계대전파)되는 전염성 질병인 추백리/가금티푸스, 닭마이코플라즈마증(MG, MS), 전염성 빈혈증, 조류아데노바이러스 감염증에 대한 항원 및 항체 검사를 실시하였다. 조사 기간은 2009년 8월부터 12월까지 5개월간 원종계 45계군, 종계 1,018계군, 백세미씨 알 생산 54계군에 대한 성적이다. 추백리/가금티푸스 항원검사에서는 모든 계군이 음성으로 확인되었으나, 항체 검사결과 종계 3.2%, 백세미씨알 생산계군 3.0%의 항체 양성율이 관찰되었다. 계종별 가금티푸스의 발생률은 종계군의 항원 검사 결과와 상반되어 육계 44.3.7%, 백세미 26.2%, 산란계 15.7%, 토종닭 12.6%, 육용 종계 1.08%였다. MG 항체 검사 결과, 원종계 71.1%, 종계 및 백세미씨알 생산계군 각 88.7% 항체 양성율이 확인되었으며, MS 항체 검사 결과도 비슷한 수준으로 나타났다. 닭 전염성 빈혈 바이러스 검사 결과, 원종계 42.2%, 종계 18.0%가 바이러스를 갖고 있는 것으로 나타났으며, 항체 양성율도 86% 이상이었다. 이와 함께 조류 아데노바이러스 항원 검사 결과에서는 원종계 4.4%, 종계 2.7%, 백세미씨알 생산계군 9.35%가 바이러스를 보유하고 있는 것으로 조사되었다. 결론적으로 국내 종계군은 닭 마이코플라즈마증과 닭 전염성 빈혈에 상당히 높은 수준으로 감염되어 있는 것으로 판명되어 질병별 적절한 예방책이 필요한 것으로 나타났다.
The objectives in the present study were to evaluate the therapeutic effect of a sodium salts mixture against Salmonella gallinarum infection in broiler. Of this study, sixty broilers at two weeks of age (body weight, $450{\pm}35$ g) were used to estimate the efficacy of a sodium salts mixture (3.25 mg sodium azide, 2.45 mg sodium cyanide, 0.8 g sodium chlorate) against Salmonella gallinarum infection in broiler with drinking water. Broilers challenged with S. gallinarum were administered with ${\times}$ 1(group I) and ${\times}$ 2(group II) sodium salt mixture for seven days, and cecal content samples were collected at the gate of treatment and on 1st, 3rd, 5th, and 7th day after administration. Changes in body weight and cecal shedding of S. gallinarum were monitored during the experimental period. All groups treated with the sodium salt mixture slightly increased body weight compared to control group but there is no significant difference. At 7th day after administration, the number of S. gallinarum in group I and II was significantly decreased compared to control group (p < 0.001). In the hematological and blood biochemical analysis, values of parameters were not significantly different between the treated groups and control group. From results of the present study, the sodium salt mixture had therapeutic effect on S. gallinarum infection in broilers.
The present study was done to investigate the effect of dietary supplemental freezer dry powder fermentation soybean by Bacillus subtilis (natto) on the growth performance and intestinal microflora, prevention of fowl typhoid infection, the uptake of vegetative diet in broiler chickens. The chickens were fed control diet (supplement antibiotics) and fermentation soybean (0.75, 1.5, 3%) diets. A total of 280 one day old broiler chickens with randomly mixed sexes were fed the four diets for 6 weeks. Body weight gain of chicken fed 1.5% fermentation soybean by B subtilis tend to increase higher than the control from 6th week. Chickens fed diets containing 1.5% fermentation soybean by B subtilis had higher intake than those fed the other levels to the 3th weeks, but lower than control from the 4th week to the 6th week. Feed conversion also improved significantly in the supplemental 1.5% fermentation soybean by B subtilis from the 4th week to the 6th week. The number of B subtilis and Lactobacillus spp in the ileum and cecum tend to increase in the supplemental fermentation soybean by B subtilis at 6 week of age, but was not significantly different. In the nutrient digestibility, the feed conversion on the supplemental 1.5% fermentation soybean by B subtilis was better than the control and the weight of drying feces lower than the control. In test of S. gallinarum intramuscular inoculation, reisolation rate of S gallinarum in liver and feces 1.5% the fermentation soybean by B subtilis 75% (liver), 17% (feces) had decreased than the control.
