Effect of Viscous Materials Removal from Sea Tangle Extracts on Volatile Flavor Constituents (점성물질 제거가 다시마 추출액의 휘발성 향미성분에 미치는 영향)

  • Volatile flavor compounds of a sea tangle powder and two kinds of extract were analyzed by GC/MS. Extract I was prepared by boiling for 2 hours and centrifugation, while extract II by a sequential procedure of enzymatic hydrolysis, boiling in 1.5% NaCl solution. centrifugation and ultrafiltration to remove viscous materials. Fifty six volatile compounds from the dried sea tangle powder and the extracts were identified. The GC profiles of the extract II were different from those of the dried powder and the extract 1, indicating most volatile compounds were lost during removing viscous materials. Particularly those compounds in the initial and later parts of the GC profiles were significantly decreased and some of the compounds such as fatty acids. 3,5-nonadien-2-ol and 1-penton-3-ol were not detected.

Nutritional Components and Physiological Activity of 4 Wild Vegetables (Salvia plebeia R. Br, Angelica acutiloba, Gynura procumbens and Saururus chinensis Baill) Cultivated in Chungbuk Province (충북산 산채 4종(배암차즈기, 일당귀, 명월초 및 삼백초)의 영양성분 및 생리활성)

  • This study investigated on the nutritional components and physiological activity of four wild vegetables namely Salvia plebeia R. Br., Angelica acutiloba, Gynura procumbens and Saururus chinensis Baill for the development of representative wild vegetables in Chungbuk. Salvia plebeia possessed the highest radical scavenging activity and beta-carotene, but exhibited the lowest α-glucosidase inhibitory activity compared to the other three vegetables. Angelica acutiloba showed high crude protein content and α-glucosidase inhibitory activity, but contained low total polyphenol content, radical scavenging ability and beta-carotene compared to the other three vegetables. Gynura procumbens showed high mineral content, beta-carotene, vitamin K1 content and α-glucosidase inhibitory activity, but showed the lowest total polyphenol content and radical scavenging ability compared to the other three vegetables. Saururus chinensis showed high crude fiber content and total polyphenol content, but contained the lowest mineral and vitamin K1 content. To conclude, it is suggested to use Salvia plebeia or Saururus chinensis as antioxidant food materials and Angelica acutiloba and Gynura procumbens as food materials and sources of α-glucosidase inhibitors. In particular, it is believed that Saururus chinensis, which possessed high content of crude fiber, is suitable for low-calorie food materials such as diet products.

Anti-osteoporotic effects of Boswellia serrata gum resin extract in vitro and in vivo

  • BACKGROUND/OBJECTIVES: This study evaluated the beneficial effects of an ethanol extract of Boswellia serrata gum resin (FJH-UBS) in osteoporosis. MATERIALS/METHODS: MC3T3-E1 osteoblastic cells and RAW 264.7 osteoclastic cells were treated with FJH-UBS. The alkaline phosphatase (ALP) activity, mineralization, collagen synthesis, osteocalcin content, and Runt-related transcription factor 2 (RUNX2) and Osterix expression were measured in MC3T3-E1 cells. The actin ring structures, tartrate-resistant acid phosphatase (TRAP) activity, and the nuclear factor of activator T-cells, cytoplasm 1 (NFATc1) expression were evaluated in RAW 264.7 cells. Ovariectomized ICR mice were orally administered FJH-UBS for eight weeks. The bone mineral density (BMD) and the serum levels of osteocalcin, procollagen 1 N-terminal propeptide (P1NP), osteoprotegerin, and TRAP 5b were analyzed. RESULTS: FJH-UBS increased the ALP activity, collagen, osteocalcin, mineralization, and RUNX2 and osterix expression in MC3T3-E1 osteoblastic cells, whereas it decreased the TRAP activity, actin ring structures, and NFATc1 expression in RAW 264.7 osteoclastic cells. In ovariectomy-induced osteoporosis mice, FJH-UBS positively restored all of the changes in the bone metabolism biomarkers (BMD, osteocalcin, P1NP, osteoprotegerin, and TRAP 5b) caused by the ovariectomy. CONCLUSION: FJH-UBS has anti-osteoporotic activity by promoting osteoblast activity and inhibiting osteoclast activity in vitro and in vivo, suggesting that FJH-UBS is a potential functional food ingredient for osteoporosis.

  • In the management activities of head office in supporting franchisees in domestic food-service enterprises, the raw material supply plays a positive role to support franchisees' operation activity and it comes into conflict with them at the same time. In this paper, I have tried to find out the relationship between raw material supplying and the satisfaction of the franchisees by experimental research in order to research the route of head office's raw material supplying. To sum up the results, it was found out there is no meaningful relationship between the raw material supplying and the franchisees satisfaction (intention to renew the agreement), It indicates it would be more effective for the franchisees to buy the raw materials from the suppliers developed by the franchisees except some key materials in order to reduce the conflicts with the franchisees.

