식품 살균을 위한 X선 조사 기술의 활용 및 전망 (The potential of X-ray irradiation as a new pasteurization technology for food)

  • Ionizing radiation is one of the efficient non-thermal pasteurization methods. The US Food and Drug Administration (FDA) allows the use of ionizing radiation to a dose up to 10 kGy for controlling foodborne pathogens and extending the self-life of foods. Recently X-rays, generated on absorption of high energy electrons in an appropriate metal target, have been used commercially for sterilization purposes. X-rays have the advantages of higher penetration power than E-beams and absence of harmful radioactive sources, such as Cobalt-60 or Cesium-137 associated with gamma-rays. That is why it has continued to receive attention as an attractive alternative to gamma-ray or E-beam irradiation. In this article, the potential of X-ray irradiation for controlling foodborne pathogens in various food products and necessary pre-requisite knowledge for the introduction of X-ray irradiation to the Korean food industry will be provided.

  • 식품 매개 병원균에 의한 문제는 식품산업뿐 아니라 세계 공공 보건에서도 문제가 된다. 최근 몇 년 간, 발효기술은 식품 내 병원성 미생물의 불활성화 및 이를 조절하기 위한 값 싸고 안전한 방법이라는 것이 밝혀졌다. 유산균 발효는 병원성 세균 및 바이러스에 대해 유의적인 항균효과를 갖는 과학적 증거를 보였다. 유기산, 박테리오신 및 과산화수소와 같은 유산균 대사체는 식품 매개 병원균에 대해 악영향을 미치고 이는 이들의 저해작용으로 이어진다. 이 화합물들은 물리적 결함만을 야기하는 것이 아니라 병원균의 유전자 발현에 대해서도 유의적인 저해 효과를 나타낸다. 게다가, 식품 내 유산균의 존재는 병원균에 대해 영양적인 경쟁을 제공하며 모든 요인이 그 성장을 억제한다. 본 연구는 유산균의 항균력, 분자생물학적 메커니즘 및 식품 매개 병원균의 불활성화를 위한 응용에 대하여 우리의 현 지식을 검토한다.

식중독 유발 세균의 증식에 미치는 겨우살이 추출물의 영향 (Antimicrobial Effect of Viscum album var. coloratum Extracts on Food-Borne Pathogens)

  • This study was performed to investigate the antimicrobial effect of the Viscum album var. coloratum extracts against food-borne pathogens. First, the Viscum album var. coloratum was extracted with methanol at room temperature and the fractionation of the methanol extracts was carried out by using petroleum ether, chloroform, and ethyl acetate, and methanol, respectively. The antimicrobial activity of the Viscum album var. coloratum extracts was determined by using a paper disc method against food-borne pathogens and food spoilage bacteria. The petroleum ether extracts of Viscum album var. coloratum showed the highest antimicrobial activity against Bacillus cereus and Shigella dysenteriae. Synergistic effect in inhibition was observed when Viscum album var. coloratum extract was mixed with Perillae folium extract as compared with each extract alone. Finally, the growth inhibition curves were determined by using petroleum ether extracts of Viscum album var. coloratum against Bacillus cereus and Shigella dysenteriae. The petroleum ether extract of Viscum album var. coloratum had strong antimicrobial activity against Bacillus cereus at the concentration of 5,000 ppm. At this concentration, the growth of Bacillus cereus was retarded more than 24 hours and up to 12 hours for Shigella dysenteriae. In conclusion, the petroleum ether extracts of Viscum album var. coloratum inhibit efficiently Bacillus cereus and Shigella dysenteriae.

식중독 유발 세균의 증식에 미치는 느릅나무 뿌리껍질 추출물의 영향 (Antimicrobial Effect of Ulmus davidiana var. japonica Extracts on Food-Borne Pathogens)

  • This study was performed to investigate the antimicrobial effect of Ulmus davidiana var. japonica extracts against food-borne pathogens. First, Ulmus davidiana var. japonica was extracted with methanol at room temperature and the fractionation of the methanol extracts was carried out by using petroleum ether, chloroform, and ethyl acetate, and methanol respectively. The antimicrobial activity of Ulmus davidiana var. japonica extracts was determined by using a paper disc method against food-borne pathogens and food spoilage bacteria. The methanol extracts of Ulmus davidiana var. japonica showed the highest antimicrobial activity against Staphylococcus epidermidis and Escherichia coli. Finally, the growth inhibition curves were determined by using methanol extracts of Ulmus davidiana var. japonica against Staphylococcus epidermidis and Salmonella typhimurium. The methanol extract of Ulmus davidiana var. japonica had strong antimicrobial activity against Staphylococcus epidermidis at the concentration of 4,000 ppm. At this concentration, the growth of Salmonella typhimurium was retarded more than 48 hours and up to 12 hours for Staphylococcus epidermidis. In conclusion, the methanol extracts of Ulmus davidiana var. japonica inhibit efficiently Staphylococcus epidermidis and Salmonella typhimurium.

  • Bovine mastitis was examined by use of the rolling ball viscosity test in Kwang-ju area. Antibiotic sus-ceptibility of isolated pathogens from raw milk was investigated by the disc method. Positive rate of so-matic cell counts (over 500,000 cells/ml) was decreased from 11.1% in 1991∼1993 to 8.0-% in 1995∼1997. Isolated mastitis-causing-pathogens were identified as Staphylococcus sp. (49.7%) Escherichia coki(23.2%) Streptococcus sp. (10.4%) and Bacillus sp(7.2%) Antibiotic susceptibility of isolated pathogens to almost antibiotics was decreased.