On account of the recent improvement in performance of brown layers, the market share taken by brown layers has increased to about 50% in the world and to almost 100% in Korea. There are several other reasons why the industry has moved from white to brown, such as : brown layers are used to be more robust, more docile and easier to manage ; e brown layers are easier to sex at the hatchery ; brown layers lay less second grade eggs, due to a better shell Quality ; brown eggs seem to be more attractive than white ; and a clear consumer preference, thus a better price per egg. More recently, however, the trend towards brown eggs has been slowing down. The main reasons for this lie in that white layers can still produce an egg at a lower cost and that white eggs have better de-shelling properties, easier candling and higher yolk and solid content of the liquid egg which are benefits for egg processing industry. Although the performance of the brown layers is still improving, there are increasing opinions in the poultry industry that the market portion of white layers should be increased based on the following reasons, such as : shell color has no effect on the nutritive value of eggs ; . brown layers consume more feed ; the percentage of meat spots is significantly higher in brown eggs than in white eggs ; . brown layers are less efficient in the second cycle of production than in the first ; white layers are more resistant to the disease of fowl typhoid. In order to increase the market share of white layers in Korea, it may be needed to enlighten the consumers not to prefer the brown and large eggs and to inform the excellencies of white eggs widely.
Clostridium perfringens, Salmonella thyphimurium and S. Montevideo isolated from the intestines of dead broiler chickens in Korea were tested for antibacterial effects to purifed bee venom. Purified bee venom from Apis mellifera L. has been used as natural antimicrobial compounds in pigs, cows, dairy cattle and chicken farms in Korea. To investigate antibacterial effect of purified bee venom was evaluated by agar well diffusion method, minimum inhibitory concentraion (MIC), minimum bactericidal concentration (MBC), and postantibiotic effect (PAE). Purified bee venom exhibited significant inhibition of bacterial growth of C. perfringens, S. thyphimurium and S. Montevideo with MIC value of 0.85, 0.68 and $0.69{\mu}g/mL$, respectively. The MBC value of purified bee venom against C. perfringens, S. thyphimurium and S. Montevideo were 3.33, 2.66 and $2.86{\mu}g/mL$. Furthermore, the results of PAE values against C. perfringens, S. thyphimurium and S. Montevideo showed the bacterial effect with 3.5, 4.0 and 3.5 hr. Stability of pufifed bee venom at acidity from pH 1 to pH 8 for 24 hr was the antibacterial activity for C. perfringens, S. thyphimurium and S. Montevideo and melittin contents. Also purified bee venom processed through the heating for 15 min, there was no signification loss of the antibacterial activity and melittin at below $100^{\circ}C$. These results obtained in this study suggest that purified bee venom might be utilized as a feed additive in poultry diets.
Fowl typhoid caused by Salmonella gallinarum has increased dramatically since 1992 and has caused a great economic losses in chicken industry by characterizing with high mortality. In these studies, we investigated the immunogenicity and protectivity in chickens which were immunized with outer membrane protein(OMP) extracted from isolates of S gallinarum against challenge with live microorganism. Outer membrane proteins were composed of various sizes of molecular weight including 14K, 22K, 31K, 36K, 40K and 55K and the most of them responded strongly against rabbit antisera in immunoblot analysis. The chickens vaccinated with OMP or vaccinated with whole-cell combined with OMP($200{\mu}g$/chickens) complex showed higher delayed type hypersensitivity(DTH) response than that of whole-cell vaccinated group. The protective rates of OMP or whole-cell combined with OMP complex group against challenge of S gallinarum were higher (above 75%) than those (45~50%) of whole-cell vaccinated group. All vaccines were safe and the body weight-gains of all vaccinated groups were not significantly different (p<0.05) from those of nonvaccinated control group. In vitro tests, OMP stimulated both the proliferation of lymphocytes and T-lymphocytes, and OMP-induced lymphocyte proliferation was higher in the cells of the immunized chickens with OMP than in those from the control chickens.