  • Our Country are called as a suzerain State of fermentation food, and preservative Technique was developed, because the specific character and the form of pottery were the best superior utensil of storage. The form couldn't give an explanation with scientifically, but we have used it for a few thousand years, which must get transformed from most compatible and reasonable form of fermentation. At present, a pottery was disappeared by the growth of the industrial society, and even the scientific form concealed it looks. For that reason, we apply the form into looks of a modernistic mechanism, and it intends to suggest a new direction to better the pottery of a formentation food.

Research on Allergy-Causing Materials of Allergy Diseases (알레르기 의심환자의 알러겐 발생빈도에 대한 연구)

  • Among the inhalant and foods which caused allergy, which is increasing nowaday, this research has a purpose of studying allergy-causing materials (allergen) which most frequently cause allergy. In order to conduct this research, we selected 100 hundred patients (M : F = 65 : 35) from in Kwangju Christian Hospital, who were tested through allergy multiple antigen simultaneous test (MAST) immunoblot assay (RIDA$^{(R)}$ X-Screen; r-Biopharm Co., Darmstadt, Germany). According to the test, the total IgE positive rate (above Class 2) from the inhalant is 58.3%, and that from food is 35%, respectively. This research showed that the first most allergen was the tick (Derm. farinae/Derm. pteronyssinus), the second most was housedust, and the third was the hair of pets such as cats and dogs; 65%, 35%, 26.3% in inhalant panel, and 52.5%, 40%, 50% in food panel, respectively. Thease allergens newly added to inhalant and food panels were not contributable to the detection of specifical allergen. We needs further improvement to be used as primary screening or quantitative test for allergy.

  • The extracts and pure saponins from the roots of Platycodon grandiflorum (PG) are reported to have a wide range of health benefits. Platycosides (saponins) from the roots of PG are characterized by a structure containing a triterpenoid aglycone and two sugar chains. Saponins are of commercial significance, and their applications are increasing with increasing evidence of their health benefits. The biological effects of saponins include cytotoxic effects against cancer cells, neuroprotective activity, antiviral activity, and cholesterol lowering effects. Saponins with commercial value range from crude plant extracts, which can be used for their foaming properties, to high purity saponins such as platycodin D, which can be used for its health applications (e.g., as a vaccine adjuvant). This review reveals that platycosides have many health benefits and have the potential to be used as a remedy against many of the major health hazards (e.g., cancer, obesity, alzheimer's) faced by populations around the world. Methods of platycoside purification and analysis are also covered in this review.

A New Method for Transduction of Mesenchymal Stem Cells Using Mechanical Agitation

  • Applications of bone marrow-derived mesenchymal stem cells in gene therapy have been hampered by the low efficiency of gene transfer to these cells. In current transduction protocols, retrovirus particles with foreign genes make only limited contact with their target cells by passive diffusion and have short life spans, thereby limiting the chances of viral infection. We theorized that mechanically agitating the virus-containing cell suspensions would increase the movement of viruses and target cells, resulting in increase of contact between them. Application of our mechanical agitation for transduction process has increased the absorption of retrovirus particles more than five times compared to the previous static method without changing cell growth rate and viability. The addition of a mechanical agitation step increased transduction efficiency to 42%, higher than that of any other previously-known static transduction protocol.

  • Flow cytometric techniques were used to investigate cell size, protein content and cell cycle behavior of recombinant Saccharomyces cerevisiae strains producing human lysozyme (HLZ). Two different signal sequences, the native yeast $MF\alpha1$ signal sequence and the rat $\alpha-amylase$ signal sequence, were used for secretion of HLZ. The strain containing the rat $\alpha-amylase$ signal sequence showed a higher level of internal lysozyme and lower specific growth rates. Flow cytometric analysis of the total protein content and cell size showed the strain harboring the native yeast signal sequence had a higher total protein content than the strain containing the rat $\alpha-amylase$ signal sequence. Cell cycle analysis indicated that the two lysozyme producing recombinant strains had an increased number of cells in the $G_2+M$ phase of the yeast cell cycle compared with the host strain SEY2102.

Comprehensive Analysis on Wrinkled Patterns Generated by Inflation and Contraction of Spherical Voids

  • We comprehensively investigated the wrinkles of a stiff layer covering a spherical void embedded in a rubber matrix after the void experienced inflation or contraction. We developed an easy experimental way to realize the inflation and contraction of the voids. The inflation took place in a void right beneath the surface of the matrix and the contraction happened in a void at the bottom of the rubber matrix. In the inflation, the wrinkle at the center of the deformation was random, and the pattern propagated into rabyrinthine, herringbone, and then oriented parallel lines as the position was away from the center of the inflation to the edge. The cracks were concentric, which were perpendicular to the parallel wrinkled pattern. In the contraction, the wrinkle was simply concentric around the surface of the void without any crack. The cracks were found only near the center of the deformation. The strain distribution in the stiff layer after the inflation and contraction was theoretically analyzed with simulations that were in excellent agreement with the experimental results.