김치부재료의 유산균 증식 및 유해균 억제효과 (Effect of Kimchi ingredients on the growth of pathogenic and lactic acid bacteria)

  • When ingredients of Kimchi were mixed and stored in $18^{\circ}C$, lactic acid bacteria, such as Leuconostoc mesenteroides and Lactobacillus plantarum, were selectively grown up. Herefore, to understand why lactic acid bacteria were selectively cultured in Kimchi, antibacterial activities of Kimchi ingredients against some pathogens and Kinlchi lactic acid bacteria were investigated. Kimchi mixed with all ingredients significantly inhibited the growth of all tested pathogens: S. typhimurium, S. sonnei, and E. coli. Kimchi without green onion, garlic or ginger inhibited the growth of S. typhimurium, but did not E. coli and S. sonnei. However, Kimchi without red pepper powder did not inhibit the growth of all tested pathogens. All ingredients of Kimchi did not inhibit the growth of L. plantarum and L. mesenteroides. These results suggest that Kimchi ingredients can synergistically inhibit the growth of pathogens and Kimchi may be a selective medium for lactic acid bacteria.

Antibacterial effect of Ishige okamurae extract against cutaneous bacterial pathogens and its synergistic antibacterial effect against Pseudomonas aeruginosa

  • Background: Cutaneous bacterial pathogens including Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, and Propionibacterium acnes are often involved in acne vulgaris. The currently available therapeutic option for these skin pathogens is an antibiotic treatment, resulting in the emergence of antibiotic-resistant bacteria. The objective of this study was to discover an alternative antibacterial agent with lower side effect from marine algae. Results: The ethanolic extract of edible brown algae Ishige okamurae exhibits potent antibacterial activity against cutaneous bacterial pathogens. Among the ethanol soluble fractions, the n-hexane (Hexane)-soluble fraction exhibited the strongest antibacterial activity against the pathogens with MIC values ranging 64 to $512{\mu}g/mL$ and with minimum bactericidal concentration values ranging 256 to $2048{\mu}g/mL$. Furthermore, the combination with Hexane fraction and antibiotics (ceftazidime, ciprofloxacin, and meropenem) exhibited synergistic effect. Conclusion: This study revealed that the I. okamurae extract exhibited a synergistic antibacterial effect against acnerelated cutaneous bacterial pathogens acquired antibiotic resistant. Thus, the results of the present study suggested that the edible seaweed extract will be a promising antibacterial therapeutic agent against antibiotic-human skin pathogens and its infections.

광바이오센서를 이용한 우유 및 유제품의 식중독균 신속검출법 (Rapid Detection Methods for Food-Borne Pathogens in Milk and Dairy Products using an Optical Biosensor)

  • Milk and dairy products are not only excellent foods for humans, providing plentiful varied nutrients, but are also a good medium for detrimental food-borne pathogens. Although the food safety field has stabilized due to standardization of food processing, such as the hazard analysis critical control point (HACCP), outbreaks and cases caused by food-borne pathogens still occur at high rates. In approximately 30% of cases, the disease-causing pathogenic organism is undetermined. Recently, a biosensor was developed that has a simple and fast response and overcomes the problems of conventional methods such as cultivation, immuno-assay, polymerase chain reaction, and microarray. Due to the high selectivity and sensitivity of optical biosensors, it is a suitable method for the immediate detection of food-borne pathogens in milk and dairy products.

해양 유해세균에 대한 감태 추출물의 항균특성 (Antibacterial Property of Ecklonia cava Extract against Marine Bacterial Pathogens)

  • 사람과 어류에게 감염증을 유발하는 Edwardsiella sp., Streptococcus sp. 및 Vibrio sp. 의 해양 유해세균에 대한 해조류 추출물의 항균 활성을 조사하였다. 연구에 사용된 4종의 식용 해조류 중에서 감태 MeOH 추출물이 본 연구에서 사용 된 6종의 모든 해양 유해세균에 대해 넓은 범위의 항균 활성을 나타내었다. 감태의 MeOH 추출물의 유기용매 분획 추출물들 중에서, EtOAc 분획 추출물이 가장 높은 항균활성을 나타내었으며 6종의 해양 유해세균에 대하여 $128{\mu}g/mL$에서 $256{\mu}g/mL$의 MIC 값을 나타내었다. 또한, HPLC 분석에 의해 감태 EtOAc 분획 추출물에 phlorotannin 화합물인 dieckol이 다량 존재하고 있는 것이 확인 되었다. 결론적으로, 감태 추출물의 phlorotannin 화합물이 여러 유해세균에 대한 강한 항균활성을 나타내는 것처럼, 해양 유해세균에 대해서도 강한 항균활성을 나타내는 것으로 판단된다.

비단풀 추출물의 식중독세균에 대한 항균활성 (Antibacterial Activity of Euphorbia humifusa Extracts on Food-Borne Pathogenic Bacteria)

  • This study was performed to investigate the antimicrobial effects of Euphorbia humifusa ethanol-extract against food-borne pathogens. The growth inhibitory effects of the extract at a concentration of 250, 500, 1,000 or 2,000 mg/L on food poisoning microorganism were determined against Salmonella typhimurium, Listeria monocytogenes, Yersinia enterocolitica, Escherichia coli O157:H7 and Staphylococcus aureus. The microorganisms growth was not affected by the extract at the concentration up to 250 mg/L, but was significantly (p<0.05) inhibited by the extract at a concentration higher than 1,000 mg/L. The extract of Euphorbia humifusa had strong antimicrobial activity against all test strains at a concentration of 2,000 mg/L. The results in the present study demonstrate antimicrobial effects of Euphorbia humifusa ethanol-extract against food-borne pathogens, suggesting that Euphorbia humifusa could be an effective natural antibacterial agent in food.