Salmonella Gallinarum (SG) is known as an important pathogen that causes fowl typhoid in chickens. To investigate SG outer-membrane proteins (OMPs) as a vaccine candidate, we used proteomic mapping and database analysis techniques with extracted OMPs. Also, extracted OMPs were evaluated in several aspects to their safety, immune response in their host and protective effects. Our research has established a proteomic map and database of immunogenic SG-OMPs used as inactive vaccine against salmonellosis in chickens. A total of 22 spots were detected by 2-dimensional gel electrophoresis and immunogenic protein analysis. Eight spots were identified by Matrix-Assisted Laser Desorption/Ionization-Time of Flight-Mass spectrometry (MALDI-TOF-MS) and peptide mass fingerprinting (PMF) and categorized into four different types of proteins. Among these proteins, OmpA is considered to be an immunogenic protein and involved in the hosts' immune system. To estimate the minimum safety dose in chickens, 35 brown layers were immunized with various concentrations of OMPs, respectively. Consequently, all chickens immunized with more than a $50{\mu}g$ dose were protected against challenges. Moreover, intramuscular administration of OMPs to chickens was more effective compared to subcutaneous administration. These results suggest that the adjuvanted SG-OMP vaccine not only induces both the humoral and cellular immune response in the host but also highly protects the hosts' exposed to virulent SG with $50{\mu}g$ OMPs extracted by our method.

The poultry industry, which produces excellent sources of protein, suffers enormous economic damage from diseases. To solve this problem, research is being conducted on the early detection of infection according to the behavioral characteristics of poultry. The purpose of this study was to evaluate the potential of a non-movement behavior observation method to detect sick chickens. Forty 1-day-old Ross 308 males were used in the experiments, and an isolator equipped with an Internet Protocol (IP) camera was fabricated for observation. The chickens were inoculated with Salmonella enterica serovar Gallinarum A18-GCVP-014, the causative agent of fowl typhoid (FT), at 14 days of age, which is a vulnerable period for FT infection. The chickens were continuously observed with an IP camera for 2 weeks after inoculation, chickens that did not move for more than 30 minutes were detected and marked according to the algorithm. FT infection was confirmed based on clinical symptoms, analysis of cardiac, spleen and liver lesion scores, pathogen re-isolation, and serological analysis. As a result, clinical symptoms were first observed four days after inoculation, and dead chickens were observed on day six. Eleven days after inoculation, the number of clinical symptoms gradually decreased, indicating a state of recovery. For lesion scores, dead chickens scored 3.57 and live chickens scored 2.38. Pathogens were re-isolated in 37 out of 40 chickens, and hemagglutination test was positive in seven out of 26 chickens. The IP camera applied with the algorithm detected about 83% of the chickens that died in advance through non-movement behavior observation. Therefore, observation of non-movement behavior is one of the ways to detect infected chickens in advance, and it appears to have potential for the development of remote broiler management system.
Protective efficacy of trivalent Salmonella inactivated vaccine containing Chlorhexidine-inactivated S. Enterltidis (SE), S. Typhimurium (ST), and S. Gallinarum (SG) strains, was evaluated in this study. A total of 70 brown nick layers were divided into 7 groups, A to G, containing 10 hens per group. All hens in groups B to D were intramuscularly immunized with approximately 7×108 cells (3×108 cells of SE+1×108 SE+1×108 cells of ST+3×108 cells of SG) of the trivalent vaccine in 0.5 mL of PBS. All chickens in groups E to G were injected with sterile PBS. All hens of groups B and E, groups C and F, and groups D and G were orally challenged with approximately 2 ×109 CFU of wild-type SE, ST, and SG, respectively. Serum IgG titers and CD3+CD4+ T-cells, and CD3+CD8+ T-cells levels of groups B to D significantly higher than those of group A. In addition, all animals in groups A to C, E and F showed no clinical symptoms and survived after the virulent challenges, whereas one chicken in group D died and all chickens in group G died following the challenge. The protection against wild-type SE and ST in liver, spleen, cecum, and cloaca of groups B and C chickens was significant effective as compared with those in groups E and F. These indicate that the trivalent inactivated vaccine can be an effective tool for prevention of Salmonella infections by inducing robustly protective immune responses and cellular immune response in chickens.
